A Simple Behavioral Paradigm to Measure Impulsive Behavior in an Animal Model of Attention Deficit Hyperactivity Disorder (ADHD) of the Spontaneously Hypertensive Rats

  • Impulsiveness is an important component of many psychiatric disorders including Attention-deficit/hyperactivity disorder (ADHD). Although the neurobiological basis of ADHD is unresolved, behavioral tests in animal models have become indispensable tools for improving our understanding of this disorder. In the punishment/extinction paradigm, impulsivity is shown by subjects that persevere with responding despite punishment or unrewarded responses. Exploiting this principle, we developed a new behavioral test that would evaluate impulsivity in the most validated animal model of ADHD of the Spontaneously Hypertensive rat (SHR) as compared with the normotensive "control" strain, the Wistar Kyoto rat (WKY). In this paradigm we call the Electro-Foot Shock aversive water Drinking test (EFSDT), water-deprived rats should pass over an electrified quadrant of the EFSDT apparatus to drink water. We reasoned that impulsive animals show increased frequency to drink water even with the presentation of an aversive consequence (electro-shock). Through this assay, we showed that the SHR was more impulsive than the WKY as it demonstrated more "drinking attempts" and drinking frequency. Methylphenidate, the most widely used ADHD medication, significantly reduced drinking frequency of both SHR and WKY in the EFSDT. Thus, the present assay may be considered as another behavioral tool to measure impulsivity in animal disease models, especially in the context of ADHD.

Antihypertensive effect of Korean Red Ginseng by enrichment of ginsenoside Rg3 and arginine-fructose

  • Background: Ginsenoside Rg3 and arginine-fructose (Arg-Fru) are known as the hypotensive compounds of Panax ginseng; however, their efficacy on antihypertension has not been reported yet to our best knowledge. Thus, hypotensive components-enriched fraction of red ginseng (HCEF-RG) was prepared from fine root concentrate (FR) and their antihypertensive effects were investigated in spontaneously hypertensive rats (SHR). Methods: Male SHRs were divided into six groups: control (Wistar Kyoto, SHR); FR 500; FR 1,000; HCEF-RG 500; and HCEF-RG 1,000; samples (mg/kg body weight) were orally administered every day for 8 wk. Blood pressure was monitored at 1 wk, 2 wk, 3 wk, 4 wk, 6 wk, and 8 wk by tail cuff method. At 8 wk after samples administration, mice were killed for the measurement of renin activity (RA), angiotensin-I converting enzyme inhibition, angiotensin II, and nitric oxide (NO) levels in plasma. Results: HCEF-RG with four-fold more Rg3 and 24-fold more Arg-Fru contents was successfully prepared from reacted mixtures of FR and persimmon vinegar (12 times against FR, v/v) at $80^{\circ}C$ for 18 h. Both FR 1,000 and HCEF-RG 1,000 showed lowered systolic blood pressure than SHR control group and HCEF-RG 1,000 group exhibited a significant decrease in diastolic blood pressure. RA was significantly lowered in all treated groups, while angiotensin II did not affect by FR and HCEF-RG treatment. However, angiotensin-I converting enzyme inhibition and NO in FR 1,000 and HCEF-RG 1,000 were significantly increased compared with SHR control group. Conclusion: HCEF-RG is more effective and useful for alleviating hypertension than FR, implying the health benefit of Rg3 and Arg-Fru.

Isaria sinclairii Extract Reduces Body Weight and Ameliorates Metabolic Abnormalities

