Comparison of blood electrolyte and biochemical parameters between single infections of rotavirus and Cryptosporidium parvum in diarrheic Hanwoo calves

  • Background: Neonatal calf diarrhea is a major problem in the cattle industry worldwide. Rotavirus and Cryptosporidium parvum are the primary causative agents, especially during the first three weeks of the calf's life. Objectives: This study investigated the differences in acid-base, electrolytes, and biochemical parameters of diarrheic calves with infection of either rotavirus or C. parvum. Methods: A total of 61 Korean native calves (≤ 20 days old) were divided into two groups based on rotavirus or C. parvum infections: rotavirus infection (n = 44) and C. parvum infection (n = 17). The calves with at a specific blood pH range (pH 6.92-7.25) were chosen for comparison. The acid-base, electrolyte, chemistry, and serum proteins were analyzed, Further, fecal examinations were performed. Results: Compared to C. parvum-infected calves, the rotavirus-infected calves showed lower levels of total carbon dioxide, bicarbonate (HCO3-), anion gap, total protein, and albumin/globulin ratio, and significantly lower levels of potassium, globulin, and α2-globulin (p < 0.05). The C. parvum-infected calves (r = 0.749) had stronger correlations between pH and HCO3- than the rotavirus-infected calves (r = 0.598). Compared to rotavirus-infected calves, strong correlations between globulin and α2-globulin, α2-globulin and haptoglobin were identified in C. parvum-infected calves. Conclusions: This study is the first to investigate acid-base, electrolyte, and biochemical parameters in calves in response to infections of rotavirus and C. parvum. Although rotavirus and C. parvum cause malabsorptive and secretory diarrhea in similar-aged calves, blood parameters were different. This would help establish the diagnostic and treatment strategies.

  • A total of 420 chicken sera from various regions were tested for the presence of antibodies to avian rotavirus using indirect immunofluorescence assay (IFA). In broiler farms, rotavirus antibodies were detected from 20 farms among 30 farms tested and the positive rates were above 50% in 9 farms. In parent stock farms, rotavirus antibodies were detected from 5 farms among 14 farms tested. From sera collected in 7 layer farms rotavirus antibodies were not detected.

  • Purpose : Acute diarrhea in one of the most important diseases in children with high morbidity and mortality worldwide. Most of acute diarrhea is induced by viruses. Rotavirus and adenovirus are leading causes of severe gastroenteritis among infants and young children worldwide. Studies for adenovirus gastroenteritis in Korea are limited. We studied the prevalence of rotavirus and adenovirus gastroenteritis from March 1998 to June 1999 in Chungju area. Methods : Stool samples were collected from 143 children with acute diarrhea. Specimens were tested for group A rotavirus antigen and for adenovirus type 40 and 41 by using available commercial kits. Results : Among 143 samples, 37% were positive for rotavirus and 16% were positive for adenovirus. Rotavirus was most prevalent from January to March, 1999 and adenovirus was prevalent during September 1999. The greatest number of rotavirus infections occurred under 24 months of age, followed by 2~4 years of age. Adenovirus was most common in 2~24 months of age. Conclusion : Rotavirus was most prevalent in winter and early spring. In our study, rotavirus was prevalent in early spring and adenovirus was in autumn.

Studies on the Infection Condition and Formaldehyde Inactivation of Human Rotavirus Wa (사람 로타바이러스 Wa의 감염 조건 및 formaldehyde에 의한 inactivation에 대한 연구)

  • Effects of protease, $CaCl_2$ EGTA, polybrene, medium pH, and formaldehyde on the infection and inactivation of human rotavirus Wa were investigated using T-flask culture of monkey kidney MA-104 cells. Rotavirus titer was improved by the addition of trypsin or clostripain. Rotavirus titer was increaeed 8 and 10 time sin the infection medium supplemented with 300 $\mu\textrm{g}$/mL of $CaCl_2$ and in the medium adjusted its pH to 8, respectively. However, addition of EGTA or polybrene to the medium decreased rotavirus titer. Rotavirus titer was reduced to 53-95% of the initial value at 1 hr after formaldehyde treatment. Furthermore, rotavirus was inactivated more than 98% at 12 hrs after formaldehyde treatment.

