궁귀탁리산(芎歸托裏散)의 L1210과 S-180이 이식된 마우스에 대한 항암(抗癌) 작용(作用) 연구(硏究) (Study of Gungguitakli-San on the Anti-Cancer in L1210 and S-180 cells Transplanted Mice)

  • Objective : The purpose of this study was to investigate effect of Gungguitakli-San(GTS) on the anti-tumor, immunocytes. Methods : This study estimated the proliferation of L1210 and S-180 cell lines, mouse splenocytes and thymocytes in vitro, and estimated the proliferation of L1210 cell, S-180 cell, thymocytes and splenocytes and body weight in S-180 cells-transplanted mice. The cytotoxicity and proliferation of cells were tested using a colorimetric tetrazoliun assay(M1T assay). Results : The results of this study were obtained as follow ; 1. GTS was significantly increased in the proliferation of thymocytes and splenocytes In vitro. 2. GTS was significantly showed cytotoxicity on the L1210 cell lines and 8-180 cell lines in vitro. 3. GTS was significantly showed cytotoxicity on the L1210 cell lines in vivo. 4. GTS was significantly increased in the weight of mice and decreased weight of sarcoma, in S-180 cells transplanted mice. 5. GTS was significantly increased in the period of survive, in S-180 cells transplanted mice. Conclusions : The author thought that GTS had action of anti-cancer by becoming immunocytes activity and by cytotoxicity of cancer cells.

Effect of Low Dose Mutagens on Adoptive Response and Plasma Membrane Glycoconjugates in Sarcoma 180 Cells

  • The present investigation was performed to elucidate the effect of pretreatment with low dose ultraviolet radiation (UV) and ethyl methanesulfonate (EMS) on cell survival by trypan blue dye exclusion method and plasma membrane glycoconjugates by lectin-cytochemistry in sarcoma 180 (S180) cells. Pretreatment with 2 J/$m^2$ UV or 2 mM EMS increased the percentage of survival of cells subsequently treated with high dose UV (10 or 20 J/$m^2$) or EMS (10 or 20 mM), respectively. Staining intensity of concanavalin A (Con A) of the cells pretreated with 2J/$m^2$ UV or 2 mM EMS and subsequently treated with 10 or 20 mM EMS was stronger than that of the cellstreated with 10 or 20 mM EMS. These results suggest that there is an adaptive response on cell survival to EMS or UV in S180 cells. And the results show a change in mannose-containing glycoconjugates of plasma membrane in S180 cells pretreated with EMS or UV and subsequently treated with EMS.

Sarcoma-180 Cell로 유발한 Mouse 복수암에 대한 Astaxanthin 함유 난황의 효과 (Inhibition of Sarcoma-180 Cell-induced Mouse Ascites Cancer by Astaxanthin-containing Egg Yolks)

  • Anticarcinogenic activity of astaxanthin-contatining egg yolk(designate AEY) was investigated for mouse ascites carcinogenesis induced by mouse Sarcoma-180(S-180) cells. Female ICR mice(8mice/treatment, 7∼8weeks of age, 25±1g) were injected, i.p. with S-180 cells(1×107cell/ml PBS). Two days later, each mouse was given 0.1ml PBS containing AEY(10, 25 or 50ug/g body weight) or control egg yolk (CEY; 50ug/g body weight) every other day for 7 times. Control mice were only given 0.1ml S-180 cells and 0.1ml PBS. Mice treated with 25ug/g body weight of AEY showed 24.8 days of life, which was equivalent to 138% of control mice's life(180.0 dyas). Based on dose-dependant experiment of AFY, mice treated with 10ug/g body weight showed slightly longer life(19.4 days) relative to mice treated with control mice, and mice treated with 50ug/g body weight exhibited 21.9 days of life. Mice treated with any dose of AEY exhibited longer life than mice with CEY 50ug/g body weight. Body weight of mice treated with AEY was reduced relative to that of control mice CEY-treated mice. These results suggest than AEY inhibits the carcinogenesis of mouse ascites induced by S-180 cells.

