The difrrrenlial display reverse transcription polymerase chain reaction (DDRT-PCR) aniilysis roils performed to identify the pathogellir strain specific amplicons. mRNAs were purified from the trophozoites of the pathogenif strain YS-27 and the non-pathogenic strain S 16. respectively. Three kinds of rirsl stranded rDNAs were reverse transcribed from the mRNAs by one base anchored oligo-dT 11M (M: A. C, or G) primers. Each cDNA lemplatr was used for DDRT-PCK analysis. A total of 144 pathogenic strain specific amplicons was observed in DDRT-PCR analysis using primer combinations of the 11 arbitrary primers and the 3 one base anchored oli해-dT11M primers. Of these 31 amplit'tons were verified as the amplirons amplified only from the mRNAs of the pathogenic strain by DNA slots biol llybridizatioil. Furthel cklaracleization of the 31 pathogenic strain sprcifil amplicons by DNA slot blot hybridlnation analysis using biotin labeled Probes or the PCR amplified DNA of rysteine proteinase genes revealed that 21 of them were amplliried from the maNAs of the cysteine proteinase genes. Four randomly selected amplirons out of the rest 10 amplirons were used fur screening of cDNA library followed by immunoscreening and all of them were turned outs to be amplified from the mRNA.
Degradation of deoxyribonucleic acid(DNA) and ribonucleic acid(RNA) by cell-free extract of mixed rumen protozoa of buffalo rumen was investigated. DNA was observed to be degraded rapidly during an initial incubation period of 2 hr with simultaneous appearance of degradation products. RNA on the other hand recorded a rapid degradation during an initial incubation period of 1 hr. RNA degradation products appeared upto an incubation period of 2 hr. DNA was observed to degrade into oligo- and mononucleotides. pyrimidine nucleosides, purine nucleoside adenosine and bases xanthine, hypoxanthine and thymine. Degradation products of RNA comprised of pyrimidine nucleosides, purine nucleoside, adenosine and bases xanthine, hypoxanthine and uracil besides oligo- and mononucleotides.
Cross-linked network solid polymer electrolytes were prepared by means of in situ hydrosilylation between poly[hydromethylslioxane-g-oligo(ethylene oxide)] and diallyl or triallyl group-containing poly(ethylene glycols). The conductivities of the resulting polymer electrolytes were greatly enhanced upon the addition of poly(ethylene glycol) dimethyl ether (PEGDME) as an ion-conducting plasticizer. Conductivities of the cross-linked polymer electrolytes were more dependent on the molecular weight of PEGDME than on the cross-linkers. The maximum conductivity was found to be 5.6${\times}$10$\^$-4/ S/cm at 30$^{\circ}C$ for the sample containing 75 wt% of PEGDME (M$\_$n/ =400). These electrolytes exhibited electrochemical stability up to 4.5 V against the lithium reference electrode. We observed reversible electrochemical plating/stripping of lithium on the nickel electrode.
The functional properties of novel sugar derivative sweeteners, fructo-oligosaccharide, maltitol, sorbitol and high maltose syrup(HMS) were examined for their humactant effect, lactic acid bacterial growth, Streptococcus mutants growth and relative sweetness compared to sucrose. Sorbitol exhibited remarkably high water activity reducing capacity, whereas fructo-oligosaccharide and maltitol showed the same level as sucrose. Maltitol showed distinct anti-bacterial(bacteriocidic) effect against Stc. mutants and most of lactic acid bacteria tested except for L. plantarum. The molar basis relative sweetness of sugar derivatives in comparison with 1%(w/w) level of sucrose were 0.69 for Neosugar(fructo-oligosaccharide), 0.21 for sorbitol, 0.50 for maltitol and 0.27 for HMS.
Rhamnose-rich (RROP-s) and fucose-rich (FROP-s) oligo-and polysaccharides were prepared and extensively characterised by physical and chemical procedures [1,2] and compared to L-fucose. Their biological properties were then studied on human skin fibroblast cell cultures, human skin explant cultures and on hairless rat skin, using a variety of cell-biological, biochemical and computerised morphometrical procedures. Among the most important properties we could establish, the following are of particular interest for the tretment and prevention of age-dependent modifications of human skin (loss of skin-tissue, cells and matrix, wrinkle formation and others) : stimulation of cell proliferation (by $^3$[H]-thymidine incorporation and the MTT test), scavenging of reactive oxygen species (ROS) using several different procedures, and protease (MMP-2 and MMP-9) down-regulation. A topical preparation, using RROP-s and FROP-s, and/or L-fucose, was shown to increase cell proliferation, dermal matrix synthesis, efficient scavenging of ROS-s and to increase also the thickness of dermal tissue when applied for 4 weeks on hairless rat skin, accompanied by the densification of collagen bundles as well as by an increase of elastin synthesis. Using fluorescent labeled FROPs, it could be shown that these oligosaccharides react with cell-membrane receptors and especially with the elastin-laminin-receptor and the fucose-mannose receptor, but they penetrate also in the cell nucleus, suggesting the possibility of a direct action on the regulation of gene expression. When applied to the human skin of a team of voluntary women encompassing all age-groups, the efficiency of FROP-containing preparation could be confirmed using indentometry and computerised evaluation of skin micro-relief, as well as evaluation of periorbital wrinkles. It appears therefore that these preparations correspond to all the requirements of active anti-aging principles.

