한국산 양태속 어류의 분류학적 검토 (Taxonomic Review of the Genus Platycephalus (Platycephalidae) from Korea)

  • 한국산 Platycephalus속 어류 2 종의 분류학적 위치관계를 면밀히 검토하기 위하여 이들의 주요 형태적 특징과 근단백질 전기영동 및 LDH, MDH isozyme 등을 분석하였다. 그 결과 가슴지느러미의 연조수에서 P. indicus 에서는 18~20개 (P. sp.에서는 16~19개), 측선 린수는 70~80개 (83~100개), 두장에 대한 안경의 비율은 10.2~13.1% (10.9~15.5%)이었고, 양안간격은 14.5~18.1% (11.8~17.8%)였으며, 이 외에도 문단의 형태, 체색 그리고 가슴 및 배지느러미의 색깔 등에서 잘 구분되었다. 한편 전기 영동상에 의한 근 단백질 band 수는 P. indicus에서 16개, P. sp.에서 18개로 분리되었다. LDH isozyme은 P. indicus에서는 2개, P. sp.에서는 1개로 나타났고, MDH isozyme은 P. indicus에서 5개, P. sp.에서 4개로 분리되었다. 이 외에도 각 band의 이동거리, 활성도 등에서도 두 종간에 상이하게 나타났었다. 따라서 한국에 서식하고 있는 양태속 어류에 속하는 P. indicus와 P. sp.는 서로 다른 종으로 간주된다.

만경 동진강 하구계의 동물플랑크톤의 분포와 염분 (Salinity and Distribution of Zooplankton in the Estuarine System of Mankyong River and Dongjin River)

  • 만경 동진강 하구계의 표층에 분포하는 동물플랑크톤을 1989년 10월과 1990년 3 월, 5월 그리고 7월에 표준 네트로 수평 채집하였다. 우점하였던 주요 분류군은 10월 과 3월은 요각류, 5월과 7월은 야광충이었다. 동물플랑크톤의 출현량은 개체수가 $12~9,256ind./m^3$, 습증량이 $2.8~1,889.8{\;}mg/m^3$ 범위로 상당한 시공간적인 연동을 보 였다. 요각류의 우점종은 10월에 Pseudodiaptomus inopinus와 Paracalanus indicus, 3 월에 Acartia hudsonica와 P. indicus, 5월에 A. hudsonica와 Centropages abdominalis 그리고 7월에 A. pacifica P. indicus와 Tortanus spinicaudatus로 변하 였다. 7월에는 강물의 유입량 증가로 정점간 염분변화가 심하게 나타났으며 ($0.7~28.9\textperthousand$), 이것은 요각류의 군집구조를 변화시켰다. 중염기수역($5.0~28.0\textperthousand$)에서는 Acartia pacifica, A. hudsonica, Centropages tenuilemis, C. abdominalis, Labidocera euchaeta, Tortanus spinicaudatus, Corycaeus affunis 그리고 Paracalanus indicus 8종이 분포 하였다. 저염분에 강한 요각류가 빈염기수역($0.5~5.0\textperthousand$)에 분포하였으며, 이들은 Thermocyclops hydlinus, Pseudodiaptomus inopinus 그리고 Sinocalanus tenellus 의 3종이었다. 고염기수역($>28.0\textperthousand$)에서는 Labidocera bipinnata, Oithona similis, Calanus sinicus, Acartia omorii 그리고 Tortanus forcipatus가 나타났다.

Genetic Variation of the Major Histocompatibility Complex DRB3.2 Locus in the Native Bos indicus Cattle Breeds

  • The major histocompatibility complex (MHC) plays well-defined roles in eliciting immune responses and combating infectious diseases. The major histocompatibility complex of cattle is referred to as BoLA (Bovine Lymphocyte Antigen). This genetic system is among the most polymorphic. In the present study, polymorphism of the BoLA- DRB3.2 locus in three Bos indicus breeds viz., Sahiwal, Rathi and Hariana was studied by polymerase chain reaction restriction fragment length polymorphism technique using the enzymes RsaI, Bst Y1 and Hae III. Both Sahiwal and Rathi are good Indian dairy breeds and survive under tough tropical conditions, while Hariana is a prominent dual-purpose breed reared both as a dairy animal and for bullock production. A total of 30 different BoLADRB3.2 alleles were observed to be present in the 3 Bos indicus breeds. Certain alleles were common amongst the three breeds while there were others that were unique to each breed. Allelic distribution amongst the three breeds showed that each breed had a unique allelic distribution pattern that was different from each other and also different from the earlier breeds studied so far for the existence of allelic variation at this locus. A dendogram was constructed based on the frequencies of the BoLA-DRB3 alleles using the UPGMA method. The Rathi and Hariana animals were genetically the most apart. The Hariana animals clustered on a different branch from the other two breeds viz. the Rathi and the Sahiwal. The smallest genetic distances for the DRB3 alleles were those between Sahiwal and Rathi (0.5461) while genetic distance between Hariana and Sahiwal was 0.6123. A comparison of the allelic frequencies of the BoLADRB3.2 locus in these 3 breeds viz. Sahiwal, Hariana and Rathi with the allelic frequencies present in the previously characterized Bos indicus Kankrej breed, which is a dual purpose breed reared both as a draught and a dairy animal, showed that the Bos indicus Sahiwal and Rathi breeds clustered into one group while the Hariana and Kankrej breeds formed another group. The Rathi and Sahiwal showed the least genetic distance of 0.5461 amongst the breeds whereas the Rathi and Kankrej, with a Nei''s genetic distance of 1.1622, were genetically the most distant apart.