마늘중 지용성 성분의 암세포증식 억제효과 연구 (A Study on the Cytotoxic Activity of Garlic(Allium Sativum) Extract Against Cancer Cells)

  • 본 연구는 유기용매로 추출한 마늘의 항암성 성분을 in vitro에서 흰생쥐의 백혈병성 임퐈모 세포인 L$_{1210}$과 P${388} 및 인체 직장암 세포인 HRT-18과 인체 결장암 세포인 HCT-48과 HT-29 또한 invivo에서 흰쥐 복수 육종암세포인 sarcoma-180세포를 대상으로 선별 시험한 것이다. S-180을 제외한 각 암세포는 in vitro에서 마늘추출물을 농도별로 첨가한 배양액에서 배양시험하였다. 암세포의 증식억제효과는 마늘의 석유에텔추출물이 알코올추출물보다 높았다. 마늘의 지용성 성분은 in vitro에서 흰생쥐의 백혈병성 임퐈모 세포, 인체 직장암 및 결장암 세포에 대해 항암효과물을 나타내었다. 각 암세포의 증식률은 첨가한 마늘추출물의 농도가 증가할수록 감소한 영향을 보였다. Petroleum ether/ethyl ether/acetic acid(90:10:1, v/v/v)의 전개용매로 사용한 TCL에서 분리한 유효한 활성성분의 Rf치는 0.18이었고, 정제하지 않은 마늘추출물보다 L$_{1210}$세포에 대해 2.3배 높은 활성을 나타냈다. S-180 암세포로 유발한 흰생쥐에 마늘 추출물을 투여한 군이 투여하지 않은 군보다 생존 기간이 1.5내지 2배 가량 연장효과를 보였다. S-180 암세포를 함유하고 있는 흰 생쥐에 마늘추출물(3ug/head)을 복강내 주사하고 3시간 후에 관찰한 결과 S-180 암세포의 뚜렷한 형태변화를 관찰하였다.

神授衛生湯의 抗腫瘍 效果 (Effects of Shinsuwuisaeng-Tang on the Anti-Tumor)

  • Shinsuwuisaeng-Tang was a drug that treated carbuncle and cellulitis. So, the purpose of this Study was to investigate effect of Shinsuwuisaeng-Tang on the anti-cancer and nitric oxide(NO) production of peritoneal macrophages. We used Shinsuwuisaeng-Tang extract(SWT) with freeze-dried, 8wks-old male mice. and cancer cell lines(L1210, sarcoma-180) for this Study. The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). The results of this Study were obtained as follow; SWT was showed cytotoxicity on the L1210 and sacoma-180(S-180) cell lines, SWT inhibited significantly proliferation of L1210 cells in L1210 cells transplanted mice, SWT accelerated NO production of peritoneal macrophages in L1210 cells transplanted mice. And SWT inhibited significantly tumor weight, increased significantly body weight and mean survival days in S-180 cells transplanted mice. This results suggest that SWT has anti-cancer by producing NO of peritoneal macrophages.

감초로 배양한 표고버섯 균사체 추출물이 항암 효과 및 알레르기 억제 효과 검증 (Anti-Cancer and Anti-Allergy Activities of Mycelia Extracts of Lentinus edodes Mushroom-Cultured Glycyrrhiza radix)

  • This study investigated the effects of mycelia of Lentinus edodes mushroom-cultured Glycyrrihiza radix(LMG) on cancer cell lines and sarcoma 180(S-180), as well as on human mast cells. In an anti-cancer tests using Hep3B(hepatic cancer cell), MCF-7(breast cancer), and HeLa(uterine cancer) cells, LMG extract exhibited greater anti-proliferation effects than Glycyrrihiza glabra(GG) extract. LMG extract multiplication restraining effects were 60% that of ethanol at 3 mg/mL extract also displayed tumor suppressive effects in mice injected with S-180 cells. The growth-inhibition rates against tumor cells were 56% for LMG and 37% for GG. When LMG was added to human mast cells, the Intensity of RT-PCR products using primers($FC{\varepsilon}RI\;c-kit$) decreased. significantly compared with that of control. These results suggest that Lentinus edodes Mushroom-Cultured Glycyrrhiza glabra has an anti-proliferation effects against cancer cell lines(Hep3B, MCF-7 and HeLa) and S-180 tumors and will be also beneficial in treating allergic reactions.

인삼의 지용성 성분과 사포닌 유도체의 항암작용 연구 (Astudy on the Anticancer Activies of Lipid Soluble Ginseng Extract and Ginseng Sapongin DErivatives Against Some Cancer Cells)

