Integrated Quantitative Phosphoproteomics and Cell-Based Functional Screening Reveals Specific Pathological Cardiac Hypertrophy-Related Phosphorylation Sites

  • Cardiac hypertrophic signaling cascades resulting in heart failure diseases are mediated by protein phosphorylation. Recent developments in mass spectrometry-based phosphoproteomics have led to the identification of thousands of differentially phosphorylated proteins and their phosphorylation sites. However, functional studies of these differentially phosphorylated proteins have not been conducted in a large-scale or high-throughput manner due to a lack of methods capable of revealing the functional relevance of each phosphorylation site. In this study, an integrated approach combining quantitative phosphoproteomics and cell-based functional screening using phosphorylation competition peptides was developed. A pathological cardiac hypertrophy model, junctate-1 transgenic mice and control mice, were analyzed using label-free quantitative phosphoproteomics to identify differentially phosphorylated proteins and sites. A cell-based functional assay system measuring hypertrophic cell growth of neonatal rat ventricle cardiomyocytes (NRVMs) following phenylephrine treatment was applied, and changes in phosphorylation of individual differentially phosphorylated sites were induced by incorporation of phosphorylation competition peptides conjugated with cell-penetrating peptides. Cell-based functional screening against 18 selected phosphorylation sites identified three phosphorylation sites (Ser-98, Ser-179 of Ldb3, and Ser-1146 of palladin) displaying near-complete inhibition of cardiac hypertrophic growth of NRVMs. Changes in phosphorylation levels of Ser-98 and Ser-179 in Ldb3 were further confirmed in NRVMs and other pathological/physiological hypertrophy models, including transverse aortic constriction and swimming models, using site-specific phospho-antibodies. Our integrated approach can be used to identify functionally important phosphorylation sites among differentially phosphorylated sites, and unlike conventional approaches, it is easily applicable for large-scale and/or high-throughput analyses.

A Study on the Reduction Plan of Dust Dispersion at Small-Scale Construction Sites in Downtown Seoul (서울 도심지 내 소규모 공사현장 비산먼지 감소방안에 관한 연구)

  • As public concern for fine dust has increased, so too has the interest in fugitive dust. Fugitive dust generated at construction sites, which is then scattered by vehicles moving around the sites, accounts for a significant portion. Small construction sites, which comprise the majority of construction sites, are often not adequately managed, because there are no regulations on fugitive dust for such sites. Therefore, in this study, in order to contribute to the establishment of fugitive dust management regulations on small-scale construction sites and to prepare measures to reduce fugitive dust, we measured and analyzed fugitive dust generated at small-scale building sites in Seoul. The fluctuations of fugitive dust generated at small-scale sites over time were checked, and the relatively low concentration when measured behind fences and railings suggested that fence installation is an appropriate method of reducing fugitive dust.

  • Due to the explosive growth of online social network users, large numbers of users discover these social network sites are a place where they can be able to spend their spare time, share feelings, ideas freely, and to search for new friends or partners. These web sites give an opportunity for its users to socialize with new people and to keep in touch or reconnect with current or old friends and families across disperse continents via these web sites, which traditionally replace the traditional methods. These social network web sites need careful investigations and findings on the usability for effective interactivity and more usability. However, little research might have previously invested on the usability of these on social network web sites. Therefore, we propose a new framework to study the usability of these social network sites. We namely call our framework "Interactivity". This framework will enable us to assess the usability of the social network sites. It will provide an overview of the user's behavior while interacting in these social network web sites. Measurement of the interactivity will be measured using Camtasia software. This software will entirely capture the interactivity of users including the screen and the movements, which the screen and the motion of the user action will undergo to analysis at the end of our research.

Web Accessibility Comparison between Handicapped Person Related Web Sites and School Web Sites (장애인 관련 웹 사이트와 학교 웹 사이트의 웹 접근성 비교)

  • Importance of Web accessability for handicapped persons is ever increasing in the information society with increasing role of the Web. The Web accessibility of handicapped person related Web sites were compared with the school Web sites in order to figure out the realities of handicapped person related Web site's accessibilities. According to the experiment, the Web accessibility of handicapped person related Web sites got lower scores than general school Web sites. The research result shows that more improvement efforts for the Web accessibility of handicapped person related Web sites are required.

Characteristics of $PM_{2.5}$ Particles Measured in the Background Sites of Korea (우리나라 청정 지역에서 측정한 $PM_{2.5}$ 입자의 특성)

  • Atmospheric fine particles $(PM_{2.5})$ were collected at the background sites, Kangwha, Taean, and Kosan and characterized to understand their behaviors at the sites. Daily samples of $PM_{2.5}$ mass were measured and ionic species, carbonaceous species, and gaseous species were analyzed. Four-day backward trajectory analysis was also carried out. The mean concentrations of anthropogenic species were highest at Kangwha among three sites, while contributions from sea salts wree highest at Taean during the measurement period due to higher wind speed at Taean. Major chemical components in fine particles were sulfate, organic carbon, nitrate, and ammoniu. Most of the non-sea-salt (nss) sulfates in $PM_{2.5}$ might be present as ammonium sulfates at these sites. Most air parcels arriving at Kangwha and Taean were from northern China. Therefore, both sites were thought to be affected by the same air parcel. At Kosan, during the measurement period, air parcels were from either northern China or sourthern China. The nss sulfate concentration in the air parcels from southern China was higher, while the nss calcium, nitrate, and ammonium concentrations were higher when the air parcels were from northern China.

