Production of Feather-Sexing Korean Native Commercial Chickens (깃털 성감별 상업용 토종 실용닭 생산)

  • The feather-sexing method is widely used commercially for chick sex identification. However, for feather-sexing to be industrially practical, the early-feathering (EF) and late-feathering (LF) genes must existed within the foundation stock, a suitable feather-sexing lines must be established, and the accuracy of sex identification by feather-sexing must be ensured. Therefore, this study introduces the method of constructing the Korean native chickens (KNC) feather-sexing lines using EF and LF genes and evaluates the effectiveness of feather sex determination on commercial chicks produced from the constructed KNC lines. The results showed that both EF and LF chickens existed within the foundation stock, with the frequency of LF genes ranging from 0 to 0.205. In feather-sexing line establishment, the paternal strain of the grandparent stock (GPS) was fixed as EF (kk) for both sexes, while the maternal strain was composed of males with LF homozygotes (ZKZK) and females with EF (ZkW). Thus, in the parent stock (PS), male breeder had EF (ZkZk) and female breeder had LF (ZKW), resulting in chicks produced from their crosses having LF (ZKZk) for males and EF (ZkW) for females, allowing sex determination based on feather development. Additionally, to evaluate the effectiveness of feather-sexing for the produced commercial chicks, a study was conducted on 1,000 samples of the produced chicks to investigate the concordance between vent-sexing and feather-sexing, showing a matching rate of 93.1%.

Skeletochronological Age Determination and Comparative Demographic Analysis of Two Populations of the Gold-spotted Pond Frog (Rana chosenica)

  • To obtain demographic information on threatened gold-spotted pond frog (Rana chosenica Okada, 1931) populations, we determined the ages of 45 male and 13 female frogs (20 males and 9 females from Cheongwon and 25 males and 4 females from Tae-an) and compared the age structures and growth patterns of the two populations in 2006. The snout-vent length (SVL) and body weight of female frogs were greater than those of male frogs in both populations. Male frogs' ages ranged 2 to 7 years old and females' ages ranged 3 to 6 years old. In both populations, 4 years old male frogs were the most abundant age-sex class. The age structures of the two populations were significantly different and the growth coefficients of male frogs from the Cheongwon population were greater than those from the Tae-an population. The mean age of males from the Tae-an population was higher than that from the Cheongwon population. However, the SVL and body weights of male frogs were not different between two populations and there was no difference between the two populations in the mean male SVL at any age. The results could increase our understanding of the life-history of this threatened frog and may be useful in conservation planning.

  • Recently, through the development of the primer extension preamplification(PEP) method which amplifies the whole genome, simultaneous multiple DNA analysis has become possible. Whole genome from each single cell can be amplified using 15 base oligonucleotide random primer. The greatest advantage of PEP-PCR is the ability to investigate several loci simultaneously and confirm results by analysing multiple aliquots for each locus. This technique led to the development of preimplantation genetic disease diagnosis using blastomere from early embryo, sperm, polar body and oocyte. In this study, we applied PEP-PCR in 20 cases of single amniocyte and 20 cases of single chorionic villus cell for the clinical application of the prenatal and preimplantational genetic diagnosis. We analysed 7 gene loci simultaneously which are 46, 47 exons related to dystrophin gene, two VNTR (variable number tandem repeat) markers using 5'dysIII, 3'CA related to dystrophin gene and DYZ1, DYZ3, DYS14 regions on chromosome Y. In all the tests, 97.5% of PEP-PCR amplifications with single cells were successful. We obtained 38/40 (95%) accuracy in gender determination through chromosome analysis comparison. Therefore, these results have significant implications for a sperm or oocyte analysis and prenatal or preimplantational genetic diagnosis.

  • The purpose of this research is the estimation of a transfer ratio according to transfer time reflecting passenger attributes such as sex, age, income, job, the car ownership, and other variables with the assumption that a transfer ratio would be different depending on each passenger attribute. This research tested transfer time through a question-survey, estimated transfer time according to the passenger attributes using a data sample, and presented some applicable models about marginal transfer time for the case of the determination of transfer ratios according to transfer time. In this research the sample which had been surveyed for passengers walking to access a transfer station was used to test and estimate and the question was present walking time to the transfer time and the marginal transfer walking time.

  • Sexing from bovine embryos fertilized in vitro implicates a possibility of the sex controlled cattle production. This study was carried out to produce the sex determined cattle through the embryonic sexing by fluorescence in situ hybridization (FISH) technique. FISH was achieved in in vitro fertilized bovine embryos using a bovine Y-specific DNA probe constructed from the btDYZ-1 sequence. Using this probe, a male-specific signal was detected on 100% of Y-chromosome bearing metaphase specimens. The analyzable rate of embryonic sexing by FISH technique was about 93% (365/393) regardless of embryonic stages. As tested single blastomere by FISH and then karyotype with their biopsied embryos, the accuracy of sex determination with FISH was 97.6%. We tried the embryo transfer with sex determined embryos on 15 cattle. Among them, the 5 cattle delivered calf with expected sex last year.

