Cytotoxic Activities of 6-Arylamino-7-halo-5,8-quinolinediones against Human Tumor Cell Lines

  • 6-Arylamino-7-halo-5,8-quinolinediones (4a-4k, 5a-5b) were tested for in vitro cytotoxicity against human solid tumor cell lines such as A 549 (non-small cell lung). SK-OV-3 (ovarian), SK-MEL-2 (melanoma), HCT-15 (colon) and XF 498 (CNS) by SRB assay. The arylamino-7-chloro-5,8-quinolinediones 4 were also evaluated for cyclin-dependent kinase (CDK2 and CDK4) inhibitory effect. Among them, the 5,8-quinolinediones 4a and 5a with 7-(4-fluorophenyl) amino group were found to be potent cytotoxic against HCT 15, SKOV-3 and XF 498, and the compounds 4f and 4i showed inhibitory activities for the CDK4.

  • In the course of screening for new bioactive compounds from oligotrophs in soil, a microorganism, designated as SA-246 and now identified as Micromonospora sp., has been shown to produce a strong antibacterial compound. The active compound was purified from broth filtrate by ethylacetate extraction, silica gel column chromatography, preparative TLC and HPLC, and was identified as crisamicin A based on mass and NMR spectral data. The compound SA- 246 exhibited not only strong antibacterial activity against Gram-positive bacteria but also cytotoxicity against cancer cell lines such as A549 (lung), SK-OV-3 (ovarian), SK-MEL-2 (melanoma), XF498 (central nervous system) and HCT15 (colon).

Study on Antitumor Activity of Samiyeongeontanggamibang(SYTG) (사미연견탕가미방(四味軟堅湯加味方)이 항암(抗癌) 및 항전이(抗轉移) 효과(效果)에 미치는 영향)

  • To evaluate the antitumor and antimetastatic effects of Samiyeongeontanggamibang(SYTG), We have examined whether SYTG can inhibit the growth of several tumor cell lines, tumor cell adhesion, experimental tumor metastasis and increase survival rate of tumor-bearing mice by inhibition of DNA topoisomrase activity. The results were obtained as follows: 1. SYTG extracts revealed an efficient cytotoxicity against A549, SK-OV-3, B16-F10, and SK-MEL-2 cell lines. 2. SYTG extracts inhibited DNA topo-isomerase I from calf thymus. 3. The T/C% in S-180 bearing ICR mice treated with SYTG was 115.8% 4. SYTG extracts significantly inhibited adhesion of A549 cell to complex extracellular matrix. 5. In pulmonary colonization assay, SYTG suppressed lung metastases in tumor cell-injected mice. 6. In CAM assay, SYTG extracts inhibited angiogenesis at $15{\mu}g/egg$ concentration as compared with control. These results suggested that SYTG extracts might be a potent inhibitor of cancer.

  • The activity-guided fractionation of some medicinal plants led to yield five kinds of natural stilbene compounds namely 3, 5-dihydroxy-4'-methoxystillbene(I), rhapontigenin(II), reveratrol(III), rhaponticin(IV) and piceid(V) and two common flavonoids, apigenin(VI) and luteolin(VII) as active principles of the antitumor property, in vitro, against five kinds of human tumor cell lines, A-549, SK-OV-3, SK-MEL-2, XF-498 and HCT15.

  • Bioassay-guided fractionation of the aerial parts of Pilea mongolica(Urticaceae) afforded two cytotoxic triterpenoids, epi-oleanolic acid (I) and oxo-oleanolic acid (II). The structures of the compounds were confirmed by spectral and synthetic evidences. Compound I and compound II exhibited cytotoxicity against cultured human tumor cell lines, A549 (non small cell lung adenocarcinoma), SK-OV-3 (ovarian), SK-MEL-2 (skin melanoma), XF498 (CNS) and HCT15 (colon) with $ED_{50}$ values of $3.2-8.1{\mu}g/ml$ and $0.7-6.8 {\mu}g/ml$, respectively.

  • To evaluate the antitumor activity and antimetastatic effects of Bujeongyangeumtang(BJYET), studies were done experimentally. The results were obtained as follows: 1. BJYET extracts exhibited a significant cytotoxicity against A549, SK-MEL-2, SK-OV-3 and B16-BL6 cell lines. 2. The T/C% was 118.2% in BJYET treated group in S-180 bearing ICR mice. 3. BJYET extracts exhibited inefficient adhesive effect of A549, B16-BL6 cell to complex extracellular matrix. 4. BJYET extracts showed a significant inhibition of lung metastasis of B16-BL6 cells in C57BL/6. 5. In vitro neovascularization assays, angiogenesis was significantly inhibited in BJYET treated group than control group. These results suggested that BJYET extracts might be usefully applied for prevention and treatment of cancer.

  • In order to elucidate the cytotoxicity-structure correlation of ginseng-derived components, several prosapogenins and sapogenins were prepared from Korean red ginseng (Panax ginseng) saponins by acid hydrolysis or alkaline cleveage, and their chemical structures were identified by a combination of spectral and physical methods. Some of these degradation products showed the cytotoxic activities against various cancer cell lines, A549, SK-OV-3, SK-Mel-2, P388, L1210 and K562. The significant difference in cytotoxicity between stereoisomers was not found and the activity was inversely proportional to the number of sugars linked to sapogenins. Diol-type prosapogenins and sapogenins showed higher cytotoxicity than triol-type ones.

  • Six bioactive alkaloids, aristolactam B(1), poperolactam A(2), aristolactam A(3), norcepharadione B(4), cepharadione B(5) and splendidine(6) were isolated by bioactivity-guided fractionalton of a methanolic extract of the aerial part of Houttuynia cordata. Several of them exhibited significant cytotoxicity against five human tumor cell lines (A-549,SK-OV-3,SK-MEL-2, XF-498 and HCT-15) in vitro.

  • A bioassay-guided fractionation of the stem bark extract of Eucommia ulmoides Oliver (Eucommiaceae) led to the isolation of three iridoid constituents, genipin (1), geniposide (3), geniposidic acid (4) as well as (${\pm}$)-guaiacylglycerol (2) and fatty acid mixtures as active ingredients of the extract responsible for the antitumoral property. The EtOAc soluble part and BuOH soluble part of the extract demonstrated a significant inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT-15 (colon) in vitro, whereas the remaining water soluble part exhibited a poor inhibition.   • The genus Styrax (Styracaceae) is different from other genera of this family due to the production of resinous material, usually secreted when the barks and trunks are injured by sharp objects. This resin, in the past considered a miraculous remedy in several parts of Asia and America, has been used in traditional medicine to treat inflammatory diseases. The CH$_2$Cl$_2$ fraction of Styrax japonica showed significant cytotoxic activities by SRB method against five human tumor cell lines (A549, HCT-15, MES-SA, SK-OV-3, and SK-MEL-2). (omitted)
