This study was conducted to diagnose the health of street trees with physiological characters, and to figure out the relation of physiological characters and cambial electrical resistances. Ginkgo biloba, Prunes serrulata and Salix koreensis were chosen in the Cheonan City. Soils under trees were collected to analyze dehydrogenase activities, and chlorophyll content, nitrate reductase and superoxide dismutase activities were analyzed from leaves sampled at the edge of crown in July. Cambial electrical resistances were measured in May, July and September, Soils with low dehydrogenase activity reflected the level of pollution. Chlorophyll content was the lowest in the leaves of P. serrulata at the Dongseo-street. Nitrate reductase activity of Ginkgo biloba was higher than P. serruluta and Satix koreensis. Nitrate reductase activity showed higher activity in the city than control(Independence Hall and Yonam College), but superoxide dismutase activity in the city lower than control. P. serruzatu in the Dongseo-street that cambial electrical resistance increase continuously during the growing season, showed the loss of vitality Cambial electrical resistance was negatively or positively correlated with nitrate reductase($r^2$=-0.566) and superoxide dismutase activity($r^2$=0.579). It was concluded that cambial electrical resistance might be suitable for diagnosing the tree health.
Objectives : The objective of this research was to investigate the antibacterial and antioxidant activities of bio-fermented Sanguisorbae Radix extract.Methods : The Sanguisorbae Radix extract was fermented byStreptococcus thermophilus,and their products was tested for antibacterial activity against pathogenic microorganisms namely,Bacillus cereus,Bacillus subtilis,Staphylococcus aureus,Escherichia coli,Vibrio parahaemolyticusandSalmonella typhimuriumby paper disc diffusion method and the antioxidant activities of extract was evaluated by five different assays as electron donating ability(EDA), superoxide dismutase(SOD)-like activity, polyphenol, flavonoid contents and nitrite scavenging ability.Results : The bio-fermented Sanguisorbae Radix extract was safe from heat. Antibacterial activity of fermented Sanguisorbae Radix extract appeared relatively highly againstBacillus cereusandStaphylococcus aureusand didn't show any difference. EDA in comparison to Vitamin C showed over 90% activity at about the same time of Sanguisorbae Radix extract expressed highly. SOD activity showed 15% in fermentation before and after. The nitrite scavenging ability of Sanguisorbae Radix extract before and after fermentation showed higher numerical value over 70% in pH 2.5 than that of butylated hydroxytoluene(BHT). But SOD activity, EDA and nitrite scavenging ability were not different between the Sanguisorbae Radix extract before and after fermentation. Total polyphenol content expressed over about 20 mg/g, and that of the Sanguisorbae Radix extracts was increased than that of the fermented Sanguisorbae Radix extracts.Conclusions : The results suggest the usefulness of developing functional materials using antioxidant active Sanguisorbae Radix extract was fermented bySalmonella typhimuriumwith high polyphenol contents and nitrite scavenging ability.

This study was conducted to evaluate the effect of anti-obesity and antioxidant enzyme activities in vitro by different solvent fractions from the roots of Codonopsis lanceolata. The cytotoxicity of different solvent fractions of C. lanceolata on 3T3-L1 preadipocytes were evaluated using the MTT assay, the rate of cell survival progressively decreased in a dose-dependent manner. Butyl alcohol fraction at $200{\mu}g/mL$ exhibited a pronounced cytotoxic effect (75.73%) on 3T3-L1 cell comparable to that of the hexane fraction (79.82%), methylene chloride fraction (84.02%), ethyl acetate fraction (87.62%) and DW fraction (86.30%) at the same concentration. The Oil Red O solution was used to determine whether different solvent fractions of C. lanceolata induce adipocyte differentiation in 3T3-L1 preadipocytes. Confluent 3T3-L1 cells were treated with $50{\mu}g/mL$ concentration of solvent fraction extracts from C. lanceolata. Inhibitory degree of lipid accumulation against solvent fraction extracts showed a significant level compared with the control. Both lipid accumulation and adipocyte differentiation showed relatively high effect on methyl chloride fraction. The root extract of C. lanceolata had the highest SOD enzyme activity of 84.5% in ethyl acetate partition layer and while water partition layer of diploid showed the lowest SOD enzyme activity of 57.9%. The activity of CAT, APX and POD showed a significantly higher activity in ethyl acetate partition layer compared with the other fraction. These results suggested that the roots of C. lanceolata may assist in the potential biological activity on anti-obesity and antioxidant capacity.
