이온교환 크로마토그래픽을 이용한 Glutathione 분리 (Separation of Glutathione by Ion Exchange Chromatography)

  • 미생물 효소에 의해 생산된 생산모액 내의 글루타치온(L-${\gamma}$-glutamyl-L-cysteinylglycine, GSH)을 액체크로마토그래피를 이용하여 분리하였다. GSH와 결합하는 수지를 선택하기 위해 여러 수지와 GSH 수용액을 사용하여 회분식 흡착실험을 한 결과, pH 8.0에서 음이온 교환수지인 Q-sepharose와 QAE- sephadex에 GSH가 결합하였으나, QAE-sephadex는 수지와 결합된 GSH를 이탈시키기 위해 사용된 salt에 의해 부피가 줄어들어 부적합하였다. GSH 분리를 위한 기초실험을 위하여 GSH, cysteine, glutamate, glycine, $\gamma$-glutamylcysteine, ATP, glucose의 혼합액에서 GSH와 $\gamma$-glutamylcysteine를 다른 물질로부터 1차 분리할 수 있었다. NaCl의 농도를 조절하여 두 물질이 중첩되는 현상을 제거하여 분리하고자 하였으며, GSH의 tailing현상을 줄이도록 노력하였다. NaCl(35mM)을 용해시킨 Tris buffer를 사용함으로써 두 물질의 분리가 가능하였고, 생산모액을 사용하여 실험한 결과, 혼합 시료와 유사한 분리결과를 얻을 수 있었다. Standard solution에서의 GSH 분리결과 72.6%의 회수율을 보였으며, 생산 모액에서는 84.4%의 회수율을 각각 보였다.

전기그릴과 전자레인지 재가열이 소금농도와 인산염 첨가수준에 따른 분쇄 돈육 패티의 품질 특성에 미치는 영향 (The Effects of Electric Grill and Microwave Oven Reheating Methods on the Quality Characteristics of Precooked Ground Pork Patties with Different NaCl and Phosphate Levels)

  • The objective of this study was to determine the effects of different reheating methods on the quality characteristics of precooked ground pork patties with different combinations of salt (1, 2%) and phosphate (0, 0.3%). The cooking and reheating methods used were the electric grill and the microwave oven. The cooking method and the addition of phosphate had detectable effects on the quality characteristics of ground pork patties. The reheating rate by microwave oven was faster than by electric grill, and decreased with increases in salt and the addition of phosphate. Cooking loss and reduction in patty diameter after reheating by microwave oven were higher than by electric grill, and these values decreased with increasing of salt/phosphate levels. The pH of patties with added phosphate was significantly higher than patties without phosphate, and no significant difference in pH was observed among patties reheated by each method. The patties reheated by electric grill had higher moisture contents than those reheated by electric grill, and the addition of phosphate resulted in higher moisture contents. The hardness of patties reheated by electric grill was lower than patties reheated by microwave oven, and the addition of phosphate increased the hardness with both reheating methods.

  • Nanoscale Chevrel $Mo_6S_8$ powders are synthesized by molten salt synthesis. Synthesized $Mo_6S_8$ powders have different mean particle sizes which are dependent on a ratio of salt to precursor. The particle sizes of $Mo_6S_8$ powders changes along with the ratio increase. $Mo_6S_8$ (6:1) demonstrates the best electrochemical characteristics among the synthesized $Mo_6S_8$ powders although the $Mo_6S_8$ (4:1) has the smallest particle size. $Mo_6S_8$ (6:1) shows a reversible capacity of 83.9 $mAhg^{-1}$, which is 27.5% and 33% improved value over $Mo_6S_8$ (2:1) and $Mo_6S_8$ (4:1) at a current density of 0.2C, respectively. The superior electrochemical properties of $Mo_6S_8$ (6:1) are attributed to the balanced particle size which provides proper contact area with electrolyte and the shortened $Mg^{2+}$ diffusion length. The $Mo_6S_8$ (4:1) has the smallest particle size but further reduction of particle size from $Mo_6S_8$ (6:1) is not advantageous.