COI 기반 제한효소 절편 길이 다형성(RFLP)을 이용한 새우젓 분석 (Identification of Salted Opossum Shrimp Using COI-based Restriction Fragment Length Polymorphism)

  • 국내 유통되고 있는 새우젓은 다양한 작은 새우의 집합체로, 최근 어획량 감소로 인해 국내산 새우젓의 경우 수입산에 비해 두배 이상 높은 가격에 거래되고 있으며, 이에 중국 및 베트남 등으로부터 수입된 새우젓이 국내산으로 둔갑되어 판매되는 사례가 빈번하게 발생되고 있다. 본 연구에서는 새우젓 Acetes japonicus, A. chinensis (Korea, China), A. indicus (I, II), Palaemon gravieri 6종류에 대하여 PCR-RFLP 마커를 개발하였다. 새우젓에서 에탄올로 염을 제거한 후 gDNA를 추출하였고, 새우젓 COI 유전자의 특이 프라이머를 제작, PCR을 진행하여 519 bp의 증폭시켰다. 증폭된 PCR산물의 염기서열분석을 토대로 Acc I, Hinf I 두 가지의 제한효소를 마커로 선정하였고, 전기영동을 통해 결과를 확인하였다. Acc I을 처리한 결과, A. japonicus, A. chinensis (Korea), A. indicus (II)는 272, 247 bp로, P. gravieri는 271, 202, 46 bp, A. chinensis (China), A. indicus (I)는 519 bp로 band를 형성하는 것을 확인할 수 있었다. 또한 Hinf I을 처리한 결과로는 A. chinensis (Korea, China), P. gravieri는 519 bp로 잘리지 않은 것을 확인한 반면, A. japonicus와 A. indicus (I)는 2 band, A. indicus (II)는 3 band를 형성하는 것을 확인할 수 있었다. 따라서 위의 결과는 국내산과 수입산이 혼합되어있는 새우젓에 있어 보다 신속한 종판별을 할 수 있음을 시사한다.

Genetic Relationships of Cattle Breeds Assessed by PCR-RFLP of the Bovine Mitochondrial DNA D-loop Region

  • To investigate the genetic relationships among various cattle breeds, bovine mtDNA D-loop region was used in 411 animals of 18 cattle breeds, including 8 Asian Bos taurus, 7 European Bos taurus, 1 Asian Bos indicus, and 2 African Bos indicus. The size of amplified PCR products from mtDNA D-loop region was 964 bp and the products were digested by 15 different restriction enzymes. Two different band patterns were identified in eight restriction enzymes (BstXI, Hae III, Msp I, Apa I, Taq I, Alu I, BamH I, EcoN I) and the rest of restriction enzymes showed more than 3 different band patterns among which Apo I and MspR9 resulted in 7 different restriction patterns. The genotypes, number of haplotype, effective number of haplotype, and degree of heterozygosity were analyzed. Based on all the PCR-RFLP data, different haplotypes were constructed and analyzed for calculating genetic distances between these breeds using Nei's unbiased method and constructing a phylogenetic tree.

Improvement in Clonal Propagation of Hemidesmus indicus R. Br. through Adenine Sulphate

  • A protocol has been developed for rapid large scale clonal propagation of an aromatic endangered medicinal plant, Hemidesmus indicus R. Br. with the elimination of the problems such as premature leaf fall and callus formation during caulogenesis and rhizogenesis. Multiple shoots were induced from shoot tip and nodal explants on Murashige and Skoog (MS) medium supplemented with 1 mg/L Benzylaminopurine (BAP) and 0.5 mg/L Napthaleneaceticacid (NAA). Addition of 15 mg/L adenine sulphate to the above medium checked leaf abscission completely, reduced the time required for caulogenesis and restored morphogenetic potential after several subcultures. The in vitro grown propagules were rooted in 1/2 MS medium supplemented with 2 mg/L Indolebutyric acid (IBA) +1 mg/L NAA and sucrose 0.7% (w/v). Addition of charcoal at 100 mg/L to the rooting medium quickened root initiation with a complete check on callus formation. The effect of sucrose concentration on both caulogenesis and rhizogenesis was also studied. The resultant plantlets were acclimatized and grown in fields where ninety eight percent of the rooted shoots survived and grew normally. The estimation of the secondary metabolite content in the shoots of the regenerated plant and the mother plant indicated that the concentration of the three secondary metabolites lupeol, vanillin and rutin was similar.

