Cloning of a 6.6-kb BamHI digested chromosomal DNA from S. griseus IFO13350 revealed the presence of an additional gene encoding a novel trypsin-like enzyme, named SprU. The SprU protein shows a high homology ($79\%$ identity, $88\%$ similarity) with the SGT protease, which has been reported as a bacterial trypsin in the same strain. The amino acid sequence deduced from the nucleotide sequence of the sprU gene suggests that SprU is produced as a precursor consisting of an amino-terminal presequence (29 amino acid residues), prosequence (4 residues), and mature trypsin consisting of 222 amino acids with a molecular weight of 22.94 kDa and a calculated pI of 4.13. The serine, histidine, and aspartic acid residues composing the catalytic triad of typical serine proteases are also well conserved. When the trypsin activity of the SprU was spectrophotometrically measured by the enzymatic hydrolysis of the artificial chromogenic substrate, N-${alpha}$-benzoyl-DL-arginine-p-nitroanilide, the S. lividans transformant with pWHM3-U gave 3 times higher activity than that of control. When the same recombinant plasmid was introduced into S. griseus, however, the gene dosage effect was not so significant, as in the cases of other genes encoding serine proteases, such as sprA, sprB, and sprD. Although two trypsins, SprU and SGT, have a high degree of homology, the pI values, the gene dosage effect in S. griseus, and the gene arrangement adjacent to the two genes are very different, suggesting that the biochemical and biological function of the SprU might be quite different from that of the SGT.
In this study, the nanosized ZnS thin films that can be used for fabrication of blue light-emitting diodes, electro-optic modulators, and n-window layers of solar cells were grown by the solution growth technique (SGT), and their surface morphology and film thickness and grain size dependence on the growth conditions were examined. Based on these results, the quantum size effects of ZnS were systematically investigated. Governing factors related to the growth condition were the concentration of precursor solution, growth temperature, concentration of aq. ammonia, and growth duration. X-ray diffraction patterns showed that the ZnS thin film obtained in this study had the cubic structure ($\beta$-ZnS). With decreasing growth temperature and decreasing concentration of precursor solution, the surface morphology of film was found to be improved. Also, the film thickness depends largely on the ammonia concentration. In particular, this is the first time that the surface morphology dependence of ZnS film grown by SGT on the ammonia concentration is reported. The energy band gaps of samples were determined from the optical transmittance values, and were shown to vary from 3.69 eV to 3.91 eV. These values were substantially higher than 3.65 eV of bulk ZnS. It was also shown that the quantum size effect of SGT grown ZnS is larger than that of the ZnS films grown by most other growth techniques.
Background: This was a randomized controlled clinical trial that aimed to evaluate the anesthetic efficacy of 2% lidocaine combined with different concentrations of epinephrine (plain, 1:200,000 and 1:80,000) during endodontic treatment of maxillary molars with symptomatic irreversible pulpitis. Methods: The trial included 144 adult patients who were randomly allocated to three treatment groups. All patients received buccal-plus-palatal infiltration. After 10 min, pulp sensibility testing was performed using an electric pulp test (EPT). If a tooth responded positively, anesthesia was considered to have failed. In the case of a negative EPT response, endodontic access was initiated under rubber dam isolation. The success of anesthesia was defined as having a pain score less than 55 on the Heft Parker visual analog scale (HP VAS), which was categorized as 'no pain' or 'faint/weak/mild' pain on the HP VAS. Baseline pre-injection and post-injection maximum heart rates were recorded. The Pearson chi-square test was used to analyze the anesthetic success rates at 5% significance. Results: Plain 2% lidocaine and 2% lidocaine with 1:200,000 epinephrine and 1:80,000 epinephrine had anesthetic success rates of 18.75%, 72.9%, and 82.3%, respectively. Statistical analysis indicated significant differences between the groups (P < 0.001, 𝛘2 = 47.5, df = 2). The maximum heart rate increase was seen with 2% lidocaine solution with epinephrine. Conclusion: Adding epinephrine to 2% lidocaine significantly improves its anesthetic success rates during the root canal treatment of maxillary molars with symptomatic irreversible pulpitis.

This study was performed by using experiment to minimize behavior difference of Monolithic and segmental Girder and to prove the design concept of the PSI (Precast PSC-Segmental I Grider). A full scale girder test was performed in four different cases, the monolithic girder, the segmental girder type-1, the segmental girder type-2 and the segmental girder type-3. The monolithic girder that was produced in one body 25 m span and the segmental girder that was jointed 5-sliced 5 m segment. The girder was built by as one body prestressing the tendons after manufacturing the segmental girder, and second prestressing after the casting of the slab concrete. The test result shows that the measured values were almost same or slightly bigger than the theoretical values which means that the PSI girder bridges concept came out to be reliable.
This study was carried out to understand the effects of Saengchinyanghyoltang-gamibang (SGT) on pancreatic islets of the mice induced with streptozotocin (STZ). In the control group, two times injected with 50 mg/kg 572 at 24-hour intervals, a few number of insulin immunoreactive-cells are observed at the pancreatic islets of the mice. In the experimental group which administered with extract of SGT during 21-day, a number of immunoreactive-cells are observed at the pancreatic islets. According to the electron microscopic observation, $\beta-cells$ of the control group were contained a few of secretory granules, but also these granules were contained electro-lucent materials. In the experimental group, a lot o( secretory granules and well developed cell organelles are observed at the $\beta-cells$. The level of glucose was significantly decreased in the experimental group compared with control group, but the level of BUN was similar in these two groups. These results suggest that administration of SGT to the mice improved the damage of $\beta-cells$ from injected with STZ.
Field emergence of Azuki bean is poor due to hard seed coat as compared to other legumes. In this study, an attempt was made to develop prediction method with regression analysis based on various seed vigor tests in laboratory for field emergence of azuki bean. Azuki bean seeds artificially aged to provide various levels of seed quality were evaluated by the standard germination test (SGT), cold germination test (CT), cool germination test (CGT), complex stressing vigor test (CSVT), tetrazolium(TZ) vigor test and electroconductivity test. The SGT was suitable for predicting the field emergence in the unaged high vigor seeds. The abnormal seedling percentage and shoot length in the CGT were highly correlated with field emergence of moderate vigor seeds artificially aged for 2 days. Electroconductivity, seed viability in the CSVT, and vigor and predicted germinability in the tetrazolium vigor test were also useful for predicting field emergence. Percent of ungerminated seed in the CSVT was correlated with field emergence in the low vigor seeds artificially aged for 4 days. In a stepwise multiple regression analysis, seed viability in the SGT, normal seedling percentage and dry matter weight in the CGT accounted for 86.9% of the predicted value of field emergence in azuki bean.

