Effect of Rootstock on Rooting and Early Yield of Stenting-propagated Cut Roses

  • The study was conducted to investigate the effect of rootstock on growth and early yield of stenting-propagated cut roses (Rosa hybrida Hort.) 'Pink Aurora' and 'Yellow King'. The scions, prepared as single-node cuttings, each with a five-leaflet leaf, were grafted onto cuttings of Rosa indica 'Major', Rosa multiflora 'Chille Wonye No. 1', Rosa multiflora 'K-1', or Rosa multiflora 'Burr' as the rootstock. The rootstock cuttings were removed of all leaves and buds before grafting. The base of scion and the top of rootstock were held together and simultaneously cut at a $45^{\circ}$ angle for ease of grafting. Scion-rootstock unions were stuck in rockwool cubes and placed on a misted glasshouse bench for rooting before being transplanted into a rockwool slabs for cultivation. Rooting was the greatest in the 'Pink Aurora' and 'Yellow King' grafted on the rootstock Rosa indica 'Major'. In 'Pink Aurora', stem length, stem diameter, five-leaflet leaves per stem, and stem fresh weight of the harvested cut flowers were not affected by the rootstock. The greatest total yield of 'Pink Aurora' was obtained in plants grafted onto the Rosa indica 'Major' rootstock. Overall growth of 'Yellow King' was the greatest in plants grafted onto Rosa multiflora 'Burr' rootstock, although total yield was not affected by the rootstock. These results suggest that Rosa indica 'Major' is the most effective rootstock not only for rooting, but also for early yield and growth for stenting propagation of these cut roses.

Antioxidant and Antimicrobial Activity of Rosa multiflora Thunberg Fruits Extracts

  • In this study, we selected some material to have potential bioactivity from natural plants, confirmed as basic data for industrializing and tried to develope the food materials using them. DPPH, ABTS, antioxidant protection factor, TBARs and antimicrobial activity of extracts from Rosa multiflora Thunberg fruits were determined. The total phenolics extracted from Rosa multiflora were 12.08, 11.82, 11.1 and 12.6 mg/g when using water, 70% ethanol, 70% methanol and 70% acetone as the solvent, respectively. The optimum conditions for extracting the phenolic compounds were 70% ethanol over for 12 hrs(11.82 mg/g). The electron donating ability and inhibition rate on ABTS of the 70% ethanol extracts were 97% and 92.2%, respectively while the antioxidant protection factor(PF) of the water extracts and 70% ethanol extracts were 1.79 and 1.34 PF, respectively. The TBAR (thiobarbituric acid reactive substance) value were $1.3{\mu}M$ for the control and $0.15{\mu}M$ for the 70% ethanol extracts. The inhibitory activity against ${\alpha}$-amylase was 26% for the 70% ethanol extracts. The 70% ethanol extracts from Rosa multiflora Thunberg fruits exhibited antimicrobial activity against H. pylori, S. epidermidis, S. aureusand and E. coli with clear zone diameters of 14, 25, 14 and 13 mm, respectively when using $200{\mu}g/mL$ of the phenolic compounds. An HPLC analysis identified 6 major phenolic metabolites in the Rosa multiflora Thunberg fruits extracts: rosmarinic acid, caffeic acid, chlorogenic acid, courmaric acid, protocatechuic acid and quercetin. In particular, the content of rosmarinic acid was the highest in the 70% ethanol extracts. Therefore these results indicate that 70% ethanol extracts from Rosa multiflora Thunberg fruits can be useful as a natural antioxidant and in functional foods.

Lignans of Rosa multiflora Roots

  • Five known lignans, (+)-pinoresinol (1), (+)-8-hydroxypinoresinol (2), (-)- dehydrodiconiferyl alcohol (3), (+)-trans-dehydrodiconiferyl alcohol (4), and (-)-olivil (5), were isolated from the roots of Rosa multiflora for the first time. Their structures were determined using spectroscopic data.

