Enhancing Protein Content in Wild-Type Saccharomyces cerevisiae via Random Mutagenesis and Optimized Fermentation Conditions

  • Single-cell protein (SCP) derived from microorganisms is widely recognized as a viable alternative protein source for the future. Nevertheless, the commercialization of yeast-based SCP is hampered by its relatively low protein content. Therefore, this study aimed to enhance the protein content of Saccharomyces cerevisiae via random mutagenesis. To achieve this, S. cerevisiae KCCM 51811, which exhibited the highest protein concentration among 20 edible S. cerevisiae strains, was selected as a chassis strain. Subsequently, a KCCM 51811 mutant library was constructed (through UV irradiation) and screened to isolate mutants exhibiting high protein content and/or concentration. Among the 174 mutant strains studied, the #126 mutant exhibited a remarkable 43% and 36% higher protein content and concentration, respectively, compared to the parental strain. Finally, the #126 mutant was cultured in a fed-batch system using molasses and corn-steep liquor, resulting in a protein concentration of 21.6 g/l in 100 h, which was 18% higher than that produced by the parental strain. These findings underscore the potential of our approach for the cost-effective production of food-grade SCP.

Preparation of Nanocrystalline ZnO Ultrafine Powder Using Ultrasonic Spraying Combustion Method (초음파분무 연소법에 의한 나노결정 ZnO 초미분체 제조)

  • For mass product of nanocrystalline ZnO ultrafine powders, self-sustaining combustion process(SCP) and ultrasonic spray combustion method(USCM) were applied at the same time. Ultrasonic spray gun was attached on top of the vertical type furnace. The droplet was sprayed into reaction zone of the furnace to form SCP which produces spherical shape with soft agglomerate crystalline ZnO particles. To characterize formed particles, fuel and oxidizing agent for SCP were used glycine and zinc nitrate or zinc hydroxide. Respectively, with changing combustion temperature and mixture ratio of oxidizing agent and fuel, the best ultrasonic spray conditions were obtained. To observe ultrasonic spray effect, two types of powder synthesis processes were compared. One was directly sprayed into furnace from the precursor solution (Type A), the other directly was heated on the hot plate without using spray gun (Type B). Powder obtained by type A was porous sponge shape with heavy agglomeration, but powder obtained using type B was finer primary particle size, spherical shape with weak agglomeration and bigger value of specific surface area. 9/ This can be due to much lower reaction temperature of type B at ignition time than type A. Synthesized nanocrystalline ZnO powders at the best ultrasonic spray conditions have primary particle size in range 20~30nm and specific surface area is about 20m$^2$/g.

An Evaluation of the Settlement and Stability in the Reclaimed Revetment by Field Monitoring Method (현장계측을 이용한 호안의 침하 및 안정성의 평가)

  • In case of construction of the final refuse disposal site on the ground where 20m soft clay layer is deposited, Sand Compaction Pile(SCP) was driven with a view to increase the strength and reduce the settlement when the reclaimed revetment intend to be constructed first in a short time. Field monitoring method is carried out in order to overcome the problems of settlement and stability in the construction of the reclaimed revetment and the assumed problems in the design of composite ground. In this paper the observed data from monitoring sections are analysed, fedback to the desigh and field, and compared with FEM analysis. Conclusions are as follow: in case of 70% replacement the use of modified soilparameters makes the FEM analysis of SCP possible. In case of 27% replacement, n(stress concentration ratio)=0.2-0.3, B(measured settleme reduction coefficient)=0.43 are evalated. Also, horizontal displacement is remarkably happened around the ground.

  • RTDs and thermocouple sensors are broadly used to measure its temperature in industry and research. The contents of this standard calibration procedure(SCP) describe procedures related to the calculation of Electrical Temperature Calibrator and show different ways to indicate the calibration results like the uncertainty in measurement. As of current, SCP of electrical temperature calibrator has not been established yet and we have some inconveniences and difficulties in the standard calibration work. To solve these problems, we have studied the calibration technique for RTD and thermocouple of temperature calibrator. In this paper, we present the mathematic model of its data and variations of measurement with the results of calibration data.

Influence of Spot Weld Pitches on Collapse Characteristics for SCP1 Vehicle Members (차체구조용 SCP1 강도부재의 점용접간격이 압궤특성에 미치는 영향)

  • Front-side members are structures with the greatest energy absorbing capability in a front-end collision of vehicles. This paper was performed to analyze initial collapse characteristics of spot welded hat and double hat-shaped section members, which are basic shape of side members, on the shift of flange weld pitches. The impact collapse tests were carried out by using home-made vertical air compression impact testing machine, and impact velocity of hat-shaped section members is 4.17m/sec and that of double hat-shaped section members is 6.54m/sec. In impact collapse tests, the collapsed length of hat-shaped section members was about 45mm and that of double hat-shaped section members was about 50mm. In consideration of these condition, axial static collapse tests(0.00017m/sec) of hat and double hat-shaped section members were carried out by using UTM which was limited displacement, about 50mm. As the experimental results, to obtain the best initial collapse characteristics, it is important that stiffness of vehicle members increases as section shapes change and the progressively folding mode induces by flange welding pitch.

