The Metabolizable Energy Value, Standardized Ileal Digestibility of Amino Acids in Soybean Meal, Soy Protein Concentrate and Fermented Soybean Meal, and the Application of These Products in Early-weaned Piglets

  • Three experiments were conducted to evaluate the metabolizable energy (ME) value, standardized ileal digestibility (SID) of amino acids (AA) of soybean meal (SBM), soy protein concentrate (SPC) and fermented soybean meal (FSBM), and the application of these products in early-weaned piglets. In Exp. 1, four barrows with initial body weight (BW) of $14.2{\pm}1.4$ kg were used in a $4{\times}4$ Latin square design. The diet 1 contained corn as the only energy source. The other three diets replaced 25% of corn in diet 1 with one of the three soybean products, and the digestable energy (DE) and ME contents were determined by difference. In Exp. 2, four barrows (initial BW of $18.2{\pm}1.5$ kg) were fitted with ileal T-cannulas and allotted to a $4{\times}4$ Latin square design. Three cornstarch-based diets were formulated using each of the soybean products as the sole source of AA. A nitrogen-free diet was also formulated to measure endogenous losses of AA. In Exp. 3, ninety six piglets (initial BW of $5.6{\pm}0.9$ kg) weaned at $21{\pm}2$ d were blocked by weight and assigned to one of three treatments for a 21-d growth performance study. The control diet was based on corn and SBM, the two treatments' diets contained either 10% SPC or FSBM and were formulated to same SID lysine to ME ratio of 3.6 g/Mcal. The results showed that the ME content of SPC was greater than SBM (p<0.05). The SID of most AA in SPC was greater than the SID of AA in SBM (p<0.05). For the essential AA, the SID of histidine, isoleucine, leucine, lysine and threonine in FSBM were greater than in SBM (p<0.05). Even though they were fed same SID lysine to ME ratio of 3.6 g/Mcal diets, pigs fed SPC and FSBM diets had greater weight gain, G:F (p<0.05) and better fecal score (p<0.05) than pigs fed SBM diet. In conclusion, SPC showed a higher ME content and SID of AA than the SBM. SID of some essential AA in FSBM was higher than SBM and was similar with SPC. But the lower antigenic proteins and anti-nutritional factors content in SPC and FSBM may be the main factors affecting the performance of early-weaned piglets rather than the increased ME content and SID of AA.

박테리오파지 P2-P4 시스템을 위한 벡터 플라스미드 개발과 안정성 (Development of Selectable Vector Plasmid in Bacteriophage P2-P4 System and Its Stability)

  • 박테리오파지 P2-P4 시스템은 바이러스 조립과정 기작 연구를 위한 뛰어난 실험 소재로 연구되어 왔으나, 이 시스템에 유전자 조작상 필수적인 유용한 플라스미드가 없는 관계로 그의 필요성이 대두되고 있다. 본연구에서는 도움파지가 없을 때 플라스미드 상태로 존재하는 P4 ash8 (sid71)을 시작물질로, 항생제 내성 유전자를 도입하여 선택성을 줄 목적으로 P4 파지 증식에 불필요한 P4 DNA 단편을 pUC4-K 플라스미드의 kmr(kanamycin resistance) 유전자로 치환하여 P4 ash8(sid71) kmr을 생성하였다. 이 플라스미드는 박테리오파지 P2로 induction하여 박테리오파지 P4 상태로 증식시킬 수 있게 그 genome 크기를 조정하였으며, 파지 상태로 전환된 것을 burst size 결정 및 CsCl 부양 균등밀도 편차실험을 통해 입증하였다. 생성된 P4 플라스미드 유도체를 유전자 조작하여 P4의 integrase 기능을 잃은 변이체를 쉽게 만들 수 있었으며, 역시 박테리오파지 P4 상태로 증식시킬 수 있었다. 이러한 변이체 플라스미드의 안정성 실험을 수행하여, P4의 integrase 기능이 지속적인 플라스미드 상태 유지를 위해 필요하다는 것을 보여주었다.

Predicting standardized ileal digestibility of lysine in full-fat soybeans using chemical composition and physical characteristics