  • The study examined the characteristics of internet shopping mall sites including on-line market places and integrated internet stores from a perspective of consumer protection. Specifically, the study investigated 1) whether sufficient information is provided on the internet shopping mall sites, 2) whether shopping mall sites abide by internet advertising principles, and 3) specific examples of consumer misleading potentialities from internet shopping sites. Further, on-line market places and integrated internet stores were compared based on their informativeness and potential consumer misleading factors. Total 50 shopping mall sites were analyzed for examining the types of information provided and consumer misleading factors. To verify study questions, descriptive statistics were used. For comparison between on-line market places and integrated internet stores, Chi-square test and independent-sample t-test were employed. The results of this study were as the following: First, not a few of the internet shopping mall sites did not provided important information such as safety and warrantees. Second, important disclosures were not noticeable on the web pages and distracting factors were frequently used. Third, integrated internet stores were desirable compared to on-line marketplaces based on informativeness and consumer misleading potentialities. As the result of this study, internet shopping malls need to advised to provide sufficient information and reduce potential misleading factors.

Mercury Contents of Medicinal Plants and the Cultivated Soils in Korea

  • This study was conducted to identify transition characteristics of mercury in several selected medicinal plants and to find the appropriate management for production of safety food. Cultivated soils and medicinal plants were collected at 29 sites for Angelica gigas (Korean angelica root), 68 sites for Platycodon grandiflorum (Balloon flower), 35 sites for codonopsis lanceolata (Deoduck), 36 sites for Dioscorea batatas (Chinese yam), 32 sites for Rehmannia glutinosa (Foxglove), 16 sites for Cnidium officinale makino (cnidium), and 26 sites for Astragalus membranaceus (milk vetch root) during the harvest season of 2013. Mercury in the soils and medicinal roots were analyzed with a Direct Mercury Analyzer. Average content of mercury in soils cultivated medicinal plants was $0.023mg\;kg^{-1}$ (range: from 0.003 to $0.074mg\;kg^{-1}$) and average content of mercury in medicinal plants was $0.003mg\;kg^{-1}$ (range: from 0.001 to $0.011mg\;kg^{-1}$), indicating that mercury in the surveyed soils and medicinal plants were not exceeded the Korean regulation.

A Study on the Intensive Safety Management Items of Small-Sized Construction Sites (소규모 건축공사 중점안전관리항목에 관한 연구)

  • Almost accidents in construction sites are occurred by unsafe work conditions and failure making safe environment. As recent construction projects become more complex, various and diversified, So the potential of construction-related accidents is increased as well. Comparing the frequency of construction-related accidents in large-sized construction sites, small-sized construction sites are more vulnerable because of lack of workforce and less effective technology system. Furthermore, the absence of safety committee, poor financial condition leads to lack of safety education, installation of safety facilities, and provision of individual protective equipment, which indicates difficulties handling even basic safety management. Therefore, in order to lower the hazard rate at small-Sized construction sites, it is necessary to proceed with more structural and thorough actions for preventing accidents. In other words, it is necessary to analyze the causes of the accidents at small-sized construction sites, set criteria for safety management, and suggest safety management items according to work classification by evaluating the relative importance of the items.

Characteristics of Air Pollutants at Three Background Sites in Korea in the Winter of 1996/1997 (1996/1997년 겨울 우리나라 배경측정소에서의 대기오염 물질 특성)

  • The daily mean concentrations of PM2.5 mass ionic species carbonaceous species and HNO3, HCl, and NH3 were analyzed at three background sites in Korea : Kangwha, Kosan, and Dongbaik during two intensive field studies between December 1996 and January 1997, Four-day backward trajectory analysis was also carried out. The mean concentrations of anthropogenic species were higher at Kangwha and Dongbaik than Kosan. At these two sites themean concentrations of nitrate and ammonium/ammonia were higher than those of Kosan. It was suggested that these two sites be considered as the background sites of the Seoul Metropolitan Area and Pusan respectively. Major chemical components in fine particles were sulfate organic carbon nitrate and ammonium for all the sites. More than 90% of sulfate were non-sea-salt(nss) sulfates and most of the nss sulfate in PM2.5 might be present as ammonium sulfates at all sites. Most of air parcels arriving a Kangwha and Dongbaik were from the northern China. At Kosan during the measurement period half of air parcels were from the northern China and the other half from the southern China. At Kosan the concentrations of anthropogenic air pollutants originated from the southern China were higher than those typically observed from the northern China.

Applications of Satellite Imagery to Surveying Archeological Sites and Remains

  • Satellite imagery was applied to locating archeological sites and remains around northeastern areas of China, called as Manchuria, and Korean peninsular, such as Mountain Fortress of Goguryeo Dynasty era($37BC{\sim}771AD$), and firing torch and smoke beacon signal sites at mountain tops in Josun Dynasty era($1392{\sim}1910AD$) as well as burial sites below the ground level in the modern era. Information on archeological sites, fire posts and burial places could be found in various literatures, but real figures of such cultural assets have been disappearing due to land development programs and human activities in recent years. Some of these historical sites were identified in satellite images using GPS(Geographical Positioning System). Real locations of these sites would be further necessary to be verified.
