Isolation of Flavone Glycoside from Circium japonicum var ussuriense and Biological Activity on the Cardiovascular System (엉겅퀴 지상부의 심혈관 작용활성 및 후라본 배당체의 분리)

  • A flavone glycoside was separated from the aerial part of Circium japonicum var ussuriense Kitamura and the effect on cardiovascular system was investigated. The cadiovascular contractility of this compound was observed in the artria and aortae from normal rats. It increased the spontancous beat in right atria and the contractile force in left atria, and caused the contraction of thoracic aortae. For the blood pressure, it had ascending effect. The effective component, which acted on stimulation and contraction in the artria and aortae of rats was fractionated with n-BuOH, separated and identified by column chromatography, UV, IR, $^1H-NMR$ and $^{13}C-NMR$. The chemical structure for this component was determined to be $hispidulin-7-{\alpha}-rhamnopyranosyl(1{\rightarrow}2)-{\beta}-D-glucopyranoside$.

Preventive effects of Injinho-tang and Haninjinho-tang on hyperlipidemia and liver damage induced by alcohol (인진호탕(茵蔯蒿湯)과 한인진호탕(韓茵蔯蒿湯)이 알코올 투여(投與)로 유발(誘發)된 흰쥐의 고지혈증(高脂血症)과 간(肝) 손상(損傷)의 예방(豫防)에 미치는 영향(影響))

  • Objectives : This study was performed to compare the preventive effect of water extracts from Injinho-tang and Haninjinho-tang on hyperlipidemia and liver damage induced by alcohol. Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And each extract was administrated for the same period. We measured the serum component in rat's blood, body weight and weight of liver. Results : 1. At first, we observed effects of Injinho-tang and Haninjinho-tang on hyperlipidemia induced by alcohol. Injinho-tang group showed significant decrease of triglyceride, body weight in comparison with those of the control group. Haninjinho-tang group showed significant decrease of total cholesterol, triglyceride in comparison with those of the control group. I 2. At second, we observed effects of Injinho-tang and Haninjinho-tang on liver damage induced by alcohol. lnjinho-tang group showed significant decrease of AST (aspartate aminotransferase), ALT (alanine aminotransferase), ALP (alkaline phosphatase), liver weight in comparison with those of the control group. Haninjinho-tang group showed significant decrease of AST (aspartate aminotransferase), ALT (alanine aminotransferase) in comparison with those of the control group. Conclusion : Reviewing these experimental results, it appears that water extracts from Injinho-tang and Haninjinho-tang have pharmaceutical preventive efficacy on hyperlipidemia and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of these.

Study on the Performance of Laser Welded Joint of Aluminum Alloys for Car Body

  • Considering the fuel consumption of car, a light structure of aluminum alloys is desired fer car body nowadays. However, fusion welding of aluminum alloys has some problems of reduction of joint efficiency, porosity formation and hot cracking. In the present work, investigation to improve the joint performance of laser welded joint has been carried out by addition of Cu, Ni, and Zr to A6NO 1 alloy welds. Aluminum alloy plate of 2.Omm in thickness with filler metal bar was welded by twin beam Nd: YAG laser facility (total power: 5kW). The filler metals were prepared by changing the chemical compositions for adding the elements into the weld metal. Thirteen filler metal bars were prepared and pre-placed into the base metal before welding. Ar gas shielding with a flow rate of 10 1/min was used. The defocusing distance is kept at 0 mm. At travel speeds off 3 to 9 and at laser power of 5kW (front beam 2kW rear beam 3kW), full penetration welds were obtained, whereas at travel speeds of 12 to 18 m/min and same power, partial penetration was observed. The joint efficiency of laser-welded joint was improved by the addition of Cu, Ni, and Zr due to the solid solution hardening, grain refining and precipitation hardening. The type of hardening has been further considered by metallurgical examination.

Antibacterial Substances of the Flower of Chrysanthemum zawadskii Herbich var. latilobum Kitamura (구절초 꽃의 항균성 물질)

  • In orther to isolate antibacterial substances from the flower of Chrysanthemum zawadskii Herbich var. latilobum Kitam., the chloroform fraction was fractionated according to the activity against B. subtilis, S. aureus and V. parahaemolyticus. Two antibacterial substances were isolated and purified by preparative TLC and recrystallization. Compound I showed activity against all the tested bacteria and compound II exhibited the activity against B. subtilis and v. parahaemolyticus except S. aureus. Compared to benzoic acid and sorbic acid which are being used as food preservatives, compounds I and II showed about five-fold stronger antibacterial activity against B. subtilis and V. parahaemolyticus. On the basis of spectrometric studies including $^1H-NMR,\;^{13}C-NMR,\;DEPT,\;^1H-^1H\;COSY,\;^{13}C-^1H\;COSY$ and Mass, compounds I and II were identified as angeloylcumambrin B and cumambrin A, respectively. This is the first report of the isolation of angeloylcumambrin B and cumambrin A from this plant.

