Potential prevention effects of Rubus occidentalis seed on UVB-induced MMP-1 production and procollagen degradation in CCD-986sk cells

  • UV exposure induces matrix metalloproteinases (MMPs) and extracellular matrix-degrading enzymes expression. We studied the protective effect of Rubus occidentalis seed against UVB-generated skin photoaging using human fibroblast cells (CCD-986sk). We used an ELISA kit to measure the supernatents of procollagen type I and MMP-1 in CCD-986sk cells after they were exposed to UVB irradiation. The CCD-986sk cells that were used with RC-E/E after the UVB irradiation caused higher levels of type I procollagen and lesser levels of MMP-1 compared with the control group. Furthermore, the RC-E/E treated group showed lesser MMP-1 levels and higher procollagen type I levels than the untreated counterpart. Therefore, it can be concluded that Rubus occidentalis seed can prevent from skin photoaging.

Effect of Unripened fruits and Ripened fruits of Rubus coreanus Miquel on Murine Peritoneal Macrophages (복분자 미숙과 및 성숙과가 생쥐의 복강 Macrophages에 미치는 영향)

  • The purpose of this research was to investigate the effects of unripened fruits and ripened fruits of Rubus coreanus Miquel on murine peritoneal macrophages. The 70% ethyl alcohol extracts (20 or 100 mg/kg) of unripened fruits (RCE-I) and of ripened fruits (RCE-II) were administered p.o. once a day for 7 days to mice. RCE-I and RCE-II decreased the phagocytic activity of murine peritoneal macrophages and the production of nitric oxide. Also, RCE-I and RCE-II increased the production of tumor necrosis factor- a from peritoneal macrophages. In general, the immuno-suppressive action of RCE-I on macrophages was more potent than those of RCE-II. These results suggest that the fruits of Rubus coreanus Miquel regulates the non-specific immune response via decrease of phagocytic activity and increase of production of tumor necrosis factor- a from murine peritoneal macrophages.

Internal Transcribed Spacer (ITS) Regions Reveals Phylogenic Relationships of Rubus Species Cultivated in Korea (Internal Transcribed Spacers (ITS)로 분석한 국내 Rubus 재배종의 계통학적 유연관계)

  • The bramble cultivated widely in South Korea, which is known as Bokbunja-ddal-gi, is regarded as having originated from Korean native Rubus coreanus. This study was carried out to obtain basic phylogenetic information on Korean cultivated bramble (KCB) by comparing the internal transcribed spacer (ITS) regions with those of R. coreanus, blackberry (R. lanciniatus), black (R. occidentalis) and red (R. idaeus) raspberry. Sequences of the ITS 1 suggest that some KCB accessions share a significant similarity with both R. occidentalis and R. coreanus in the ITS 1 region. The ITS 2 sequences of the three KCB accessions clustered more closely to those of two R. occidentalis accessions than to those of R. coreanus. These results suggest that there exist variations in the sequences of ITS among KCB accessions and KCB accessions are more closely related to black raspberry than R. coreanus in the ITS regions.

Evaluation of Anti-inflammatory activity of Fissistigma poilanei and Rubus laxus Focke

  • The aim of the study was to determine the anti-inflammatory activities of the plants with origin of China. The Fissistigma poilanei, which is a species of plant in the family Annonaceae and Rubus laxus Focke, which is a species of plant in the family Rosaceae from China were tested for anti-inflammatory activities. Samples were prepared using 95% ethanol using nitric oxide (NO) assay for assessing the anti-inflammatory activity. NO assay experiment showed that extracts of the Fissistigma poilanei give 28.3% increases in anti-inflammatory activity and extracts of Rubus laxus Focke give 57.1% decreases in anti-inflammatory activity, compared to control. To determine the cell toxicity, MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay was used. MTT assay experiment showed that Fissistigma poilanei and Rubus laxus Focke might have less toxicity of 60.5% and 62.3%, respectively, compared to control. Taken together, these experiments showed that Fissistigma poilanei extracts might have significantly higher anti-inflammatory activities and relatively lower toxicity, compared to control. This implies that this study might give a tool to develop a new potential anti-inflammatory therapeutic candidate.