  • Objective: The present work was conducted to evaluate suitable variables and develop prediction equations using chemical composition and physical characteristics for estimating standardized ileal digestibility (SID) of lysine (Lys) in full-fat soybeans (FFSB). Methods: The chemical composition and physical characteristics were determined including trypsin inhibitor activity (TIA), urease activity (UA), protein solubility in 0.2% potassium hydroxide (KOH), protein dispersibility index (PDI), lysine to crude protein ratio (Lys:CP), reactive Lys:CP ratio, neutral detergent fiber, neutral detergent insoluble nitrogen (NDIN), acid detergent insoluble nitrogen (ADIN), acid detergent fiber, L* (lightness), and a* (redness). Pearson's correlation (r) was computed, and the relationship between variables was determined by linear or quadratic regression. Stepwise multiple regression was performed to develop prediction equations for SID of Lys. Results: Negative correlations (p<0.01) between SID of Lys and protein quality indicators were observed for TIA (r = -0.80), PDI (r = -0.80), and UA (r = -0.76). The SID of Lys also showed a quadratic response (p<0.01) to UA, NDIN, TIA, L*, KOH, a* and Lys:CP. The best-fit model for predicting SID of Lys in FFSB included TIA, UA, NDIN, and ADIN, resulting in the highest coefficient of determination (R2 = 0.94). Conclusion: Quadratic regression with one variable indicated the high accuracy for UA, NDIN, TIA, and PDI. The multiple linear regression including TIA, UA, NDIN, and ADIN is an alternative model used to predict SID of Lys in FFSB to improve the accuracy. Therefore, multiple indicators are warranted to assess either insufficient or excessive heat treatment accurately, which can be employed by the feed industry as measures for quality control purposes to predict SID of Lys in FFSB.

Wrist PA-grip view에서 먼쪽노자관절의 안정성 정도 측정 시 왜곡도 평가 (Evaluation of Distortion in Measuring the Stability of Distal Radio-ulnar Joint in Wrist PA-Grip View)

  • Wrist PA-grip view는 주변 인대들의 손상을 쉽게 진단할 수 있기 때문에 삼각섬유연골 파열 진단에 시행한다. 그러나 의료장비 기술의 발전에도 불구하고 기하학적 요소에 따른 영상의 왜곡은 아직까지도 많은 제한점이 있다. 본 논문에서는 정확한 Wrist PA-grip view 검사를 위해 현재 사용되고 있는 검사방법에서 발생되는 왜곡을 분석하여 영상의 왜곡을 최소한으로 감소시킬 수 있는 검사방법을 제시하고자 한다. Wrist PA-grip view를 재현하기 위하여 인체모형 팬텀을 사용하였고 source of image distance (SID)는 110 cm와 임의로 정해놓은 130 cm, 150 cm로 설정하였다. 각 SID에 따라서 손목 팬텀을 x축 방향 및 y축 방향으로 각각 0, 2, 4, 6, 8, 10 cm 이동하였다. 정량적 평가를 하기 위해 손목 팬텀의 이동 시 마다 자뼈와 노뼈의 먼쪽관절면의 거리의 차이를 picture archiving and communication system (PACS) system을 통해 측정하였다. 정성적 평가로는 방사선사 20명을 대상으로 설문조사를 실시하였다. SID에 따라 나뉜 세 그룹 간의 x축과 y축 방향의 움직임에 따른 왜곡도를 비교하기 위해 Kruskal Wallis test 통계기법을 시행하였으며 사후검정으로 Tukey test를 시행하였다. 정량적 평가 결과에서 x축 방향으로 이동하여 얻은 측정값은 모든 그룹에서 유의한 차이를 보이지 않았다 (p>0.05). y축 방향으로 이동하여 얻은 측정값은 대부분의 그룹에서 유의한 차이를 보였다 (p<0.05). 영상에서 왜곡이 임상적 진단과 수술에 많은 영향을 미치는 검사의 경우, 보다 정확한 진단을 위해 영상 후처리로 영상의 질을 유지하는 한도 내에서 기존의 SID인 110 cm 보다는 150 cm에서 검사하여 왜곡을 감소시키기를 제안하는 바이다.

박테리오파아지 P2-P4 시스템을 위한 tetracyclin resistance marker 함유 P4 유도체 벡터 플라스미드 조성 (Construction of New P4-Derived Vector Plasmid Containing Tetracyclin Resistance Marker for the Bacteriophage P2-P4 System)

  • 바이러스 조립 과정 기작 연구를 위한 좋은 재료인 박데리오파아지 P2-P4시스템에 이용될 벡터 플라스미드를 개발하기 위하여, P4 ash8 sid71을 출발 물질로 삼아 새로운 P4 유도체 벡터를 조성하였다. 유전자 재조합 기법을 써서 쉽게 선택 가능한 tetracyclin내성 유전자(tetR)를 도입하고 플라스미드P 4의 크기를 조절하였다. 이를 통해 얻어진 P4 ash8(sid7l) tetR은 12.09 kb의 크기를 가지며, 필요할 때 P2로 induction하면 생물학적 활성을 가지는 박데리오파아지로 전환 가능하였다. 전환된 파아지의 burst size를 결정하고, CsCi 부양균등밀도 편차실험을 수행하였다. 균등밀도 실험 분포도에서 P2크기 파아지 머리의 packaging 상한을 추정할 수 있었다.