  • 양태, Platycephalus indicus 정자의 동결보존을 위해 적정 동해방지제와 동결높이에 따른 예비동결(0, 2, 4, 6cm), 그리고 해동온도에 따른 정자의 운동성과 생존율로 비교하였다. 적정 동해방지제로 dimethylsulphoxide(DMSO), glycerol, methanol에 대해 실험하였으며, DMSO와 glycerol에서 methanol에 비하여 높은 생존율과 운동성을 나타내었다. 또한, 동결보존에 있어 예비동결은 액체질소표면으로부터 2 cm 높이에서 높은 생존율과 운동성을 보였고, 해동온도로는 $20^{\circ}C$가 가장 적합하였다.

Muscle-specific color stability in fresh beef from grain-finished Bos indicus cattle

  • Objective: To investigate the color and oxidative stabilities of longissimus lumborum (LL) and psoas major (PM) muscles from grain-finished Bos indicus cattle in Brazil. Methods: The LL and PM muscles were obtained 24 h post-mortem from eight (n = 8) Nellore bull carcasses, fabricated into 1.5-cm steaks, aerobically packaged, and stored at $4^{\circ}C$ for nine days. Steaks were analyzed for myoglobin concentration, pH, instrumental color, metmyoglobin reducing activity (MRA) and lipid oxidation. Results: The LL steaks exhibited greater (p<0.05) redness, color stability, and MRA than their PM counterparts on days 5 and 9. The LL and PM steaks demonstrated similar (p>0.05) lightness and yellowness on days 0, 5, and 9. On the other hand, PM steaks exhibited greater (p<0.05) myoglobin concentration, pH, and lipid oxidation than their LL counterparts. Conclusion: These results indicated that muscle source influenced the color and oxidative stabilities of beef from grain-finished Bos indicus animals. These results highlighted the necessity of muscle-specific strategies to improve the color stability of beef from grain-fed Bos indicus cattle.

Induction of Sexual Stage and Colony Morphology of Some Isolates of Sclerotium rolfsii Causing Spotted Leaf Rot in Plants

  • Twenty-two isolates of Sclerotium rolfsii causing spotted leaf rot from Varanasi, India were grown on 6% Cyperus rotundus rhizome meal agar (CRMA) medium for the induction of athelial stage (Athelia rolfsii). Only one isolate obtained from Sphaeranthus indicus formed basidial stage on CRMA medium while the other 21 isolates did not. Basidial stage was also produced in S. indicus isolate at different concentrations (5.5, 6.0 and 6.5% w/v) of CRMA medium. Size of basidia, sterigmata and basidiospores of this isolate was measured. Basidia clavate, hyaline and measured $10{\sim}12{\times}4{\sim}5\;{\mu}m$ in size, basidiospores hyaline, unicellular, subglobose to ellipsoid produced on sterigmata and measured $3{\sim}5{\times}2{\sim}4\;{\mu}m$ in size, sterigmata hyaline and measured $4{\sim}5{\times}1.5{\sim}2\;{\mu}m$ in size. The results of the present study revealed wide variation in spotted leaf rot isolates of S. rolfsii. A reddish zone around the colony of S. rolfsii isolate from Vernonia sp. was observed on CRMA medium. HPLC analysis of the zone revealed the presence of gallic and ferulic acid which were also thought to be responsible for reduced mycelial growth of the isolate on CRMA medium.

PREVALENCE OF HELMINTHIC INFESTATIONS IN ZEBU CATTLE (Bos indicus) AT SAVAR, BANGLADESH

  • Rectal fecal samples from zebu cattle (Bos indicus) at Savar, Dhaka, were collected in every two months of the year from March, 1987 to February, 1988 and were examined using Stoll's dilution technique. Out of a total of 737 fecal samples examined in six periods, 589 (79.9%) samples were found to be positive for one or more helminths. Overall prevalences for fasciola, paramphistome, schistosome, strongylids, ascaris, strongyloides, trichuris and capillaria infestations were respectively 19.5%, 48.0%, 5.9%, 52.2%, 1.5%, 2.5%, 5.5% and 12.7%. Prevalences of fasciola, paramphistome and schistosome infestations were significantly (p<0.01) higher from middle of August to December. Strogylids, trichuris and capillaria infestations were significantly (p<0.01) higher from July to October, although strongylids infestation also increased during January-February. Significantly (p<0.01) higher fasciola and paramphistome infestations were observed in animals after one year of age, whereas strongylids, ascaris, strongyloides and trichuris infestations were significantly (p<0.01) higher in cattle upto one year of age. Fasciola infestation was significantly (p<0.01) higher in female cattle and significantly (p<0.05) higher strongylids infestation was found in males.
