Growth Inhibition of Extract from Sulfur fed Duck Carcass against Various Cancer Cell Lines (유황오리 추출물의 각종 암세포에 대한 생육억제 효과)

  • This study was carried out to investigate the anticancer effect of extracts from sulfur fed duck carcass. Growth inhibition of cancer cell lines was measured by MTT assay. Eleven cancer cell lines, such as Calu-3(human lung carcinoma), SK-MES-1(human lung carcinoma), HL6O(human leukemia), KB(human epidermoid of mouth carcinoma), Farrow(human melanoma), HEP-2(human larynx carcinoma), SNU-1(human stomach carcinoma), K-562 (human leukemia), WiDr(human colon carcinoma), P388(mouse leukemia) and 3LL(mouse lung carcinoma) showed the growth inhibition higher than 50%, but those, such as SF-188(human brain carcinoma), A-549(human lung carcinoma) and HEC-lB(human uterus carcinoma) showed the growth inhibition lower than 50% in the extract of sulfur fed duck carcass at the concentration of 10 mg/㎖. The sulfur fed duck carcass extract had better growth inhibition than the normal counterpart against various cancer cell lines at the concentration of 10 mg/㎖. When the effect of growth inhibition of an effluent by different concentrations of methyl alcohol(25, 50, 75 and 100%) tested on Diaion HP-20 column chromatography, an effluent by concentration of 100% methyl alcohol showed the most strong effect of growth inhibition against HEP-2(human larynx carcinoma).

Psychosocial Stress Levels and Related Factors in some Rural Residents (일부 농촌지역 주민에서 사회심리적 스트레스 수준과 관련요인)

  • Objectives: This study was performed to investigate the levels of psychosocial stres and to identify its Methods: The study subjects were 1,806 adults aged 40-70 years living in Naju City, Jeollanamdo. Sociodemographic characteristics, health-related behavior, social suport and personality were collected for statistical analysis. The measurement instrument of psychosocial stress was psychosocial well-being index. The regression analysis. Results: The results showed that 16.6% of subjects were categorized as high stress, 63.4% as moderate, and 20.0% as normal. Overall, the mean value of PWI was 17.29.8 and significantly diferent by socioeconomic characteristics, health-related behavior, social suport and personality. In the case of men, the income, decreased positive social suport, increased negative social suport, type A behavior pattern, decrease of internal locus of control or poorer self-esteem. In the case of women, the PWI was significantly deteriorated in the subjects with no job, no spouse, decreased positive social suport, increased negative social support, type A behavior pattern, decrease of internal locus of control or poorer self-esteem.Conclusions: The psychosocial stres for rural residents was related with social suport and personality. Thus, these results should be considered to reduce stres levels in rural adults.

Nucleotide Sequence and Cloning of sfs4, One of the Genes Involved in the CRP-Dependent Expression of E. coli mal Genes. (CRP 의존성 maltose 대사 촉진 유전자 sfs4의 클로닝 및 염기배열 결정)

  • In Escherichia coli, CRP forms a complex with cAMP and acts as a transcriptional regulator of many genes, including sugar metabolism operons. The E. coli MK2001, which is introduced the altered crp, is functional in the expression of lac, ara and man, in the absence of cAMP. However, the expression of mal gene is fully activated by the addition of cAMP or cGMP. The object of the study is cloning of the sfs (sugar fermentation stimulation) genes, which was involved in regulation of mal gene expression with the altered crp gene, and structural analysis and characterization of the genes at the molecular level. We have cloned 5 different E. coli genes which stimulate the maltose metabolism in a crp, cya::km (MK2001) background. Newly identified genes were designated as sfs. One of the sfs genes (pPC1), located at the 53.2 min map position on the E. coli chromosome, was further analyzed. Expression of the genes, which is involved in maltose metabolism, malQ (amylomaltase), was increased to 5.8-fold in the presence of a plasmid, pAP5, containing the subcloned sfs4 gene. The nucleotide seguence of a common 2,126 bp segment of the pPCM1 was determined and two open reading frames (ORF1 and ORF2) were detected. The ORF1 encodes the sfs4 gene and ORF2 encodes a truncated protein. Potential CRP binding site is located in the upstream of the putative promoter in the regulatory region. Expression of the cloned sfs4 gene was positively regulated by the cAMP-CRP complex.