Protection by Chrysanthemum zawadskii extract from liver damage of mice caused by carbon tetrachloride is maybe mediated by modulation of QR activity

  • Our previous study demonstrated that methanolic extract of Chrysanthemum zawadskii Herbich var. latilobum Kitamura (Compositae) has the potential to induce detoxifying enzymes such as NAD(P)H:(quinone acceptor) oxidoreductase 1 (EC 1.6.99.2) (NQO1, QR) and glutathione S-transferase (GST). In this study we further fractionated methanolic extract of Chrysanthemum zawadskii and investigated the detoxifying enzyme-inducing potential of each fraction. The fraction (CZ-6) shown the highest QR-inducing activity was found to contain (+)-(3S,4S,5R,8S)-(E)-8-acetoxy-4-hydroxy-3-isovaleroyloxy-2-(hexa-2,4-diynyliden)-1,6-dioxaspiro [4,5] decane and increased QR enzyme activity in a dose-dependent manner. Furthermore, CZ-6 fraction caused a dose-dependent enhancement of luciferase activity in HepG2-C8 cells generated by stably transfecting antioxidant response element-luciferase gene construct, suggesting that it induces antioxidant/detoxifying enzymes through antioxidant response element (ARE)-mediated transcriptional activation of the relevant genes. Although CZ-6 fraction failed to induce hepatic QR in mice over the control, it restored QR activity suppressed by $CCl_4$ treatment to the control level. Hepatic injury induced by $CCl_4$ was also slightly protected by pretreatment with CZ-6. In conclusion, although CZ-6 fractionated from methanolic extract of Chrysanthemum zawadskii did not cause a significant QR induction in mice organs such as liver, kidney, and stomach, it showed protective effect from liver damage caused by $CCl_4$.

Immune Response of Olive flounder, Paralichthys olivaceus against Miamiensis avidus (Ciliophora: Scuticociliatida) (스쿠티카충 Miamiensis avidus에 대한 넙치의 면역반응)

  • Miamiensis avidus is a scuticociliate causing mortality in olive flounder Paralichthys olivaceus. To evaluate immune response of olive flounder against M. avidus, 2.6×106cells/fish of Formalin killed cell (FKC) was intraperitoneally (i.p.) injected, and 2.4 × 106cells/㎖ of sonicated FKC was immersion immunized to 14.9 cm (26.8g) fish. Fish were immunized 2 times with 2 weeks intervals. Antiserum from immunized fish caused agglutination and immobilization of the ciliate. In ELISA test, immunized group exhibited higher titers than control group. In addition, i.p. immunized fish showed higher phagocytic ratio, phahgocytic index and chemotaxic activity. To evaluate in vivo efficacy of FKC on fish, 1.43 × 105cells/fish was i.p.. injected, and 2.2 × 105 cells/㎖ of sonicated cell was immersed into 8.6cm (6.3g) fish. After 2 times immunization with 2 weeks intervals, fish were infected with 2.0 × 104 and 2.0 × 103 cells/ml of live ciliates by immersion. After 3 weeks, cumulative mortality was lower in the i.p. immunized group and mortality was delayed in immersion immunized group. In conclusion, specific immune response of oliver flounder against M. avidus was elevated after immunization and these immune response may prevent and/or delay for the M. avidus infection to olive flounder.

Effects of Concentration of Nutrient Solution and Irrigation Frequency on Growth and Flower Quality of Cut Chrysanthemum Grown Hydroponically in Perlite (국화의 펄라이트 양액재배시 양액농도 및 관주주기가 생육과 절화 품질에 미치는 영향)

  • This study was carried out to investigate the effects of nutrient solution concentration, irrigation frequency on growth, flowering, and cut flower longevity of Dendranthema grandiflorum (Ramat.) Kitamura 'Shuhouno-chikara' hydroponically grown in perlite media. Not only stem length but also mineral contents of each plant organ in nutrient solution culture were much higher than those in soil culture. 1/2S of nutrient solution was good at early stage, but 1S of nutrient solution was better as chrysanthemum growth progressed. Among different concentrations of nutrient solution, mineral contents of each plant part showed no significant difference. 1S treatment of nutrient solution increased the vase life by 3 days than 2S treatment. The growth and mineral contents of each plant organ were great in plants irrigated 8 times a day, because of high moisture contents of medium and high water availability, followed by more frequent nutrient replenishment near roots. There was no correlation between nutrient solution concentration and vase life of cut flower grown in nutrient solution culture. Cut flowers irrigated twice per day had the longer vase life than other treatments (4, 8 times).

