To make sure of the usefulness of extract of Rubus coreanus Miquel for producing functional feeding honey, we compared some antioxidative indicators of feeding honey using extract of Rubus coreanus Miquel with acacia honey, SueBee Clover honey(USA), feeding honey on the market. The water content of four honeys were 16.6~26.5%, pH were 3.18~3.70, and titratable acidity ranged 0.018~0.022%. The phenolic compound contents of SueBee Clover honey and feeding honey using extract of Rubus coreanus Miquel were 8.3 mg/100 g and 7.3 mg/100 g, respectively, and were significaltly higher thgheacacia honey and feeding honey on the market. The flavonoid contents per 100 g of honey ranged from 2.3 mg(acacia honey) to 15.0 mg(SueBee Clover honey). DPPH anion scavenging activity of four honeys were not high. 0.5~2.0 g/ml of feeding honey using extract of Rubus coreanus Miquel was 16~36% and showed a concentration-reliant figure. At the concentration of 0.25~0.75 mg/ml, the reducing power of four honeys increased concentration-dependently, and the power of 0.25 mg/ml of feeding honey using extract of Rubus coreanus Miquel was corresponding to thgt of $150{\mu}g$/ml of a vitamin C solution. ABTS radical scavenging activity of feeding honey using extract of Rubus coreanus Miquel was 43.3~68.4%; the highest activity amongst all samples. When plotting the dose-response curve, ABTS radical scavenging activity also increased as their concentration increased from 62.5 mg/ml to 500 mg/ml. The heat treatment reduced the phenolic compound contents of acacia honey and feeding honey using extract of Rubus coreanus DPPH anion scavenging activity of feeding honey on the market and feeding honey using extract of Rubus coreanus Miquel did not changed significaltly after heating for 20 min, and the three honey except SueBee Clover honey maintained the reducing power with the same treatment.
Yukwa samples, made with additions of 0, 10, 20, and 30% Rubus coreanus Miquel extract, were puffed by different puffing methods(convection oven, microwave oven, and fryer) and were then examined for quality characteristics such as moisture content, expansion rate, color, hardness, and sensory qualities, in order to determine the optimal ratio of Rubus coreanus Miquel extract in the formulation and the optimal expansion of Yukwa. The moisture contents of Yukwa base increased as the level of Rubus coreanus Miquel extract increased. And the Yukwa puffed by convection had higher moisture than the samples puffed by microwave oven or frying, with the exception of the sample containing 20% Rubus coreanus Miquel extract. When comparing the expansion rates of samples, no significant differences were found between the control and extract-containing samples puffed by convection, microwave, and frying methods, respectively. However, the sample puffed by frying had the highest expansion rate, whereas the convection-, microwave-puffed samples showed no significant differences. In the convection-, microwave-, and fryer-puffed samples, lightness and yellowness decreased, but redness increased, as the level of Rubus coreanus Miquel extract increased. And the sample puffed by microwave oven had greater lightness as compared to the samples puffed by convection and frying. In terms of hardness, no significant differences were found the control and extract-containing samples puffed by convection and microwave methods. However, hardness was maximal in the sample puffed by convection and lowest in the sample puffed by frying, in which it increased according to the addition of Rubus coreanus Miquel extract. In the consumer acceptance evaluations and characteristics intensity rating tests, the samples puffed using convection and microwave methods showed higher scores for the majority of evaluated characteristics as compared to the samples puffed by frying. And the samples containing Rubus coreanus Miquel extract obtained fairly good scores. In conclusion, the results indicate that additions of 10$\sim$20% Rubus coreanus Miquel extract are optimal for Yukwa that is puffed by convection and microwave methods, as this range provides good physiological properties and reasonably high overall consumer acceptability.
For the purpose of application to new natural dyestuff, the dyeability of Rubus coreanus Miquel extract was analyzed. It was dyed in silk fabric according to various dye concentration, dyeing temperature, dyeing time, and dyeing repetition. And the effects of mordanting conditions were examined as color differences and color fastnesses. The maximum absorbance of Rubus coreanus Miquel extract was at 521 nm. The dyeaffinities were increased gradually as were increased dye concentration, dyeing temperature, dyeing time, and dyeing repetition. The dyeaffinity was increased at pre-mordanted condition, and color difference was increased distinctly at post mordanted condition. The hues of mordanted silk fabrics were RP, P, PB, BG where as non mordanted silk fabric was R. Lightfastness was the best at pre-Sn mordanted and the worst at Post-Al mordanted. Washing and perspiration fastnesses were improved on pre and post-Sn mordanted.
