Genes expression monitoring using cDNA microarray: Protocol and Application

  • The major issue in the post genome sequencing era is determination of gene expression patterns in variety of biological systems. A microarray system is a powerful technology for analyzing the expression profile of thousands of genes at one experiment. In this study, we constructed cDNA microarray which carries 2,304 cDNAS derived from oligo-capped mouse cDNA library. Using this hand-made microarray we determined gene expression in various biological systems. To determine tissue specific genes, we compared Nine genes were highly-expressed in adult mouse brain compared to kidney, liver, and skeletal muscle. Tissue distribution analysis using DNA microarray extracted 9 genes that were predominantly expressed in the brain. A database search showed that five of the 9 genes, MBP, SC1, HiAT3, S100 protein-beta, and SNAP25, were previously known to be expressed at high level in the brain and in the nervous system. One gene was highly sequence similar to rat S-Rex-s/human NSP-C, suggesting that the gene is a mouse homologue. The remaining three genes did not match to known genes in the GenBank/EMBL database, indicating that these are novel genes highly-expressed in the brain. Our DNA microarray was also used to detect differentiation specific genes, hormone dependent genes, and transcription-factor-induced genes. We conclude that DNA microarray is an excellent tool for identifying differentially expressed genes.

한국산 장마(Dioscorea oppasita cv. Jang-Ma)에서 분리한 Chinese yam necrotic mosaic virus (Chinese yam necrotic mosaic virus Isolated from Chinese Yam in Korea)

  • 경상북도 안동, 의성, 군위, 대구의 장마 재배포장에서 모자이크와 괴저반점을 보이는 장마잎을 채집하였다. 이들 이병 잎 조직을 시료로 하여 D N 법 및 ISEM법에 의해 투과전자현미경으로 바이러스 입자를 관찰한 결과, 각각의 시료에서 ChYNMV 항혈청에 반응한 660nm의 사상형 입자가 확인되었다. 장마 잎에서 부분 정제한 바이러스에서 RNA를 추출하여 이것을 주형으로 ChYNMV 특이적 프라이머와 oligo-dT 프라이머를 이용하여 외피단백질 유전자와 3‘-말단 비전사부위를 포함하는 약 1.2kbp의 3’-말단을 증폭하였다. 외피단백질 아미노산서열은 Macluravirus로 알려진 ChYNMV (A B044386)와 97.9%의 상동성을 보여 장마에서 분리한 바이러스를 ChYNMV로 동정하였다. ChYNMV와 다른 Macluavirus의 외피 단백질 아미노산서열을 비교한 결과, M말단 영역에서 가장 많은 변이를 확인하였고 Macluravirus 속에는 잘 보존된 영역이 존재하였다.

Pyrococcus furiosus의 β-1,3-glucanase를 처리한 laminarin 분해 산물을 이용한 바이오 에탄올의 생산 (Application of β-1,3-Glucanase from Pyrococcus furiosus for Ethanol Production using Laminarin)

  • 갈조류 유래의 다당류인 laminarin을 기질로써 호열성 미생물인 Pyrococcus furiosus의 $\beta$-1,3-glucanase와 반응 시킨 뒤, 분해산물을 yeast를 이용한 알코올 발효과정을 통하여 에탄올을 생산하고자 하는 연구를 수행하였다. 33 kDa (297 a.a, 894 bp)의 재조합 $\beta$-1,3-glucanase를 대장균에게 발현 후 순수하게 정제 하였으며, 정제한 $\beta$-1,3-glucanase와 laminarin을 반응시킨 결과 단당을 포함하여 oligo당 형태로 분해됨을 TLC와 HPLC로써 확인하였다. 그리고 이러한 분해산물을 에탄올 생산 배지의 유일한 탄소원으로써 첨가하여 yeast를 배양한 결과 48시간뒤에는 세포 외로 최소 0.3%의 알코올을 생산함을 gas chromatography로써 확인하였다. 따라서 $\beta$-1,3-glucanase와 laminarin의 최적 분해반응 및 yeast의 최적 알코올 발효 조건을 확립한다면 본 연구의 방법을 이용한 해조류로부터의 bio-ethanol의 생산을 성공적으로 수행 할 수 있으리라고 판단된다.