Characteristics of Rubus coreanus Miq. Fruits at Different Ripening Stages (숙성에 따른 토종 복분자 딸기의 특성)

  • To develop a healthy functional food containing Rubus coreanus fruit (RCF) powder with different degrees of ripening, the composition, physicochemical properties, and antioxidant activity of native RCF during ripening were investigated and compared with Rubus occidentalis fruit (ROF). Ripened RCF was bright red in color, whereas ripened ROF was dull and dark red in color. The seeds of RCF were softer and smaller than those of ROF. Crude protein, crude lipid, and total sugar content of RCF powder increased with increased ripening time, whereas ash and total dietary fiber contents decreased with increased ripening time. Color differences increased with Increased ripening. Eighteen types of amino acids were analyzed from RCF, and glutamic acid had the highest content. RCF powder had the highest level of potassium and calcium and did not contain any hazardous metals. Mineral and organic acid contents decreased according to ripening.

Quality changes in Rubus coreanus Miquel during Frozen Storage (냉동 저장 중 복분자딸기(Rubus coreanus Miq.)의 품질 변화)

  • Ripened Rubus coreanus Miquel harvested at Gochang was stored for 8 months at $-20{\pm}2^{\circ}C$, and quality changes were investigated at intervals of 2-3 months. Soluble solid concentration increased slightly, but acidity fell during frozen storage. The lightness (L value) and red color index (a value) of ripened R. coreanus Miquel decreased during storage. The vitamin C and ellagic acid contents were 30.79 mg/100 g and $155.12\;{\mu}g/100\;mg$ mg before storage, respectively, but fell to 17.03 mg/100g and $140.11\;{\mu}g/100\;mg$ after 8 months of storage. However, the total anthocyanin content was 39.45 mg/100 g before storage, and this remained unchanged from 2 to 8 months of storage.

  • The purpose of this research was to investigate inhibitory activity of Sasang medicines on diabetes. Reduce 36 kind of medicines to powder, abstract add 100 times methanol. Examine inhibitory activity on α-amylase and α-glucosidase in vitro. Thirty six oriental herbs were screened for inhibitory activities against carbohydrate digesting enzymes in vitro. Methanol extract of Pleuropterus multiflorus Turez, Ephedra sinica, Moutan cortex Radicis, Magnolia obovata, Alpinia officinarum, Amomi Semen, Rubus coreanus, and Cinnamomi cortex inhibited yeast α-glucosidase activity and extract of Moutan cortex Radicis and Rubus coreanus inhibited porcine α-amylase activity by more 50%. Rubus coreanus showed the strongest inhibition against both α-glucosidase and α-amylase. Therefore, Rubus coreanus was the most effective medicice of 36 kind of medicines.

  • This study was carried out to evaluate growth, morphological characteristics, and sugar content among fifty six blackberry (Rubus fructicosus) genotypes derived from gamma-ray treatment (fifty two lines), MNU treatments (three lines), and cross breed R. fructicosus${\times}$R. parvifolius(one line). While 56 genotypes had similar stem diameter with origin variety, the most of mutants showed reduced leaf size. Also, they showed a wide range of variation in fruit size and one hundred fruit weight compared with origin variety. In fruit size and sugar content, two mutant lines showed large fruit sizes and ten mutant lines showed 20% higher sugar content than the original variety. In the correlation analysis, positive relationships were determined between sugar content and stem diameter, fruit length, leaf wide and fruit length/fruit wide ratio. However, leaf length/leaf wide ratio showed a negative correlation with sugar content. The results will be used as fundamental data for Rubus fructicosus breeding program.

  • A new quercetin 3-O-trisaccharide was isolated from the leaves of Rubus crataegifolius (Rosaceae). The structure of this compound was determined as quercetin $3-O-{\alpha}-L-arabinofuranosyl-(1{\rightarrow}2)-{\beta}-D-xylopyranosyl-(1{\rightarrow}2)-{\beta}-D-galactopyranoside$ on the basis of 2D-NMR spectroscopic data. This flavonol glycoside was isolated for the first time from a natural source.

  • Fruits of Rubus coreanum (Korean name: Bog-bun-ja) have been used in oriental traditional medicine as the remedies for impotence, pollution, premature ejaculation and frequency of urination etc. It is known to have phenolic compounds as an astringent. By means of chromatographic methods, four hydrolyzable tannins were isolated from the fruits of R. coreanum. The structures of these compounds were established as gallic acid, 2,3(S)-HHDP-D-glucopyranose, sanguiin H-4 and sanguiin H-6 on the basis of physicochemical and spectroscopic evidences.
