대구.경북지역 오리 유래 Salmonella속 균의 혈청형, 항균제 내성 및 S. Enteritidis와 S. Typhimurium의 plasmid profiles, phage types 및 PFGE (Serotypes, antimicrobial resistance of Salmonella spp. and plasmid profiles, phage types, PFGE of S. Enteritidis and S. Typhimurium isolated from ducks in Daegu-Gyeongbuk province)

  • Salmonella spp. is of increasing public health concern as causative pathogens of food poisoning. The aim of this study was to investigate the serotypes and antimicrobial resistance pattern of Salmonella spp. isolated from duck farms in Daegu-Gyeongbuk province. Also, S. Enteritidis and S. Typhimurium isolates were further examined for plasmid analysis, phage typing and pulsed-field gel electrophoresis (PFGE). A total of 34 Salmonella spp. (16.4%) were isolated from duck farms and ten serotypes were identified. The predominant serotypes were S. Typhimurium (23.5%) S. Fyris (17.6%) and S. Haardt (11.8%), S. Agona and S. Enteritidis (respectively 8.8%). Of 34 Salmonella isolates, 15 (44.1%) isolates were resistant to at least one antimicrobial agent and multiple resistance (resistance to more than 4 drugs) was observed in 9 strains (26.5%). The high resistance was found to streptomycin (32.4%), tetracycline (29.4%), ampicillin, kanamycin and nalidixic acid (respectively, 26.5%), all Salmonella isolates were susceptible to cefoxitin, cefotaxime, gentamicin, amikacin and ciprofloxacin. All S. Enteritidis and S. Typhimurium isolates were found to contain only one plasmid (ca. 54 or 55kb, respectively). Among the S. Enteritidis isolates, two phage types were found, PT32a and PT1c, respectively, one isolates did not react with any of the phages used. Whereas, all S. Typhimurium isolates were RDNC (reacts but does not conform). PFGE showed to be a useful typing method better than plasmid analysis and phage typing for discrimination of isolates especially, S. Typhimurium isolates. Our results indicated that the serotypes of Salmonella isolates are widely distributed in duck farms, further epidemiological studies should be carried out.

우리나라의 가금과 환경에서 분리한 Salmonella species의 특성 (Characteristics of Salmonella species isolated from domestic poultry and environmental samples in Korea)

  • This study was conducted to investigate the isolation prequency, serotypes, and related epidemiological properties of 341 Salmonella spp from domestic poultry and environmental samples during the period 1993-1995. A total of 1,918 samples was collected during the three years period in nationwide. Most of Salmonella spp were isolated from the intestinal contents of poultry, especially cecal(46.0%) and rectal(35.8%) contents. Among the tested samples, rat(28.5 %) was the most predominant Salmonella reservoirs and followed by duck(24.8%), broiler(18.8%), layer(14.8%) and feed(7.1%), in order. More than twelve Salmonella serovars were identified among the 341 Salmonella isolates. The most prevalent serotypes isolated from non-human sources were S enteritidis (22.3%) and S pullorum (21.9%), S muenchen (13.9%), S typhimurium (12.6%), S gallinarum, S meleagridis, S heidelberg, and S senftenberg were followed, in order. In layer chickens, S pullorum (26.0%) was the most predominant serotype but S muenchen (33.0%) was in broiler chickens, S enteritidis (28.4%) was in ducks, and S typhimurium (60.0%) was in rats, respectively. As a results, S enteritidis was identified as the most prevalent serotype in non-human Salmonella isolates in Korea during the period 1993-1995. A preliminary study on the phage typing of 19 S enteritidis selected from the nationwide scale was shown that S enteritidis phage type(PT) 4 was the most predominant PT, and SEPT 1, SEPT 6a, SEPT 7 and SEPT 7a variant were also found in the same period.

반응표면 분석을 이용한 Salmonella enteritidis의 증식 효능 평가 (Assessment of the Effectiveness of a Natural Antimicrobial substance on Salmonella enteritidis using Response Surface Analysis)

  • This study was performed to investigate the single and combined effect of concentrations of garlic juice according to the pH and temperature on the growth of Salmonella enteritidis in brain heart infusion broth, and to develope Response surface model for the effect of concentrations of garlic extract. The results of electric conductibility of Salmonella enteritidis growing in the range of incubation temperature ($25~42^{\circ}C$), pH (5.5~9.0) and concentration of garlic Juice (0~0.8%) showed that a badge with high temperature had low D.T.value and concentration of garlic extract were significantly correlated with D.T.value (p<0.01). Growth of Salmonella enteritidis in the condition of $37^{\circ}C$ and pH 7.0 presented the lowest D.T.value.

