Three Terpenes and One Phenolic Compound from Sasa borealis

  • Four compounds of previously known structures, friedeline (1), 3-hydroxyglutinol (2), p-hydroxy-benzealdehyde (3), and squalene (4) were obtained from the n-hexane- and EtOAc-soluble fractions of the dried whole plants of Sasa borealis. The structures of 1-4 were identified by the interpretation of their spectroscopic data including 1D and 2D NMR as well as by comparison with previously reported values. This is the first report on the isolation of compounds 1-4 from S. borealis.

  • As part of study of the constituents of bamboo grasses, the leaves of Sasa borealis (Hackel) Makino (Gramineae) were examined. Friedelin, glutinol, isoorientin and isovitexin have been reported as constituents of bamboo grasses. In this study, tricin and two flavone glycosides, tricin $7-O-{\beta}-D-glucopyranoside$ and luteolin $6-C-{\alpha}-L-arabinopyranoside$ have been isolated from EtOAc extract of S. borealis, by consecutive silica gel, Sephadex LH-20 column chromatography and a repetitive HPLC. The structures of these compounds were determined by IR, $^1H-NMR,\;^{13}C-NMR,\;^{13}C-^1H\;COSY,\;^1H-^1H\;COSY,\;HMBC$ and Mass spectral data.

Pharmacognostical Studies on the Folk Medicine 'JoRitDae' (민간약 조릿대의 생약학적 연구)

  • Korean folk medicine 'JoRitDae' has been used mainly to cure vomiting and cancer. With regard to the botanical origin of 'JoRitDae', it has been considered to designate the rhizome of Sasa species (Gramineae), but there was no pharmacognostical confirmation on it. To clarify the botanical origin of 'JoRitDae', the anatomical characteristics of the rhizome of Sasa species growing wild in Korea and Japan, Sasa borealis, S, kurilensis, S. quelpaertensis, S. charatacea and S. senanensis were studied. As a result, it was found the morphological criteria for the five Sasa species that could discriminate them by the number of cell layers in cortex, the ratio of thickness of cortex to rhizome radius, the number of cell layers in the vascular bundle sheath, the total number of vascular bundle and thickness of vascular bundle sheath. According to these criteria, it was clarified that the commercial folk medicine 'JoRitDae' was the rhizome of Sasa borealis.

Inhibitory Effects of Sasa borealis on Mechanisms of Adipogenesis (조릿대 에틸아세테이트 분획물의 지방세포에서 분화전사인자 조절을 통한 지방형성 저해 효능)

  • Sasa borealis is a major source of bamboo leaves used for traditional medicine in Korea. Obesity is a serious health problem in industrialized countries that has been implicated in various diseases, including type 2 diabetes, hypertension, cancer, and coronary heart disease. Recent reports have proposed mechanisms to reduce obesity by decreasing preadipocyte differentiation, and proliferation in 3T3-L1 preadipocyte. The preadipocytes play a key role by differentiation into mature adipocytes and increasing fat mass. In this study, we investigated whether ethanol-soluble extracts and ethyl acetate-soluble fractions from Sasa borealis inhibits intracellular accumulation of lipid droplets in a dose-dependent manner in 3T3-L1 cells (an important model system for studying adipogenesis). The down-regulation of PPAR${\gamma}$ and C/EBP${\alpha}$ (key adipogenic transcription factors) were confirmed by the reverse transcription polymerase chain reaction (RT-PCR). Ethyl acetate-soluble fractions from Sasa borealis attenuated the expression of PPAR${\gamma}$ and C/EBP${\alpha}$. These results suggest that Sasa borealis inhibits adipogenic differentiation by regulating adipogenic transcription factors in 3T3-L1 cells. Therefore, Sasa borealis extracts may be a good candidate for the management of obesity.

Comparative Study for Anti-inflammatory and Anti-obesity Effect of Fractions from Leaf and Stem of Sasa Borealis (조릿대의 잎과 줄기 추출물 분획의 염증 및 비만 억제 효과 비교)

  • Sasa borealis has long been used in folk remedies and traditional Korean medicine for analgesic, sedative and anticarcinogenic purposes. To excavate novel natural compounds for treating obesity, we separated leaf and stem and fractionated. We screened phenolic contents, anti-oxidative and anti-inflammatory properties with the fractions. Dichloromethane (DCM) and ethyl acetate (EA) fraction from both leaf and stem exhibited high anti-oxidative properties demonstrated by DPPH, ABTS assay in accordance with their phenolic contents. In addition, DCM and EA fraction from leaf inhibited nitric oxide (NO) production as well as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in Raw 264.7 cell. Finally, DCM and EA fraction from both leaf and stem significantly inhibited 3T3-L1 preadipocyte differentiation and decreased intracellular lipid level. These results suggest that DCM and EA fraction from Sasa borealis may contain therapeutic agent for obesity by attenuating inflammation and oxidative stress.

