Objectives : The roots of Dipsacus asper had been used as the herbal medicine "Cheon-Sokdan" in Korea. Moreover, the roots of Phlomoides umbrosa were used as "Han-Sokdan." In the present study, a comparative anatomical comparison of Cheon-Sokdan, Han-Sokdan, and Ilbon-Sokdan were conducted, because Ilbon-Sokdan, the roots of Dipsacus japonicus, was regarded as substitute of Cheon-Sokdan. Methods : For this study, permanent preparations were made using a paraffin embedding method. Anatomical features of these three Sokdans were observed using a light microscope. Results : The starch grains of parenchyma cells and the amounts of calcium oxalate crystals hardly differed among the three plants. Particularly, the longitudinally-sectioned vessels of the three plants showed a wide variety depending on the focal depth of the light microscope. Therefore, these features could not be considered as obligate criteria for distinguishing these plants. The shape of the xylem was linear in Cheon-Sokdan and Ilbon-Sokdan, whereas that in Han-Sokdan was wedge-shaped. The phloem of Cheon-Sokdan and Ilbon-Sokdan were rhomboid, whereas that of Han-Sokdan was thimble-like. Therefore, the shape of xylem and phloem appeared as good criteria for distinguishing Han-Sokdan from the other plants studied. Cheon-Sokdan and Ilbon-Sokdan showed characteristics similar in many parts. However, in the xylem of Ilbon-Sokdan, fiber bundles were more developed than those of Cheon-Sokdan. Therefore, the development of fiber bundles in xylem was considered suitable for distinguishing between Cheon-Sokdan and Ilbon-Sokdan. Conclusions : The identification-keys established in this study would be helpful for identifying microscopic features among the three Sokdans.

Electrophoretic method was utilized to classify 100 varieties of sweet potato germplasm maintained at the National Crop Experiment Station of Korea in 1993. The esterase isozyme patterns in the leaves were classified into 14 different types. Type Ⅸ included the most of the varieties (46) tested and Ⅶ, I, III, Ⅷ, II and V types of all included 47 varieties in order. The other 7 varieties had different band pattern with each other. Type I having many kind of band pattern included Shinyulmi, Beniastma and High starch which had the dry type of tuberous roots varieties. The esterase isozymes pattern in the tuberous roots were classified with 18 kinds of types. The C type included 22 varieties and B, K, A, E, I and N in order. The proteins pattern in the tuberous roots were classified with 7 kinds of types. I type included 36 varieties, and IV type included 27 varieties and II, III, Ⅶ and Ⅵ types in order.

Colonization of Pseudomonas chlororaphis O6, a nonpathogenic rhizobacterium, on the roots induced systemic resistance in cucumber plants against tai-get leaf spot, a foliar disease caused by Corynespora cassiicola. A cDNA library was constructed using mRNA extracted from the cucumber leaves 12 h after inoculation with C. cassiicola, which roots had been previously treated with O6. To identify the genes involved in the O6-mediated induced systemic resistance (ISR), we employed a subtractive hybridization method using mRNAs extracted from C cassiicola-inoculated cucumber leaves with and without previous O6 treatment on the plant roots. Differential screening of the cDNA library led to the isolation of 5 distinct genesencoding a GTP-binding protein, a putative senescence-associated protein, a galactinol synthase, a hypersensitive-induced reaction protein, and a putative aquaporin. Expressions of these genes are not induced by O6 colonization alone. Before challenge inoculation, no increase in the gene transcriptions could be detected in previously O6-treated and untreated plants but, upon subsequent inoculation with the pathogenic fungus, transcription levels in O6-treated plants rose significantly faster and stronger than in untreated plants. Therefore, the O6-mediated ISR may be associated with an enhanced capacity for the rapid and effective activation of cellular defense responses which becomes apparent only after challenge inoculation on the distal, untreated plant parts, as suggested by Conrath et al. Metabolomics is the study of global metabolite profiles in a system (cell, tissue, or organism) under a given set of conditions. Metabolomics has its roots in early metabolite profiling studies but is now a rapidly expanding area of scientific research in its own right. In this study, the applications of metabolomics in natural product studies are explored. Ginseng is a well-known herbal medicine and has various pharmacological effects, which include antiaging, anticancer, antifatigue, memory enhancing, immunomodulatory, and stress reducing effects. Metabolomic analysis of organic acids has not been performed for evaluation whether ginseng has been cultivated using conventional or environmental-friendly farming methods. In this study, profiling analysis was conducted for organic acids (OAs) in ginseng roots produced using conventional or environmentfriendly farming methods at five locations in each of five regions. In OA profiles, lactic acid was the most abundant OA in all regions, with the exception for environmentally friendly farmed ginseng in two of the five regions, in which glycolic acid was most abundant OA. OA profiles in all regions showed isocitric acid levels were increased by environment-friendly cultivation, which suggests metabolic differences associated from farming method, and that isocitric acid might be a useful discriminatory biomarker of environmental-friendly and conventional cultivation. The results of the present study suggest metabolomic studies of OAs in ginseng roots might be useful for monitoring whether ginseng has been cultivated using conventional or environmentally friendly farming methods.
