Cyclic AMP response element binding (CREB) protein acts as a positive regulator of SOX3 gene expression in NT2/D1 cells

  • SOX3 is one of the earliest neural markers in vertebrates, playing the role in specifying neuronal fate. In this study we have established first functional link between CREB and human SOX3 gene which both have important roles in the nervous system throughout development and in the adulthood. Here we demonstrate both in vitro and in vivo that CREB binds to CRE half-site located -195 to -191 within the human SOX3 promoter. Overexpression studies with CREB or its dominant-negative inhibitor A-CREB indicate that this transcription factor acts as a positive regulator of basal SOX3 gene expression in NT2/D1 cells. This is further confirmed by mutational analysis where mutation of CREB binding site results in reduction of SOX3 promoter activity. Our results point at CREB as a positive regulator of SOX3 gene transcription in NT2/D1 cells, while its contribution to RA induction of SOX3 promoter is not prominent.

A Novel Glycine-Rich Region in Sox4 is a Target for the Proteolytic Cleavage in E. coli (전사활성 인자인 Sox4의 단백질 분해효소에 의한 표적 부위에 관한 연구)

  • Sox4, a transcription factor, consists of three functional domains: an HMG-box domain as a DNA binding domain, serine rich region as a transactivation domain and glycine rich region (GRR), an unknown functional domain. Although Sox4 is known to be functionally involved in heart, B-cell and reproductive system development, its physiological function remains to be elucidated. We used pGEX expression system to develop a simple and rapid method for purifying Sox4 protein in suitable forms for biochemical studies of their functions. Unexpectedly, we observed that full-length Sox4 appears to be protease-sensitive during expression and purification in E. coli. To map the protease-sensitive site in Sox4, we generated various constructs with each of functional domains of Sox4 and purified as the GST-Sox4 fusion proteins using glutathione beads. We found that the specific cleavage site for the proteolytic enzyme, which exists in E. coli, is localized within the novel GRR of Sox4. Our study suggest that the GRR of Sox4 may a target for the cellular protease action and this cleavage in the GRR may be involved in regulating physiological function of Sox4. Additionally, our study may provide a useful method for investigating the proteolytic cleavage of the target molecule in E. coli.

Overexpression of SOX15 Inhibits Proliferation of NT2/D1 Cells Derived from a Testicular Embryonal Cell Carcinoma

  • SOX (Sry-related HMG box) family proteins, which have an evolutionarily conserved DNA binding domain, have crucial roles in cell differentiation. However, their target genes remain enigmatic. Some members of the SOX family may have roles in regulation of cell proliferation. We established stable NT2/D1 cell lines overexpressing SOX15 (SOX15-NT2/D1), and a modified 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay showed that the SOX15-NT2/D1 cells exhibited significantly slower growth than the controls. Flow cytometry analysis revealed that an increased fraction of the SOX15-NT2/D1 cells were in G1-G0. In addition, a microarray analysis identified 26 genes that were up-regulated in the SOX15-NT2/D1 cells, but none that were down-regulated genes. Among the up-regulated genes, IGFBP5, S100A4, ID2, FABP5, MTSS1, PDCD4 have been shown to be related to cell proliferation and/or the cell cycle.

Influence of $1{\alpha}$, 25-dihydroxyvitamin $D_3$ [1, $25(OH)_2D_3$] on the expression of Sox 9 and the transient receptor potential vanilloid 5/6 ion channels in equine articular chondrocytes

  • Background: Sox 9 is a major marker of chondrocyte differentiation. When chondrocytes are cultured in vitro they progressively de-differentiate and this is associated with a decline in Sox 9 expression. The active form of vitamin D, 1, 25 $(OH)_2D_3$ has been shown to be protective of cartilage in both humans and animals. In this study equine articular chondrocytes were grown in culture and the effects of 1, 25 $(OH)_2D_3$ upon Sox 9 expression examined. The expression of the transient receptor potential vanilloid (TRPV) ion channels 5 and 6 in equine chondrocytes in vitro, we have previously shown, is inversely correlated with de-differentiation. The expression of these channels in response to 1, 25 $(OH)_2D_3$ administration was therefore also examined. Results: The active form of vitamin D (1, 25 $(OH)_2D_3$ when administered to cultured equine chondrocytes at two different concentrations significantly increased the expression of Sox 9 at both. In contrast 1, 25 $(OH)_2D_3$ had no significant effect upon the expression of either TRPV 5 or 6 at either the protein or the mRNA level. Conclusions: The increased expression of Sox 9, in equine articular chondrocytes in vitro, in response to the active form of vitamin D suggests that this compound could be utilized to inhibit the progressive de-differentiation that is normally observed in these cells. It is also supportive of previous studies indicating that $1{\alpha}$, 25-dihydroxyvitamin $D_3$ can have a protective effect upon cartilage in animals in vivo. The previously observed correlation between the degree of differentiation and the expression levels of TRPV 5/6 had suggested that these ion channels may have a direct involvement in, or be modulated by, the differentiation process in vitro. The data in the present study do not support this.

  • Carbon dioxide reduction technologies using photosynthetic microorganism were suggested to overcome environmental destruction caused by $CO_2$ in flue gases from power plant and other industries. However, there are many toxic constituents in flue gas including CO, NOx, SOx. Continuous and Excessive supply of these noxious gases to cells will leads to inhibition of microalgal growth along with partial cell death. In this study, we tested the noxious effect of SOx and NOx on the pH and microalgal growth under photoautotrophic culture in three microalgae of Neochloris oleoabundans, Chlorella vulgaris and Haematococcus pluvialis. As a result, SOx concentration more than 50 ppm led to the rapid reduction of pH, thereby inhibiting of the growth in Neochloris oleoabundans and Chlorella vulgaris. NOx concentration more the 100 ppm reduced the exponential growth of N. oleoabundans and C. vulgaris. And H. pluvialis exhibited low sensitivity to SOx and NOx. Consequently, the three microalgae of N. oleabundas, C. vulagaris and H. pluvialis showed the normal vegetative growth in 25 ppm of NOx and SOx. Above all, H. pluvialis was useful for the $CO_2$ sequestration of the flue gas including high concentrations of NOx and SOx.

