Study of a 13-weeks, Repeated, Intramuscular Dose, Toxicity Test of Sweet Bee Venom in Sprague-Dawley Rats

  • Objectives: This study was performed to analyze a 13-week repeated dose toxicity test of Sweet Bee Venom (SBV) extracted from bee venom and administered in Sprague-Dawley (SD) rats. Methods: Male and female 5-week-old SD rats were treated once daily with SBV (high-dosage group: 0.28 mg/kg; medium-dosage group: 0.14 mg/kg; or low-dosage group: 0.07 mg/kg) for 13 weeks. Normal saline was administered to the control group in a similar manner (0.2 mL/kg). We conducted clinical observations, body weight measurements, ophthalmic examinations, urinalyses, hematology and biochemistry tests, and histological observations using hematoxylin and eosin (H&E) staining to identify any abnormalities caused by the SBV treatment. Results: During this study, no mortality was observed in any of the experimental groups. Hyperemia and a movement disorder were observed around the area of in all groups that received SBV treatment, with a higher occurrence in rats treated with a higher dosage. Male rats receiving in the high-dosage group showed a significant decrease in weight during the treatment period. Compared to the control group, no significant changes in the ophthalmic parameters, the urine analyses, the complete blood cell count (CBC), and the biochemistry in the groups treated with SBV. Compared to the control group, some changes in organ weights were observed in the medium-and the high-dosage groups, but the low-dosage group showed no significant changes. Histological examination of thigh muscle indicated cell infiltration, inflammation, degeneration, and necrosis of muscle fiber, as well as fibrosis, in both the medium- and the high-dosage groups. Fatty liver change was observed in the periportal area of rats receiving medium and high dosages of SBV. No other organ abnormalities were observed. Conclusion: Our findings suggest that the No Observed Adverse Effect Level (NOAEL) of SBV is approximately 0.07 mg/kg in male and female SD rats.

  • Recombinant human granulocyte-macrophage colony stimulating factor (hGM-CSF) is a glycoprotein and hematopoietic growth factors that regulates the proliferation of myeloid precursor cells and activates mature granulocytes and macrophages. In a previous study, we reported that hGM-CSF could be produced in transgenic rice cell suspension culture, termed rhGM-CSF. In the present study, we examined the repeated dose toxicity of rhGM-CSF in SD rats. The repeated dose toxicity study was performed at each dose of 50 and 200 ${\mu}g/kg$ subcutaneous administration of rhGM-CSF everyday for 28-days period. The results did not show any changes in food and water intake. There were also no significant changes in both body and organ weights between the control and the tested groups. The hematological and blood biochemical parameters were statistically not different in all groups. These results suggest that rhGM-CSF may show no repeated dose toxicity in SD rats under the conditions.

  • The purpose of this study is to see the radiation protection effect of the oral injected Dendranthema zawadskii var. latilobum (Maxim.) Kitam. extracts on the small intestine and uterus of female SD Rat as a natural radiation protection agent. The experimental group was divided into four groups: Normal Control group (NC group), Injected Dendranthema zawadskii var. latilobum (Maxim.) Kitam. extracts group (DZ group), irradiated group after injecting Dendranthema zawadskii var. latilobum (Maxim.) Kitam. extracts (DZ+IR group). The whole body of SD Rat was irradiated with gamma-ray 10Gy, and the administration of oral Dendranthema zawadskii var. latilobum (Maxim.) Kitam. Extract was 2 cc (71.56 mg/day/kg) once a day for 2 weeks. For this study, chages in blood cell levels, SOD assay, small intestine and uterus were observed. In the 21st white blood cell level, the DZ+IR group recovered to a normal level, and the IR group didn't. The IR group villus length was lower than other groups on Day 1. IR group was partially recovered, and DZ+IR group was recovered like the NC group on Day 21. In the case of the first-day endometrium, the IR group was thin and the boundary was cloudy, and the DZ+IR group was thicker and the boundary was clearer than the IR group. Day 21 IR group still did not recover, and DZ+IR group recovered like NC group. This is believed to have radiation protection effects in the blood cells and small intestine and uterus of the irradiated female SD Rat, and is expected to be useful for the study of natural radiation protection materials.

