Adult stem cells can make identical copies of themselves for long periods of time. They also give rise to many differentiated mature cell types that have characteristic morphology and specialized function. Human adult stem cells are the attractive raw materials for the cell/tissue therapy, however, it is not easy to get from the adult tissues. In the present study, we tried to isolate a cell population derived from human umbilical cord vein which has been discarded after birth. The cells were isolated after treatment of the umbilical vein with collagenase or trypsin. After 3 days of culture, two kinds of cell populations were found consisting of adherent cells with endothelial cell-like and fibroblast-like morphology, respectively. When these cells were subcultured 12 times over a period of 3 months, almost cells appeared uniformly to exhibit fibroblastoid morphology which was different from that of mesenchymal stem cells obtained from human bone marrow The results of RT-PCR analyses showed distinct expression of BMP-4, oct-4, and SCF genes but not of GATA, PAX-6 and Brachyury genes. On immunohistochemical staining, the cells were negative for the von Willebrand factor(vWF), alpha-smooth muscle actin and placental alkaline phosphatase. From these observations, it is suggested that stem-like cells might be present in human umbilical cord vein.

In this study, a spectral fatigue analysis was performed for the topside structure of an offshore floating vessel. The topside structure was idealized using beam elements in the SACS program. The fatigue analysis was carried out considering the wave and wind loads separately. For the wave-induced fatigue damage calculation, motion RAOs calculated from a direct wave load analysis and regular waves with different periods and unit wave heights were utilized. Then, the member end force transfer functions were generated covering all the loading conditions. Stress response transfer functions at each joint were produced using the specified SCFs and member end force transfer functions. fatigue damages were calculated using the obtained stress ranges, S-N curve, wave spectrum, heading probability of each loading condition, and their corresponding occurrences in the wave scatter diagrams. For the wind induced fatigue damage calculation, a dynamic wind spectral fatigue analysis was performed. First, a dynamic natural frequency analysis was performed to generate the structural dynamic characteristics, including the eigenvalues (natural frequencies), eigenvectors (mode shapes), and mass matrix. To adequately represent the dynamic characteristic of the structure, the number of modes was appropriately determined in the lateral direction. Second, a wind spectral fatigue analysis was performed using the mode shapes and mass data obtained from the previous results. In this analysis, the Weibull distribution of the wind speed occurrence, occurrence probability in each direction, damping coefficient, S-N curves, and SCF of each joint were defined and used. In particular, the wind fatigue damages were calculated under the assumption that the stress ranges followed a Rayleigh distribution. The total fatigue damages were calculated from the combination with wind and wave fatigue damages according to the DNV rule.
The equilibrium structures for the ground and transition states of $C_7H_7^+$ isomers have been investigated using sophisticated ab initio quantum mechanical techniques with various basis sets. The structures of tropyrium and benzyl cations have been fully optimized at the DZP CCSD(T) levels of theory. And the structures of o-, m-and p-tolyl cations are optimized fully up to the DZ CCSD(T) levels of theory. The geometries for the transition states between three isomers of tolyl cations have been optimized up to DZP CISD level of theory. The SCF harmonic vibrational frequencies for tropylium, benzyl, and three isomers of tolyl cations are all real numbers, which confirm the potential minima and each unique imaginary vibrational frequencies for TS1 and TS2 confirm the true transition states. The relative energy of the benzyl cation with respect to the tropyrium cation is predicted to be 28.5 kJ/mol and is in good agreement with the previous theoretical predictions. The 0 K heats of formation, ${\Delta}H^{\circ}_{f0}$, have been predicted to be 890, 1095, 1101, and 1110 kJ/mol for tropylium, ortho-, meta-, and para-tolyl cations by taking the experimental value of 919 kJ/mol for the benzyl cation as the base level. Human eyelid adipose-derived stem cells (hEAs) and amniotic mesenchymal stem cells (hAMs) are very valuable sources for the cell therapeutics. Both types of cells have a great proliferating ability in vitro and a multipotency to differentiate into adipocytes, osteoblasts and chondrocytes. In the present study, we evaluated their stem cell characteristics after long-time cryopreservation for 6, 12 and 24 months. When frozen-thawed cells were cultivated in vitro, their cumulative cell number and doubling time were similar to freshly prepared cells. Also they expressed stem cell-related genes of SCF, NANOG, OCT4, and TERT, ectoderm-related genes of NCAM and FGF5, mesoderm/endoderm-related genes of CK18 and VIM, and immune-related genes of HLA-ABC and ${\beta}$2M. Following differentiation culture in appropriate culture media for 2-3 weeks, both types of cells exhibited well differentiation into adipocyte, osteoblast, and chondrocyte, as revealed by adipogenic, osteogenic or chondrogenic-specific staining and related genes, respectively. In conclusion, even after long-term storage hEAs and hAMs could maintain their stem cell characteristics, suggesting that they might be suitable for clinical application based on stem cell therapy.
We investigated the toxic effects of difenoconazole on the development in the African clawed frog, Xenopus laevis. To test the toxic effects, frog embryo teratogenesis assays using Xenopus were performed. Embryos were exposed to various concentrations of difenoconazole (0-30 ${\mu}M$). $LC_{100}$ for difenoconazole was 30 ${\mu}M$, and the $LC_{50}$ determined by probit analysis was 27.19 ${\mu}M$. Exposure to difenoconazole concentrations ${\geq}$5 ${\mu}M$ resulted in 10 different types of severe external malformation. Histological examinations revealed dysplasia of the eye, heart, liver, somatic muscle, and swelling of the pronephric ducts. The tissue-specific toxic effects were investigated with an animal cap assay. Blood cells were normally induced at a high frequency by mSCF and activin A. However, the induction of blood cells was strongly inhibited by the addition of difenoconazole. Electron micrographs of tested embryos showed the degeneration of somatic muscle and the shrinkage of microvilli on pronephric duct. The gene expression of cultivated animal cap explants was investigated by reverse transcriptase-polymerase chain reaction (RT-PCR). It revealed that the expression of the blood-specific marker(${\beta}$-globin II) and muscle-specific marker (XMA) were more strongly inhibited than the neural-specific marker(XEn2) by the addition of difenoconazole.