  • Forty-five Salmonella typhimurium isolates were encountered 8 phage types in which DT197 and U302 were the predominant types. The DT104 type which was first found from pig in Korea, and was resistant to chloramphenicol, streptomycin, sulfamethoxazole/trimethoprim, tetracycline, gentamicin and nalidixic acid. Twenty-two S enteritidis isolates were encountered 5 phage types in which PT4 were the representative (predominant). S enteritidis isolates were susceptible to all antimicrobial agents. As a result of PFGE analysis for S typhimurium and S enteritidis, PFGE patterns was better than phage typing in discriminating of strains. PFGE patterns were not in accord with phage type even though some strain had the same phage types.

  • YS-medium(0.1% Yeast extract +10% Skimmilk)중에 유산간균 L.casei, L. acidophilus , L. bulgaricus 와 병원성 E, coli, Sal. enteritides를 혐기적으로 혼합배양하면서 각각의 생육과 저해를 조사한 결과 , E. coli와 Sal. enteritideis의 생육이 각 유산균에 의하여 현저하게 저해되었다. 그러나 L.casei와 L.acidophilus의 각 유산균은 E.coli와 Sal. enteritidis에 의하여 생육이 전혀 저해되지 않았고 , L. bulgaricus는 E. coli 에 의하여 약간 저해를 받았다. E. coli와 Sal. enteritidis 의 단독배양액의 pH는 72시간째에 각각 5.08과 5.70이었으며, 유산균혼합배양액의 pH는 3.75~4.48이었고, 병원성균의 생육억제작용은 주고 pH 저하에 기인하였다.

  • The in visto inhibitory activities of essential oils of the Rosmarinus officinalis as well as its main constituents were evaluated against antibiotic-susceptible and -resistant strains of Staphylococcus aureus, streptococcus pneumoniae, Salmonella enteritidis and S. typhimurium. The essential oil fraction of R. officinalis and its main components, 1,8-cineole and camphor, exhibited significant inhibitory activities against most of the tested strains in this study, with MICs(minimum inhibitory concentrations) racing from 0.5mg/ml to 16mg/ml. The total oil fraction showed higher activity than its main components, 1,8-cineole and camphor against S. aureus strains. No remarkable differences were evident between MICs of the susceptible and resistant strains of S. aureus. Among the tested strains, S. pneumoniae CCARM 3523, the resistant strain to norfloxacin, oxacillin and erythromycin exhibited significantly lower sensitivity to the tested oils than antibiotic-susceptible strain. The oils revealed mostly higher inhibitory activity against S. typhimurium than against S. enteritidis.

  • The common pathogen Salmonella enteirtidis (S. enteritidis) is the major cause of foodborne disease. Protein identification by peptide mass fingerprinting using the matrix-assisted laser desorption ionization time of fight (MALDI-TOF) mass spectrometry (MS) can analysis unambiguously identity the spots from 2-dimensional electrophoresis (2-DE) gel. In this report, we examined protein components from patterns of S. enteritidis proteins. In addition, antigens that are recognized by sera can be identified by immunoblotting. This study that 2-DE analysis of S. enteritidis yields useful information concerning S. enteritidis proteome, the results that have been obtained led to a more detailed understanding of Salmonella pathology and open further interesting fields for future work.

Antibacterial activity of supernatant obtained from Weissella koreensis and Lactobacillus sakei on the growth of pathogenic bacteria

  • This study was carried out to obtain basic data for the industrial use of Weissella koreensis and Lactobacillus sakei. The antibacterial activity of supernatants obtained from W. koreensis and L. sakei were tested against pathogenic bacteria such as Escherichia coli KCCM 11234, Salmonella enteritidis KCCM 3313, Salmonella enteritidis KCCM 12021, Salmonella typhimurium KCCM 40253, and Salmonella typhimurium KCCM 15. The supernatant of L. sakei showed antibacterial activity against E. coli KCCM 11234, S. enteritidis KCCM 12021, and S. typhimurium KCCM 15, while the supernatant of W. koreensis showed antibacterial activity against E. coli KCCM 11234 and S. enteritidis KCCM 12021. The effect of pH changes and heat treatment on antibacterial activity of the supernatants was examined using the sensitive pathogenic bacteria (E. coli KCCM 11234, S. enteritidis KCCM 12021 and S. typhimurium KCCM 15). Antibacterial activity against sensitive pathogenic bacteria was maintained under heat treatment at all temperatures, but there was no antibacterial activity associated with pH modification. Furthermore, it was confirmed that the antibacterial activity of the supernatants obtained from W. koreensis and L. sakei was a result of organic acids including, lactic, acetic, phosphoric, succinic, pyroglutamic, citric, malic, and formic acids. Therefore, the present study showed that the organic acids produced by L. sakei and W. koreensis exhibited a strong antibacterial activity against pathogenic bacteria. Moreover, in the food industry, these organic acids have the potential to inhibit the growth of pathogenic bacteria and improve the quality of stored food.