Standardized ileal digestible lysine requirement of pregnant sows under commercial conditions

  • Objective: The present experiment aimed to determine standardized ileal digestible (SID) lysine (Lys) requirements for pregnant sows individually housed under commercial farm conditions. Methods: Two hundred multiparous sows (parity = 5.1±2.0) on day 42 of gestation were randomly allocated to five dietary treatments with a balanced parity. Experimental diets were formulated to contain 0.22%, 0.32%, 0.42%, 0.52%, and 0.62% of SID Lys for the mid-gestation period (days 42 to 76) and 0.36%, 0.46%, 0.56%, 0.66%, and 0.76% of SID Lys for the late gestation period (days 77 to 103). All indispensable amino acids except Lys were provided at 110% of their requirement estimates. Daily feed allowance per sow was determined based on the back-fat thickness and body condition score at the second pregnancy check and on day 90 of gestation. Three different statistical models were used to estimate the SID Lys requirement. Results: Total born piglets alive per litter increased linearly and quadratically (p<0.001) as dietary SID Lys increased. For total born piglets alive per litter, the SID Lys requirement estimates ranged from 9.69 to 12.4 g/d for the mid-gestation period (1.19 to 1.52 g/Mcal metabolizable energy; 0.39% to 0.49%) and 14.6 to 17.4 g/d for the late gestation period (1.62 to 1.93 g/Mcal metabolizable energy; 0.52% to 0.62%). Conclusion: The mean values of the SID Lys requirement for the mid-gestation period and the late gestation period are 11.1 and 16.1 g/d (1.36 and 1.79 g/Mcal metabolizable energy; 0.44% and 0.58%), respectively, for maximal total born piglets alive per litter.

Effect of increasing levels of threonine relative to lysine on the performance and meat quality of finishing pigs

  • Objective: The present study aimed to evaluate the effects of varying standardized ileal digestible lysine:threonine (SID Lys:Thr) ratio in the diet on the performance and meat quality of finishing pigs. Methods: In total 192 crossbred pigs ([Landrace×Yorkshire]×Duroc, 17 weeks old), with an initial body weight (BW) of 70.6±3.9 kg were used in an 8-wk trial. Pigs were randomly allotted to one of six dietary treatments based on their initial BW and sex (8 replications; 4 pigs per pen, 2 barrows and 2 gilts). The pigs in the 6 treatments were fed diets having different SID Lys:Thr ratios such as 1:0.65, 1:0.66, 1:0.67, 1:0.68, 1:0.69, and 1:0.70. Results: A linear increment (p<0.05) in average daily gain (ADG) and trends in reduction in feed conversion ratio (FCR) were observed during day 29 to 56 of the experiment and the apparent total tract digestibility (ATTD) of dry matter tended to increase linearly (p = 0.094) at the end of the experiment (day 56) with the increase in the dietary SID Lys:Thr ratios. The backfat thickness and lean percentage increased (linear effect, p<0.05) on day 28. In addition, at day 56, a linear (p<0.05) increment in lean percentage was observed. Significant quadratic responses (p = 0.02) for pH and drip loss at day 7 (p = 0.02), a linear increase (p<0.05) in cooking loss and drip loss at day 7, and a trend in quadratic response (p = 0.07) in the lightness of meat color (L*) were observed, whereas other meat quality indices were unaffected by varying the SID Lys:Thr ratios. Conclusion: The SID Lys:Thr ratio for maximum ADG, minimum FCR and enhanced digestibility was found to be 0.70. However, for carcass trait and meat quality, the SID Lys:Thr ratio of 0.65 was enough.

P2 sir3-sized head에 packaging 되기 용이한 크기의 박테리오파지 P4 유도체 조성과 정성 연구 (Construction and characterization of the bacteriophage P4 derivatives whose genome size suitable for packaging into a P2sir3-sized head)

  • "P2 sir-관련 도움파지 비효율성"이라는 용어는 P2 sir 변이체가 그들의 위성 박테리오파지인 P4에 대해 도움파지 역할을 충분히 못하는 것을 가리키는 용어이다. 이 연구의 목적은 이러한 P2 sir-관련 도움파지 비효율성을 극복하는 요인들을 조사하는 것이다. 우선 P2의 cos 영역을 함유하는 P4 sid71 cosP2가 P2 sir3의 도움파지 비효율성을 극복하는지를 조사하였다. 그 결과 P4 sid71 cosP2는 P2 sir3의 도움파지 비효율성을 극복하지 못하였다. P2의 cos 영역 대신에 $P2_{sir}$-sized head에 packaging되는 DNA 크기가 도움파지 비효율성을 극복하는 중요한 요인으로 나타났다. 이 연구에서는, DNA 조작을 통해 packaging 되는 DNA 크기가 P4 ost1과 P4 ost2 사이가 되는 세 종류의 P4 유도체를 조성하였다. 그 중 하나인 P4 sid71 delRI::apr의 CsCl 균등밀도차실험을 거쳐 packaging되는 DNA의 크기를 확인하였다. P4 유도체들의 후손방출량 실험에 따르면 그들은 모두 P2sir3-관련 도움파지 비효율성을 극복하였다. $P2_{sir3}$-sized head에 잘 packaging되는 P4 유도체의 크기는 28-29 kb로 나타났다.

