아프리카 난 Aerangis confusa의 향기성분 methyl 3-methyloctanoate의 합성 (Synthesis of Methyl 3-methyloctanoate, the Key Perfume Component of African Orchid Aerangis confusa)

  • Branched methyl group을 가지는 아프리카산 난(Aerangis confusa, Aerangis kirkii) 꽃의 천연향기성분인 methyl 3-methyloctanoate를 값싼 미생물 발효산물인 itaconic acid를 출발 물질로 하여 제조한 branched metyl group을 부분구조로 가지는 2-methyl-1,4-butandiol를 중간체로 하여 전 공정 9단계로 합성하였다. 최종 향기성분 metyl 3-methyloctanoate의 methyl ester대신 다양한 알킬 ester 화합물을 합성하여 그 향을 비교한 결과 서로 상이한 향 특성을 나타내므로 화장품이나 식품산업에 있어서 유용한 향료조성물의 첨가소재로 개발 가능하리라 사료된다.

해양 원형 규조류 Cyclotella meneghiniana 성장 연관 미생물 군집구조 분석: 배양단계에 따른 증거 (Associated Bacterial Community Structures with the Growth of the Marine Centric Diatom Cyclotella meneghiniana: Evidence in Culture Stages)

  • There are a number of pieces of evidences that suggest a link between marine diatoms and microorganisms, but knowledge about related microbial communities is greatly lacking. The present study investigated the microbial community structures related to the growth of the marine diatom Cyclotella meneghiniana. We collected free-living bacteria (FLB) and particle-associated bacteria (PAB) at each growth stage (e.g., lag, exponential, stationary and death) of the diatom, and analyzed their bacterial 16S rDNA using pyrosequencing. Metagenomics analysis showed that community structures of FLB and PAB differed considerably with the progress of growth stages. FLB showed higher diversity than PAB, but variation in the different growth stages of C. meneghiniana was more evident in PAB. The proportion of the genus Hoeflea, belonging to the order Rhizobiales, was dominant in both FLB and PAB, and it gradually increased with the growth of C. meneghiniana. However, Enhydrobacter clade tended to considerably decrease in PAB. In addition, Marinobacter decreased steadily in FLB, but first increased and then decreased in PAB. These results suggest that Hoeflea, Enhydrobacter, and Marinobacter may be closely related to the growth of diatom C. meneghiniana.

Cloning of the Alkaline Phosphatase Gene from Kluyveromyces fragilis

  • In order to clone the gene coding for alkaline phosphatase in the yeast Kluyveromyces fragilis, a genomic library was constructed using the yeast-E. coli shuttle vector pHN114 as a cloning vector. From the genomic library, a clone carrying the gene was isolated and the plasmid was designated as pSKH101. A restriction enzyme map was made using this plasmid. Subcloning experiments and complementation studies showed that alkaline phosphatase was active only in the original 3.1 kb insert. Southern hybridization analysis confirmed that the cloned DNA fragment was derived from K. fragilis genomic DNA. Using a minicell experiment, the product of the cloned gene was identified as a protein with a molecular weight of 63 KDa. A 0.6 kb HindIII fragment, which showed promoter activity, was isolated using the E. coli promoter-probe vector pKO-1.

  • Streptomyces속 균이 생산하는 trypsin inhibitor의 trypsin에 대한 반응성을 조사해 본 결과 본 저해물질은 crystalline trypsin (20.000 unit, hog pancreas)에 대하여 1/8량에서 약 50%의 저해률을 나타내었으며 trypsin에 대한 저해양상은 mixed noncompetitive-competitive inhibition type이었으며 enzyme-inhibitor complex를 빨리 형성하는데 반응액중 isoleucine이 공존하면 활성이 증가되였으며 Ag$_{+}$ Hg$_{++}$등의 금속ion은 강하게 본 저해물질의 작용을 억제하였다. 저해률은 사용한 기질의 종류에 따라 차이가 나서 albumin을 사용하였을 때는 casein이나 hemoglobin을 사용하였을 때보다 저해률이 높았다. 그러고 혈액의 응고에 대해서도 저해작용을 나타내었다.

  • Gibberellins (GAs) are a group of phytohormones that control many developmental processes in higher plants. We report the cloning and expression pattern of gibberellin biosynthesis genes from a new GA-producing fungus, Fusarium proliferatum (strain KGL0401). These genes sequences are deposited in the National Center for Biotechnology Information (NCBI) under accession numbers EF119831, EF119832, DQ313173, DQ313174, DQ313175, DQ313176, and DQ313177. The expression level of these genes was maximal at a 0.5 M : 0.17 M carbon : nitrogen ratio, and minimal at a 0.25 M : 0.47 M carbon : nitrogen ratio.