Changes in Kimchi Quality as Affected by the Addition of Sasa borealis Makino Extract (조릿대(Sasa borealis Makino) 추출물 첨가가 배추김치의 품질에 미치는 영향)

  • This study was focused on finding the potential of hot water extract of bamboo shoot (Sasa borealis Makino) on the fermentation of Kimchi made with Chinese cabbage. The properties of Kimchi were examined up to 28 days of storage. The pH and acidity decreased regardless of treatments and showed no significant difference between treatments. There was a decreasing tendency of both total and reducing sugars in kimchi but the addition of bamboo extract did not affect the soluble sugar levels. Interestingly, bamboo extracts affected the lactic acid fermentation and ripening, resulting in the increase of lactic acid in bamboo extract treatment. Number of total bacterial cell of additive group is higher than control one, probably due to the stimulative effect of bamboo extract on bacterial growth. Level of lactic acid bacteria was also higher in the additive group, thus, it is considered that bamboo extract appeared to enhance the proliferation of lactic acid bacteria. The acceptability of treated Kimchi was higher in general. And results of intensity evaluation in color and texture were higher as well by addition of bamboo extract.

Effects of Sasa Borealis Leaf Extract on the Glucose Tolerance of Major Foods for Carbohydrate (조릿대 잎 추출물이 탄수화물 급원 식품의 당 내성에 미치는 영향)

  • Sasa borealis leaf has been known to have anti-diabetic properties. In this study, we tried to evaluate the effects of Sasa borealis leaf extract (SBE) on the inhibition of $\alpha$-glucosidase activity and postprandial glycemic response following ingestion of four carbohydrate-rich foods; cooked rice, ramen (instant noodle), noodle, and bread. Fourteen healthy female adults consumed 50 g of glucose (control) or one of the four foods containing 50 g of available carbohydrate with or without 2,000 mg of SBE. The activity of $\alpha$-glucosidase was inhibited dose-dependently by SBE. With SBE, blood glucose concentration at 15 min and the positive area under the curve (AUC) of postprandial glycemic response at 15 min and 30 min after consuming each of the four foods were reduced significantly. As the result, total positive AUC during 120 min was decreased in case of taking cooked rice or bread. Glycemic index and glycemic load of the four foods were declined from 13% to 23% with SBE. The results of this study suggest that SBE may be effective for postprandial glucose control by inhibiting $\alpha$-glucosidase activity.

Isolation and Structural Identification of Minor Constituents from Sasa borealis

  • Compounds of (+)-5,5'-dimethoxylariciresinol (1) and (3S,5R,6S,7E)-5,6-epoxy-3-hydroxy-7-megastigmen-9-one (2) were isolated from an EtOAc extract of the whole plant of Sasa borealis (Hack.) Makino (Gramineae) for the first time in the present investigation. The structures of compounds were identified by analysis of spectral data including 1D- and 2D-NMR spectra as well as by comparison of their data with the published values. These compounds have never been isolated previously from the family Gramineae.

Evaluation of Forage Production and Feed Value of Sasa borealis in the Jeju Area (제주지역 조릿대의 사초생산성 및 사료가치평가)

  • This study was conducted to evaluate the forage production and feed value of Sasa borealis (S. borealis) in Jeju Island in order to improve the utilization of Sasa borealis and to help mitigate the problem of reduced plant species diversity caused by S. borealis in Hanlla Mountain. To investigate the forage production, three quadrat structures were installed in the S. borealis natural community in the middle part of Hanlla Mountain. From May to October 2017, S. borealis in quadrats was cut at a fixed time of each month, and then forage production and regenerated acidity per kg/ha were evaluated. For the evaluation of feed value, compositional analysis was performed on the monthly samples. In vitro digestion experiments were carried out using cannula mounted Hanwoo. In vitro neutral detergent fiber digestibility(IVNDFD) and in vitro acid detergent fiber digestibility(IVADFD) were measured after the experiment. Forage production of S. borealis showed relatively good regeneration ability in May and June, but the regeneration ability decreased as the cutting was repeated. In order to use S. borealis as a forage, it is considered efficient to feed black goats with good fiber decomposition or horses good palatability to S. borealis and relatively good digestibility.

Reverse-Phase HPLC Method for Identification of Diastereomeric Constituents from Sasa borealis (Sasa borealis의 Diastereomeric 성분들의 역상 고속액체크로마토그래프 분석방법)

  • Reiterated normal-phase column chromatography lead to the isolation and purification of six known compounds but for the first time from the whole plant of Sasa borealis (Hack.) Makino (Gramineae): tricin 4'-O-(erythro-${\beta}$-guaia-cylglyceryl) ether (1), tricin 4'-O-(threo-${\beta}$-guaiacylglyceryl) ether (2), tricin 4'-O-[erythro-${\beta}$-guaiacyl-(9'-O-acetyl)-glyceryl] ether (3), tricin 4'-O-[threo-${\beta}$-guaiacyl-(9'-O-acetyl)-glyceryl] ether (4), (-)-pinoresinol (5), and vanillin (6). The structures of the compounds (1-6) were established based on interpretation of high resolution NMR (COSY, HSQC, HMBC, and NOESY) spectral data. In particular, compounds 1 and 3 were diastereomers of compounds 2 and 4, respectively. These two sets of diastereomers were able to be simultaneously identified and quantified by a gradient reversed-phase HPLC method with UV photodiode array, This sensitive HPLC method is noteworthy as a simultaneous separation and identification method to test the extract of the family Gramineae which contains these compounds.