  • Obesity is a major risk factor for cardiovascular disease. In our case study using animal models for disease states such as obesity or hypertension, we found that, Isaria sinclairii remarkably reduced body weight and ameliorated metabolic abnormalities in Zucker and SHR rats. Genetically obese (fa/fa) Zucker rats were one animal model chosen for this study. Four groups of rats received a standard diet and were treated orally with the following test samples daily for 8 weeks: saline (negative control), ethanol extracts of I. sinclairii, hot water extract of I. sinclairii, or $Xenical^{(R)}$ (30 mg/kg, positive control). Mild reductions (6.3%) in body weight gain were observed in the groups treated with the hot water extract of I. sinclairii compared to the control after 8 weeks. Interestingly, organ weight was greatly reduced by this Dongchunghacho (I. sinclairii), in parallel with the mild reductions in body weight gain and reductions in abdominal fat (adipose tissue). Also observed was a 4.1 % decrease in the ratio of heart weight/body weight compared to the control group. As a hypertensive animal model, SHR (spontaneously hypertensive rat) and WKY (Wistar Kyoto) rats were also administered these extracts for one month. Treatment with the hot water extract of I. sinclairii caused greater reductions in body weight gain for the SHR group (10.9%) compared to the WKY group's (5.2%). Based on these results, I. sinclairii extracts contain selective action for anti-obesity activity, naturally occurring candidate for regulation of body weight increase, as demonstrated in the present study.

Rg3-enriched Korean Red Ginseng enhances blood pressure stability in spontaneously hypertensive rats

  • Background: Korean Red Ginseng (Panax ginseng) has been shown to exert antihypertensive effects. In particular, ginsenoside Rg3 is thought to be a potent modulator of vascular function. The present study was performed to examine the antihypertensive efficacy of Korean Red Ginseng (KRG) extract and Rg3-enriched KRG (REKRG) extract. Methods: Spontaneously hypertensive rats (SHRs) andWistar-Kyoto rats (WKYs) were divided into six groups (WKY control, WKY-KRG, WKY-REKRG, SHR control, SHR-KRG, and SHRREKRG), and systolic blood pressure (SBP) and diastolic blood pressure (DBP) were measured at the carotid artery, followed by injection of 3mg/kg KRG or 3mg/kg REKRG. Results: REKRG treatment significantly decreased SBP and DBP 3hours post-treatment in the SHR group compared with SHR control group. However, SBP and DBP were not significantly different in KRG-treated SHRs compared with control SHRs. REKRG treatment did not significantly alter SBP or DBP 3hours post-treatment in the WKY group compared with WKY control group. Similarly, there were no differences in SBP or DBP with KRG treatment in the WKY group and WKY control group. Both KRG and REKRG increased endothelial nitric oxide synthase phosphorylation levels in the aorta, and the increases in endothelial nitric oxide synthase phosphorylation levels by REKRG treatment were higher than those with KRG treatment. Similarly, nitric oxide production in plasma from WKYs and SHRs was also increased by both KRG and REKRG. Conclusion: These results suggest that REKRG has a more beneficial effect on blood pressure control than KRG in SHRs.

오령산(五苓散)의 본태성 고혈압 동물모델에서 혈압강하 및 혈관 이완 효과 (Effect of Oryeongsan on Spontaneously Hypertensive Rat decrease of Blood Pressure and Vasodilatory)

  • Oryeongsan (ORS), a formula composed of five herbal medicines, has long been used to treat impairments of the regulation of body fluid homeostasis. The purpose of this study was to determine the antihypertensive and renal protective effects of ORS in rats with hypertension. Spontaneously hypertensive rats (SHR) were divided into two groups with similar mean baseline systolic blood pressure (SBP). Then, 1 mL/kg of vehicle (distilled water) or 1.5, 3 g/kg of ORS extract were administered orally once a day for 4 weeks. SBP and diastolic blood pressure (DBP) were measured at weeks 1, 2, 3 and 4. At the end of the experiment, blood was collected, and heart were removed for histology. By the 2 weeks after initiation of treatment, the ORS treated group had significantly lower SBP than SHR rats. The ORS treatment significantly improved blood pressure and echocardiogram parameters compared to hypertensive rats. Additionally, the left ventricular (LV) remodeling and LV dysfunction were significantly improved in ORS treated group hypertensive rats. Furthermore, an increase in fibrotic area has been observed in SHR rats compared with Wistar-Kyoto rats (WKY). Furthermore, administration of ORS significantly attenuated cardiac fibrosis in hypertensive rats. Therefore, these findings suggest that ORS has a protective effect on heart failure by alleviating hypertensive heart disease and cardiovascular dysfunction in SHR.