  • Purpose: This study aimed to investigate the changes in the occurrence of rotavirus gastroenteritis (RGE) after the introduction of rotavirus vaccine and estimate rotavirus vaccine effectiveness in hospitalized children. Methods: We compared the retrospective data of 671 patients with acute gastroenteritis (AGE) admitted to the Department of Pediatrics, Hanyang University Seoul Hospital from January 1, 2014, to December 31, 2015, with retrospective data of 1,243 patients admitted to the same institution with AGE from January 1, 2004, to December 31, 2005. The vaccine effectiveness was estimated using a case-positive control test-negative study. Results: The proportion of RGE in AGE was significantly lower in 2014 to 2015 (9.0%, 48/531) than in 2004 to 2005 (22.7%, 282/1,243) (P<0.001). In particular, there was a significant decrease in the 6- to 11-, 12- to 23-, and 24- to 35-month-old groups (P<0.001), whose rotavirus vaccination rates were higher than the remaining age groups. The monthly distribution of patients with RGE in 2004 to 2005 was higher from November to May, peaked in January, followed by December and February. In 2014 to 2015, the monthly distribution of patients with RGE slightly peaked in January. In 2014 to 2015 study, the complete rotavirus vaccination rate was 66.0% (332/503) and incomplete vaccination rate was 6.2% (31/503). Presumed rotavirus vaccine effectiveness was 83.3% (95% confidence interval [CI], 60.5% to 92.9%) in the complete vaccination group and 27.4% (95% CI, -163.7% to 80.0%) in the incomplete group. Conclusions: The proportion of RGE in AGE has markedly decreased since the introduction of rotavirus vaccines, and improving vaccination coverage will further reduce the burden of RGE in Korea.

  • Rotavirus is the major cause of gastroenteritis in children under the age of 5. Rotavirus infection may lead to several neurological complications as meningitis, encephalitis, convulsion, encephalopathy, hemorrhagic shock, central pontine myelinolysis, Guillain-Barre syndrome, and Reye's syndrome. Further, some reports have described diffuse cerebral white matter lesions on diffusion-weighted magnetic resonance imaging (MRI) in neonates with rotavirus induced seizures. Here, we report on three neonates with rotavirus induced seizures with cerebral white matter abnormalities on MRI.

Inhibitory Effect of Ginseng Polysaccharides on Rotavirus Infection

  • Polysaccharides and saponins were isolated from the root of Panax ginseng C.A. Meyer (Family Araliaceae), treated at various temperatures, and their inhibitory effects on rotavirus were investigated. As the temperature of processing increased, the molecular weight of the polysaccharides decreased, but the yields of water extracted increased. These polysaccharides inhibited rotavirus infection in MA104 cells, but there were no significant differences in rotavirus infection-inhibitory potency. However, ginseng saponins did not exhibit rotavirus infection-inhibitory activity.

  • Rotavirus is a most common etiologic viral agent of pediatric gastroenteritis. Most newborns infected by rotavirus show no specific symptoms, yet, some of neonates can develop serious complications such as intestinal perforation, necrotizing enterocolitis or even death. And rotavirus infection may cause only neurologic manifestation such as seizure without gastrointestinal manifestations in some neonates. We experienced a case of neonatal rotavirus infection with benign convulsion without gastrointestinal manifestations, and report this case with literature review as followings.

Effect of Betulinic Acid Isolated from Ziziphus jujuba Lam on Infection of Rotavirus in MA-104 Cell (대추나무 껍질 유래 Betulinic acid의 in vitro rotavirus 감염억제 효과)

  • This study was conducted to evaluate the inhibitory effects of betulinic acid isolated from Ziziphus jujuba on various human rotavirus, such as KU, S2 and YO. The results obtained are summarized as follows: At the concentration of betulinic acid 0.1/2%, all human rotavirus is showed a maximum effect on their growth even though it's evaluated on the in vitro test. The each inhibitory rate of MA-104 cells infected by human rotavirus KU, S2 and YO was 62.1%, 59.7% and 65.2%, respectively, at the concentration of 0.1/2%. The anti-virus activity of betulinic acid was showed as the dose-dependent manner at the used dosages except at the dose of 0.1%.