  • 사람들에게 계란과 관련하여 일어나는 S. enteritidis(SE)와 S. typhimurium(ST)의 전파를 줄이기 위해서 감염된 상업적인 산란계군을 찾는 것은 매우 중요한 일이다. 그러므로 본 연구는 국내 산란계군에서 생산되는 계란의 주된 식중독균인 SE와 ST의 청정화 관리를 위한 기초 연구로서 SE와 ST에 대한 산란계군의 감염 여부를 파악하기 위해 시중에 유통하는 계란의 난황에 존재하는 SE와 ST에 대한 특이 항체를 ELISA방법으로 조사하였다. 시중에 유통되는 3개 제품의 계란의 난황을 1:100부터 1:1,600까지 두배 희석법으로 희석한 후 SE균체와 ST균체 및 ST의 OMP항원에 대해 특이 항체 역가를 ELISA로 측정한 결과 대조군에 비해 희석곡선의 형태가 차이가 나는 것을 알 수 있었다. 시중에 유통중인 2개 제품의 계란을 1:200과 1:1,000으로 희석한 후 ST항원, OMP항원과 SE항원에 대한 각각 ELISA를 실시한 결과 각각의 항원에 대해 난황특이 항체에 대한 역가는 같은 제품일지라도 차이가 있었다. 21개 제품의 계란을 각 회사마다 5개씩 무작위로 추출하여 계란난황을 1:1,000으로 희석한 후 3가지 항원 ST균체, SE균체 및 ST의 OMP 항원에 대해 ELISA을 실시한 결과 SE와 ST균체 항체보다 OMP항원에 대해 전반적으로 높은 항체 역가를 나타냈으며, ST는 30%, SE는 28.5%, OMP는 28.5% 수준에서 대조구와 비슷한 낮은 역가의 항체 값을 나타내었다. 본 논문의 결과는 난황항체 역가를 측정함으로서 산란계군에서 ST와 SE에 오염을 파악하거나 ST와 SE의 오염을 줄이는데 효과적으로 이용될 수 있는 민감한 방법이라고 생각된다.X>$10^{8}$ g ~ 3.05$\times$$10^{8}$ g 분포를 나타냈으며, 생녹각김치, 유산처리 녹각김치, 대조김치, 열수추출 녹각김치의 순으로 많았다. 녹각 김치를 사람들에게 맛보게 하여 짠맛, 신맛, 향기, 색, 조직감을 평가시킨 결과 열수추출 녹각김치와 생녹각김치가 가장 높은 값을 나타냈고 이어서 대조김치와 유산처리 녹각김치의 순을 나타냈다.TEX>0.13)은 무첨가 초콜릿(3.25$\pm$0.15)에 비해 모두 유의적(P<0.001)으로 낮은 점수를 나타냈으나, 2% 첨가수준에서는 맛, 향, 조직감 및 기호도에서 유의적인 차이를 보이지 않았다.의 소규모 와, 강한조류 및 지형적 특성(불규칙한 해저지형 및 고립도서)에 의해 이루어지는 왕성한 수평 및 연직 혼합 등과 같은 어장학적 호조건에 의해 마라도 주변에 체류하게 되고, 이들 체류어군은 조류방향에 따라 섬의 조상 측에 농밀군을 형성하는 섬의 조상측 어장이다.향을 주지 않는 것으로 나타났으므로 청소년 각자의 식습관 및 식품 섭취에 대한 관심을 고취시킬 필요가 있다고 생각된다.d with an MR peak in the vicinity of the coercive field. Rapid, Sensitive, and Specific Detection of Salmonella Enteritidis in Contaminated Dairy Foods using Quantum Dot Biolabeling Coupled with Immunomagnetic Separation

  • Colloidal semiconductor CdSe-ZnS core-shell nanocrystal quantum dot (Qdot) are luminescent inorganic fluorophores that show potential to overcome some of the functional limitations encountered with organic dyes in fluorescence labeling applications. Salmonella Enteritidis has emerged as a major cause of human salmonellosis worldwide since the 1980s. A rapid, specific, and sensitive method for the detection of Salmonella Enteritidis was developed using Qdot as a fluorescence marker coupled with immunomagnetic separation. Magnetic beads coated with anti-Salmonella Enteritidis antibodies were employed to selectively capture the target bacteria, and biotin-conjugated anti-Salmonella antibodies were added to form sandwich immune complexes. After magnetic separation, the immune complexes were labeled with Qdot via biotin-streptavidin conjugation, and fluorescence measurement was carried out using a fluorescence measurement system. The detection limit of the Qdot method was a Salmonella Enteritidis concentration of $10^3$ colony-forming units (CFU)/mL, whereas the conventional fluorescein isothiocyanate-based method required over $10^5CFU/mL$. The total detection time was within 2 h. In addition to the potential for general nanotechnology development, these results suggest a new rapid detection method of various pathogenic bacteria from a complex food matrix.
