The Study of Seunggal-tang Gamibang on the Anti-allergy Effects

  • Objectives The purpose of this study is to investigate effects of Seunggal-tang gamibang (SGT) on the PMA plus A23187-induced RBL-2H3 cells. Methods The effect of SGT on ${\beta}$ -hexosamonidase release, passive cutaneous anaphylaxis, $TNF-{\alpha}$ production and IL-4 secretion was studied. Results SGT inhibited the release of ${\beta}$ -hexosamonidase in RBL-2H3, without affecting cell viability. The passive cutaneous anaphylaxis (PCA) was inhibited by the SGT. Moreover SGT also reduced production of $TNF-{\alpha}$ and secretion of IL-4 in a dose-dependent manner. Conclusions SGT has anti-allergy effects on the RBL-2H3 cells.

The Anti-Oxidative Effect of Suhwagije-decoction on the Liver and Spleen Cells of Aged Rats (수화기제탕 약침액이 노화 흰쥐의 간세포 및 비장세포의 항산화능에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the anti-oxidative effect of Suhwagije-decoction (SGT) pharmacopuncture solution on the liver and spleen cells of aged rats. Methods : The liver and spleen cells of 10, 52, and 72 week old Sprague Dawley (SD) rats were divided into three groups: distilled water (DW), Vit.C, and SGT groups, which were treated with DW, Vit.C and SGT pharmacopuncture solution respectively. The levels of superoxide dismutase (SOD), glutathione (GSH) and nitric oxide (NO) were then measured. Results : 1. Treatment of SGT pharmacopuncture solution significantly decreased NO concentration in spleen cells of 52 week old rats as compared with that of other groups. 2. Treatment of SGT pharmacopuncture solution significantly increased SOD activity in spleen cells of 72 week old rats as compared with that of other groups. Conclusions : Suhwagije-decoction (SGT) pharmacopuncture solution may have an anti-oxidative effect in the spleen cells of aged rats.

  • This study was clinically performed with the 73 outpatients of urticaria who had visited the Dept. of Dermartology, Oriental Medical Hospital, Dong-eui University from January 1st 2000 to June 30th 2003. They have all taken the oriental herb medicine as Sungmagalguntanggamibang(SGT, 升麻葛根湯加味方) more than 1 time. Urticaria is a common disease and a well known reaction pattern of the skin charaterized by erythema, wheals and edema, that is the results of various causes. Urticaria is classified into acute or chronic type. The etilogy of acute uricaria has been determined in many cases, but the cause of chronic urticaria has been determined in 10$\%$ to 20$\%$ of cases. Character of chronic type urticaria is recurrent pruritus and erythematous skin disease by stimulating factor. Through checking up the present patient state, we verified the effect of SGT in various ways. The results were summarized as follows; 1. An increase of taking a medicine(SGT) and attending a hospital in number let down repeated urticaria occurence. 2. SGT is more effective when the symptoms with the severe pruritic wheals are spread on the whole body in daily occurrence. 3. In urticaria treatment method, gradually reducing a western medicine and going side by side SGT, that is good at symptoms improvement. 4. Etiology of urticaria is classified into intestine-stomach accumulated heat by indigestion(食滯腸胃積熱), wind-moisture(風濕), wind-heat(風熱) etc. The SGT is available in the order of frequency of use.

The Effects of Ssanggang-tang($Shu\bar{a}ngji\grave{a}ng-t\bar{a}ng$) on Rat with Induced Hyperlipidemia (쌍강탕(雙降湯)이 고(高)cholesterol 식이(食餌)로 유발(誘發)된 고지혈증(高脂血症) 백서(白鼠)에 미치는 영향(影響))

  • Objective : Ssanggang-tang(SGT) is noted as an effective method to treat hyperlipidemia in China. The main purpose of this study was analysis of the effects of Ssanggang-tang(SGT) on rats induced with caused hyperlipidemia. Methods : We observed 3 experimental groups: normal, control, and SGT. Rats were provided a normal diet in the normal group, and the other groups were provided a hyperlipidemic diet to induce hyperlipidemia. After 2 weeks, SGT was treated in the SGT group. For 6 weeks, values related to hyperlipidemia were observed in the 3 experimental groups. Results : SGT decreased some values related to hyperlipidemia like total cholesterol, low density lipoprotein cholesterol(LDL cholesterol), triglyceride, acetocoenzyme A acetyltransferase(ACAT), and 3-hydroxy-3-methylglutaryl coenzyme A reductase(HMG-CoA reductase). However, it showed no effect on weight change or high density lipoprotein cholesterol(HDL cholesterol). Conclusion : These results suggest that SGT might be effective in treatment and prevention of hyperlipidemia.

