Influences of mineral contents in decoction water on the extraction efficiency of bioactive compounds from Eucommiae Cortex (전탕용수의 미네랄 함량이 두충의 유효성분 추출에 미치는 영향 분석)

  • Objective : Eucommiae Cortex is bark of Eucommia ulmoides Oliver, a deciduous tree native to China. Today, teas or herbal drugs made from E. ulmoides leaves are widely consumed as a healthy food especially in Eastern Asia including Japan, China and Korea. This study was aimed to verify the extraction efficiency of bioactive compounds from Eucommiae Cortex depending on characteristics of decoction water. Method : Six types of decoction water with various origin were prepared and the contents of minerals in water were analyzed by ICP-AES and ICP-MS. The contents of extracted bioactive compounds were analyzed by HPLC and the relationship between mineral contents and extracted bioactive compounds in the decoction water were analysed by multiple factor analysis. The decoction waters could be classified in three groups by hierarchical cluster analysis. Result : Extracted amounts of compounds in Eucommiae Cortex showed a positive relationship with Na, Si, K concentration in decoction water, while a negative relationship with Ca, Mg concentration. Conclusion : The study proved that the minerals in water influenced the extraction efficiency of bioactive compounds from Eucommiae Cortex.

Development of Schizogenous and Lysigenous Aerenchyma in Rice Root

  • Aerenchyma development in rice (Oryza sativa L.) roots is quite important for adaptation to waterlogged or reduced soil conditions. Anatomical observations were carried out to clarify the development of schizogenous and lysigenous aerenchyma in elongating crown roots of rice. The crown roots of 3rd and 4th phytomer were taken from rice plants of the 8th leaf stage grown by hydroponic culture. The schizogenous intercellular spaces in the cortex of crown root tip were observed using a light microscope with semi ultra-thin sections and the lysigenous aerenchyma in mature tissue of crown root were observed using a cryo scanning electron microscope (cryo-SEM) with freezing fracture method. The schizogenous intercellular spaces in the root tip exist obviously in the middle portion of cortical cell layers close to the root-root cap junction, but not in root cap, stele and outer cell layers of cortex. The air spaces were formed at the junction of four neighbouring cells of inner cortex in the transverse sections, and between longitudinal cell layer connected along the root axis. Although many of those spaces were filled with liquid, some spaces seem to exist as air spaces. The lysigenous aerenchyma in the cortex, which hardly filled with liquid, emerged at 3-4 cm segment from the root tip and increased toward the basal region of root axis. The developing process of lysigenous aerenchyma was primarily separation of a radial row of cells caused by the shrinking and collapsing of cortical cells and then formation of septa along the radial cell rows by the fusion of cell wall with each other. These results suggest that the schizogenous and lysigenous aerenchyma playa role as a passage for the movement of oxygen into the root tip region where oxygen is required for respiration.

A Review on Brain Imaging Studies of Suicide in Youth (청소년기 자살에 대한 뇌영상 연구)

  • Suicide is a leading cause of death worldwide, especially among adolescents and young adults. Considering this fact, it is imperative that we understand the neural mechanisms underlying suicidal thoughts and behaviors in youth from a neurodevelopmental perspective. In this review, we focused on the magnetic resonance imaging studies that examined the neural correlates of suicidal ideations (SI) or attempts (SA) in youth. We reviewed twenty-three cross-sectional studies reporting the structural and functional alterations in association with SI or SA among adolescents and young adults with various mental disorders. The previous literature suggests that the dorsolateral prefrontal cortex, anterior cingulate cortex, and ventral frontolimbic circuit, may play an important role in the pathophysiology of suicidal behavior in youth through altered top-down control over emotion and impulsivity. Future studies with a longitudinal design and using multimodal imaging techniques may be of help to identify novel therapeutic targets specific for youth with suicidal thoughts and behaviors.

Rusty-Root Tolerance and Chemical Components in 4-year old Ginseng Superior Lines (4년생 인삼계통의 적변내성 및 화학성분 특성)

  • Experiments were carried out to examine the rusty tolerance in 61 inbred lines of ginseng cultivated in field, and chemical components were analyzed to clarify the difference between healthy and rusty ginseng roots. Among them, 10 lines showed rusty tolerance (RT) while 10 lines showed rusty sensitivity (RS). The content of phenolic compound in RT was lower than that in RS in cortex, epidermis and branch & fine roots, but it was not difference between RT and RS in stele. The contents of K, Ca, Na in RT were lower than RS in cortex, and the content of Mg, Fe, Na, Mn, AI, Si in RT were lower than RS in epidermis, and the content of Fe in RT were lower than RS in branch & fine roots, but mineral contents were not difference between RT and RS in stele. The content of phenolic compound in healthy cortex was lower than that in rusty cortex in same 6-year roots, but the mineral contents were not difference between healthy and rusty cortex in same 6-year roots. In root of seedlings, the contents of phenolic compound, K and Na in RT were lower than RS. It was suggested that the contents of phenolic compound, K and Na might be marker to select rusty tolerance ginseng lines.