Smoking can increase oxidative stress and thereby change the antioxidant defense system in the body. To investigate the relationship between male adolescent smoking and antioxidant status, we surveyed the eating habits and dietary intake of 82 smokers and 44 nonsmokers recruited from a male technical high school. In addition, antioxidant enzyme activity and lipid peroxide values were determined in both the plasma and the erythrocytes. Although the frequency of food intake was not significantly different, most nutrient intake was unexpectedly higher in smokers than in nonsmokers. In comparison with the Korean RDA, especially the average intake of Ca, Fe and vitamin $B_2$ didn t reach 75% of the Korean RDA in either smokers or nonsmokers. An analysis of antioxidant enzyme activity showed that plasma catalase. superoxide dismutase (SOD), glutathione peroxidase (GSH-px), erythrocyte catalase and GSH-px activities showed no significant difference between smokers and nonsmokers. However, the erythrocyte SOD activity of smokers (1.57 unit/mgHb) was significantly lower than that of nonsmokers (2.00 unit/mg Hb). In addition, the plasma ceruloplasmin concentration of smokers (28.68 mg/$d\ell$) was significantly higher than that of nonsmokers (26.30 mg/$d\ell$), whereas the specific ceruloplasmin ferroxidase activity of smokers (0.31 unit/mg) was lower than that of nonsmokers (0.35 unit/mg). The plasma and erythrocyte thlobarbituric acid reactive substance (TBARS) of smokers (2.57 $\mu$mol/L, 0.32 $\mu$mol/gHb) were also significantly higher than those of nonsmokers (2.25 $\mu$mol/L, 0.27 $\mu$mol/gHb). The overall data indicate that adolescent smoking might decrease the antioxidant capacity of the body, in part, by lowering the erythrocyte SOD activity and the specific ceruloplasmin ferroxidase activity.
Objectives : The present study was designed to investigate effects of mixed extract from korean medicinal herbs (MIX) on oxidation/reduction reaction-related and aging-related enzyme in vitro. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase inhibition assay. Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. The MIX showed 97% inhibition of collagenase activity, and 64% inhibition of elastase activity at 1 mg/ml concentration of MIX, next only to positive control, which indicate good efficacy for anti-wrinkle ingredient. Also it's treatment showed 34% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of MIX. Antioxidant activities were determined by DPPH radical scavenging, xanthine oxidase (XO) inhibiting activity and SOD-like activity. Also these scavenging, XO-inhibiting and SOD-like activities were measured in 91%, 80%, and 63% inhibition, respectively, at a treated dose of 1 mg/ml, compare to control. Conclusions : These results suggest that possibility of mixed korean medicinal herbs as a functional ingredient for anti-wrinkle and whitening, anti-oxidation and anti-aging cosmetic formula.

This study was conducted to observe the association between antioxidant enzyme activity, free iron content and lipid oxidation of Korean native chicken (KNC) meat during refrigerated storage. Four lines of KNC (Yeonsan ogye, Hyunin black, Hoengseong yakdak and Hwangbong) were raised under similar conditions. A total of 16 roosters were randomly sampled and slaughtered at the age of 12 mon. The breast and thigh meats were stored aerobically for 10 d at 4℃. Although thigh meat had higher antioxidant enzyme activity, it was more susceptible to lipid oxidation and released more iron during storage than breast meat. Aerobic refrigerated storage for 10 d significantly decreased the activity of antioxidant enzymes and increased the amount of free iron and malondialdehyde. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were negatively correlated with lipid oxidation, whereas that of catalase was not. The amount of free iron was positively associated with lipid oxidation. We concluded that chicken line did not affect strongly on antioxidant enzyme activity and lipid oxidation in breast meat of KNC. However, the thigh meat of Hwangbong and Hyunin black had higher SOD and GSH-Px activity, respectively, and lower malondialdehyde contents than that of other chickens. SOD, GSH-Px and free iron play significant roles in meat lipid oxidation during refrigerated storage.
The purpose of this study was to investigate the effect of vitamin C and vitamin E supplementation on the iron contents and oxidative stress of the rats. Rats were fed 18g ascorbic acid and 300IU $\alpha$-tocopherol/kg diet, respectively. Rats were sacrificed at 1, 3, 5 and 7 month of age. The blood, liver and brain were selected for the quantitation of iron and malondialdehyde(MDA) contents, glutathione peroxidase(GSHPx), superoxided dismutase(SOD) and catalase(CAT) activity. Iron and MDA contents and GSHPx activities were increased with aging. Vitamin C and Vitamin E supplementation increased iron contents of the plasma. Vitamin C raised iron contents, but vitamin E decreased iron contents of the liver. In the brain vitamin C and vitamin E did not affect the iron level. MDA levels were decreased with vitamin C and vitamin E supplementation in the erythrocyte and liver, and vitamin C supplementation elevated MDA levels in the brain. GSHPx activity was increased with vitamin C and vitamin E supplementation. SOD activities of erythroucyte and brain were not affected with age, but in the liver, SOD activity was raised with age and vitamin C supplementation. Vitamin C and vitamin E supplementation promoted CAT activity of erythroucyte and liver, and CAT activity of brain was eleveated with vitamin addition but was decreaed with vitamin E addition. Vitamin C and vitamin E decreased iron contents of blood plasma, MDA contents of plasma and liver, and CAT activity of erythrocyte. Above results indicated that iron contents and biomarkers of oxidative stress were more affected by age than antioxidant action of vitamin C and vitamin E.