The purpose of this study was to verify the effect of dietary treatment of Hovenia dulcis thunb extract and Rubus coreanus Miquel extract combined with full-body nerve massage on fatigue recovery. The results are as follows. Homogeneity between groups was secured due to the general characteristics of the study subjects, and the abdominal group showed a significant decrease in fatigue and pain changes after full nerve massage. Changes in fatigue substances in the blood showed a significant decrease in the Hovenia dulcis thunb extract group, and changes in blood levels before and after the stress hormone experiment showed a significant decrease in cordicol in the Hovenia dulcis thunb extract group. Through the results of this study, we were able to confirm that oxidative stress was reduced in adult women in their 20s to 50s through a full-body nerve massage after eating Rubus coreanus Miquel extract and Hovenia dulcis thunb extract, using an objective and systematic experimental method for changes in oxidative stress.
천연에서 유래한 항산화 및 항용혈 소재개발을 위해 복분자 에탄올 추출물의 생리활성 효능 평가를 실시하였다. 복분자를 에탄올 추출한 후, 획득한 추출물을 섬유아세포주인 HS68에 처리하여 DPPH 라디칼 소거능과 SOD 유사 항산화 효능을 평가하였다. 또한 Sprague Dawley 렛트 적혈구 세포에 과산화수소로 산화를 유도한 후 추출물의 항용혈 효능을 살펴보았다. 그 결과 추출물은 농도 의존적으로 DPPH 라디칼 소거능과 SOD 유사 활성이 증가하는 것으로 나타났다. 렛트 적혈구를 이용한 항용혈 평가에서는 과산화수소를 단독으로 처리한 양성대조군보다 추출물을 처리한 군에서 항용혈율이 농도 의존적으로 유의하게 증가하였다. 이들 결과는 복분자 추출물이 항산화효능과 항용혈 약용 식물 소재로서의 가능성을 내는 것으로 사료된다.
통상적으로 식물의 열매, 잎, 뿌리 등의 추출물이 가지는 다양한 효능이 밝혀짐에 따라 점차 천연소재에 대한 관심이 증가하고 있는 추세이다. 본 연구에서는 복분자 딸기로부터 얻은 4종(미숙과, 완숙과, 잎/줄기, 뿌리의 에탄을 추출물)의 각기 다른 추출물에 대한 항산화, 미백, 항주름 효과를 비교하였다. 복분자 딸기의 항산화 및 항염 효과에 대한 몇몇의 보고가 있지만 효능에 대한 연구는 충분하지 않다. 따라서 복분자 추출물의 화장품적 효능 효과를 비교한 결과, 미백효과 및 주름개선효과에 있어, 미숙과부위의 추출물이 다른 부위의 추출물에 비하여 효과가 높은 것으로 관찰되었다. 이 결과를 바탕으로 복분자 딸기의 미숙과는 완숙과나 잎/줄기, 뿌리보다 화장품 원료로 더 높은 응용 가능성을 가지고 있음을 알 수 있었다.
The potential biological activities of water and ethanol extracts from two varieties of Rubus coreanus Miquel fruits produced in the Gochang-gun (GR) and Hoengseong-gun (HR) regions of Korea were examined. The hydroxyl radical ($\cdot$OH) scavenging activity, reducing power, lipid peroxidation inhibitory activity, and antiproliferative activity on cancer cells by these extracts were examined, and $\alpha$-amylase and $\alpha$-glucosidase inhibition assays were also performed. The EtOH extract from GR showed high hydroxyl radical scavenging activity ($EC_{50}=119.47{\pm}5.13\;{\mu}g/mL$), lipid peroxidation inhibitory activity ($EC_{50}=213.45{\pm}3.14\;{\mu}g/mL$) and a concentration dependence, with OD values ranging from 0.15 to 0.56 (50 to 200 ${\mu}g/mL$), for reducing power. The EtOH extract from GR has the highest antiproliferative activities on MDA-MB-231 and HepG2 cancer cells among four extracts. Meanwhile, all extracts showed certain $\alpha$-amylase and $\alpha$-glucosidase inhibition activities. These results indicate that extracts from two varieties of R. coreanus fruits have significant antioxidant, anti-diabetic and anti-tumorigenic activities, which suggest these extracts could be a potential source for pharmaceutical.