Helicobacter pylori 억제효과를 가지는 영실(Rosa multiflora Thunberg) 추출물을 첨가한 식빵의 품질특성 (Quality Characteristics of White Bread Amended with Rosa multiflora Thunberg Extracts with Inhibitory Activity against Helicobacter pylori)

  • Whole fruit 영실로부터 페놀성 물질의 추출은 70% 에탄올을 용매로 하여 추출하였을 때가 최적이었다. 영실로부터 제조한 70% 에탄올 추출물의 Helicobacter pylori에 대한 항균활성은 $200{\mu}g/mL$의 phenolic compoun d 함량에서 13 mm의 저해환을 나타내었다. 영실 추출물을 첨가한 식빵의 품질 특성을 알아본 결과 영실 추출물을 첨가한 반죽은 밀가루 수화와 반죽 형성, 시각적인 반죽 내부평가에서 품질 열화는 관찰되지 않았다. 영실 추출물을 첨가한 반죽으로 구운식빵 제품의 특성을 알아본 결과 대조구에 비하여 수분 함량이 높아 촉촉한 조직감을 주었으며, 불규칙한 기공이 많았으나 대조구에 비해 유의적인 차이는 없었다.

찔레나무뿌리(Rosa multiflora root)의 항산화 및 항염증효과 (Anti-Oxidant and Antiinflammatory Effects of Rosa multiflora Root)

  • 찔레나무뿌리를 열수, 에탄올, 메탄올, 아세톤에서 추출하여 항산화 및 항염증 효능효과를 확인하였다. 그 결과 아세톤추출물에서 가장 많은 67.05${\pm}$0.56 mg/g의 폴리페놀 함량을 확인하였으며, SOD 유사활성능을 제외한 DPPH, ABTS, superoxide anion 라디컬소거능 확인 결과 우수한 항산화 효과를 확인하였다. 찔레나무뿌리 추출물의 HAase 저해능을 측정한 결과 에탄올, 메탄올, 아세톤 추출물 500 ${\mu}g/ml$ 에서 60% 이상의 높은 HAase 저해 효과를 확인할 수 있었다. NO 생성량을 확인한 결과, RAW 264.7 cell에서 LPS에 의해 유도된 NO 생성을 농도의존적으로 뚜렷하게 감소시키는 것을 확인하였으며, 에탄올 추출물에서 NO 생성억제 효과가 가장 우수한 것을 확인할 수 있었다. 위의 사실에 기초하여 NO 생성저해의 기전을 알아보기 위하여 iNOS의 발현을 분석한 결과, 에탄올추출물 100 ${\mu}g/ml$에서 40%의 iNOS protein 발현 감소를 확인하였으며, iNOS의 발현억제가 NO생성억제와 유사한 경향을 나타냄으로 NO생성억제는 iNOS의 발현저해를 경유한 것임을 확인할 수 있었다. 이러한 결과들은 찔레나무뿌리 추출물이 항산화 및 항염증 연구의 기초 자료로 활용될 것으로 예상된다. 또한 추후 산업적 응용도 가능함으로 기능성화장품의 가능성을 제시하고 있다.

우리나라 중부지방에서의 자폐수종 선발에 관한 연구 (A Study on Selecting Barrier Trees in the Middle Temperature forest Zone of Korea)

  • To select suitable siecies for barrier trees in the middle tmperature forest zone of Korea, sprout ratio, verdi cal density ratio and impenetrability were analyzed for 41 species including 9 evergreen conifer trees which had been transplanted 18 years ago and maintained as a hedge at Pocheon - Gun, Kyonggi -Do, Korea. 1. The species with high sprout ratio of hedges were Rosa multiflora, Lonicera maackii, Chamaecyparis pisifera, Thuja occidentalis in order at the upper portion, Thuja occidentalis, Rosa multiflora, Chamaecyparis pisifera, in order oa the middle portion, and Thuja occidentals, Chamaecyparis pisigera, Rosa multiflora, Cercdiphyllum japonicum, Cornus kousa, Ulmus pumila, Robinia pseudoacacia in order at the lower portion of hedges. 2. the species with high vertical density ratio were Thuja occidentalis, Chamaecyparis pisifera, Weigela subsessilis, Acer ginnala in order. 3.The species with hight impnetrability were Thuja occidentalis, Chamaecyparis pisifera, Forsythia Koreana, Juniperus rigida, Juniperus chinensis, Pinus strobus in order. 4. the values of correlation coefficents between the sprout ratio and impenetrability are given in table 3 and positive significant correlations can be observed in conifer trees.5. The species with high suitability for tree barrier were Thuja occidontalis, Chamaecyparis pisifera, Acer ginnala, Juniperus chinensis, Rosa multiflora, Counus kousa, Cercidiphyllum japonocum, Weigela subsessilis, Cornus officinalis, Philadelphus schrenckii in order.