천마 Extracts가 백서의 국소적 관상순환기능에 미치는 영향 (Effects of Gastrodia Rhizoma Extracts on Global Coronary Circulation in Rats)

  • 본 연구는 고혈압을 비롯한 순환기 질환의 예방이나 치료의 목적을 위해 민간요법으로 전수되어 사용되고 있는 천마의 효능에 대한 사실성을 확인하고, 나아가서는 이러한 효능에 대한 천마의 작용기전을 유추하려고 하였다. 현재까지 천마 extracts에 대한 유효성분의 분석이 이루어져 있지 않기 때문에, 본 연구는 SD 실험쥐와 선천성 고혈압의 소질을 지닌 SHR 실험쥐를 이용하여 천마의 water-extracts나 ethanol-extracts의 심장 및 관상 순환기능에 대한 약리적인 효능을 과학적으로 구체적인 실험방법을 통해서 조사하였다. 천마 extracts는 추출과정에 사용된 용매, 추출기간 및 실험쥐 model에 따라 관상 순환기능에 대한 작용효과가 상반된 현상으로 나타났다. 물이나 50% ethanol에 16시간 추출된 extracts는 사용된 농도에서 SD 실험쥐의 관상 혈관저항을 저하시켜 혈관확장의 효과를 가져왔지만, SHR 실험쥐에서는 ethanol에 추출된 extracts는 사용된 농도에서 심장기능 그 자체에 유의적인 독성효과를 주지 않는다는 것을 말해준다. 본 연구결과는 천마의 water-extracts에 의해서 나타난 혈관확장작용이 일반적으로 국소적인 혈관확장에 관여한다는 lactate 생성의 증가에 기인하는 것보다는 오히려 extracts에 의한 대사적 acidosis 현상에 기인한다는 사실을 제시하고 있다. 본 연구에서 천마의 수용성 extracts가 혈관확장작용 또는 혈압강하작용을 나타낸다는 사실이 제시된 것은 심장이나 순환기 질환을 예방하고 치료하기 위한 천마의 임상적인 활용측면에서 매우 중요한 성과이다. 그러나, 천마의 순환기능에 대한 약리적 효능을 나타내는 성분추출은 조사되어야 할 과제로 아직 남아 있다. 또한, 천마를 한방에서 약재로써 이용되는 반면에 천마를 식이로 이용하기 위해서 식이성 천마에 대한 효능을 연구해야 할 필요가 있다. 이러한 연구결과로써, 천마는 혈관순환기질환의 예방이나 치료에 이용되는 생약으로써 활용될 뿐만 아니라 안전성을 가진 건강식품으로 개발되어 국민보건 향상에 이바지하리라 기대된다.

BSHR/2 네트워크를 위한 APS 프로토콜 분석 (Analysis of the APS protocol for BSHR/2 networks)

  • SDH(synchronous digital hierarchy)를 기본으로 한 SHR(self-healing ring) 네트워크는 장애가 발생할 경우 프로세스를 재구성할 수 있으며 APS 프로토콜을 사용하고 있다. 본 연구에서는 양방향으로 트래픽을 전송할 수 있고 두 개의 선로를 갖는 BSHR/2(two fiber bidirectional SHR)네트워크에서의 장애 복구 시간을 측정하였다. 이를 위해 APS(automatic protecti switching) 프로토콜을 분석하고 ITU-T G.841이 제시하는 최대 장애 복구 시간인 50㎳내에 장애를 복구하기 위해 필요한 각 노드들의 프로세싱 시간 영역을 제시하였다. 또한 다중 장애에서 SD(signal degrade) 장애가 선행된 후 다시 SF(signal failure) 장애가 발생하는 경우와 SF 장애가 선행된 후 다시 SF 장애가 발생하는 경우에 대해서도 분석하였으며, 어떠한 경우에도 50㎳내의 장애를 복구할 수 있었다.