  • We investigated the interaction of serotonin and galanin (GA) by a double immunostaining method in the Japanese quail. Serotonin-immunoreactive (IR) cells were located in the paraventricular organ (PVO) and infundibular nucleus (IF). The number of the cells under short-day photoperiod (SD) was less in the dark phase than in the light phase. GA-IR cells were found in the PVO, IF and median eminence. The number of GA-IR cells in SD was significantly greater than that in long-day photoperiod (LD). Numerous GA- IR varicose fibers ran along serotonin- IR cell bodies and nerve fibers in the PVO and IF of the same sections. Very few serotonin-IR fibers ran along GA-IR cell bodies and GA-IR nerve fibers in the ventral part of the IF. The present results suggest that the possibility of functional interaction takes place between serotonin- and GA- IR neurons in the PVO and IF.

  • For this investigation, author isolated Streptococcus mitis strain SD-9 from the bacterial flora in the human dental plaque, which was incubated in brain-heart infusion media containing 5% sucrose at 37℃ for 24 hours. For the cytochemical demonstration of polysaccharide produced by this strain, a modified thiosemicarbazide osmium method (Critchley et al., 1967) was used. After fixation with this reagent, the harvested cells was suspended in 1% agar for the higher concentration of cells(Kellenberger et al., 1964). And they were dehydrated in the various concentration of ethanol, and embedded in Epon 812(Luft, 1961). Sectioning was done with the Sorvall MT-2 Porter Blum ultramicrotome by means of a glass knife, and the sections were stained with saturated uranyl acetate and lead citrate (Raynolds, 1963). All preparations were examined in a electron microscope, Hitachi HU-ll E-1 type. The morphological features of extracellular polysaccharide produced by Streptococcus mitis strain SD-9 were appeared in 3 structurally different forms, those are, electron dense fibrillar material linearly arranged adjacent to the outer surface of cell wall, highly electron dense globular material adjacent to the outer surface of cell wall, and strutureless fluffy meshwork of possible very fine filament.

황금의 간보호활성 및 Cytochrome P450 발현조절에 관한 연구 (Study on the Hepatoprotective Effect and Cytochrome P450 Regulation of Scutellaria Radix)

  • In this study, the liver protective effect of the hot water extracts of Scutellaria radix (SR) was investigated. The SR exhibited a hepatoprotective activity against $CCl_4$-induced liver damage in Sprague-Dawley (SD) rats and Chang cell. And the SR also showed significant decrease of malodialdehyde (MDA) and increase of glutathion (GSH), catalase activity in rat liver homogenate. The expression of cytochrome P450 2E1 (CYP2E1), measured by RT-PCR and western blot, was significantly decreased in the SR treated SD rats and Chang cell. But $CCl_4$ and SR has no significant effect on 1A1 and 3A1 isoform of cytochrome P450. Based on these findings, it is suggested that hepatoprotective effects of SR possibly related to antioxidative effects and downregulation of CYP2E1 expression.

보폐원탕(補肺元湯)이 노화쥐의 간장과 폐장 세포의 항노화 및 항산화능에 미치는 영향 (Anti-aging and Anti-Oxidant Effect of Bopyewon-tang in liver and Lung Cell of SD Rats)

  • 1. Objective The purpose of this study is to investigate the anti-aging and anti-oxidant effects of Bopyewon-tang(BPW) in Liver and Lung cell. 2. Methods This experiment was used the tissue of liver and lung cells of 6, 52 and 68 weeks old SD rats. Each age group was again divided into three groups. One group, as normal group, was not-treated cells, another group, as control group, was saline-treated cells, and the last group, as experimental group, was BPW -treated cells. After culture for 48 hours, each groups measured the level of SOD, GSH, MDA and NO in the tissue of liver and lung cells. 3. Results and Conclusion 1. The activity of SOD in liver of in 52 w-BPW group, 68 w-BPW group, and in lung of 68 w-BPW were significantly increased in comparison with those of the normal groups. 2. The levels of NO in liver of in 68 w-BPW group was significantly decreased in comparison with those of the normal groups. 3. The levels of MDA in lung of in 68 w-BPW group was significantly decreased in comparison with those of the normal groups.