Reaction Characteristics of SOx/NOx Removal Using CuO/γ-Al2O3 Sorbent/Catalyst (CuO/γ-Al2O3 흡수제/촉매를 이용한 SOx/NOx 제거 반응특성)

  • Reaction characteristics of simultaneous removal of SOx and NOx have been investigated in a thermogravimetric analyzer and tubular fixed bed reactor using the $CuO/{\gamma}-Al_2O_3$ sorbent/catalyst. Sulfur removal capacity of $CuO/{\gamma}-Al_2O_3$ sorbent/catalyst is largely enhanced above both the temperature of $450^{\circ}C$ and the loading of 6wt% due to the participation of alumina support in a sulfation reaction. The NO reduction efficiency of 8wt% $CuO/{\gamma}-Al_2O_3$ sorbent/catalyst shows the maximum value at $370^{\circ}C$ and then decreases with the increase of reaction temperature due to the oxidation of $NH_3$ gas. The presence of sulfate on the surface of sorbent/catalyst enhances the optimum reaction temperature showing the maximum deNOx efficiency. In the simultaneous removal of SOx and NOx at $250^{\circ}C$. deNOx activity of $CuO/{\gamma}-Al_2O_3$ sorbent/catalyst is rapidly decreased due to the formation of ammonium salts such as $NH_4HSO_4$. In the simultaneous removal reaction of SOx and NOx, the optimum temperature showing the maximum deNOx efficiency increases to $400^{\circ}C$ due to the presence of $SO_2$ gas.

Automated Determination of SOx in Air by Diffusion Scrubber-lon Chromatography (확산포집-이온크로마토그래프법을 이용한 대기중 SOx의 자동정량)

  • An automatic method is developed for the determination of SOx in atmosphere. The method involves SOx sampling in diffusion scrubber followed by ion chromatographic analysis. Filtered air is withdrawn at 1.8.ell./min through a diffusion scrubber of which inner tube is made of PTFE(Gore-tex) membrane tubing. 1mM $H_{2}$ $O_{2}$ is used as absorbing solution so that SOx is oxidized to S $O_{4}$$^{2-}$. The scrubbered solution is automatically injected into ion chromatograhpy eith conductivity detection for sulphate determination. Replacement of commonly used polyproplene membrane with PTFE gives several merits such as easy preparation of diffusion scrubber, better collection efficiency. No measurable memory effect is experienced, and this isin contrast to previous work for ammonia. Detection limit of this method defined by three times standard deviation is 0.56ppbv. The precision is 0.4% RSD at SOx concentration of 7.3ppbv Results for Seoulatmosphere ate presented.

  • Experimental Investigations were carried out to remove NOx, SOx simultaneously from simulated flue gas[NO(0.02%)-SO$_2$(0.08%)-$CO_2$-Air-$N_2$] by using a plasma chemical reaction. Ammonia gas(14.81%) balanced by argon was diluted by all and was Introduced to mall simulated flue gas duct through NH$_3$ Injection system which is in downstream of reactor. The NH$_3$ molecular ratio(MR) was determined based on (NH3) to [NO+S0$_2$]. MR is 1, 1.5, 2.5. The NOx removal rate significantly increased with increasing NaOH bubble quantity. The SO$_2$ removal rate was not significantly effected by applied voltage, however it fairly Increased with increasing NH$_3$ molecule ratio. By-product aerosol particle was observed by XRD(X-ray diffraction) after sampling, The NOx, SOx removal rates, when H2O vapour bubbled by dry all was injected to plasma reactor, were better than those of other cases. When aqueous NaOH solution(20%) bubbled by 2.5( ι /min) of $N_2$ and 0.5 ( ι /min) NH$_3$(MR=1.5) were injected to simulated flue gas, The NOx. SOx removal rate was 95 ~ 100[%]

DDX53 Regulates Cancer Stem Cell-Like Properties by Binding to SOX-2

  • This study investigated the role of cancer/testis antigen DDX53 in regulating cancer stem cell-like properties. DDX53 shows co-expression with CD133, a marker for cancer stem cells. DDX53 directly regulates the SOX-2 expression in anti-cancer drug-resistant $Malme3M^R$ cells. DDX53 and miR-200b were found to be involved in the regulation of tumor spheroid forming potential of Malme3M and $Malme3M^R$ cells. Furthermore, the self-renewal activity and the tumorigenic potential of $Malme3M^R$-CD133 (+) cells were also regulated by DDX53. A miR-200b inhibitor induced the direct regulation of SOX-2 by DDX53 We therefore, conclude that DDX53 may serve as an immunotherapeutic target for regulating cancer stem-like properties of melanomas.

  • Activated carbon fibers were prepared from the petroleum isotropic pitch and organometallic compounds. The metalsvwere dispersed uniformly in the ACFs. The specific surface area and pore size distributions of metal containing ACFsvwere measured. The mesopores of ACFs were developed by Co, Ni, and Mn metals addition and the catalytic reactivityvof ACFs'SOx removal was increased by adding Ni and Pd metals. It was found that the mesopores did not work forvthe improvement of catalytic reactivity of ACFs' SOx removal with the blank experiment using the metal removedvACFs.