Biological activities of Rosa multiflora Ethanol Extract as Cosmetic Material

  • The Rosa multiflora, a well-known plant belonging to Rosacea, is widely used in orthodox medicine in worldwide. However, its biological activity as a functional ingredient for cosmetic products have not yet been studied. Accordingly, an investigation of the above mentioned atrributes was performed on a 50% ethanol extract of Rosa multiflora. The antioxidant activities were determined by DPPH. Additionally, the contents of total phenols and flavonoids were analyzed. Also, the phenolic compounds were detected using HPLC. The melanogenesis regulatory effect was evaluated using melanin content and cellular tyrosinase activity in B16F10 melanoma cells. The elastase inhibitory activity assay was performed for anti-wrinkle effect. The antimicrobial activity was assessed using the disc diffusion assay. The DPPH radical scavenging ability, denoted by the $SC_{50}$ value was found to be $123.1{\mu}g/ml$, whereas that of positive control (ascorbic acid) was $27.5{\mu}g/mL$. The content of total polyphenol and flavonoid content were 202 mg/g and 86.77 mg/g, respectively. In addition, astragalin and gallic acid were identified in the extract. Also, the ethanol extract significantly inhibited ${\alpha}$-MSH-induced melanogenesis in B16F10 cells. For anti-wrinkle effect, elastase inhibition activity of the ethanol extract was 53.2% at a concentration of $100{\mu}g/ml$. The antimicrobial activity of the extract against S. aureus and E. coli was observed to be 0.5 - 5%, and no significant activity was noted against C. albicans. Therefore, the ethanol extract of Rosa multiflora can be used effectively for development of functional cosmetic materials.

Rosa multiflora Thunberg fruit 추출물로부터 Helicobacter pylori 억제물질의 분리 및 동정 (Isolation and Identification of Inhibitory Compounds on Helicobacter pylori from Rosa multiflora Thunberg Fruit Extracts)

  • 영실에 존재하는 페놀성 화합물의 추출 최적 조건을 알아보고자 추출용매를 달리하여 phenol성 물질의 함량을 비교한 결과, 70% ethanol을 용매로 하여 24시간 교반 추출하였을 때 phenol성 물질의 용출량이 13.8 mg/g 이상으로 가장 높았다. Helicobacter pylori에 대한 항균효과는 영실추출물을 150 ${\mu}g$/ml의 농도로 첨가하였을 때 생육 저해환이 12 mm, 200 ${\mu}g$/ml 일 때 14 mm의 생육저해환이 나타나 항균활성이 우수한 것을 알 수 있었다. H. pylori에 대한 저해물질을 정제하기 위하여 영실을 70% 에탄올로 추출한 후 Sephadex LH-20 column과 $C_{18}$ cartridge column을 이용하여 항균활성을 가지는 물질을 순수 정제하고 구조를 동정한 결과 protocatechuic acid, chlorogenic acid 및 quercetin으로 동정하였으며, 단일물질에 의한 H. pylori에 대한 저해효과 보다 혼합물질에 의한 synergy 효과에 의해 저해 효과가 높아지는 것을 알 수 있었다.