  • The anticancer activities of petroleum ether extract of Panax ginseng root(crude GX) and its partially purified fraction from silicic acid column chromatography (7:3 GX) were studied with Sarcoma 180(S-180) or Walker carcinosarcoma 256 (Walker 256) in vivo and with L1210 leukemic lympocyte in vitro. Potential cytotoxic activities of the crude GX and against L1210 cells were compared with those of 5-Fluorouracil (5-FU) and saponin derivatives (Panax-diol, Panax-triol, Diol saponin, Triol saponin) in vitro. In order to observe the physiological effects of the crude GX and 7:3 GX on the animals with cancer, hemoglobin(Hb), red blood cell(R.B.C) and white blood cell after treatment with each GX in comparison with corresponding control groups, respectively. The anticancer effects of the crude GX and 7:3 GX were estimated by measuring the survival time of S-180 bearing mice after treatment with them. The experimental results obtained are summarized as follows; 1. The one unit of cytotoxic activity against L1210 cells was equivalent to 2.54$\mu\textrm{g}$ and 0.88$\mu\textrm{g}$of the crude GX and 7:3 GX per ml of culture medium, respectively. 2. The cytotoxic activities of Panax-diol, Panax=triol, Diol saponin and triol saponin against L1210 cells were not detected. 3. The anticancer activities of 5-FU against L1210, S-180 and Walker 256 were very effective in vivo and vitro tests. 4. The significantly increased W.B.C values of mice after inoculation with S-180 cells were reduced to normal range by the crude GX treatment. 5. The significantly decreased Hb values of rats after inoculation with Walker 256 were recovered to normal range by oral administration of the crude GX. 6. The survival times of mice inoculated with S-180 cells were extended about 1.5 to 2 times by the 7:3 GX treatment compared with their control group.

Effects of Ethyl methanesuifonate and Ultraviolet light on Induction of the Adaptive Response in Chinese Hamster Ovary and Sarcoma 180 Cells

  • This study was performed by the sister chromatid exchanges (SCEs) and micronuclei (MN) assays to investigate the adaptive response to ultraviolet light (UV) or ethyl methanesulfonate (EMS) in Chinese hamster ovary (CHO) and Sarcoma 180 (S180) cells. The pretreatment with 1 J/m$^2$ UV or 2 mM EMS decreased the frequency of SCEs induced by the treatment with 5 J/m$^2$ UV or 8 mM EMS in CHO cells. The pretreatment with UV (1 or 2 J/m$^2$) or EMS (1, 2 or 3 mM) did not affect the SCEs induced by the treatment with 7 J/m$^2$ UV or 10 mM EMS in S180 cells. On the other hand, the pretreatment with 1 J/m$^2$ UV or 2 mM EMS decreased the frequency of MN induced by the treatment with 5 J/m$^2$ UV or 8 mM EMS in CHO cells. The pretreatment with UV (1 or 2 J/m$^2$) or EMS (1, 2 or 3 mM) did not affect the frequency of MN induced by the treatment with 7 J/m$^2$ UV or 10 mM EMS in S180 cells. It is suggested that there are adaptive responses at the level of chromosome and micronuclei to UV and EMS in CHO cells.

GMSGS was significantly decreased in the Weight of Sarcoma in S-180 cells transplanted mice. 5. GMSGS was significantly increased in the Period of Survive in S-180 cells transplanted mice. The present author thought that GMSGS had action of anti-cancer by becoming immunocytes activity(proliferation of thymocytes and splenocytes).

Anticancer activity of subfractions containing pure compounds of Chaga mushroom (Inonotus obliquus) extract in human cancer cells and in Balbc/c mice bearing Sarcoma-180 cells

  • The Chaga mushroom (Inonotus obliquus) has been used in folk medicine to treat cancers. However, limited information exists on the underlying anticancer effects of the major component of I. obliquus in vivo. We hypothesize that the pure compounds ($3{\beta}$-hydroxy-lanosta-8,24-dien-21-al, inotodiol and lanosterol, respectively) separated from I. obliquus would inhibit tumor growth in Balbc/c mice bearing Sarcoma-180 cells (S-180) in vivo and growth of human carcinoma cells in vitro. To test this hypothesis, the growth inhibition of each subfraction isolated from I. obliquus on human carcinoma cell lines (lung carcinoma A-549 cells, stomach adenocarcinoma AGS cells, breast adenocarcinoma MCF-7 cells, and cervical adenocarcinoma HeLa cells) was tested in vitro. Then, after S-180 implantation, the mice were fed a normal chow supplemented with 0, 0.1 or 0.2 mg of subfraction 1, 2 or 3 per mouse per day. All of the subfractions isolated from I. obliquus showed significant cytotoxic activity against the selected cancer cell lines in vitro. Subfraction 1 was more active than subfraction 2 and subfraction 3 against the A549, AGS and MCF-7 cancer cell lines in vitro. In in vivo results, subfraction 1 isolated from I. obliquus at concentrations of 0.1 and 0.2 mg/mouse per day significantly decreased tumor volume by 23.96% and 33.71%, respectively, as compared with the control. Subfractions 2 and 3 also significantly inhibited tumor growth in mice bearing S-180 as compared with the control mouse tumor. Subfraction 1 isolated from I. obliquus showed greater inhibition of tumor growth than subfractions 2 and 3, which agrees well with the in vitro results. The results suggest that I. obliquus and its compounds in these subfractions isolated from I. obliquus could be used as natural anticancer ingredients in the food and/or pharmaceutical industry.