Antioxidant activity and biological properties in the MeOH extracts from different plant parts of Euonymus alatus (THNUB.) were measured by DPPH free radical scavenging ability and inhibition ability against xanthine/xanthine oxidase and proliferation in HL-60 cells. DPPH free radical scavenging activities in extracts of plant parts were high such as leaf, wing, root, seed and stem, respectively. The EtOAc fractions of plant parts were purified through LH-20 column chromatography and identified by GC/MS. LH-4 fraction and LH-5 fraction of leaf, stem and root showed stronger activities than other fractions in the inhibitor activity of DPPH and xanthine/ xanthine oxidase. $IC_{50}$ values of LH-4 fraction eluted from stem extracts showed such as 2.38 and 5.32 in DPPH and xanthine/ xanthine oxidase assay. Polyphenolic compounds were identified in purified LH-20 fractions showed highest $IC_{50}$ value in DPPH and xanthine/xanthine oxidase assay The activity of POD according to sampling time was high in root harvested in May and leaf harvested in September, respectively. The activity of SOD showed only in the extracts of stem in plant parts. SOD and POD in leaf were similar in the patterns of isozyme to those of stem. The purified extracts from Euonymus alatus (THNUB.) exerted inhibition ability of proliferation in HL-60 cells.
Physiological activities of unripened fruit, ripened fruit and leaf of Rubus coreanus Miquel were examined. Total polyphenolic compound content, electron donating ability (EDA), nitrite scavenging activity and SOD-like activity were examined using extracts of Rubus coreanus Miquel extracted with various extraction solvents such as 80% methanol, 75% acetone and water. The leaf part of Rubus coreanus Miquel included higher contents of total polyphenol compound compared with those of the other sample groups, unripened and ripened fruit. The total polyphenol compound content of leaf (100 g, dry base) extracted with 75% acetone showed the highest value of $5.06{\sim}5.87g$. As for EDA, unripened fruit showed over 90% of electron donating ability. No significant difference in EDA was found among the extracts prepared with various extraction solvents, 80% methanol, 75% acetone and water. Rubus coreanus Miquel extracts showed different nitrite scavenging abilities under different pH conditions. The nitrite scavenging abilities at pH 1.2 were in the range of $41.25{\sim}63.24%$ whereas they were $1.59{\sim}10.99%$ at pH 4.2 and $-2.84{\sim}7.94%$ at pH 6.0. The high levels of SOD-like activities were found in ripened fruit when different extracting methods were applied. Agar diffusion tests were accomplished to examine the antimicrobial activities of the extracts prepared from unripened fruit, ripened fruit and leaf with various extraction solvents. All of the extracts revealed antimicrobial activity against Bacillus cereus whereas no antimicrobial activities were observed against Escherichia coli, Salmonella typhimurium and Staphylococcus aureus.

In order to study on SOD like activity and Electric donor ability of Hedyotis diffusa $W_{ILLD}$, testing antibacterial effect on E. coli O157 which causes infectious inflammatory disease; measunng inhibitory effect on tyrosinase which stimulates melanin formation; and measuring the influence of it to SOD like activity, DPPH and TBARS which are related to ageing and carcinogenesis. The results of these experiments, are as follows. 1. In the antibacterial experiment with the extract of Hedyotis diffusa $W_{ILLD}$ on E. coli O157, there was no change in proliferation from the beginning of culture to two hours after, but proliferation inhibiting effect on E. coli O157 was detected from three hours after the beginning of culture. 2. Tyrosinase inhibitory effect was measured as $0.39{\pm}0.026%$. Compared with the control group, the effect was very slight. 3. The SOD like activity of the extract of Hedyotis diffusa $W_{ILLD}$. was measured as 21.33${\pm}$ 4.264%. Compared with the extracts of several other herbs, it was much more significant. 4. The DPPH of the extract of Hedyotis diffusa $W_{ILLD}$. was $53.3{\pm}0.91%$ when 0.02g was used and $83.5{\pm}1.82% $ when 0.05g was used. The result when 0.05g was used was more significant than 0.02g was used. 5. The TBARS of the extract of Hedyotis diffusa $W_{ILLD}$. was measured as 0.724 ${\pm}$0.O04MDAppm. Compared with the extracts of several other herbs, the result was more significant. From these results, we found that Hedyotis diffusa $W_{ILLD}$. can be used in therapy of dysentery with bloody stool and fever which is caused by infection with E. coli O157 and that it can be also used effectively as age resister or anticarcinogen.