  • 콘크리트의 열화 및 철근부식에 대응하기 위하여 다양한 보수재료가 존재하나 이러한 보수재료를 평가하는 방법은 단순히 염해, 중성화 등의 단독열화만을 대상으로 하고 있어 여러 가지 열화인자가 복합적으로 작용하는 실제 환경과는 차이를 나타내게 된다. 따라서 본 연구에서는 폴리머시멘트계 보수용 단면복구재가 KS 기준을 만족하는지 확인한 후 복합열화 환경 하에서 염해, 중성화. 철근부식을 평가하였다. 실험결과 각각의 보수재료들은 KS 기준을 만족하였지만 내구성능은 상이하게 나타나 향후 이에 대한 고려가 필요할 것으로 사료된다.

시판소금이 김치발효 미생물의 생육에 미치는 영향 (Effects of Commercial Salts on the Growth of Kimchi-Related Microorganisms)

  • 천일염, 한주 소금, 구운 소금 및 죽염이 김치 발효에 중요한 영향을 미치는 미생물인 Leuconostoc mesenteroides, Lactobacillus plantarum과 산막효모인 Pichia membranaefaciens 및 식품오염의 지표균인 E. coli의 생육에 미치는 영향을 조사하였다. 각각의 소금을 3%, 5% 농도로 첨가하여 김치 발효 조건에 따라 배양 온도를 각각 1$0^{\circ}C$, 18$^{\circ}C$, 37$^{\circ}C$로 나누어 비교하였다. 그 결과 Leuconostoc mesenteroides는 배양 온도가 낮을수록, 소금 농도가 높을수록 성장이 저해되었으나 소금 종류별로는 특이할 만한 저해 효과는 나타나지 않았다. Lactobacillus plantarum도 마찬가지로 배양 온도가 낮을수록, 소금 농도가 높을수록 성장이 억제되었으며, 특히 18$^{\circ}C$, 5% 소금농도에서 죽염에 대해 저해 효과가 두드러졌고 1$0^{\circ}C$배양에서는 소금 종류와 농도에 관계없이 생육이 상당히 저조했다. Pichia membranaefaciens는 배양 온도에 관계없이 죽염에서 상당한 저해효과를 보였으며, 특히 18$^{\circ}C$, 5$^{\circ}C$ 소금농도에서는 전혀 자라지 못해 완전한 억제 효과를 보였다. E. coli도 다른 소금에 비해 1$0^{\circ}C$와 18$^{\circ}C$에서 죽염에 대해 저해 효과가 두드러졌다. 따라서 본 실험 결과 김치발효에도 어느 정도 관여하나 과다한 산 생성으로 김치의 가식기간을 단축시키는 Lactobacillus plantarum과 김치 표면에 산막을 형성하여 부패를 촉진시키는 Pichia membranaefaciens 그리고 식품오염의 지표균인 E. coli의 성장에 죽염이 다른 소금에 비해 상당한 저해 효과를 나타내었다.

고정식 부력을 이용한 염도 및 수위 측정 방식에 대한 연구 (Salinity and water level measuring device using fixed type buoyancy)

  • 염전의 자동화 시스템 구성을 위해서는 증발지의 염도 및 수위 측정이 필요하다. 이를 위해 본 논문에서는 2개의 고정된 부력체의 부력을 측정하여 함수의 염도 및 수위를 동시에 측정할 수 있는 방법을 제안하였다. 제안된 측정방식은 측정 장치가 함수에 잠기게 될 때 주부력체와 비교부력체의 부력을 측정하고, 두 부력의 합과 차를 통해 염도와 수위를 동시에 측정하는 것이다. 부력의 측정에 기계적인 움직임이 없어서 뻘이 포함된 염전의 환경에서 측정 오류와 유지보수가 필요성을 줄일 수 있다. 제안된 방법을 적용하여 염전의 증발지에서 원격으로 염도 및 수위를 동시에 측정할 수는 있는 시스템을 개발하였다. 여러 수준의 염도를 갖는 기준함수를 사용한 측정 실험을 통하여 염도 오차 0%, 수위 오차 2mm의 결과를 얻고, 제안된 염도 및 수위 측정 장치의 유효성을 확인하였다. 개발된 염도 및 수위 측정 장치를 활용하여 자동화 시스템을 구성할 경우 노동력 절감 및 작업 환경 개선과 생산성 증진이 기대된다.