RAPD analyses were performed to investigate genetic relationships and useful molecular maker for 3 species and their 17 regional populations of the Pueraria lobata and related taxa. The length of amplified DNA fragments ranged from 200 to, 2,800 bp. Two hundred and eight scorable polymorphic makers and three scorable monomorphic makers were found from the PCR reactions with 15 random oligo primers, and those were analyzed by Nei's genetic distance coefficient. Based on the UPGMA phenogram from RAPD analyses, two major groups (9 populations from Korea; 3 populations from foreign countries) were recognized. And it showed distinct genetic differences from related taxa. The RAPD results was very useful to define the samples by geographical distribution and to discuss the relationships among the populations and their related taxa of the Pueraria lobata.

The purpose of this study is to assess naturalness of each and use type for Daegu and provide the basic data for urban green space planning. The results are as follows: 1) As a result of periodic analysis, It is known that in the 1400´s that the rate of a-/oligo-/meso-hemeroby was over 90% of that area, in which naturalness was very high. By the 2020´s, the population will increase to about 3,500,000 and the occupation rae of meat-and poly-hemeroby, where naturalness is very low, is expected to increase rapidly. 2) The recent land uses of Daegu are divided ito 17 types. In the survey of a occupation rate of each land use type, a forest are occupies 48.8% of the entire area. The military installation zone is lowest with 0.07%. 3) Investigation of a dominant hemeroby grade in each land use type showed that the urban complex area in characterized by meta-hemeroby : and the space for pavement and building construction where naturalness is lowest. On the other hand, oligo-hemeroby that is worth being preserved was surveyed to be dominant in woodlands and natural forests. 4) In the analysis of hemeroby mapping in the process of a green space planning, it is clear that hemeroby will be used a s a tool that assesses the suitability for recreation activity of each sample case. For example, it enables a poly-hemeroby region to be utilized as a near-natural recreation area including $\beta$-and $\alpha$-euhemeroby region in Daegu. Besides, hemeroby can provide basic data to make a value judgement of each sample case with a view to preservation of nature.
For the olefin recovery from polyolefin off-gas, the permeation behaviors of olefins and nitrogen were investigated through three kinds of PDMS membranes - cross-linked PDMS membranes, a polysiloxaneimide membrane, and oligo-PDMS modified PDMS membranes. Their pure gas permeabilities were measured as a function of operation temperature(-20 to $50^{\circ}C$) and pressure(1 to 25 atm) with ethylene($C_2\;H_4$), propylene($C_3\;H_6$), butylene($C_4\;H_8$), and nitrogen($N_2$) gases. The permeabilities of olefins and nitrogen highly depended upon the nature of PDMS membranes. Among these membranes, cross-linked PDMS membranes showed stable and high olefin/nitrogen selectivities over a wide operation pressure range and further study in various test conditions. Their permeability of olefin and nitrogen were governed by the condensation temperature(solubility selectivity) and plasticization, not the order of the size(diffusivity selectivity) of gases, which matched well with the general permeation behavior of rubbery polymeric membranes for condensable and non-condensable gases. With increasing feed pressure or decreasing feed temperature, the permeabilities of more condensible olefins increase highly, presumably due to high solubility and plasticization, but that of non-condensible nitrogen decreases slightly and thus, the selectivities of olefin/nitrogen increase highly.

In the field of designing of nano-fillers of polyimide nanocomposites, the two strategic points are the heat-resistance and compatibility with polyimide, a matrix polymer. In this study, we designed oligo(amic acid) having alkyl side chains and terminal amine groups to satisfy previous requirements and studied the modification of surface of layered silicates. Oligo(amic acid)s were prepared by the reaction of diamine monomers and PMDA and their molecular weight was controlled in about 2000g/mol. After that, acidification and ion exchange reaction led to the high-temperature organophilic layered silicate (OLS). XRD pauerns of OLS showed the more increased gallery spacing by $4{\AA}$ than that of the pristine layered silicate and the initial decomposition temperatures of OLS were in above $280^{\circ}C$. The polyimide nanocomposite films based on heat resistant OLS showed that the OLSs were well dispersed through the matrix and their CTEs showed a decrease of $26\%$ compared with pristine polyimide films.