Effects of space allowance on the social behavior of Korean native cattle (Bos taurus coreanae) steers (한우 거세우의 사회 행동에 공간 허용이 미치는 영향)

  • This study was carried out to find out how space allowance affect the social behavior of Korean native cattle (Bos taurus coreanae) steers. Twelve Korean native cattle (Bos taurus coreanae) steers were used as subjects, each of which was 30-month-old and observation period was from June to July 2003. Five (T1) and seven (T2) steers were allotted to two pens of $5m{\times}10m$ in a building with slate roof and open sides respectively. They were fed at 09:00 h and 16:00 h, twice a day. The behaviors of steers were recorded from 06:00 h to 17:00 h, using two color CCD cameras (Samsung SDC-411, Korea), one B/W CCD cameras (Samsung SBC-340, Korea), one multiplexer (Samsung SDM-081, Korea) and a time lapse VCR (Samsung SRV-30, Korea). The behaviors of each steer were recorded every 2 min using an instantaneous point sampling method. While the mean percentage of time budget in WA of T1 was lower than that of T2 (p<0.05), the mean percentage of time budget in SF of T1 was higher than that of T2 (p<0.05). When it gets hot, steers in T1 rested from 10:00 h to 14:00 h when it gets cool, showing 40~80% of LD rate while steers in T2 rested from 12:00 h, when it very hot to 17:00 h, showing 20~50% of LD rate, which is relatively low. Steers in T1 were fed from 06:00 h to 08:00 h when it was cool and from 16:00 h to 18:00 h, showing 20~45% of EA rate while steers in T2 were fed from 08:00 h to 14:00 h when it was hot, showing 25~50% of EA rate. In conclusion, it turned out that the number of steers affected their social behavior, and T1 was better environment than T2 in terms of welfare.

THE SEALING ABILITY OF OBTURATION TECHNIQUES IN OPEN APEX (개방 근첨 치아의 근관 충전방법에 따른 치근단 폐쇄효과에 관한 연구)

  • The purpose of this study was to compare the leakage of four different obturation techniques in conjunction with immediate apical barrier of ${\beta}$-tricalcium phosphate(TCP) in teeth with open apex. Eighty single-rooted human premolar teeth were prepared and sectioned horizontally, so maximum diameter in apex was 4mm. Apical defects that were similar to open apex, were created with #1/2 round bur and SF104R bur. The apical foramen were opened to a size 80 file extended 3mm beyond the apex. The teeth were placed into the oasis block soaked saline to simulate periapical tissue often associated with pulpless teeth and received apical barriers consisting of TCP followed by obturation using lateral condensation technique, vertical condensation technique, continuous wave technique and thermoplasticized gutta-percha injection technique. Two unobturated teeth served as positive and negative controls. Teeth were immersed in resorcinol-formaldehyde resin for S days at $4^{\circ}C$, and the resin was allowed to polymerize completely for 4 days at room temperature. Teeth were then sectioned horizontally at 1.5mm(level 1), 2.5mm(level 2) and 3.5mm(level 3) from the apex, and examined under a stereomicroscope at ${\times}40$ magnification. The photographs were taken at ${\times}40$ magnification of the filling in each level and scanned. The leakage length in tooth/resin interface was measured at each of the three levels. Each ratio of leakage was obtained by calculating the ratio of the leakage length of canal wall infiltrated with resin to the total length of the canal and was analyzed statistically(One-way ANOVA and Scheffe test). The result were as follows : 1. At the level 1, there was the least leakage in the thermoplasticized gutta-percha injection technique group(group 4), but there was statistically significant(p<0.05). 2. At the level 2, there was the least leakage in the thermoplasticized gutta-percha injection technique group(group 4), and the most leakage in the continuous wave technique group(group 3). There was statistically significant difference between the thermoplasticized gutta-percha injection technique group and the continuous wave technique group(p<0.05). 3. At the level 3, there was the least leakage in the thermoplasticized gutta-percha injection technique group(group 4), but there were no statistically significant differences between other groups(p>0.05). These results suggest that thermoplasticized gutta-percha injection technique which had 1mm apical gutta-percha matrix after the formation of TCP apical barrier, can demonstrate favorable apical sealing.

Improving Initial Abstraction Method of NRCS-CN for Estimating Effective Rainfall (유효우량 산정을 위한 NRCS-CN 모형의 초기손실량 산정방법 개선)

  • In order to improve the runoff estimation accuracy of the Natural Resources Conservation Service (NRCS) curve number (CN) model, this study incorporated rainfall and maximum potential retention as contributors for initial abstraction. The modification was proposed based on 658 rank-order data of rainfall and subsequent runoff from 15 watersheds. The NRCS-CN model (M1), one of its inspired modified model (M2), and the proposed model (M3) were analyzed employing different CN approaches. Using tabulated, calculated and least squares fitted CNs ($CN_T$, $CN_C$, $CN_{LSF}$, respectively), the models' performances were evaluated based on Root Mean Square Error (RMSE), Nash-Sutcliffe Efficiency (NSE), and Percent Bias (PBIAS). Applications of model M1, M2, and M3, respectively exhibited watershed cumulative mean [RMSE (23.60, 18.12, 16.04), NSE (0.54, 0.73, 0.79), and PBIAS (36.54, 20.25, 12.00)]. Similarly, using CNC (for M1 and M2 model) and $CN_{LSF}$ (for M3 model), the performance of three models respectively were assessed based on watershed cumulative mean [RMSE (17.17, 15.88, 13.82), NSE (0.76, 0.80, 0.85), and PBIAS (3.06, 4.47, 0.11)]. The proposed model (M3) that linked all of the NRCS-CN variants showed more statistically significant agreement between the observed and estimated data.

