Effect of Glycyrrhiza glabra Extracts on Immune Response (감초 추출물이 면역응답에 미치는 영향)

  • This study was conduced to investigate on immune response of the hot water extract(PHE), 50% methanol extract(PME) and acetone extract(PAE) from Glycyrrhiza glabra. The experiment was carried out by phagocytosis, plaque forming cell(PFC), hemalysin titration(HY) and rosette forming cell(RFC) assay by using BALB/c mice. The results obtained from this study are as follow ; The effects of Glycyrrhiza glabra extracts on phagocytosis was tended to be slight increase in GME and GAE groups compared to the control group, but not significant. In the experiment of PFC and HY, the results of experiment groups which was given each samples were significantly higher than the control group. The result of rosette forming cell in GME and GAE groups were significantly higher than control group.

Effect of Polyphenol Compounds from Korean Pear on Immunofunctional Activity (한국산 배의 Polyphenol 화합물군이 쥐의 면역기능에 미치는 영향)

  • This study was conducted to investigate immunofunctional activity of the polyphenol fractions isolated from Korean pear. In the experimental of Rosette forming cell, the results showed that all the polyphenol fractions enhance the cell count compared with the control group. Especially polyphenol fraction II and III showed highly significant effect on Rosette forming cell, and allergy inhibition. After antigen challenge, histamine content of blood of the polyphenol groups was lowered to near the normal group.

Immunopotentiating Effect of Polysaccharides Extracted from Agrocybe cylindracea (Agrocybe cylindracea로부터 추출한 다당류의 면역증강활성)

  • The immunopotentiating effects of the polysaccharides, both intracellular and extracellular, was examined by an animal feeding test. The results are summarized as follows. The oral administration of intracellular and extracellular polysaccharides of Agrocybe cylindracea for 10 days resulted in the enhanced phagocytic activity of peritoneal exudate cells(PEC), spleen cells(SC), and monolymphocytes(ML). In the experiment of PFC(plaque forming cell) and RFC(rosette forming cell), the results showed that all the polysaccharide fractions enhanced the immune related cells. The EAC II group(the extracellular polysaccharide of Agrocybe cylindracea 10 mg/0.2 ml distilled water/day/ mouse) increased the PFC and RFC by 46~50% and 43%, respectively, compared to the control group. On the other hand, the IAC I group(the intracellular polysaccharide of Agrocybe cylindracea 1 mg/0.2 ml distilled water/day/mouse) increased the PFC and RFC by 49~70% and 91%, respectively. In terms of the mitogenic activity, the extracellular polysaccharides of A. cylindracea showed a higher activity than the intracellular polysaccharides.

  • Effects of Allolobophora caliginosatrapezoides (Ac) polysaccharide fractions on the inflammation and hypersensitivity were studied in vivo. It showed that Ac polysaccharide fractions have the significant inhibitory activities of inflammation and hypersensitivity; They inhibited significantly the carrageenin-induced paw edema and acetic acid-induced writhing syndrome. They also inhibited significantly the Arthus reaction and delayed hypersensitivity in the sheep red blood cell-sensitized mice in accordance with the inhibition of haemaglutinin titer, haemolysin titer, plaque-forming cells and rosette-forming cells. They also improved markedly the oxazolone-induced dermatitis in rats dose-dependently. As the above results, it exhibited that Ac polysaccharide fraction inhibited not only humoral immune response, but also cell-mediated immune response. It seemed that methanol and ether extracts have also another physiological active agents.

