Inhibitory Effects of Maesaengi (Capsosiphon fulvescens) Extracts on Angiotensin Converting Enzyme and α-Glucosidase (매생이 추출물의 angiotensin converting enzyme 및 α-glucosidase 활성 저해 효과)

  • Physiological activities of hot water (MHW) and 80% ethanol (MEH) extracts from Maesaengi (Capsosiphon fulvescens) were investigated in this study. For the evaluation of antioxidant activities for MHW and MEH, 2,2-diphenyl-1-pic-rylhydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD)-like activity were measured. DPPH radical scavenging activity and SOD-like activity of MHW, and MEH were increased weekly in a dose-dependent manner, and were about 10.8, 13.8, 62.4, and 27.1% at 10 mg/ml, respectively. The angiotensin converting enzyme (ACE) inhibitory activities of MHW and MEH were about 5.9% and 49.7% at 1 mg/ml, respectively. The ${\alpha}$-glucosidase inhibitory effect of MHW and MEH were about 1.4% and 67.3% at 1 mg/ml, respectively. To determine the influence of MHW and MEH on alcohol metabolizing activity, the generating activities of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) were measured. Facilitating rates of ADH activity by MHW and MEH were increased weekly in a dose-dependent manner and ALDH activities were not detected. Elastase inhibitory activities of MHW and MEH were 75.9% and 51.2% at 10 mg/ml, respectively.

A Study on Seaweed Sea Staghorn(Codium fragile) Ethanol Extract for Antioxidant (해조류 청각(Codium fragile) 에탄올 추출물의 항산화에 관한 연구)

  • Seaweeds are rich in in minerals, vitamins, proteins, and fiber, and are classified as green algae, and they are distributed on the coasts of Korea, East Asia, Oceania, etc. and are used as a health function material as well as food ingredients in our countries and countries. In this study, Codium fragile was extracted from ethanol and concentrated to confirm DPPH radical scavenging activity, SOD activity, FRAP and ABTS cation radical scavenging ability. As a result, DPPH scavenging activity was 0.83, 22.83, 38.27, 40.93, 45.60% at 6.25, 12.50, 25, 50 and $100{\mu}g/mL$, SOD-like activity were 23.13, 33.63, 33.93, 44.07 and 59.07%. FRAP and ABTS showed antioxidant activity in a concentration-dependent manner, as in the previous experiment. Therefore, this study confirmed that it can be used as a cosmetic material using Codium fragile, a natural material.

Chemical Composition and Antioxidant Activities of Prunus salicina Formosa Produced in Gimcheon (김천산 자두 후무사의 화학적 성분과 항산화 활성)

  • The present study was designed to investigate the chemical composition and antioxidant activities of Prunus salicina Formosa produced in Gimcheon. The free sugar amounts in Prunus salicina Formosa were fructose>glucose>sucrose>maltose, and malic acid contents in Prunus salicina Formosa were the highest among all the organic acids found in the present study. Each Prunus salicina Formosa extract was obtained by treating Prunus salicina Formosa with distilled water, 80% ethanol, 60% acetone, and 80% methanol at 25 and $50^{\circ}C$. The highest contents of total polyphenol and flavonoid were observed in the 60% acetone extract and 80% ethanol extract, respectively. The 60% acetone extract exhibited the greatest DPPH radical scavenging ability, reducing power, and nitrite scavenging ability. However, SOD-like activity was not considerably different for all the extracts studied. The results from the present study could be helpful for developing antioxidant products using Prunus salicina Formosa produced in Gimcheon.

Antioxidant Activities of Extracts from Different Parts of Sasa borealis (조릿대의 부위별 추출물의 항산화 활성)

  • Objectives : This study was performed to investigate the antioxidant activity of water and ethanol extracts from Sasa borealis leaves, stems and roots. Methods : Sasa borealis leaves, stems and roots extract were prepared using water and ethanol. The total polyphenol and total flavonoid contents were analyzed. 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activity, SOD like activity, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)(ABTS), hydroxyl radical scavenging activity, and Nitrite scavenging activity assays were carried out to determine the antioxidant activities. Results : The antioxidant activities of the Sasa borealis appeared higher in ethanol extract than water extracts. Total polyphenol and total flavonoids contents in ethanol extracts of leaves were $24.6mg/m{\ell}$ and $14.3mg/m{\ell}$, respectively, which were much higher than those of any other parts. SOD like activity was 70% ethanol extract of the leaves was highest with 15.68%. Electron donating ability was 70% ethanol extract of the leaves had the highest 59.07%. It exhibited high electron donating ability than BHT(45.68%). Nitrite scavenging activity of 70% ethanol extract was higher than the water extract at pH 2.5 and pH 4.2. Nitrite scavenging activity was 70% ethanol extract of the leaves was the highest 75.2%. Hydroxyl radical scavenging activity was 70% ethanol extract of the leaves was highest with 16.16%, showed very low activity than BHT(61.56%). Conclusions : These results suggest that 70% ethanol extracts from Sasa borealis leaves, exhibited higher antioxidant activities than those of root and stem, and can be potentially used as proper natural antioxidants.

