Detection of Avidin Based on Rugate-structured Porous Silicon Interferometer

  • Biosensor based on rugate PSi interferometer for the detection of avidin has been described. Rugate PSi fabricated by applying a computer-generated pseudo-sinusoidal current waveform has been prepared for the application as a label-free biosensor based on porous silicon interferometer. The fabrication, optical characterization, and surface derivatization of a rugate PSi has been described. The method to fabricate biotinderivatized rugate PSi has been investigated. The surface and cross sectional morphology of rugate PSi are obtained with SEM. FT-IR spectroscopy is used to characterize the oxidation and functionalization reaction of rugate PSi sample. Binding of the avidin into the biotin-derivatized rugate PSi induces a change in refractive index. A red-shift of reflectivity by 18 nm in the reflectivity spectrum is observed, when the biotin-modified rugate PSi was exposed to a flow of avidin.

Design of Rugate Filters of Inhomogeneous Refractive Index Using the Fourier transform (Fourier 변환을 이용한 불균일 굴절률 Rugate 필터의 설계)

  • Rugate filters of inhomogeneous refractive index were designed using the Fourier transform and the effect of reflectance, stop bandwidth, optical thickness, and Q function on the rugate filter was investigated. An iterative correction process using a merit function was employed to fit an initial design to the target spectrum. Three Q functions derived by Sossi, Bovard, and Fabricius, respectively, were compared in terms of the number of iteration, merit function, and optimum optical thickness. The result shows that after a number of iterations the Q functions by Bovard and Fabricius produce high rejection rugate filters closer to the target spectrum than the Sossi's Q function and the optimal optical thickness is determined by the stop-band width of the rugate filter. ilter.

Well Defined One-Dimensional Photonic Crystal Templated by Rugate Porous Silicon

  • Well defined 1-dimentional (1-D) photonic crystals of polystyrene replicas have been successfully obtained by removing the porous silicon from the free-standing rugate porous silicon/phenylmethylpolysiloxane composite film. Rugate porous silicon was prepared by an electrochemical etching of silicon wafer in HF/ethanol mixture solution. Exfoliated rugate porous silicon was obtained by an electropolishing condition. A composite of rugate porous silicon/phenylmethylpolysiloxane composite film was prepared by casting a toluene solution of phenylmethylpolysiloxane onto the top of rugate porous silicon film. After the removal of the template by chemical dissolution, the phenylmethylpolysiloxane castings replicate the photonic features and the nanostructure of the master. The photonic phenylmethylpolysiloxane replicas are robust and flexible in ambient condition and exhibit an excellent reflectivity in their reflective spectra. The photonic band gaps of replicas are narrower than that of typical semiconductor quantum dots.

Smart Particles Containing Multiple Rugate-structured Photonic Crystal

  • The rugate porous silicons containing multiple photonic band gaps have been generated by applying a composite waveform summed three computer-generated pseudo-sinusoidal current waveforms and exhibit three sharp photonic band gaps in the optical reflectivity spectrum. Generated multiple rugate porous silicons display three rugate peaks corresponding to the each of the sine components varied from 0.42, 0.36, and 0.30 Hz, with a spacing of 0.06 Hz between each sine component. The resulting rugate PSi films have been removed from the silicon substrate by applying an lift-off current and are then made into particles by ultrasono-method in a organic solution. The sensing experiments using these particles for organic solvents such as toluene, hexane, acetone, and methanol have been achieved. Condensing of organic vapors in the pores increases the refractive indices of entire particle which results a red shift in the photonic peaks.

  • Photonic crystals containing multiple rugate structure are prepared by electrochemical etchings. Typically etched rugate PSi prepared in this study. Etching is carried out in a Teflon cell by using a two-electrode configuration with a Pt mesh counter electrode. They exhibit sharp photonic band gaps in the optical reflectivity spectrum. This reflectivity can be tuned to appear anywhere in the visible to near-infrared spectral range, depending on the programmed etch waveform. We study the method of full width half maxima and reflectivity index control by using amplitude.