Antitumor and Immunomodulator Effects of Hyangsapyungwisan in ICR-mice (향사평위산(香砂平胃散)이 항암(抗癌) 및 면역조절작용(免疫調節作用)에 미치는 영향(影響))

  • In order to investigate antitumor and immune response effect by Hyangsapyungwisan after Sarcoma-180 cells and methotrexate were treatred each other, the extract of Hyangsapyungwisan was orally administered to ICR mice for 14 days. To evaluate the effects of the Hyangsapyungwisan, 50% inhibition concentration($IC_{50}$), mean survival days, tumor weight for antitumor effects, hemagglutinin titer, hemolysin titer, rosette forming cells, natural killer cell activity and productivity of interleukin-2 for immune responses measured in ICR mice. The results were summarized as follows: 1. Mean survival time in Hyangsapyungwisan-treated group was slightly prolonged, as compared with control group(13.46%). 2. On the MTT assay, cell viability was significantly inhibited by $5{\mu}g/well,\;2.5{\mu}g/well,\;1.25{\mu}g/well,\;and\;0.625{\mu}g/well$ of Hyangsapyung-wisan concentration inhibited cell viability significantly. $IC_{50}$ for cell viability was $11.59{\mu}g/well$. 3. Tumor weight in Hyangsapyungwisan treated group was depressed, as compared with the control group(p<0.05). 4. Hemagglutinin titer in Hyangsapyungwisan-treated group was slightly increased with no significance, as compared with the control group. 5. Hemolysin titer in Hyangsapyungwisan-treated group was silightly increased, as compared with the control group(p<0.05). 6. Rosette forming cells in Hyangsapyungwisan-treated group was silightly increased, as compared with the control group(p<0.05). 7. Naural killer cell activity in Hyangsapyungwisan-treated group was significantly increased(p<0.05). 8. Production of interleukin-2 was significantly increased(p<0.05). According to the above results, Hyangsapygwisan had prominent antitumor effects, and enhance both cellular and humoral immunity in mice.

Enumeration of Korean native goat erythrocytes (KGRBC)-rosette forming cells in peripheral blood of Korean cattle (재래산양 적혈구를 이용한 한우 순환 혈액내 rosette 형성 세포 정량)

  • In order to enumerate the T-lymphocytes in bovine peripheral blood lymphocytes (PBL) by E rosette assay, KGRBC were treated with various concentrations of 2-aminoethylisothiouronium bromide(AET) and dextran(Dex), singly or in combination. To further standardize the assay, optimum concentration of AET- and/or Dex-treatment and incubation time for rosette forming cell(RFC) counts were determined. The levels of B-lymphocytes in the PBL were evaluated by erythzocyte-antibody($EA_{Fc}$)- and erythrocyte-antibody-complement (EAC)-rosetting techniques. The results obtained were as follows; The PBL from 20 clinically normal Korean cattle were formed as low percentage of spontaneous E-rosette ($6.7{\pm}2.4%$) in control group, whereas in KGRBC treated with 0.1M AET for 20 minutes and 8% Dex were formed as $37.3{\pm}2.7%$ and $45.1{\pm}2.1%$, respectively. And the synergistic effects were noted no less than $66.5{\pm}5.6%$ when the KGRBC treated with 0.1M AET and 8% Dex subsequently and rate of RFR did not change significantly between 3~24 hours incubation time at $4^{\circ}C$, EA-and EAC-RFR were $23.3{\pm}9.1%$ and $23.1{\pm}7.9%$, respectively. These results suggest that the KGRBC would be a useful agent for the enumeration of T-lymphocytes by E rosette assay and B-lymphocytes by EA- or EAC-rosette assay in cattle-PBL.

Effect of Corynebacterium parvum on Cellular and Humoral Immune Responses in Mice (Corynebacterium parvum이 마우스의 세포성(細胞性) 및 체액성(體液性) 면역반응(免疫反應)에 미치는 영향(影響))