Antioxidant Activities of Bamboo (Sasa Borealis) Leaf Extract according to Extraction Solvent (추출용매에 따른 조릿대 잎 추출물의 항산화활성)

  • This study was carried out to investigate the antioxidant activity of bamboo (Sasa borealis) leaf extract by measuring electron donating ability, superoxide dismutase (SOD)-like activity, reducing power, and lipid peroxidation inhibitory activity. Two crude extracts by water or 70% EtOH and five fractions of n-hexane, chloroform, ethyl acetate, n-butanol, and aqueous from the crude extract of 70% EtOH were prepared for this study. The crude extracts of water and 70% EtOH yielded 8.5% and 11.4%, respectively and the yields of n-hexane, chloroform, ethyl acetate, n-butanol, and aqueous fractions were 5.1% to 0.6%. Total polyphenol contents of the water and the 70% EtOH crude extracts were not significantly different; however, their total flavonoid contents were significantly greater in the 70% EtOH than in the water crude extract. Total polyphenol contents were the highest in chloroform fraction followed by ethyl acetate and n-butanol fractions and total flavonoid contents were the highest in ethyl acetate fraction followed by chloroform and n-hexane fractions. The two crude extracts as well as the five fractions showed election donating ability, SOD-like ability, reducing power, and lipid peroxidation inhibitory activity. Most of the antioxidant activities of each crude extract or fractions increased proportionally with the concentration. These results indicate that bamboo (Sasa borealis) leaf extracts show antioxidant activities due to its substantial content of polyphenol including flavonoid. Thus, it could be concluded that crude extracts by water or 70% EtOH and the fractions from the 70% EtOH extract, especially chloroform, ethyl acetate and n-butanol, would be useful as natural antioxidant substances.

Antioxidant Activity of Methanol Extracts from Cudrania tricuspidata Bureau according to Harvesting Parts and Time (이용부위 및 채취시기에 따른 꾸지뽕나무 메탄올 추출물의 항산화성)

  • To obtain the information on antioxidant activity by harvesting parts and time in Cudrania tricuspidata Bureau, total phenolic contents, flavonoid contents, superoxide dismutase (SOD) like activity and 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activity of methanol extracts from Cudrania tricuspidata were investigated. Total phenolic contents was the highest in leaf [$30.2{\sim}38.8\;mg/g$ dry weight (DW)] and followed by root bark, stem bark, fruit, root except bark and stem except bark. Among harvesting time, total phenolic contents of leaf was the highest as $38.1{\sim}38.8\;mg/g$ DW in May and June, and then decreased to October. Root bark harvested in April and May contained $26.6{\sim}27.8\;mg/g$ DW total phenolic compounds and total phenolic content fell to a very low values at 1.8 mg/g DW in February. Flavonoid contents was very high in leaf and root bark. Especially, root bark harvested in April had the highest of flavonoid contents as 23.2 mg/g DW. As a result of simple linear regression analysis of total phenolic contents on flavonoid contents according to harvesting parts, it showed a high correlation (p < 0.001) with $0.57{\sim}0.97\;R^2$ (coefficient of determination). As $RC_{50}$ value, that was, the concentration of sample required for 50% reduction of DPPH absorbance, was $13.7{\sim}20.5\;{\mu}g/mL$ in leaf so DPPH radical scavenging activity was very high. But SOD like activity of $1,000\;{\mu}g$ extract/mL MeOH was a low value of $0.3{\sim}9.2%$ in all samples.

The Skin Whitening Effect of Gypsum Extract using B16F10 cell lines (석고 추출물의 미백 효과)

  • Objectives : Recently the aesthetic, especially skin whitening, is focused on. This study was designed to investigate the effects of Gypsum Extract(GE) on skin whitening using B16F10 cell lines. Methods : In this study, we investigated effect of GE on cell viability, melanin synthesis, Superoxide dismutase(SOD)-like activity and tyrosinase activity in vitro and in vivo. Results : In results, The proliferation of B16F10 cell lines was increased by GE concentration. But there was no statistical signification. Melanin synthesis was decreased except GE 125㎍/㎖ of treated group. And the melanin synthesis ratio was increased in group being treated α-MSH. But After treating GE, it was decreased compared to Non-GE treated group. In addition, the SOD-like activity was enhanced in more than GE 125㎍/㎖ of treated group. And the activity of tyrosinase in vitro was inhibited in GE 500㎍/㎖ and 1,000㎍/㎖ of treated groups. The activity of tyrosinase in vivo was reduced in both Arbutin 500㎍/㎖ of treated group and Gypsum 500㎍/㎖ of treated group compared to controled group. After being treated α-MSH, the activity of tyrosinase was enhanced. But after treating Arbutin 500㎍/㎖ or Gypsum 500㎍/㎖, it was inhibited being compared to the group of Non-medicament treated. Conclusion : The results of this study showed the effects of GE that can inhibit melanin synthesis and the activity of tyrosinase and enhance the SOD-like activity. It means that gypsum has the skin whitening effect.