Ileal Digestibility of Amino Acids in Conventional and Low-Kunitz Soybean Products Fed to Weanling Pigs

  • An experiment was conducted to determine the standardized ileal digestibility (SID) of amino acids (AA) in four sources of full-fat soybeans (FFSB) and in one source of soybean meal (SBM). The FFSB had different concentrations of trypsin inhibitor units (TIU) and included two sources of conventional FFSB, and two sources of a soybean variety that was selected for a reduced concentration of the Kunitz trypsin inhibitor. The conventional FFSB was either low temperature-processed (LT-FFSB-CV; 37.7% CP, 35.4 TIU/mg) or high temperature-processed (HT-FFSB-CV; 40.5% CP, 4.4 TIU/mg). The low-Kunitz FFSB was also either low temperature-processed (LT-FFSB-LK; 36.2% CP, 23.5 TIU/mg) or high temperature-processed HT-FFSB-LK; (38.2% CP, 4.0 TIU/mg). The SBM contained 47.5% CP and 3.20 TIU/mg. Twelve weanling barrows (initial BW: $11.1{\pm}1.3\;kg$) were fitted with a T-cannula in the distal ileum. Pigs were allotted to a replicated $6{\times}6$ Latin square design with six diets and six periods per square. Five diets were prepared using each of the soybean sources as the only source of AA in the diet. An N-free diet was also included in the experiment to measure basal endogenous losses of AA. The two low temperature-processed FFSB had lower (p<0.05) AID and SID values for all indispensable AA than the two high temperature-processed FFSB and SBM. The SID values for all indispensible AA except Trp were greater (p<0.05) in LT-FFSB-LK than in LT-FFSB-CV, but the SID of AA in HT-FFSB-CV and HT-FFSB-LK were not different. The SID of AA in SBM were not different from the SID in HT-FFSB-CV and in HT-FFSB-LK. Results of this experiment show that a reduction of the TIU from 35.4 to 23.5 TIU/mg will improve the SID of AA, but this reduction is not sufficient to completely ameliorate the negative impact of trypsin inhibitors. Results also show that the SID of AA in high temperature-processed FFSB is similar to that in de-hulled SBM.

Effect of Different Inclusion Level of Condensed Distillers Solubles Ratios and Oil Content on Amino Acid Digestibility of Corn Distillers Dried Grains with Solubles in Growing Pigs

  • The purpose of this experiment was to determine and compare the digestibility of crude protein (CP) and amino acids (AA) in full-oil (no oil extracted) and de-oiled (oil extracted) corn distillers dried grains with solubles (DDGS) with different condensed distillers solubles (CDS) ratios. Six barrows ($29.6{\pm}2.3kg$) fitted with ileal T-cannula were allotted into a $6{\times}6$ Latin square design. Each period was comprised of a 5-d adaption period followed by a 2-d collection of ileal digesta. The five test diets contained 62% DDGS as the sole source of AA. A nitrogen-free diet was used to measure the basal endogenous losses of CP and AA. Chromic oxide (0.3%) was used as an index in each diet. The results showed that CP and AA were very similar in 5 DDGS, but the standardized ileal digestibility (SID) of lysine (from 56.16% to 71.15%) and tryptophan (from 54.90% to 68.38%) had the lowest values and largest variation within the essential AA, which suggests reduced availability of AA and different levels of Maillard reactions in the five DDGS. The apparent ileal digestibility and SID of CP and most of AA in full-oil DDGS (sources 1 and 2) were greater (p<0.05) than de-oiled DDGS (sources 3, 4, and 5). Comparing the AA SID in the 5 DDGS, full-oil with low CDS ratio DDGS (source 1) had non-significantly higher values (p>0.05) than full-oil with high CDS ratio DDGS (source 2); however, the SID of most AA of de-oiled with low CDS ratios DDGS (source 3) were non-significantly lower (p>0.05) than de-oiled with high CDS ratio DDGS (source 4); and the de-oiled DDGS with middle CDS ratio (source 5) but with different drying processing had the lowest SID AA values. In conclusion, de-oiled DDGS had lower SID of CP and AA than full-oil DDGS; a higher CDS ratio tended to decrease the SID of AA in full-oil DDGS but not in de-oiled DDGS; and compared with CDS ratio, processing, especially drying, may have more of an effect on AA digestibility of DDGS.