Bacillus subtilis가 생산하는 비특이적 $\beta$-fructofuransoidase의 부분정제 및 특성 (Partial Purification and Properties of Non-specific $\beta$ -fructofuranosidase Produced by Bacillus subtilis)

  • Bacillus subtilis의 세포내 이눌라아제가 부분정제되고 그의 작용 모드와 일반적 특성이 조사되었다. 이 효소는 gel filtration에 의하여 분자량을 추정하였을 때 49,000이었고, 등진점은 5.2 이었다. 기질에 대한 친화성의 지표인 Km값은 설탕에 대해서는 10mM, 라피노오스에 대해서는 18mM 이었다. 이 효소는 산성쪽에서는 불안정한 단백질로서 pH6.6에서 최대 활성을 보였으며 최적온도는 10분간 반응시켰을 때 50'C였다. 이 효소의 작용모든는 이눌린같은 구조를 가지는 과당 중합체를 과당 끝부분으로부터 하나씩 잘라가는 exo-cleavage 형이었다.

올리고 부틸렌 테레프탈레이트와 6-아미노 카프론산으로부터 포리에스테르 아미드의 합성 (Preparation of Polyesteramide from Oligobutylene Terephtahlate and 6-amino Caproic Acid)

  • Polyesteramide(PEA) has been synthesized with different molar ratio of 6-aminocaproic acid(ACA) and oligo butylene terephthalate(OBT) prepared from Dimethyl terephthalate(DMT) and 1, 4-butanediol(BD) by the melt polymerization. Oligomer and polymer molecular structure were characterized by IR spectra, H'-NMR spectra. Also the thermal properties were examined by thermogravimetric analysis(TGA) and differential scanning calorimetry(DSC). DSC and TGA results exhibited that the$ T_m$ s were detectable in the vicinity of $170-220^{\circ}C$ and the Ta's were detectable in the vicinity of $380-390^{\circ}C$ . X-ray diffraction analysis results show that the synthesized polyesteramide has high degree of crystallinity.

멸치볶음 저작·연하 용이 노인식 개발 (Development of Easily Chewable and Swallowable Stir-fried Anchovy for Elderly)

  • Failure of the functions involved in ingestion leads to, not only loss of enjoyment of eating, but also protein-energy malnutrition. Dysmasesis and difficulty of swallowing occurs in various diseases, but aging is a major cause. In the aging society, the proportion of elderly people with dysmasesis and difficulty of swallowing is expected to increase rapidly. Developing foods for the elderly that are easy to chew and swallow is thus required. This study was conducted to develop easily chewable and swallowable foods for the elderly who can crush foods and ingest with their own tongues, but occasionally have difficulty in drinking fluids such as water and tea. Various foods for the elderly with chewing and swallowing difficulties were used for sensory assessment. The sensory panel consisted of 10 dietitians (10 women) in nursing care facilities. The sensory optimal composite recipes were determined by central composite design (CCD). The sensory measurements were significantly different in saltiness (p<0.05), sweetness (p<0.05), and overall quality (p<0.05). The optimum formulation of stir fried anchovy calculated by numerical and graphical methods was 3.74 g of soy sauces and 30.17 g of oligo-saccharides. Stir fried anchovy had a moisture content, hardness and adhesiveness of 76.52%, 2.10, and -1.57, respectively.

Hypolipidemic Properties of Fermented Capsicum and Its Product

  • This study was conducted to investigate the effects of fermented capsicum and a capsicum product on lipid metabolism. Fermented capsicum was prepared from red pepper puree tov three months. After 9() days of fermentation, capsaicin and dihydrocapsaicin concentrations were reduced from 24.7 and 14.7 g/mL to 15.5 and 6.45 g/mL, respectively. The capsicum product was prepared from the fermented capsicum mixed with prune extract, green tea extract, neroli extract and oligo-saccharide. Thirty-two male Sprague-Dawley rats were as- signed to four dietary groups (control, high-fat control (BE-control), high-fat-fermented capsicum (HF-S-1), high- fat-capsicum product (HF-S-2)). Plasma and hepatic lipid profiles were examined after three weeks of experimental diet. Food intakes were significantly lower in the HF-S-1 and HF-S-2 groups compared to the control group (p<0.05). The weight of perirenal fat pads was lowest in animals on the control diet (low-fat) and highest in high-fat control diet. The addition of fermented capsicum to high fat diets, HF-S-1 and HE-S-2 groups, resulted in significantly lower fat pad weights compared with the HF-control group. Both fermented capsicum (HF-S-1) and the capsicum product (HF-S-2) groups had lower plasma TG levels, atherogenic-index, and liver TG levels than the BE-control group (p <0.05). Liver TC levels were significantly lower in the HF-S-2 group than the HF-control group. The results demonstrate a hypolipidemic effect of fermented capsicum and the fermented cap-sicum product.