The Treatment Effect of Neurofeedback Training on Executive Function in Attention-Deficit Hyperactivity Disorder (뉴로피드백 훈련이 Attention-Deficit Hyperactivity Disorder 아동의 실행기능에 미치는 영향)

  • Objectives : Children with attention-deficit hyperactivity disorder (ADHD) have been shown to display more inhibitory deficits and executive function deficits. This study investigated the treatment effects of neurofeedback (NF) training on executive function by comparing the results of neuropsychological tests of the trained children at pre- and post-training. Methods : Fifteen children with ADHD, aged 6 to 14 years, participated in the study. The NF treatment consisted of slow cortical potential (SCP) training and these sessions took place once a week. The ADHD children performed 20 sessions of NF training within 6 months. Pre-training and post-training assessments encompassed Continuous Performance Test (CPT), Stroop Test, Children's Color Trails Test I&II (CCTT) and Wisconsin Card Sorting Test (WCST). Results : Patients receiving NF training showed significant improvement in visual commission error and standard deviation of auditory response time on CPT ; and total errors on WCST. But there was no significant improvement in the Stroop test and CCTT. Conclusion : SCP training using NF improves the self-regulatory capacities and impulsivity in ADHD patient, especially impulsivity in visual stimulation tasks. This study showed evidence of clinical efficacy of NF on executive function in ADHD.

  • Azerbaijan, located on western part of Caspian Sea, has been developed oil and gas exploration and production business from about 200 years ago and once charged of 50% of world oil production in the early 20th century. After Azerbaijan was independent of Soviet Union in 1991, foreign oil companies, including Korean oil companies, have been investing in Azerbaijan oil business on offshore blocks. Productive capacity of downstream in the Azerbaijan is lower than productive capacity of upstream, whereas there are important pipelines, such as BTC, SCP, NREP, and WREP, provide opportunities of the oil and gas exporting to the world. Oil fields in onshore blocks in the Azerbaijan are almost mature oil fields, caused by high recovery rate. Offshore oil blocks are still high potential in oil exploration with high risk.

Reduced Volume of a Brainstem Substructure in Adolescents with Problematic Smartphone Use

  • Objectives: Despite the growing concern regarding the adverse effects related to problematic smartphone use (PSU), little is known about underlying morphologic changes in the brain. The brainstem is a deep brain structure that consists of several important nuclei associated with emotions, sensations, and motor functions. In this study, we sought to examine the difference in the volume of brainstem substructures among adolescents with and without PSU. Methods: A total of 87 Korean adolescents participated in this study. The PSU group (n=20, age=16.2±1.1, female:male=12:8) was designated if participants reported a total Smartphone Addiction Proneness Scale (SAPS) score of ≥42, whereas the remaining participants were assigned to the control group (n=67, age=15.3±1.7, female:male=19:48). High-resolution T1 magnetic resonance imaging was performed, and the volume of each of the four brainstem substructures [midbrain, pons, medulla, and superior cerebellar peduncle (SCP)] was measured. Analysis of covariance was conducted to reveal group differences after adjusting for effects of age, gender, whole brainstem volume, depressive symptoms, and impulsivity. Results: The PSU group showed a significantly smaller volume of the SCP than the control group (F=8.273, p=0.005). The volume of the SCP and the SAPS score were negatively correlated (Pearson's r=-0.218, p=0.047). Conclusion: The present study is the first to reveal an altered volume of the brainstem substructure among adolescents with PSU. This finding suggests that the altered white matter structure in the brainstem could be one of the neurobiological mechanisms underlying behavioral changes in PSU.

Shear Strength and Compressibility of Oyster Shell-Sand Mixtures for Sand Compaction Pile (SCP공법 적용을 위한 굴패각-모래 혼합토의 전단과 압축특성)

  • Strength and deformation characteristics of oyster shell-sand mixtures were investigated to utilize waste oyster shell being treated as a waste material. Standard penetration test (SPT) is a common method to obtain in-situ strength in sand. However, in case of oyster shell-sand mixtures, there was no information between SPT N-value and internal friction angle of mixture soils. In this paper SPT experiments from several large scaled model chamber tests and large scaled direct shear tests were carried out with varying unit weight of oyster shell-sand mixtures. Appropriate correlations were in tile study observed among N-value, unit weight and internal friction angle, which make it possible to estimate in-situ strength from SPT and the coefficient of volume compressibility from the confined compression tests to compute the settlement of oyster shell-sand mixtures.