The dynamic photoelastic technique had been utilized to investigate the possibillity of relieving the large local singular stresses induced at the corner of a right- angle- indenter. The indenter compressed a semi-infinite body dynamically with an impact load applied on the top of the indenter. The effects of the geometric changes of the indenter in terms of the diameter (d) and the location (1) of the stress relieving notch on the behavior of the dynamic contact stresses were investigated. The influence of stress relieving notches positioned along the edge of the semi-infinite body on the dynamic contact stresses were also studied by changing the diameter (D) and the location (L) of the notch. A multi-speak-high speed camera with twelve sparks were used to take photographs of full field dynamic isochromatic fringe patterns. The contact singular stresses were found to be released significantly by the stress relief notches both along the indenter and the edge of the semi-infinite body. The optimal position and geometry of the stress relieving notches were obtained with the aid of limited experimental results.

In this study, a method for fatigue performance estimation of deepwater steel catenary riser (SCR) under short-term vortex-induced vibration was investigated for selected S-N curves. General tendency between S-N curve capacity and fatigue performance was analysed. SCRs are generally used to transport produced oil and gas or to export separated oil and gas, and are exposed to various environmental loads in terms of current, wave, wind and others. Current is closely related with VIV and it affects fatigue life of riser structures significantly. In this regards, the process of appropriate S-N curve selection was performed in the initial design stage based on the scale of fabrication-related initial imperfections such as welding, hot spot, crack, stress concentration factor, and others. To draw the general tendency, the effects of stress concentration factor (SCF), S-N curve type, current profile, and three different sizes of SCRs were considered, and the relationship between S-N curve capacity and short-term VIV fatigue performance of SCR was derived. In case of S-N curve selection, DNV (2012) guideline was adopted and four different current profiles of the Gulf of Mexico (normal condition and Hurricane condition) and Brazil (Amazon basin and Campos basin) were considered. The obtained results will be useful to select the S-N curve for deepwater SCRs and also to understand the relationship between S-N curve capacity and short-term VIV fatigue performance of deepwater SCRs.

Turbulence plays an important role in molecular clouds. However, the properties of turbulence are poorly understood. In order to study the influence of turbulence in molecular clouds, we need to sample the turbulent properties in the full range of scales down to sonic scale. We mapped the $20^{\prime}{\times}60^{\prime}$ area covering the Orion Molecular Cloud (OMC) 1-4 region in HCN 1-0 and HCO+ 1-0 with Taeduk Radio Astronomy Observatory (TRAO) 14-m telescope as part of the TRAO key science program, "Mapping turbulent properties of star-forming molecular clouds down to the sonic scale (PI: Jeong-Eun Lee)". In addition, we combine our TRAO data with other molecular line maps ($^{13}CO$ 1-0, $C^{18}O$ 1-0, CS 1-0, $N_2H^+$ 1-0) obtained with the Nobeyama Radio Observatory (NRO) 45-m telescope. To analyze these data, we apply statistical methods, the principal component analysis (PCA) and spectral correlation function (SCF), which are known to be useful to study underlying turbulent properties and to quantitatively characterize cloud structure. We will present the preliminary results of observations and analyses.
The anti-tumor effect of monocyte-derived DC (MoDC) vaccine was studied in lung cancer model with feasible but weak Ag-specific immune response and incomplete blocking of tumor growth. To overcome this limitation, the hematopoietic stem cell-derived DC (SDC) was cultured and the anti-tumor effect of MoDC & SDC was compared in mouse lung cancer minimal residual model (MRD). Therapeutic DCs were cultured from either $CD34^+$ hematopoietic stem cells with GM-CSF, SCF and IL-4 for 14 days (SDC) or monocytes with GM-CSF and IL-4 for 7 days (MoDC). DCs were injected twice by one week interval into the peritoneum of mice that are inoculated with Lewis Lung Carcinoma cells (LLC) one day before the DC injection. Anti-tumor responses and the immune modulation were observed 3 weeks after the final DC injection. CD11c expression, IL-12 and TGF-${\beta}$ secretion were higher in SDC but CCR7 expression, IFN-${\gamma}$ and IL-10 secretion were higher in MoDC. The proportion of $CD11c^+CD8a^+$ cells was similar in both DC cultures. Although both DC reduced the tumor burden, histological anti-tumor effect and the frequencies of IFN-${\gamma}$ secreting $CD8^+$ T cells were higher in SDC treated group than in MoDC. Conclusively, although both MoDC and SDC can induce the anti-tumor immunity, SDC may be better module as anti-tumor vaccine than MoDC in mouse lung cancer.