  • 진균류의 세포벽제거 및 이에 따른 protoplast생성 등에 많이 이용되어지고 있는 chitinase를 Streptomyces sp. 115-5 균주로부터 생산하여 순수 정제한 다음, 이 chitinase의 작용을 저해하는 포도당의 저해양상을 조사하였다. 포도당 외에 D-glucuronic acid, D-sorbitol및 D-xylose등도 chitinase의 활성을 저해하였다. 그런고로, 포도당 분자에 의한chitinase의 활성 저해 효과에는 위의 분자들의 공통부분인 2번, 3번 및 4번 탄소의 hydroxyl group들의 구조위치가 중요한 영향을 가진다는 것을 알 수 있다. 그리고 포도당에 의한 chitinase의 저해양상은 competitive inhibition 과 non-competitive inhibition 과의 혼합 저해형으로 나타났다.

Characteristics of the Conditioned Medium Produced by CA-12 Lymph Node Stromal Cells

  • CS-2l lymphoma cells that preferentially metastasize to lymph nodes after s.c. inoculation into BALB/c mice were grown in vitro in the presence of CA- 12 stromal cells isolated from lymph nodes. In order to obtain fundamental data on the identification and characterization of the soluble factors produced by CA-12 stromal cells, the conditioned medium of CA-12 stromal cells that inhibited apoptosis of CS-21 cells was examined. Various analytical treatments revealed that the soluble factors in CA-12 conditioned medium are very sensitive to heat treatment and trypsinization. Moreover CA-12 conditioned medium has an affinity with heparin but not with Con-A. In addition to these, the activity of CA-12 conditioned medium was blocked by H-7, a PKC inhibitor, but the conditioned medium could not induce the differentiation of thymocytes. We concluded that CA-12 conditioned medium contains stromal cell-derived apoptosis-inhibitory molecules that play an important role in proliferation of CS-2l cells by suppressing cell apoptosis.

  • 토양으로부터 분리한 포도당 이성화효소를 강하게 생산하는 방사균을 Bergey's manual 8판에 따라 동정한 결과 Streptomyces antibioticus 근록의 균주이었다. 본 균의 배양액으로부터 균체를 모아서 해사를 넣고 파쇄 하여 증류수로 추출하고 Mn-처리를 하여 핵단백질을 제거한 후 황산 ammonia 분획침전(0.5∼0.8포화), 수석, DEAE-cellulose column chromatography, DEAE-sephadex (A-50) column chromatography 및 sephadex G-200에 의한 gel filtration을 거쳐 비활성도로 약 380배, 회수율 25% 정도로 분리 정제하였다.

저독성의 새로운 양이온성 리포좀을 이용한 유전자의 전달 (Gene Delivery using a Novel Cationic Liposome with Low Toxicity)

  • 콜레스테롤 유래의 양이온성 리피드 2-aminoethylcarbamate-cholesterol(Chol-E)를 합성하여 이의 리포좀을 제조하였다. 리포좀은 다양한 비율로 중성지방인 DOPE와 섞어서 만든 후 $100{\sim}200nm$의 membrane으로 extrusion시켜 균일한 리포좀을 제작하여 크기 및 전위를 측정하였다. 형광단백질 및 luciferase plasmid의 발현을 여러가지 세포에서 확인한 결과 우수한 발현양상을 보였으며 혈청이 있는 조건에서도 발현이 증가임을 볼수 있었으며, 합성 ODNs의 전달도 adipocyte cell 에서도 잘 이루어지는 것을 확인할 수 있었다 임상실험에 쓰이는 저독성의 DC-chol에 비교하여도 독성이 적은 리포좀임을 알 수 있으며 혈청하에서도 안정하게 유전자를 전달할 수 있는 응용성이 기대되는 새로운 리포좀을 제조하였음을 알 수 있다.

A Review of EOS Thermal Control Logic for MSC on KOMPSAT-2

  • MSC (Multi-Spectral Camera) system is a remote sensing instrument to obtain high resolution ground image. EOS (Electro-Optic System) for MSC mainly consists of PMA (Primary Mirror Assembly), SMA (Secondary Mirror Assembly), HSTS (High Stability Telescope Structure) and DFPA (Detector Focal Plane Assembly). High performance of EOS makes it possible for MSC system to provide high resolution and high quality ground images. Temperature of the EOS needs to be controlled to be in a specific range in order not to have any thermal distortion which can cause performance degradation. It is controlled by full redundant CPU based electronics. The validity of thermistor readings can be checked because a few thermistors are installed on each control point on EOS. Various kinds of thermal control logics are used to prevent 'Single Point Failure'. Control logic has a few set of database in order not to be corrupted by SEU (Single Event Upset). Even though the thermal control logic is working automatically, it can also be monitored and controlled by ground-station operator. In this paper, various ways of thermal control logic for EOS in MSC will be presented, which include thermal control mode and logic, redundancy design and status monitoring and reporting scheme.