Simultaneous Detection Properties of Organic Vapor, Pressure Difference and Magnetic Field using a Rugate-structured Free-standing Porous Silicon Film (Rugate 구조를 갖는 자립형 다공성 실리콘 박막을 이용한 유기 증기, 압력차, 자기장의 동시 감응 특성)

  • In this study, we investigated the simultaneous detection properties of organic vapor, pressure difference, and magnetic field using a single rugate-structured free-standing porous silicon (RFPS) thin film. Both the wavelength and the intensity of the rugate peaks were changed in the reflectivity spectrum measured at the thin film surface while the organic vapor was exposed to the RFPS thin film. However, when the pressure difference and the magnetic field were exposed to the film, only the rugate peak intensity was changed. Therefore, it is possible to distinguish whether or not the organic vapor is detected by simultaneously changing the rugate peak wavelength and intensity. In addition, a method of distinguishing between the pressure difference and the magnetic field detection signal has been derived by rapidly modulating the direction of the magnetic field. This study shows that it is possible to simultaneously detect and distinguish various objects using a single RFPS thin film, and it is found that porous silicon can be utilized as a sensor sufficiently.

Fabrication and Optical Characterization of Rugate-structured Polymer Replicas

  • Photonic crystals containing rugate structure result in a mirror with high reflectivity in a specific narrow spectral region and are prepared by applying a computer-generated pseudo-sinusoidal current waveform. Well defined 1-dimentional photonic polymer replicas showing a reflectivity at 534 nm have been successfully obtained by the removal of rugate porous silicon (PSi) template from the polystyrene composite film. XRD measurement indicates that the oxidized rugate PSi has been completely removed from the composite films. Polymer replicas exhibit a sharp resonance in the reflectivity spectrum. Optical characteristics of photonic polymer replicas indicate that the surface of polymer film has a negative structure of rugate PSi. These replicas are stable in aqueous solutions for several days without any degradation. The methods have been provided for the construction of photonic structures with polymers.

Fabrication and Characterization of Silole and Biotin-functionalized Rugate Porous Silicon

  • Multi-functionalized rugate porous silicon (PSi) for biosensor was developed by hydrosilylation with silole and its further reaction with biotin groups. PSi was generated by an electrochemical etching of silicon wafer in aqueous ethanolic HF solution PSi prepared by using etching conditions showed that many sharp spectral lines can be obtained in the optical reflectivity spectrum. 1,1-hydrovinyl-2,3,4,5-tetraphenylsilole was obtained from the reaction of 1,1-dilithio-2,3,4,5-tetraphenyl-1,3-butadiene with dichlorovinylsilane. Multi-functionalized PSi with silole and biotin groups was characterized by UV-vis absorption spectroscopy, Ocean optics 2000 spectrometer, and fluorescence spectroscopy. Optical characteristics such as reflectivity and photoluminescence (PL) were observed. An increase of the reflection wavelength in the reflectivity spectrum by 20 nm was observed, indicative of a change in refractive indices induced by hydrosilylation of the silole and biotin groups to the rugate PSi. This red-shift was attributed to the replacement of some of the Si-H group of fresh rugate PSi with silole and biotin group.

  • The differences of properties for both single-layered and multi-layered porous silicon were investigated. Multistructured porous silicons such as DBR or rugate porous silicon exhibit strong reflection resonances providing the reflection of a specific wavelength in the optical reflectivity spectrum. DBR PSi displays a square varying porosity gradient in the direction perpendicular to the plane of the filter but a sinusoidally varying porosity gradient was obtained for rugate PSi.

  • Multiple rugate structures can be etched on a silicon wafer and placed in the same physical location, showing that many sharp spectral lines can be obtained in the optical reflectivity spectrum. Porous silicon samples were prepared by electrochemical etch of heavily doped p-type silicon wafers. The etching solution consisted of a 3:1 volume mixture of aqueous 48% hydrofluoric acid and absolute ethanol. Galvanostatic etch was carried out in a Teflon cell by using a two-electrode configuration with a Pt mesh counterelectrode. A sinusoidal current density waveform varying between 51.5 and $74.6mA/cm^2$ is applied. The anodization current was supplied by a Keithley 2420 high-precision constant current source which is controlled by a computer to allow the formation of PSi multilayer.