Anti-allergic Effect of Seungmagalgeun-tang through Suppression of NF-${\kappa}B$ and p38 Mitogen-Activated Protein Kinase Activation in the RBL-2H3 Cells

  • In previous report, Seungmagalgeun-tang (SGT) could exert its anti-inflammatory actions in the BV-2 microglial cells. However, study on the anti-inflammatory effect of SGT in mast cells has not been identified. Therefore, we examined on the anti-inflammatory effect of SGT on the phorbol 12-myristate 13-acetate (PMA) and calcium ionophore A23187-induced rat basophilic leukemia (RBL-2H3) cells. SGT inhibited the release of ${\beta}$-hexosaminidase and secretion and expression of pro-inflammatory cytokines such as tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-4 on RBL-2H3 cells, without affecting cell viability. The protein expression level of nuclear factor (NF)-${\kappa}B$ (p65) was decreased in the nucleus by SGT. In addition, SGT suppressed the degradation of inhibitory protein $I{\kappa}B-{\alpha}$ protein, the activation of p38 mitogen-activated protein kinase (MAPK), and the expressions of cyclooxygenase (COX)-2 mRNA and protein level in RBL-2H3 cells. These results suggest that SGT could be involved anti-allergic effect by control of NF-${\kappa}B$ (p65) translocation into the nucleus through inhibition of $I{\kappa}B-{\alpha}$ degradation and suppression of COX-2 expression.

Streptomyces griseus Trypsin (SGT) Has Gelatinase Activity and Its Proteolytic Activity Is Enhanced by Manganese

  • Gelatinase is a proteolytic enzyme that hydrolyzes gelatin. Gelatinolytic activity was detected from culture broths of Streptomyces griseus IFO13350 and HH1 by paper disc assays on 0.5% agar plates containing 1% gelatin. The concentrated extracellular protein from the S. griseus was analyzed by SDS polyacrylamide gel, and two proteins, with molecular weights of 30 and 28 kDa, respectively, were identified to have gelatinase activity by gelatin zymography. The protein with a molecular weight of 28 kDa was confirmed to be S. griseus trypsin (SGT). The effects of metal ions and metal chelators on the protease activity of the SGT were studied. Of the metal ions tested, only manganese was found to enhance the protease activity, 2.6 times, however, $Co^{2+},\;Cu^{2+},\;and\;Zn^{2+}$, and metal chelators, such as EDTA and EGTA, inhibited the SGT activity. When the protease activity of the SGT was measured at various pHs, in the presence of 5 mM $MnCl_2$, its highest activity was at pH 11.0, whereas only 60% of the maximum activity was observed between pHs 4.0 and pH 6.0, and almost 80% activity between pHs 7.0 to pH 10.0. The protease activity was measured at various temperatures in the presence of 5 mM $MnCl_2$. The SGT was found to be stable up to $60^{\circ}C$ for 30 min, while only 16% of the enzyme activity remained at $60^{\circ}C$, and at $80^{\circ}C$ almost all the activity was lost. The optimal temperature for the protease activity was $50^{\circ}C$.