찔레꽃 및 찔레꽃 복합물 추출물이 고지방·고콜레스테롤 식이 흰쥐의 지질 함량 개선에 미치는 영향 (Effects of Rosa multiflora and Rosa multiflora Complex on Lipid Content in Rats Fed a High-Fat·High-Cholesterol Diet)

  • 본 연구에서는 고지방 고콜레스테롤 식이 흰쥐에서 찔레꽃과 찔레꽃 복합물이 혈청 및 간 조직의 지질대사에 미치는 영향을 관찰하였다. 실험군을 4군으로 나누어 정상 식이군(N군), 고지방 고콜레스테롤 식이군(HF군), 고지방 고콜레스테롤 식이에 찔레꽃을 5% 첨가한 군(HR군), 고지방 고콜레스테롤 식이에 찔레꽃 복합물을 첨가한 군(HRC군)으로 나누었다. 찔레꽃 복합물은 찔레꽃, 뱀딸기풀, 곰보배추, 조릿대로 구성되어 있다. 체중증가량과 식이효율은 HF군이 N군에 비해 유의적으로 증가하였고 HR군과 HRC군이 HF군에 비해 감소하였다. 혈청 중성지질은 HF군이 N군에 비해 유의적으로 증가하였고 HRC군은 유의적으로 크게 감소하였다. 혈청 HDL-cholesterol은 HR군과 HRC군이 HF군에 비해 증가하였다. 혈청 총콜레스테롤과 LDL-cholesterol은 HF군이 N군에 비해 유의적으로 증가하였고, 특히 동맥경화지수에서 HRC군은 HF군에 비해 유의적으로 감소하였다. 간 중성지질과 총콜레스테롤은 HF군이 N군에 비해 유의적으로 증가하였고 HR군과 HRC군은 HF군에 비해 유의적으로 감소하였다. 대변 중성지질과 총콜레스테롤은 HR군과 HRC군이 HF군에 비해 유의적으로 증가하였다. UDP-glucuronyl transferase 활성은 HR군과 HRC군이 HF군에 비해 증가하였다. Total bile acid는 HR군과 HRC군이 HF군에 비해 증가하였고, 특히 HRC군은 HF군에 비해 유의적으로 증가하였다. 이러한 결과 찔레꽃과 찔레꽃 복합물은 UDP-glucuronyl transferase 활성, total bile acid 및 지질 대사 조절에 효과적으로 활용될 수 있다고 사료된다.

찔레꽃 에탄올추출물의 생리활성과 화장품 방부효과 (Biological Activity and Cosmetic Preservative Effects of Rosa multiflora Ethanol Extracts)

  • Background: The Rosa multiflora, a well-known plant belonging to Rosacea, is widely used in orthodox medicine in worldwide. However, its biological activity and cosmetic preservative efficacy have not yet been studied. Thus, this species is yet to be defined as a functional cosmetic material. Accordingly, an investigation of the above mentioned atrributes was performed on a 50% ethanol extract of Rosa multiflora. Methods and Results: The antioxidant activity was assessed through free radical scavenging assays with 2,2-diphenyl-1-picrylhydrazyl (DPPH). Additionally, the contents of total phenols and flavonoids were analyzed. The phenolic compounds were detected using HPLC. The antimicrobial activity against Staphylococcus aureus, Escherichia coli, and Candida albicans was assessed using the disc diffusion assay. The preservative effect (challenge test) on a formulation of soothing gel was performed for 28days. The DPPH radical scavenging ability, denoted by the $SC_{50}$ (half maximal inhibitory concentration for DPPH radical scavenging) value was found to be $131.63{\mu}g/m{\ell}$. The content of total polyphenol and flavonoid content were 202 mg/g and 86.77 mg/g, respectively. In additon, astragalin and gallic acid were identified in the extract. The antimicrobial activity of the extract against S. aureus and E. coli was observed to be 5 - 0.5%, and no significant activity was noted against C. albicans. The ethanol extracts (5% and 10%) met the preservation standards of the Cosmetics, Toiletry, and Fragrance Association (CTFA). Conclusions: Thus the ethanol extract of R. multiflora can be used in cosmetics as a natural preservative and antioxidant.