  • Many greenhouse gases occur naturally, such as water vapor, carbon dioxide, methane, nitrous oxide, and ozone. Others such as hydrofluorocarbons (HFCs), perfluorocarbons (PFCs), and sulfur hexafluoride ($SF_6$) result exclusively from human industrial processes. Current global warming has been linked to anthropogenic greenhouse gas concentration increases. Methods to quantify greenhouse gas emissions during animal agriculture and the possibility to apply those to circumstance in the world were studied. Various chamber methods with trace gas analyzer (TGA) were used to quantify greenhouse gas emissions from stored manure. Methane fluxes from pails storing liquid swine manure were measured. Methane emissions increased a little with time and mean was 393.2 ${\mu}g\;m^{-2}\;s{-1}$ (standard error : 4 ${\mu}g\;m^{-2}\;s{-1}$).

Effect of Control of Leaf Number on Growth and Chemical Characteristics in Flue-cured Tobacco (황색종담배의 엽수조절이 생육형질 및 화학성분에 미치는 효과)

  • The study was carried out to investigate the effect of control leaf number on growth and chemical characteristics, and on yield and quality In (rue-cured tobacco The rest upper leaves of 14, 16, and 18 leaves per plant were removed at button stage, and the plants were harvested :0, 12, and 16 leaves with discard 4 leaves of the bottom, respectively. 1. There were showed a tendency to increase in length (LL), width (LW), area (LA), fresh (LFW) and dry weight (LDW) per leaf as leaf numbers were decreased, while those per plant and the ratios of increase of those per plant were decreased. 2. In compensation effect (CE), there were increase in LL, LFW, LDW, LL/LW and LDW/LFW with each decreased in leaf numbers while LW was decreased, LA unchanged. 3. There were significant Increases in LL, LW, and LA at 5th~8th stalk positions form bottom (SP), while those .at 13th~16th SF were decreased. Also, LDW at 5th~l0th, and LDW/LFW and weight Per unit leaf area at 11th 16th were Increased as leaf numbers decreased. 4. Mean per plant and CE of nicotine were increased, but those of sugar were decreased with each decrease In leaf numbers. In addition, nicotine contents at 5th~14th SP except for 9th~10th were increased, otherwise sugar contents at 7th~14th were decreased. 5. There were showed a tendency to increase in grade per Plant and in tirade at 5th~8th SP, but decrease in CE of grade as leaf numbers were decreased. There wee significant decrease in grade at 11th~14th SP. Also, ratio of 5 grade was decreased, while ratios of 3 and 4 grades were increased. 6. When the 2 leaves at 3rd~4th SP were harvested, price was decrease 2.6% and yield was increased 5.8 %. 7. When the 6 leaves from convention were removed at but ton stage, effects of apparent, compensational, and net per leaf on yield were -3.9%, +2.6%, and -6.5%, respectively, and those on price were +1.1%, -1.6% and +2.7%, respectively.

Germ Line Transformation of the Silkworm, Bombyx mori L. with a piggyBac Vector Harboring the Human Lactoferrin Gene (락토페린 유전자도입 piggyBac 벡터에 의한 누에 형질전환)

  • Lactoferrin, an ion-binding 80-kDa glycoprotein, has been suggested to have many biologic activities, such as facilitating ion absorption and having antimicrobial and anti-inflammatory effects. Several of these activities are likely to only be facilitated by human lactoferrin because they depend on the binding of human lactoferrin to specific receptor. To produce recombinant human lactoferrin to animal foods using transgenic silkworm, Bombyx mori L, we have cloned and sequenced the cDNA encoding for a human lactoferrin (HLf) from the mRNA in mammary tumor line (GI-101). As a result, the 2.5-kb fragment of HLf gene was cloned with pGEM-T vector and then this fragment was sequenced. In the nucleotide sequence analysis, single open reading frame of the 2,136-bp encoding for a polypeptide of 712 amino acid residues was detected. On the other hand, we constructed a recombinant plasmid(pPT-HLf), containing human lactoferrin gene for germ line transformation of the silkworm using a piggyBac transposon-derived vector. A nonautonomous helper plasmid encodes the piggyBac transposase. Approximately 6.7% of individuals in the G0 silkworms expressed green fluorescent protein (GFP). PCR analyses of GFP-positive silkworms (G0 and G1) revealed that independent insertions occurred frequently. Furthermore, Western blot analysis showed that the recombinant HLf expressed in hemolymph has the same molecular weight (80 kDa) as a native protein. On the basis of these experiments, expression of HLf in next generation of transgenic silkworm is now in process.