Dietary Salt Modulates the Adrenocortical Expression of P450 11Beta-hydroxylase in Mice

  • This study was conducted to determine the effect of dietary salt on the synthesis of glucocorticoids in the adrenal cortex of mice. Mice had ad libitum access to 3% sodium chloride as the only drinking fluid (high salt diet) for either 4 days or 4 weeks. Adrenocortical expression of cytochrome P450 11beta-hydroxylase, a major regulatory enzyme in the biosynthesis of glucocorticoids, was examined by immunohistochemistry and western blot analysis. Ultrastructure of adrenocortical cell and plasma level of corticosterone were analyzed as well. Size and density of lipid droplets in the cortical cell were increased by high salt diet. Four days of high salt diet decreased P450 11beta-hydroxylase in the adrenal cortex, but 4 weeks increased it. Plasma level of corticosterone changed in parallel with the Cortical level of P450 11 beta-hydroxylase. These results suggest that high salt diet may modulate the biosynthesis of glucocorticoids, at least partly, via regulating the expression of P450 11beta-hydroxylase in adrenocortical cells.

  • 3-Monochloro-1,2-propanediol (3-MCPD) is a contaminant of acid-hydrolyzed vegetable protein. Several reports have suggested that chronic exposure to 3-MCPD could produce neurotoxicity in vitro or neurobehavioral effects inaspects of experimental animals. Disturbance of the nitric oxide signaling pathway by chronic exposure to 3-MCPD may be a causal factor of neurological disorders in rats. In order to investigate the relationship between 3-MCPD administration and expression of inducibal nitric oxide synthase (iNOS), the numbers and distribution patterns of iNOS-immunoreactive neurons were examined. At the all three bregma level examined, the optical density of iNOS-postive neurons was significantly increased following exposure to 3-MCPD. The change was more severe in the upper layer than in deep layer of the cortex. These data suggest that 3-MCPD toxicity may be mediated through disturbances to the nitric oxide signaling pathway.

Histopathological Findings of Sudden Death Caused by Acute Heat Stroke in an African Lion(Panthera leo) (급성 열사병으로 폐사한 아프리카 사자의 병리조직학적 소견)

  • Heat stroke can lead multi-organ damage with hemorrhage and necrosis in the lungs, heart, liver, kidneys, brain and Ut. Heat stroke occurs when the elevation of core body temperatures induce a failure of thermoregulatory mechanism. A four-year-old male African Lion(Panthera leo) showed clinical signs such as panting, tachycardia, hyperthermia, unconsciousness and mydriasis under He hish humidity and hot weather. Clinical treatment and pouring cool water was unsuccessful. Grossly, congestion of lungs and pleura was observed. Yellowish discoloration was observed in the renal cortex. Microscopically, the coagulative necrosis in kidney and congestion of lungs and spleen were observed. In our knowledge, this case was closely associated with acute heat stroke.

Study on Pathology of Taeeumin Medicinal Herb in Sasang Constitutional Medicine (태음인 약재의 사상의학적 병리 연구)

  • Objective This study was designed to define the efficacy of medicinal herbs of Taeeumin in Sasang constitutional medicine. Method We searched and analyzed original text such as Donguisusebowon Sinchuk edition written in 1901, Donguisusebowon Gabo edition written in 1894, Donguisusebowon Sansang Chobongwon, and posthumous manuscripts left by Je-ma Lee published by the Ministry of Health of North Korea. Results Seven herbs such as Ephedrae Herba, Coicis Semen, Castaneae Semen, Fel Ursi, Mori Cortex Radicis, Ginkgonis Semen, and Farfarae Flos regulate the mechanism of Esophagus-Cold (Wiwanhan). Three herbs such as Melonis Pedicellus, Ailanthi Radicis Cortex, and Quisqualis Fructus regulate the mechanism of Cold Lung-Dry (HanpaeJo). Sixteen herbs such as Rhei Rhizoma, Angelicae Tenuissimae Radix, Puerariae Radix, Cimicifugae Rhizoma, Angelicae Dahuricae Radix, Moschus, Gleditsiae Spina, Mume Fructus, Aurum, Glycine Semen Germinatum, Ampelopsis Radix, Cornu rhinocerotis, Antelopis Cornu, Bomeolum, Bezoar Bovis, and Typhae Pollen regulate the mechanism of Liver-Heat (Ganyeol). Three herbs such as Chrysanthemi Indici Flos, Nelumbinis Semen, and Spirodelae Herba regulate the mechanism of Heat Lung-Dry (YeolpaeJo). Conclusion Forty four herbs of Taeeumin regulate the mechanisms of Esophagus-Cold (Wiwanhan), Cold Lung-Dry (HanpaeJo), Liver-Heat (Ganyeol), Heat Lung-Dry (YeolpaeJo) and correct the energy-fluid pathology of Taeeumin.

Antioxidative Role of Selenoprotein W in Oxidant-Induced Mouse Embryonic Neuronal Cell Death

  • It has been reported that selenoprotein W (SelW) mRNA is highly expressed in the developing central nerve system of rats, and its expression is maintained until the early postnatal stage. We here found that SelW protein significantly increased in mouse brains of postnatal day 8 and 20 relative to embryonic day 15. This was accompanied by increased expression of SOD1 and SOD2. When the expression of SelW in primary cultured cells derived from embryonic cerebral cortex was knocked down with small interfering RNAs (siRNAs), SelW siRNA-transfected neuronal cells were more sensitive to the oxidative stress induced by treatment of $H_2O_2$ than control cells. TUNEL assays revealed that $H_2O_2$-induced apoptotic cell death occurred at a higher frequency in the siRNA-transfected cells than in the control cells. Taken together, our findings suggest that SelW plays an important role in protection of neurons from oxidative stress during neuronal development.