Representing planar Pythagorean hodograph (PH) curves by the complex roots of their hodographs, we standardize Farouki's double cubic method to become the undetermined junction point (UJP) method, and then prove the generic existence of solutions for general $C^1$ Hermite interpolation problems. We also extend the UJP method to solve $C^2$ Hermite interpolation problems with multiple PH cubics, and also prove the generic existence of solutions which consist of triple PH cubics with $C^1$ junction points. Further generalizing the UJP method, we go on to solve $C^2$ Hermite interpolation problems using two PH quintics with a $C^1$ junction point, and we also show the possibility of applying the modi e UJP method to $G^2[C^1]$ Hermite interpolation.

This study is conducted to evaluate the effects of containerized landscape tree production methods on post-transplant stress. Two types of container such as plastic pot(pot), fabric growing bag(bag) were adopted to restrict tree roots. Each types of containers was divided into seven sub-types. One traditional production method was included as comparison. Two landscape woody plants species (Magnolia denudata, Albizzia julibrissin) were implanted in the 7 sub-types of container. After one or two growing season in the container, each types of container trees was transplanted. Half of the trees were transplanted in the mild spring season, and another half of trees were transplanted in the improper summer season. The data were collected on the crown wilting ratio and trunk die-back ratio. The result of the analysis based on these data were as follows; 1) The container production methods were lower than the traditional production methods by 3 times in the average wilting ratio of summer season's transplanting point. 2) Post-transplant stress was more successfully mitigated, in case the "pot" type as was the "bag" types of container. 3) The effective and economic way of mitigating post-transplant stress by container production methods was selecting container plants of vigorous and deep root systems. 4) The "pot" type of container was to restrict tree roots more successfully, But, winter chilling and low temperature attacked the "pot" type tree's twigs and suckers. These results indicated that "pot" grown container plants should managed carefully during the winter after transplanting. Based upon the results of this study, a subsequent research on the development of container material, growth type of the container trees, and other maintaining method will be required.es, and other maintaining method will be required.
Background: In ecosystem carbon cycle studies, distinguishing between $CO_2$ emitted by roots and by microbes remains very difficult because it is mixed before being released into the atmosphere. Currently, no method for quantifying root and microbial respiration is effective. Therefore, this study investigated the relationship between soil respiration and underground root biomass at varying distances from the tree and tested possibilities for measuring root and microbial respiration. Methods: Soil respiration was measured by the closed chamber method, in which acrylic collars were placed at regular intervals from the tree base. Measurements were made irregularly during one season, including high temperatures in summer and low temperatures in autumn; the soil's temperature and moisture content were also collected. After measurements, roots of each plot were collected, and their dry matter biomass measured to analyze relationships between root biomass and soil respiration. Results: Apart from root biomass, which affects soil's temperature and moisture, no other factors affecting soil respiration showed significant differences between measuring points. At each point, soil respiration showed clear seasonal variations and high exponential correlation with increasing soil temperatures. The root biomass decreased exponentially with increasing distance from the tree. The rate of soil respiration was also highly correlated exponentially with root biomass. Based on these results, the average rate of root respiration in the soil was estimated to be 34.4% (26.6~43.1%). Conclusions: In this study, attempts were made to differentiate the root respiration rate by analyzing the distribution of root biomass and resulting changes in soil respiration. As distance from the tree increased, root biomass and soil respiration values were shown to strongly decrease exponentially. Root biomass increased logarithmically with increases in soil respiration. In addition, soil respiration and underground root biomass were logarithmically related; the calculated root-breathing rate was around 44%. This study method is applicable for determining root and microbial respiration in forest ecosystem carbon cycle research. However, more data should be collected on the distribution of root biomass and the correlated soil respiration.