  • The effect of subcutanecus injection of Corynebacterium parvum($700{\mu}g$) on cellular and humoral immune responses when given at various time relative to sheep red blood cell(SRBC) sensitization were studied by the evaluation of Arthus, delayed-type hypersensitivity(DTH), rosette forming cell, hemagglutinin and hemolysin reactions. Arthus reactivity(3 hours) developed in control mice and test mice pretreated with C. parvum 8 days prior to intravenous sensitization with SRBC were similar. However, there was slight depression of reactivity when C. parvum was given subcutaneoutly(s.c.) 4 or 2 days prior to SRBC sensitization. Arthus reactivity was significantly depressed when C. parvum was given s.c. either at the same time as, or 2 days later than, antigen. DTH reaction was net depressed significantly when C. parvum was injected 8 or 2 days prior to SRBC sensitization or at the same time as antigen. In contrast DTH was significantly augmented when C. parvum given s.c. 4 days prier to SRBC sensitization. DTH was depressed when C. parvum was given s.c. 2 days after antigen. No significant change occurred in rosette forming percetages of spleen cell when C. parvum was given s.c. 8, 4 or 2 days before SRBC sensitization. In contrast, a significant reduction in percentages of rosette forming cell occurred when C. parvum was given s.c. either at the same time as, or 2 days later than, antigen. Serum hemaggulutinin and hemolysin titers were not significantly affected by subcutaneous injection of C. parvum regardless of time relative to SRBC sensitization. However, mercaptoethanol-resistant hemaggulutinin and hemolysin(IgG) titers were somewhat augmented when C. parvum was given 2 days after antigen. It is concluded from these results that depending on the time and route of inoculation, C. parvum can enhance or depress immune responses in mice, suggesting the time and route of C. parvum inoculation is an important point of concern about clinical use of C. parvum for the treatment of cancer.

  • Recently pathotoxicological study of lymphoid organs by administration of some phthalate ester in rats, indicated marked effect of architechure of thymus, spleen, and lymphonodes. Dioctyl phthalate (DOP), one of the phthalate ester, caused statistically significant reduction in the weights of various lymphoid organs. This senstivity of the lymphoid organ to phthalate toxicity which could lead to adverse effects on the immune response and also suppression of immune system. Therfore it is possible the presense of di-(2-ethylhexyl) phthalate (DEHP), one of the phthalate ester as well as DOP, in spleen and other organs might have some moderately effect on the function of the immune system, So our present study was proceeded to assess the effect of DEHP on the immunotoxicity in mouse. In the immune response of DEHP administered mice, HA, HY, Arthus reaction and Rosette forming cell were decreased but DTH was increased. Furthermore, in the DEHP plus ethanol group, HA, HY, Arthus reaction and Rosette forming cell were remarkably decreased and elevation of DTH was inhibited.

  • Effects of beta-carotene on the immunobiological responses were studied in ICR mice. ICR male mice were divided into 8 groups (10 mice/group), and beta-carotene at doses of 4, 20 and 100 mg/kg were orally administered to ICR mice once daily for 28 consecutive days. Cyclophosphamide (CY) was injected intraperitoneally (i.p.) to ICR mice with a single dose of 5 mg/kg body weight at 2 days before secondary immunization. Mice were sensitized and challenged with sheep red blood cells (5-RBC). Immune responses were evaluated by humoral immunity, cellular immunity and non-specific immunity. The results of this study were summarized as follows: (1) Beta-carotene significantly increased the weight ratios of liver, spleen and thymus to body weight depending on dose, and significantly increased the increasing rate of body weight and the number of circulating leukocyte. (2) Beta-carotene dose-dependently increased hemagglutination titer, Arthus reaction and hemolytic plaque forming cell related to humoral immunity. (3) Beta-carotene significantly increased delayed-type hypersensitivity reaction and rosette forming cell related to cellular immunity. (4) Beta-carotene dose-dependently increased phagocytic activity, and significantly increased natural killer (NK) cell activity. (5) Beta-carotene dose-dependently inhibited reductions in humoral immunity, cellular immunity, NK cell activity and phagocytic activity by treatment with CY.

  • Effects of squalene on cellular and non-specific immune responses and antitumor activity in mice were investigated. Cellular and non-specific immunological assay parameters adopted in the present study were delayed-type hypersensitivity reaction and resette forming cells (RFC) for cellular immunity, activities of natural killer (NK) cells and phagocyte for non-specific immunity. Squalene resulted in marked increases of cellular and non-specific immune functions and enhancement of host resistance to tumor challenge in dose-dependent manner.