Antioxidant and Antibacterial Activity of Commercially Available Herbs in Korean Markets (국내 시판되는 허브류의 항산화 및 항균효과 검색)

  • In this study, we investigated the antioxidant and antibacterial activity of methanol extracts from 6 plants which were Chrysanthemum zawadskii Herb. var. latilobum (Maxim.) Kitamura (Gu-jeol-cho), Lavandula spica L. (Lavender), Rosmainus offcinals L. (Rosemary), Cymbopogon citrates (Lemongrass), Saussureae radix (Mok-hyang), Calendular officinalis L. (Calendular). Antioxidative effects of herbal extracts were measured by polyphenols, flavonoids contents and DPPH radical scavenging activity assay. We also evaluated the antibacterial activity against Staphylococcus aureus and Escherichia coli O157:H7. Methanol extracts from Gu-jeol-cho, lavender, rosemary and lemongrass showed high polyphenols contents as well as strong DPPH scavenging activity. In particular, rosemary extract contained highest polyphenol levels as $126.69{\pm}2.62{\mu}g/mg$ compared to other herbs. As for DPPH radical scavenging activities, $IC_{50}$ values of rosemary extracts were $6.23{\pm}0.58{\mu}g/mL$. The rosemary extracts also showed higher antibacterial effects against S. aureus and E. coli O157:H7. These results indicate that rosemary could be used as natural antioxidant and antibacterial agents.

Screening of immune enhancing activities in medicinal herbs, Compositae (국화과 약용 식물의 면역증진활성 검색)

  • The biological activities of immune modulating activities of the extracts from Echinacea purpurea, Chrysanthmum indicum L. and Circium japonicum var. ussuriense KITAMURA were compared. About 70% of the growth of human hepatocarcinoma and 80% of human gastric cancer cell was inhibited in adding 0.5mg/ml of the ethanol extracts of Echinacea purpurea, Chrysanthmum indicum L. and Circium japonicum var. ussuriense KITAMURA, respectively. The growth of human breast cancer cells was also inhibited in adding 0.5mg/ml of the extracts as well as 60% of the human cancer cells. It was proved that the growth of human normal lung cell, scored as 15% for the extracts. Overall selectivity of the extracts on several human cancer cell line was over 3, which is higher than those from the conventional herbs. The growth of both human immune B and T cells was enhanced up to 1.4 to 2.0 times by adding the extracts, compared to the controls. The secretion of tumor necrosis $factor-alpha(TNF-{\alpha})$ from T cell was also increased up to 94 pg/ml in adding the Echinacea purpurea ethanol extract (0.5mg/ml). Circium japonicum var. ethanol extract also increased up to about 96 pg/ml of interleukin-6(IL-6) from B cell.

Crown Gall of Chrosanthemum Caused by Agrobacterium rubi and A. tumefaciens (Agrobacterium rubi와 A. tumefaciens에 의한 국화 뿌리혹병)

  • Crown gall on lower stem and root of chrysanthemum(Dendranthema grandiflorum Kitamura) was observed at Hwasung and Gumi in 2001 and 2004, respectively. Tumors were semi-round with rough surface texture of dark brown color. Nine isolates inducing gall formation on lower stem of chrysanthemum among twenty isolates from the tumor tissues were characterized. Their colonies were convex, glistening, circular with an entire edge and whitish or tannish cream in color on potato dextrose agar supplemented with 0.5% $CaCO_3$. The virulent isolates were rod-shaped with peritrichous flagellae, gram negative, aerobic and growing on D1M agar. Among nine virulent isolates, one isolate was identified as Agrobacterium rubi and eight isolates were A. tumefaciens based on biochemical and physiological characteristics, fatty acid profiles and substrate utilization patterns. A. tumefaciens had strong pathogenicity and broad host range compared with A. rubi. This is the first report on crown gall of chrysanthemum in Korea. To our knowledge, crown gall of chrysanthemum caused by A. rubi is first report in this study worldwide.