Corynebacterium glutamicum에서 발현된 L-Threonine Aldolase를 이용한 파킨슨병 치료제 L-threo-2,3-Dihydroxyphenylserine (L-threo-DOPS)의 합성 (Synthesis of L-threo-2,3-Dihydroxyphenylserine (L-threo-DOPS) by Thermostable L-Threonine Aldolase Expressed in Corynebacterium glutamicum R)

  • Erro-prone PCR에 의해서 열안정성이 향상된 Streptomyces coelicolor A(3) 유래의 L-threonine aldolase를 Corynebacterium glutamicum R에서 과잉발현시키기 위하여 Corynebacterium용 vector plasmid인 pCRB1의 SD배열과 개시코든사이의 1염기를 제거한 고발현용 vector plasmid인 pCG-H44(2)를 구축하였다. pCG-H (2)에 의해서 형질전환된 C. glutamicum R 균주(CGH44-2)에서 L-TA를 발현시킨 결과, 기존의 Corynebacterium용 vector plasmid인 pCRB1(CGH44-1)보다 L-TA의 발현량이 높았다. L-threo-DOPS의 합성을 위한 최적조건은 $30^{\circ}C$, 0.1 M cirtric acid buffer(pH 7.0)이었으며, 0.1% TritonX-100를 첨가하였을 경우 보다 높은 활성을 보였다. 최적조건하에서 CGH44-2를 whole cell biocatalyst로 이용한 반복회분식반응에서 재조합대장균을 숙주로 이용한 경우보다 재조합Corynebacterium을 이용하였을 경우, 목적하는 L-threo-DOPS의 합성이 안정적으로 이루어졌다.

Anti-Cancer Effect of Ginsenoside F2 against Glioblastoma Multiforme in Xenograft Model in SD Rats

  • The glioblastoma multiforme (GBM) is the most common malignant brain tumor in adults. Despite combination treatments of radiation and chemotherapy, the survival periods are very short. Therefore, this study was conducted to assess the potential of ginsenoside $F_2$ (F2) to treat GBM. In in vitro experiments with glioblastoma cells U373MG, F2 showed the cytotoxic effect with $IC_{50}$ of 50 ${\mu}g/mL$ through apoptosis, confirmed by DNA condensation and fragmentation. The cell population of cell cycle sub-G1 as indicative of apoptosis was also increased. In xenograft model in SD rats, F2 at dosage of 35 mg/kg weight was intravenously injected every two days. This reduced the tumor growth in magnetic resonance imaging images. The immunohistochemistry revealed that the anticancer activity might be mediated through inhibition of proliferation judged by Ki67 and apoptosis induced by activation of caspase-3 and -8. And the lowered expression of CD31 showed the reduction in blood vessel densities. The expression of matrix metalloproteinase-9 for invasion of cancer was also inhibited. The cell populations with cancer stem cell markers of CD133 and nestin were reduced. The results of this study suggested that F2 could be a new potential chemotherapeutic drug for GBM treatment by inhibiting the growth and invasion of cancer.

Effects of Photoperiod Treatment on Histological Changes in Testis Tissues of the Golden Hamster

  • Many mammals in temperate zones are affected by the distinctive changes of the four seasons in these zones. Their reproductive status is active in the summer climate and inactive during severe winter weather. The golden hamster (Mesocricetus auratus) is seasonal breeding animal whose sexual activities are regulated by photoperoidism. The reproduction and metabolism are activated by long summer days (LD) and inhibited by short winter days (SD). After several months of SD, animals become refractory to this inhibitory photoperiod and spontaneously revert to LD-like physiology. The suprachiasmatic nuclei (SCN) house the primary circadian oscillator in mammals. Seasonal changes in the photic input to this structure control many annual physiological rhythms via SCN-regulated pineal melatonin secretion, which provides an internal endocrine signal representing photoperiod. The aim of this study was to assess the variation in the morphology of the testis in relation to the natural photoperiod in male golden hamsters. The hamsters were castrated at different weeks (2, 5, 8, and 15). The cell numbers of tubules with spermatogonia (SG), spermatocyte (SC), spermatids (ST), and spermatozoa (SZ) were recorded in each sample. The results showed that testicular regression of golden hamsters occurred in the SD-treated animals. The present investigation determines that the effects of the photoperiod on the reproduction of male golden hamsters. It was also found that the circadian period increases the rate of reproductive inhibition in animals exposed to inhibitory photoperiods.
