Cytotoxicity of Environmental Estrogenic Compound, Bisphenol A, via Generation of Free Radicals (내분비계 장애물질인 Bisphenol A의 free radical 생성을 통한 독성발현)

  • Bisphenol A shares similarities in structure, metabolism and action with DES, a known human teratogen and carcinogen. Bisphenol A, a monomer of polycarbonate and epoxy resins, has been detected in canned food and human saliva. The purpose of the this study was to evaluate the cytotoxicity, cell proliferation of bisphenol A In the presence of a rat liver S9 mix, contaning cytochrome P450 enzymes, and Cu (II). In the present study, Bisphenol A in combination with Cu (II) exhibited a enhancement in cytotoxicity which were inhibited by free radical scavengers. The content of malondialdehyde, an end product of lipid peroxidation, was also found to increase with concentration of bisphenol A. Also, we examined the change of CuZn-SOD, Mn-SOD, catalase and GPx activities in the MCF-7 cells exposed to bisphenol A. The activities of CuZn-SOD, CPx, catalase were found to decrease with bisphenol A concentration. Meanwhile, the activity of Mn-SOD was unchanged. This indicated that elevated oxidative stress caused by imbalance between the production and removal of free radicals occurred in cells.

  • A suproxide dismutase gene of Aquifex pyroprolus, a novel marine hypenhermophilic bacterium, was cloned, expressed, and characterized. The SOD of A pyrophilus (ApSOD) is an iron-containing homo-oligomeric protein with a monomeric molecular weight of 24.2 kDa. the amino acid sequence is similar to those of known Mn- and Fe-SODs from thermophilic archaea, and metal binding residues in all SOD sequences from different species are also conserved in A. pyrophilus SOD.   • A superoxide dismutase (SOD)-based superoxide ion $(O_2^-)$ sensor was fabricated by immobilizing SOD on a self-assembled monolayer (SAM) of 3-mercaptopropionic acid (MPA) which was prepared on a gold electrode. The SAM of MPA was found to function as an effective promoter for the electrode reaction of SOD. The amperometric response to $(O_2^-)$ was monitored at 300mV and -100mV vs. Ag/ Agel in 5mM phosphate buffer solution containing $KO_2$. The sensor was proved to have a high sensitivity, selectivity and short response time (<5 s) and negligible interference.

  • Reactive oxygen species (ROS) are produced in the metabolic process of oxygen in cells. The superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) in cells systemize the antioxidant enzymes to control the oxidative stress. Genistein is one of the isoflavonoids, and its role in controlling cellular oxidative stress is presently the active issue at question. In this study; we analyzed genistein-induced survival rates of the CHO-K1 cells, activities of antioxidant enzymes, ROS levels, and expression levels of antioxidant enzyme genes in order to investigate the effect of genistein on cellular ROS production and antioxidative systems in CHO-K1 cells. As results, the survival rate of cells was decreased as the dose of genistein increases (12.5${\sim}$200 ${\mu}$M). Genistein increased cellular ROS levels, while it reduced total SOD activities and the expression of CuZnSOD. In conclusion, we suggest that genistein may induce oxidative stress via down-regulation of SOD.

The Changes of Antioxidant Enzymes in the Lung of Alloxan-induced Diabetic Rats (알록산 유도 당뇨흰쥐의 폐에서 황산화계의 변화)

  • The present study was attempted to investigate the mechanism of oxidative cellular injuries which occur in diabetic rats by determining changes of antioxidant enzymes activity in the lung of alloxan-induced diabetic rats, the contents of glutathione in the lung, liver, blood samples, and ${\gamma}$-glutamylcysteine synthetase activities in the liver. Superoxide dismutase activities (SOD), including Cu, Zn-SOD and Mn-SOD, decreased in the lung of diabetic rats compared with those of normal control rats. However, activities of catalase and glutathione peroxidase(GPX) activities were not affected in the lung of diabetic rats. In diabetic rats, glutathione contents in the lung, liver, and blood samples, as well as the activities of ${\gamma}$-glutamylcysteine synthetase in the livers which is known to be the key enzyme of glutatione biosynthesis, decreased significantly. From these experimental results, it is thought that the decrease in SOD activities in the lung, glutathione contents and ${\gamma}$-glutamylcysteine synthetase activities in some tissues in alloxan-induced diabetic rats may be the crucial cause of vullnerability to oxidative cellular injuries.

Resistance of SOD2-transgenic petunia line to oxidative stress

  • SOD2-transgenic $T_3$ petunia line (A2-36-2-1-1-35) was treated with different levels of methyl viologen (MV) to determine its resistance to oxidative stress. Four (4) levels of MV (0, 100, 200, and $400\;{\mu}M$) were applied. The SOD2-transgenic $T_3$ petunia line exhibited a very significant oxidative stress resistance at the highest MV concentration ($400\;{\mu}M$) treatment compared to non-transgenic plant. RNA and protein expression of SOD2 transgene and higher parenchyma cell density in the transgenic petunias exhibiting resistance to oxidative stress proves its contribution to the expression of its resistance to oxidative stress.

The Effects of Red Ginseng Extracts on Antioxidant Enzyme Activities and Lipid Peroxidation of the Kidney in ${\gamma}$-Postirradiated Mice (감마선 조사전 홍삼추출물 투여가 생쥐 신장에서 항산화 효소활성과 지질과산화 수준에 미치는 영향)

  • The effects of red ginseng extracts (5.5 mg/mouse: i.p.) on the activities of antioxidant enzymes (superoxide dismutase, catalase and peroxidase) and lipid peroxidation were studied in the cytosol fraction of kidney. The experiments were carried out with whole-body irradiated (6.0 Gy, $^{60}Co$) and non-irradiated ICR mice. In the red ginseng extract-treated and irradiated mice, the activities of Cu, Zn- SOD, Mn-SOD, catalase and peroxidase were significantly enhanced by 27.8, 31.9, 17.9 and 15.0%, respectively, but the contents of malondialdehyde were considerably decreased (81.OfS) after 21 days, compared with those of non-treated mice. The enhanced activities of antioxidant enzymes inhibited the increase of malondialdehyde product resulted from the ionizing radiation. These results suggest that red ginseng extracts probably play an important role in radioprotective effect. Key words Red ginseng, SOD, catalase, peroxidase, lipid peroxidation.

Anti-oxidative Effect of Piperine from Piper nigrum L. in Caenorhabditis elegans

  • Piper nigrum L. (Piperaceae), which is a well-known food seasoning, has been used as a traditional medicine for the treatment of vomiting, abdominal pain, diarrhea and anorexia in Korea, China and Japan. Methanol extract from the fruit of P. nigrum was successively partitioned as n-hexane, methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions. Among those fractions the ethyl acetate soluble fraction showed the most potent DPPH radical scavenging activity, and piperine was isolated from the ethyl acetate fraction. To know the antioxidant activity of piperine, we tested the activities of superoxide dismutase (SOD) and catalase together with oxidative stress tolerance and intracellular ROS level in Caenorhabditis elegans. To investigate whether piperine-mediated increased stress tolerance was due to regulation of stress-response gene, we quantified SOD-3 expression using transgenic strain including CF1553. Consequently, piperine enhanced SOD and catalase activities of C. elegans, and reduced intracellular ROS accumulation in a dose-dependent manner. Moreover, piperine-treated CF1553 worms exhibited significantly higher SOD-3::GFP intensity.