Desalinization Effect of Pennisetum Alopecuroides and Characteristics of Leachate Depending on Calcium Chloride (CaCl2) Concentration

  • Background and objective: Calcium chloride (CaCl2) and sodium chloride (NaCl) are commonly used as a deicing agent in South Korea and penetrate the soil on the roadside, causing damage to plants. This study was conducted to investigate the salinity reduction effect of Pennisetum alopecuroides and the chemical characteristics of soil leachate. Methods: The plants were treated with five different concentrations of CaCl2 (0, 1, 2, 5, and 10g·L-1) and were grouped into the Cont., C1, C2, C5, and C10 groups. CaCl2 of 200 m·L-1 was sprayed to each plant once every two weeks. The growth of P. alopecuroides (plant height, leaf length, leaf width and the number of leaves) was measured. The level of EC and pH, and exchangeable cations (K+, Ca2+, Na+, and Mg2+) in the leachate of soil was monitored. Results: The pH of soil leachate decreased as the CaCl2 concentration increased, and the EC increased significantly. The content of K+ did not change significantly until the concentration of CaCl2 reached 5 g·L-1, but the content of Ca2+, Na+, and Mg2+ significantly increased. The plant height, leaf length, and leaf width of P. alopecuroides showed the highest value in CaCl2 1 g·L-1 followed by CaCl2 2 g·L-1 and the control group. Root fresh weight was the highest in CaCl2 2 g·L-1. On the other hand, there was no change in the shoot fresh weight, dry weight and root dry weight, and P. alopecuroides growth inhibition at the concentration of 5 g·L-1 or higher in the plant height and leaf length. Conclusion: P. alopecuroides is relatively highly salt-tolerant and can improve the salt damaged soil by lowering the content of the salt-based exchangeable K+ ions.

Reduction of Stress Caused by Drought and Salt in Rice (Oryza sativa L.) Crops through Applications of Selected Plant Extracts and the Physiological Response Mechanisms of Rice

  • In many areas of the world, salt damage and drought have had a negative impact on human survival due to a decrease in agricultural productivity. For instance, about 50% of agricultural land will be affected by salt damage by 2050. Biostimulants such as plant extracts can not only increase the nutrient utilization efficiency of plants, but also promote plant growth and increase resistance to abiotic or biotic stress. Therefore, the objective of this study was to determine how selected plant extracts might reduce levels of stress caused by drought and salt and to better understand the physiological response mechanisms of rice plants. In this study, we used Soybean leaves, Soybean stems and Allium tuberosum, Allium cepa, Hizikia fusiforme, and Gracilaria verrucosa extracts were used. These extracts had been used in previous studies and were found to be effective. The materials were dried in a dry oven at 50℃ for 5 days and ground using a blender. Each 50 g of materials was put in 1 L of distilled water, stirred for 24 hours, filtered using 4 layers of mirocloth, and then concentrated using a concentrator. Rice (cv. Hopumbyeo) seeds were immersed and germinated, and then sown in seedbeds filled with commercial soil. In drought experiments, three rice seedlings at 1 week after seeding was transplanted into 100 ml cups filled with commercial soils and grown until the 4-leaf stage. For this experiment, the soil weight in a cup was equalized, and water was allowed to become 100% saturated and then drained for 24 hours. Thereafter, plant extracts at 3% concentrations were applied to the soils. For NaCl treatments, rice plants at 17 days after seeding were treated with either 100 mM NaCl or plant extracts at 1%+ 100 mM NaCl combinations in the growth chamber. Leaf injury, relative water content, photosynthetic efficiency, and chlorophyll contents were measured at 3, 5, and 6 days after treatments. Shoot fresh weight of rice under drought conditions increased 28-37% in response to treatments of Soybean leaf, Soybean stem, Allium tuberosum, Allium cepa, Hizikia fusiforme, and Gracilaria verrucosa extracts at 3% when compared with control plants. Shoot fresh weight of rice subjected to 100 mM NaCl treatments also increased by 6-24% in response to Soybean leaf, Soybean stem, Allium tuberosum, Allium cepa, Hizikia fusiforme, and Gracilaria verrucosa extracts at 3% when compared with control plants. Compared to the control, rice plants treated with these six extracts and subjected to drought conditions had significantly higher relative water content, Fv/Fm, total chlorophyll and total carotenoids than control plants. With the exception of relative water contents, rice plants treated with the six extracts and subjected to salt stress (100 mM NaCl treatments) had significantly higher Fv/Fm, total chlorophyll and total carotenoids than control plants. However, the type of extract used did not produce significant difference in these parameters. Thus, all the plant extracts used in this study could mitigate drought and NaCl stresses and could also contribute substantially to sustainable crop production.