지방족 에스터-카보네이트 공중합체의 합성 (Synthesis of Aliphatic Ester-Carbonate Copolymer)

  • 생분해성 지방족폴리에스터인 poly(butylene succinate) (PBS)의 기계적 물성을 변화시키기 위해서 에스터 주쇄 구조에 카보네이트 구조를 도입시킨 에스터-카보네이트 공중합체를 합성하였다. 이 공중합체는 일단계에서 succinic acid와 1,4-butanediol(BD)을 에스터 반응시켜 oligo(butylene succinate)를 합성하였고, 이 단계에서 oligohexamethylenecarbonate biol(OHMCG)을 첨가하고 고진공하에서 축중합하여 제조하였다. 촉매로는 titanium(IV) isopropoxide(TIP)를 사용하였다. 합성된 공중합체의 구조는 FT-IR과 $^1H$-NMR을 이용하여 확인하였으며, 열적, 기계적 물성은 각각 DSC와 UTM을 이용하여 조사하였다. 고분자량의 공중합체를 얻을 수 있는 적정 촉매양은 succinic acid에 대해서 1wt%였다. Diol 혼합물의 몰비(BD:OHMCG)가 249:1~149:1일 때, 높은 점도를 갖는 공중합체를 얻을 수 있었다 또한, OHMCG의 함량이 증가함에 따라 공중합체의 용융점은 계속 감소하였다. 혼합물의 몰비가 149:1인 경우의 공중합체의 물성은 PBS에 비해 초기탄성율은 약간 감소하였지만 파단신장율은 2배 정도 증가되었다.

Synthesis and Characterization of Cationic and Anionic Cyclodextrin Oligomers and Their Use in Layer-by-Layer Film Formation

  • Ionically modified ${\beta}$-cyclodextrins, which have excellent water-solubility, have been interested in purification technology as well as drug carrier system. The present study summarizes the synthesis and characterization of cationic and anionic ${\beta}$-cyclodextrin (${\beta}$-CyD) products using by polycondensation. The oligo (${\beta}$-CyD)s are synthesized from ${\beta}$-CyD, epichlorohydrin (EP) and choline chloride (CC; for cationic polymer) or chloroacetic acid (CAA; for anionic polymer) through one step polycondenstaion process. Unlike the previous studies, we successfully purified the ionic ${\beta}$-CyD condensation products from the ${\beta}$-CyD reaction mixtures and accomplished a great level of structural analysis. The detailed structural analysis of these ionic ${\beta}$-CyD compounds is done by $^1H$ NMR, MALDI-TOF as well as GPC analysis and confirms the formation of oligomers with a few units of ${\beta}$-CyD. We found that the sequence of reactant addition also could effect on the molecular weight of the resulting product as well as the molar ratio of the reactants. Finally, we used the cationic and anionic ${\beta}$-CyD oligomers for fabricating multilayer films by layer-layer process.

  • The sodium ${\alpha}-sulfo$ fatty acid vinyl ester oligomers, which are oligomer type surfactants were prepared by polymerization with fatty acid vinyl acetate. The ${\alpha}-sulfonation$ of fatty acid vinyl ester oligomers were carried by direct addition of sulfur trioxide. The dispersing performance of oligomer type anionic surfactants and sodium dodecyl sulfate(SDS) in the aqueous suspension of iron oxide and titanium dioxide particles was evaluated by particle size distribution and zeta-potential measurement. As results, the particles of iron oxide and titanium dioxide were flocculated by addition of small amount of oligomer type anionic surfactants and sodium dodecyl sulfate(SDS), then the flocks redispersed by more addition of oligomer type anionic surfactants and SDS. The flocculation, redispersion process was observed in lower concentration range of oligomer type anionic surfactants than SDS. Especially, the dispersing action of sodium ${\alpha}-sulfo$ palmitic acid vinyl ester oligomer was better than sodium ${\alpha}-sulfo$ lauric acid vinyl ester oligomer.