  • DNA typing is often used to determine identity from human remains. Recently, the molecular biological analysis of ancient deposits has become possible since methods for the recovery of DNA conserved in bones or teeth from archaeological remains have been developed. In the field of archaeology, one of the most promising approaches is to identify the individuals present in a mass burial site. We performed nuclear DNA typing and mitochondrial DNA sequencing analysis based on PCR from a Korea ancient human remain excavated from Sa-chon Nuk-island and civilian access controlline(CACL). A femur bone were collected and successfully subjected to DNA extraction, quantification, PCR amplification, and subsequently typed for several shot tandem repeat(STR)loci. 4 types of STR systems used in this study were CTT multiplex(CSF1PO, TPOX, TH01), FFv multiplex(F13A01, FESFPS, vWA), Silver STRⅢ multiplex(D16S539, D7S820, D13S317), and amelogenin for sex determination. This studies are primarily concerned with the extraction, amplification, and DNA typing of ancient human bone DNA samples. Also, it is suggestive of importance about closely relationship between both fields of archaeology and molecular biology.

Age Studies on the Butter Fish Population from Southwestern Waters of Korea

  • The present paper concerns the age determination and growth of butter fish, Pampus argenteus from the southwestern waters of Korea by otolith reading. 743 specimens taken by stow-net in the southern part of the Yellow Sea and northeastern part of the East China Sea during the period from October 1972 to September 1973 were examined. Results of the study are summarized as follows: 1. Sex ratio of females to males was found to be 2:1. 2. Ring marks on the otolith were found to be formed twice a year, once during the period of January-May and the other time in September. 3. The Lee's phenomenon was observed on the otolith sample. 4. The relationship between the radius of otolith(R) and fork length(L) was found to be as follows: R=0.3069+0.0133L 5. Calculated fork length at the time of otolith ring formation are found to be as follows:I-ring,71.67mm; II-ring, 125.05mm; III-ring, 168.65mm; IV-ring, 201.74mm; V-ring, 225.80mm; VI-ring, 240.84mm. 6. Maximum fork length calculated according to the diagram of Walford's growth transformation was found to be 281.5mm. 7. Growth curve, when related to the von Bertalanffy's equation, was laid out as $L_{t}=281.5[1-e$^{-0.674(t-0.128)}]$

  • We determined the maturity and growth of the Marbled sole, Pseudopleuronectes yokohamae based on monthly sampling in the West Sea of Korea from February 2009 to December 2010. Determination of sex of P. yokohamae was by gonadal inspection, and age and growth were determined by analyzing the otolith. The biological minimum size of P. yokohamae female was 24.5 cm. The seasonal changes in the ratio of a translucent zone to an opaque zone revealed that the end of the translucent zone of the otolith were annuli formed in May once a year. We compared 8 type growth equations with several length-at-age data. By using the length-at-monthly age data of these, the calculated von Bertalanffy growth equations were $L_t=41.7(1-e^{-0.47(t+0.24)})$ for females and $L_t=32.7(1-e^{-0.75(t+0.01)})$ for males. We estimated that 24.5 cm was the total length of 2 year old P. yokohamae spawns in the cold water season from January to April, and grows to 8 years old for females and 6 years old for males in the West Sea of Korea.

  • For the purpose of estimating the sex and age in the remains of Catholic martyrs, the authors investigated size, shape and attrition of teeth, morphology of pulp cavity, and the changes of jaws. The 22 teeth, upper and lower jaws, the 23 teeth, upper and lower jaws, the 6 teeth and the 1 tooth were referred from The Pusan Research Institute for Church History in Pusan Catholic Parish. The results were as follows : 1. The 22 teeth, upper and lower jaws presumed to be Lee, Kwan Bok were evaluated to be in the latter half of female thirties. 2. The 23 teeth, upper and lower jaws presumed to be Kim, So ja, Maria were evaluated to be in the middle of male thirties. 3. The 6 teeth presumed to be lee, Jung Suk were evaluated to be in the earlier half of male forties. 4. The 1 tooth presumed to be Lee, Sam jo was evaluated to be in the latter half of male forties.

A Study on the Distributions of Paraoxonase Activity and the Factors affecting Paraoxonase Activity of a Rural Population (일부 농촌주민 혈중 Paraoxonase 활성도의 분포 및 관련인자에 관한 조사연구)

  • The paraoxonase (E. C. 3.1.1.2) is a major enzyme to detoxicate the organophosphorus and carbamate which are the most widely used as the agricultural spraying insecticides. To investgate the distributions of plasma paraoxonase activity and the factors affecting the enzyme activity, the plasmas of 945 Korean rural population were analysed with the modified Krisch's direct sphectrphotometry method. Three indices of the enzyme activity - basal activity, stimulated activity (by NaCl), % stimulation - were obtained from the analysis. Three indicies suggested unimodal distributions, so we couldn't identify the low activity group risk group to organophosphorus & carbamate insecticides poisoning. There is no significant relation between 3 actvity indicies and sex, age, or history of insecticide use (p>0.05). The basal activity and the stimulated activity have significant relationship and high coefficient of determination with the activities of their parents ($r^2$=0.30, 0.24 ; p<0.05), but the % stimulation does not ($r^2$=0.02 ; p<0.05). These results suggest that the activity of paraoxonase is determined mainly by the genetic factor.