Effects of Suhwagije-tang Distillate on Serum and Testosterone in Aging Rats (수화기제탕(水火旣濟湯)이 주령별 흰쥐의 혈청과 테스토스테론에 미치는 영향)

  • 1. Objectives The purpose of this study is to investigate the effects of oral administration of distilled Suhwagije-tang in aging rats. 2. Methods The SD rats used in this experiment were 6, 48 and 68 weeks old. These groups were treated with distilled water(DW), 1% Vitamin C(Vit. C) and 50% Suhwagije-tang(SGT) distillate respectively, once a day for 4 weeks. Blood samples were taken from heart and the levels of various parameters in serum were analysed. 3. Results and Conclusions (1) At all concentration of SGT, survival rates of liver cells were higher than the control group. (2) The rate of which the 10W-SGT group gained weight was significantly higher than that of the control groups. (3) The levels of albumin in serum of 10W-SGT group were significantly increased in comparison to those of the DW group. The levels of ALP, total cholesterol and HDL-cholesterol in serum of 52W-SGT group were significantly decreased in comparison to those of the DW group. (4) The levels of testosterone in serum of SGT groups showed no significantly changes in comparison to those of the control groups. Testosterone levels were naturally decreased with advancing years, so 72W-DW group's testosterone level was significantly decreased in comparison to those of 10W-DW group's. In contrast, 72W-SGT group's level was the highest in comparison to those of the control groups and those of deviation from 10W, 52W and 72W in SGT groups was also smaller than control groups'. (5) The levels of glucose, total-bilirubin, ALT, AST, BUN, CRP, hs-CRP, homocystein, Na, K, Na/K ratio, Cl and Ca in serum of SGT groups showed no significantly changes in comparison to those of the control groups. These results suggest that oral administration of Suhwagije-tang distillate should be effective in promoting growth and preventing rapid decline of testosterone in aging rats does not cause any imbalance in liver, kidney and electrolyte.

Transglutaminase 2 mRNA Expression in Salivary Gland Tumor Cell Line

  • Purpose: Transglutaminase 2 (TGase 2) is expressed by tumor necrosis factor-${\alpha}$ in various carcinoma. The role of TGase 2 expression in salivary gland tumors is not clear yet. Established slaivary gland tumor (SGT)cell line has been used to study the pathogenesis of salivary gland adenocarcinoma on a cellular level in vitro. The pupose of this study were to examine mRNA expression of TGase 2 in SGT cell line compared to other tumor cell lines, and to apply these results to the pathogenesis of salivary gland tumor. Materials and Methods: After SGT, SCC-15, HN 4, and HeLa tumor cell lines were cultured under preconfl uency, and 3 days after postconfl uency, the cells were harvested for total RNA extraction and cDNA preparation. Result: Reverse transcription polymerase chain reaction for semiquantitative mRNA analysis was done. TGase 2 mRNA expression was not induced by confl uency in all the cell lines. TGase 2 mRNA expression was variable but markedly enhanced in SGT cell line. Conclusion: mRNA expression of TGase 2 should play an important role in the pathogenesis of SGT cell line originated from ductal cell.

  • Steam generator tubes (SGTs) in nuclear power plants (NPPs) are a boundary between the primary side generating heat by nuclear fission and the secondary side generating electric power by a turbine. The water inside the SGT is high temperature and high pressure. Therefore, defects and magnetic phases (MPs) are partly produced in non-magnetic SGT by high stresses and temperatures. This causes trouble regarding the safety of SGTs but it is difficult to detect the MP using the conventional eddy current technique (ECT). In particular, a circumferential defect (CD) and circumferential magnetic phase (CMP) cannot detected by ECT. Consequently, a new method is needed to detect CDs and CMPs in SGT. A new U-type yoke with two types of coils was designed and the reactance signal by the CMPs and CDs in the SGT material was simulated.

Management of patients with allergy to local anesthetics: two case reports

  • Lidocaine is the most commonly used local anesthetic (LA) agent in various dental as well as oral and maxillofacial procedures. Although rare, adverse effects and allergic reactions to lidocaine have been reported. In patients with suspected allergy to LA or a history of such reaction, careful history-taking and allergy testing should be performed to choose an alternative LA agent to avoid any adverse effects. Here, we present two cases of delayed hypersensitivity reaction to lidocaine, wherein the patients presented with erythema, edema, and itching. Intradermal testing confirmed allergic reaction to lidocaine, and the patients underwent successful dental treatment using an alternative LA agent. This report highlights the importance of allergy testing prior to LA use considering the serious consequences of allergy to these agents and describes the management of such patients using an alternative LA agent.