The endodontically treated tooth is generally restored with post & core, owing to the brittleness and the loss of large amount of tooth structure. Although there have been lots of studies about the endodontically treated teeth, the three-dimensional quantitative studies about the strees distribution of them are in rare cases. In this study, it was assumed that the coronal portion of the upper incisou had severely damaged. After the root canal therapy it was post cored, and restored with PFM crown, for this experiment nine types of model were constructed : 1); long, 2); medium, 3); short gold post for the roots supported with a narmal alveolar bone, 4); long, 5); medium, 6); short gold post for the roots supported with an alveolar bone resorbed to its 1/3 of root length, 7); long, 8); medium, 9); short base metal post for the roots supported with an alveolar bone resorbed to its 1/3 of root length. Force was applied from two directions. One was functional maximum bite force(300N) applied to the spot just lingual to the incisal edge with the angle of 45 degrees to the long axis of the tooth, and the other one was horizontal force(300N) applied to the labial surface. The results analyzed with three-dimensional finite element method were as follows : 1. Stress was concentrated on the middle portion of the labial side dentin of the root and the lingual portion of the apical dentin of the root. Stress in the post showed maximum value at 2 mm above the post apex. 2. In case of the long post and base metal post, strees was concentrated on the apex of the root and the post. 3. In case of the longer post, the displacement on the post-cement interface was lessened. The gold post was more displaceable than the base metal post. 4. In case of the alveolar bone resorption, stress concentrated on the root and the post and displacement on the post-cement interface were increased.
Identification of the origins of Panax ginseng has been issued in Korea scientifically and economically. We describe a metabolomics approach used for discrimination and prediction of ginseng roots from different origins in Korea. The fresh ginseng roots from six ginseng cooperative associations (Gangwon, Gaeseong, Punggi, Chungbuk, Jeonbuk, and Anseong) were analyzed by UPLC-MS-based approach combined with orthogonal projections to latent structure-discriminant analysis multivariate analysis. The ginsengs from Gangwon and Gaeseong were easily differentiated. We further analyzed the metabolomics results in subgroups. Punggi, Chungbuk, Jeonbuk, and Anseong ginseng could be easily differentiated by the first two orthogonal components. As a validation of the discrimination model, we performed blind prediction tests of sample origins using an external test set. Our model predicted their geographical origins as 99.7% probability. The robust discriminatory power and statistical validity of our method suggest its general applicability for determining the origins of P. ginseng samples.

Root is an important organ of plant that typically lies below the surface of the soil. Root surface determines the ability of plants to absorb nutrient and water from the surrounding soil. This study describes an application of image processing and computer vision which was implemented for rapid measurement of seed germination such as root length, surface area, average diameter, branching points of roots. A CCD camera was used to obtain RGB image of seed germination which have been planted by wet paper in a humidity chamber. Temperature was controlled at approximately 250C and 90% relative humidity. Pre-processing techniques such as color space, binarized image by customized threshold, removal noise, dilation, skeleton method were applied to the obtained images for root segmentation. The various morphological parameters of roots were estimated from a root skeleton image with the accuracy of 95% and the speed of within 10 seconds. These results demonstrated the high potential of computer vision technique for the measurement of seed germination.