흰쥐 뇌(腦)에서의 Presynaptic ${\alpha}-Receptor$와 MAO 활성의 상관 관계 (The Relationship between Presynaptic ${\alpha}-Receptor$ and Monoamine Oxidase Activity in the Rat Brain)

  • Relationship between hypertension and monoamine oxidase (MAO) activity in rat brain and the change of this relationship by presynaptic ${\alpha}-receptor$ agonist were studied. Animals were divided into three groups. Group I was composed of normotensive Sprague-Dawley rats (NR), group II of spontaneously hypertensive rats (SHR) and group III of acquired hypertensive rats induced by deoxycorticosterone acetate (DOCA) and NaCl treatment. Clonidine, a presynaptic ${\alpha}-receptor$ agonist, was administered to groups II and III. Blood pressures and MAO activities were measured in each group. MAO activities in the brain of SHR were lower than those of NR. Animals in group II received clonidine which lowered blood pressures but did not change MAO activities in the brain. DOCA and NaCl induced hypertension 21 days after these treatments in group III and did not cause any changes in brain MAO activity. Clonidine lowered blood pressures of group III but did not change MAO activities. The data from the present study suggest that abnormaly low MAO activities in SHR brain may be one of the underlying factors for the susceptibility to hypertension and that the decrease in noradrenergic neuronal activities through presynaptic ${\alpha}-receptor$ activation by clonidine may not be related to the changes of brain MAO activities.

자연발증적 고혈압 흰쥐에서 무청 건조분말의 항고혈압 효과 (Antihypertensive Properties of Dried Radish Leaves Powder in Spontaneously Hypertensive Rats)

  • The study aim was to investigate the antihypertensive effect after oral supplementation of dried radish leaves powder (DRLP). Angiotensin-converting enzyme (ACE) activity was measured by spectrophotometric assay. The systolic blood pressure (SBP) was measured in spontaneously hypertensive (SHR) and normotensive rats (Wistar) by the tail cuff method after a 4-week diet with DRLP at the level of 2.5% or 5%. The supplementation of DRLP decreased SBP of SHR although the 5% supplementation level did not show any more pronounced effect than the 2.5% level did. The decrease in the SBP observed for both 2.5% and 5% DRLP was accompanied by significant increases of the urinary Na and K excretion. The DRLP supplementation showed a potent ACE-inhibitory activity in pulmonary tissue from both hypertensive and normotensive rats. However, the DRLP supplementation did not affect the SBP in normotensive rats. These results indicated that DRLP exerted an antihypertensive effect in SHR due to the decreased ACE activity and increased urinary Na excretion.

선천성 고혈압 흰쥐에서 두릅 에탄올 추출물의 항고혈압 효과 (Antihypertensive effect of ethanol extracts of Aralia elata in spontaneously hypertensive rats)

  • Antihypertensive effects of ethanol extracts of Aralia elata (Miq.) Seem. (AE) were investigated in spontaneously hypertensive rats (SHR). SHR aged 14 weeks were treated for 8 weeks with AE (10 or 50 mg/kg/day) or amlodipine besylate (Am; 10 mg/kg/day) orally. Hypertension results in injury to several organs and can produce a significant increase in malondialdehyde (MDA) content as a result of lipid peroxidation and endothelial dysfunction. In this study, oral administration of AE and Am significantly reduced systolic blood pressure, organ weight index, and MDA content in tissues but increased significantly the plasma nitrite and nitrate concentrations. The endothelium-dependent relaxant activities of acetylcholine ($10^{-10}-10^{-3}M$) in norepinephrine (NE)-precontracted aorta were increased in AE- and Am-treated rats. Particularly strong endothelium-dependent relaxant activities were observed in AE-treated (50 mg/kg) rats. The endothelium-independent relaxant activities of sodium nitroprusside ($10^{-10}-10^{-3}M$) in NE-precontracted aorta were not changed. The results of this study suggest that AE has both antihypertensive and end-organ protective effects in SHR.