Vascular endothelial growth factor (VEGF) is a key factor involved in the induction of angiogenesis and has become an attractive target for anti-angiogenesis therapies. The purpose of this study was to elucidate the anti-angiogenic activity of Rubus coreanus Miquel water extract (RCME). Rubus coreanus Miquel has long been employed as a traditional medicine, and recent studies have demonstrated that it has measureable biological activities. Thus, we investigated for the first time the effect of RCME on angiogenesis and its underlying signaling pathways. The effects of RCME were tested on in vitro models of angiogenesis, namely, proliferation, migration, invasion and tube formation of human umbilical vein endothelial cells as well as an ex vivo model of vessel sprouting from the rat aorta in response to VEGF. We observed that VEGF-induced angiogenesis was strongly suppressed by RCME treatment compared to that of the control group. Moreover, we found that RCME inhibited VEGF-induced activation of matrix metalloproteinases and phosphorylation of extracellular signal-regulated kinase and p38, and also effectively inhibited phosphorylation of VEGF receptor 2. These results indicated that RCME inhibits angiogenesis by suppressing phosphorylation of the VEGF receptor and may be useful for the treatment of angiogenesis-dependent diseases such as cancer and diabetic retinopathy.
위염과 위궤양의 일차적 발병 인자로 알려진 Helicobacter pylori의 생육을 억제하고 urease 산물인 암모니아의 축적에 의한 위점막 손상을 완화시킬 목적으로 복분자로부터 H. pylori urease저해물질을 분리정제하고 소재화와 관련된 일부 성질을 규명하였다. 양념 채소류, 차류, 죽류, 건강채소류등의 식용식물, 약용식물, 허브 및 해조류, 총 173종으로부터 극성도에 따라 계통추출한 수용성인 냉수추출물(Fr, 1), methanol 추출물(Fr.4), 열수추출물(Fr.5) 519점을 대상으로 H. pylori urease 저해활성을 검색하였다. 1차 및 2차 저해활성 검색 결과 복분자의 70% acetone추출물이 약 24%의 가장 높은 저해활성을 나타내었으며, 이 추출물을 ethyl acetate와 butanol을 사용하여 ethyl acetate/DW layer(RCE/RCWl)와 butanol/DW layer(RCB/RCW2)로 순차 분획한 후 활성획분인 RCW2 내의 활성 본체를 확인할 목적으로 periodate oxidation과 pronase digestion을 실시한 결과 펩타이드 또는 단백질성 물질로 판명되었다. 저해활성 물질은 DEAE-Toyopearl 650C, Butyl-Toyopearl 650M 및 Sephadex LH-20순의 column chromatography에 의 해 분리 정제되었다. 분리 urease 저해물질, RCW2-IIIc $\alpha$는 HPLC의 gel permeation chromatography에 의해 비교적 순도 높은 분자량 약 13 kDa의 단일 물질임이 확인되었다. 저해활성물질은 열에 안정성을 보이 는 내열성 단백질임을 알 수 있었고 위내 단백분해효소인 pepsin에도 가수분해 저항성을 나타냄으로써 기능성식품의 소재로 높은 소재화 적성을 보였다.
This study was aimed at investigating hepatic toxicity and immunomodulating effects of defatted methanol extracts of two kinds of medicinal plants, Rubus coreanus Miq. and Atractylodes japonica Koidz. in mice. Defatted methanol extracts of fruits of Rubus coreanus Miq. and rhizome of Atractylodes japonica Koidz. were added at the level of 0.5% or 5%(w/w) to cholesterol-supplemented AIN-76 diet. Each diet was fed to 8 ICR male mice for 30 days. Weight gain and food efficiency ratio of the mice fed 5.0% extract of Rubus coreanus Miq. were significantly lower than those of the mice fed 0.5% extract Relative liver weight and activity of plasma alanine aminotransfernse were significantly increased only in the mice fed 5% extract of Atractylodes japonica Koidz. compared with the others. Splenocyte proliferation was not significantly different between the groups fed 0.5% or 5.0% extract of Rubus coreanus Miq. However, splenocyte proliferation was significantly decreased in the mice fed 5.0% extract of Atractylodes japonica Koidz. compared with that in the mice fed 0.5% Production of interleukin-2 by splenocytes from the mice fed 0.5% extract of Atractylodes japonica Miq. was significantly higher than the control value and it became lower with 5.0% dietary level. Secretion of $interferon-\gamma$ was not significantly different among groups. In conclusion, the defatted methanol extract of Atractylodes japonica Koidz. was likely to exert immunomodulating effect at the level of 0.5% but it may exert adverse effects on immune and liver functions at the level of 5.0%.