전국 권역별, 연령별 여자 성인과 노인의 짠맛 미각판정치, 식태도 및 식행동 비교 (A Comparison of Salty Taste Assessment, Dietary Attitude and Dietary Behavior among Adult and Senior Women by Region and by Age in Korea)

  • Objectives: The purpose of this study was to compare and analyze the results of salty taste assessment, dietary attitudes, and dietary behaviors among adult and senior women by region and by age. The results generated from this was expected to provide fundamental data for implementing a nationwide salt reduction education program. Methods: The salty taste assessment tool was applied to 4,064 subjects from 15 areas in Korea. Also, a survey of dietary attitude and dietary behavior related to salt intake was conducted for all subjects participated in this study. Results: The salty taste assessment scores by region and by age were the lowest in capital (p < 0.01) and was highest among the 70+ year age group (p < 0.01). The dietary attitude scores and dietary behavior scores showed that Gyeongsang was the highest (p < 0.001) and the capital was the lowest. The dietary attitude scores were highest at 20&30's followed by 40's and 70's group (p < 0.001). Dietary behavior scores showed that 20's~30's and 40's groups were higher than the other age groups (p < 0.001). The score of 'I like kimchi' was $3.46{\pm}0.88$, which was the highest among 10 dietary behavior questions. The score of 'I eat a lot of kimchi' was $3.30{\pm}0.90$, which was the highest among 10 dietary behavior questions related to salt intake. The scores of salty taste assessment had significant positive correlations between the scores of dietary attitude (p < 0.001), dietary behavior (p < 0.001) and self-awareness (p < 0.001). Conclusions: Based on the results of salty taste assessment, we observed a tendency that older people and regions except the capital had higher preference for salty taste. Our results suggested the necessity for a nationwide salty reduction education program tailored for regions and for different age groups.

염분분사환경에서 냉연 304 스테인레스강의 부식거동 (The Corrosion Behavior of Cold-Rolled 304 Stainless Steel In Salt Spray Environments)

  • 염분분위기에서의 부식은 사용후핵연료의 중간저장 기간동안 304 스테인레스 강재 건식저장용기의 주 열화기구들 중 하나다. 본 연구에서는 감소정도가 서로 다른 냉연 304 스테인레스 강 시편들에 0.5wt.%의 염화나트륨 연무를 분사시키면서 느린 변형속도시험(SSRT)과 중성염 분사시험(NSS)을 $85^{\circ}C$$200^{\circ}C$ 에서 수행하였다. $85^{\circ}C$에서 2000 시간 동안 시험한 NSS시편의 무게 변화는 $200^{\circ}C$에서 시험한 시편의 무게 변화와 크게 달랐다. NSS 시편의 $85^{\circ}C$에서 무게 감량은 미미하였지만, 냉연 감소율이 증가함에 따라서 무게 변화는 점진적으로 감소하였다. $85^{\circ}C$$200^{\circ}C$에서 그리고 염분분사 환경에서 가볍게 냉연 가공된 시편의 SSRT 시험으로부터 얻은 항복강도와 극한 인장응력의 값은 공기 중의 값보다 약간 낮았다. 그러나 염분 분위기에서 부식으로 인한 20% 감소 냉연시편의 강도는 더 이상 변화하지 않았다. 예비결과는 냉연 304 스테인레스 강의 질과 성능이 건식저장용기의 제작을 위한 조건에 맞는다는 것을 증명하였다. 그러나 냉연 스테인레스 강의 장기적인 성능을 더 잘 이해하기 위해서는 염분분위기에서 이 재질의 부식거동에 관한 더 많은 연구가 필요하다.