In this study, we investigated the antioxidant activities and compared other physiological properties including anti-cancer and antimicrobial effects of several traditional Korean herb-combined remedies such as Gilgyung-tang (GGT), Daihwangmokdan-tang (DHMDT), Sagan-tang (SGT), Socheonryongtang (SCRT), Sihocheonggan-tang (SHCGT), Sipyukmiyuki-eum (SYMYKE) and Hwangheuk-san (HHS), which were recorded in “Dong-eui-bo-gam” for “Ongjeo”. Total phenolic contents of the herb medicines were in a rich order of GGT < SYMYKE < SCRT < SHCGT < DHMDT < SGT < HHS. Among them, HHS appeared highest in superoxide dismutase-like activity, ferric reducing antioxidant power, scavenging of 2,2’-diphenyl-1-picrylhydrazyl, and 2,2’-azinobis(3-ethylbenzothiazoline-6-sulfonate) cationic radical activity. Interestingly, there was a positive relationship between their total phenolic contents and their antioxidant activities. Although all of them showed anti-proliferative activities in human colon cancer HCT-116 cells, HHS was seven times higher than GGT. Antimicrobial activities against Escherichia coli and Helicobacter pylori were revealed only on SGT, SCRT, SYMYKE and HHS. Taken together, these findings reveal the potential use of traditional Korean herbal formulas as functional ingredients in antioxidant and physiological materials.

In this paper, we conducted experimental investigation on the field applicability through the verification of reinforcement effect of the steel pipe reinforcement grouting using high strength steel pipe. SGT275 (formerly known as STK400) steel pipe is generally applied to the traditional steel pipe reinforcement grouting method. However, the analysis of tunnel collapse cases applying the steel pipe reinforcement grouting shows that there are cases where the excessive bending and breakage of steel pipe occur. One of the reasons causing these collapses is the lack of steel pipe stiffness responding to the loosening load of tunnels caused by excavation. The strength of steel pipe has increased due to the recent development of high strength steel pipe (SGT550). However, since research on the reinforcement method considering strength increase is insufficient, there is a need for research on this. Therefore, in this study, we conducted experiments on the tensile and bending strength based on various conditions between high strength steel pipe, and carried out basic research on effective field application depending on the strength difference of steel pipe through the conventional design method. In particular, we verified the reinforcement effect of high strength steel pipe through the measurement results of deformed shape and stress of steel pipe arising from excavation after constructing high strength steel pipe and general steel pipe at actual sites. The research results show that high strength steel pipe has excellent bending strength and the reinforcement effect of reinforced grouting. Further, it is expected that high strength steel pipe will have an arching effect thanks to strength increase.
Three malting barley cultivars, Sacheon #6, Doosan #12, and Doosan #22 were collected from Gwangsan, Chinju and Milyang which were artificially aged to provide varying levels of seed quality. Samples were evaluated by the standard germination test (SGT), cold germination test (CT), electroconductivity test and tetrazolium vigor test (TZ). In a multiple regression analysis, percent germination in the SGT accounted for 65% of the variation in field emergence of malting barley. Vigor index of the standard germination and cold germination tests also contributed significantly to the regression equation. Grain yield was predicted by the vigor index of TZ test. Percent standard germination and percent TZ germination prediction were useful for predicting grain yield in nine lots of malting barley.

In this study, the ZnS nanosized thin films that could be used for fabrication of blue light-emitting diodes, electro-optic modulators, and n-window layers of solar cells were grown by the solution growth technique (SGT), and their structural and optical properties were examined. When the growth temperature was $75^{\circ}C$, the surface morphology and the grain size uniformity were the best. The energy band gaps of samples were determined from the optical transmittance valued, and were shown to vary from 3.69 eV to 3.91 eV. These values were substantially higher than 3.65 eV of bulk ZnS, demonstrating that the quantum size effect of SGT grown ZnS is remarkable. Photoluminescence (PL) peaks were observed at the positions corresponding to the lower energy than that to energy band gap, illustrating that the surface states were induced by the ultra-fineness of grains in ZnS films. Particularly, for the first time, it is reported for the SGT grown ZnS that the PL peaks were shifted depending on the grain size.