Skin Whitening Effect of Leonuri Herba Extracts(LHE) on Malignant Melanoma Cell (익모초(益母草) 추출물이 악성 흑색종 세포에 미치는 피부미백효과)

  • Objectives : In order to find out the whitening effects of Leonuri Herba, this study was designed to identify the effects and the action mechanism of LHE(Leonuri Herba extract) on Malignant melanoma cell lines. Methods : After treating LHE on the B16F10 cell-Malignant melanoma cell line-, the cell survival rate, melanin biosynthesis rate, intra&extracellular tyrosinase activity rate, SOD-like activity, tyrosinase mRNA gene expression were investigated. The results were compared with control group without LHE treatment or with positive control group treated with whitening efficacy substance. Results : B16F10 cell survival rate, melanin biosynthesis rate, and intra&extracellular tyrosinase activity were significantly inhibited depending on the concentration of treated LHE. Melanin biosynthesis rate and tyrosinase activity rate were also decreased when ${\alpha}-MSH$ was combined with LHE. In addition, the SOD-like activity was increased in a concentration-dependent manner in the treatment with the LHE, indicating signigicant activity at high concentrations, and the tyrosinase mRNA gene expression was decreased in both the LHE-treated group, the LHE and ${\alpha}-MSH-treated$ group. Conclusions : LHE seems to inhibit melanin synthesis through inhibition of tyrosinase activity and inhibition of tyrosinase mRNA gene expression. It also has the effect of promoting SOD-like activity and may be used clinically as a skin whitening agent in the future.

Antioxidant Activity of Solvent Extracts from Sargassum thunbergii (지충이 용매 추출물의 항산화 활성)

  • In this study, we investigated the antioxidant activity of solvent extracts from Sargassum thunbergii by measuring electron-donating ability (EDA), nitrite- scavenging ability, superoxide dismutase (SOD)-like activity, reducing power and antioxidant activity in $\beta-carotene-linoleate$ model system. Total phenolic compound contents of chloroform and methanol extracts from Sargassum thunbergii were $22.5{\pm}0.8$ mg/100 mL and $20.1{\pm}0.6$ mg/100 mL, respectively. The EDA were proportionally increased with concentration of the extracts except hexane extract. The EDAs were $70.4\%\;and\;63.2\%$ in methanol and water at concentration of $1000{\mu}g/mL$, respectively. The nitrite-scavenging ability of methanol extract (pH 2.5, $1000{\mu}g/mL$) was $89.4{\pm}0.9\%$ that was higher than those of other extracts. SOD-like ability of chloroform extract was stronger than other extracts. Reducing power of chloroform extracts at $1000{\mu}g/mL$ were 0.75 as O.D. value of 700 nm and antioxidant activities measured with $\beta-carotene-linoleate$ model system were in order of methanol $(78.4\%)>BHT\;(72.1\%)>chloroform\;(62.2\%)>water\;(51.4\%)>hexane\;(43.2\%)$. These results indicated that methanol extract from Sargassum thunbergii showed the highest activities on the EDA, nitrate- scavenging ability and antioxidant activities while the chloroform extract had the highest effect on the SOD-like activity and reducing power.

Development of Functional Beverage (SanYa) from Fermented Medical Plants and Evaluation of Its Physiological Activities (산야초를 이용한 기능성 발효음료개발 및 생리활성 연구)

  • This study was investigated to analyze the contents of flavonoid compounds and the effects of fermentation on the physiological activities of medical plants, also known as SanYa (SY). Antioxidative activity of the fermented SanYa (FSY) was measured by using DPPH radical scavenging and SOD-like activity. DPPH radical scavenging and SOD-like activity of FSY were 94.3% and 45.0%, respectively. Nitric oxide (NO) synthesis was increased 11 times through the addition of FSY. However, NO production of the macrophages RAW264.7 cells stimulated with lipopolysaccharide (LPS) was reduced to 56% through the addition of FSY. FSY showed fibrinolytic activity and indicated about 69.8% and 73.7% of xanthine oxidase and angiotensin converting enzyme inhibitory activities, respectively. These results suggested that FSY plays a significant role in fibrinolytic activity and have strong xanthine oxidase and angiotensin converting enzyme inhibitory activities.
