꾸지뽕나무(Cudrania tricuspidata Bureau)의 총 폴리페놀 함량 및 항산화 활성 (The Antioxidant Activity and Total Polyphenol Content of Cudrania tricuspidata)

  • The purpose of this paper is to describe the differences in mineral, ascorbic acid, and total polyphenol content, and also the relationship between polyphenol content and antioxidant activity(DPPH radical scavenging, FTC, and TBA methods) in three sizes of Cudrania tricuspidata leaves. For the mineral contents of the leaves, Ca was highest(17.26 mg/g.d.w.), followed by K(15.53 mg), P(2.61 mg), and Mg(1.99 mg). Ascorbic acid was found to be slightly higher(43.4 mg/100g.f.w.) in the basal parts of the medium size leaves and lowest in the whole parts(9.9 mg) of the small size leaves. Total polyphenol content was highest($1,384{\sim}1,258mg/100g.f.w.$) in the leaves, for all sizes, when extracted with 80% ethanol, followed by the stem bark(543 mg%), roots(369 mg%), stems(243 mg%), and thorns(223 g%). By determining DPPH activity, we found that antioxidant activity was higher in the ethanol extracts of the leaves than in the ethanol extracts of the thorns, stems, roots, and stem bark. While in the tea, no differences were found between the ethanol and water extracts.

추출조건이 노니 부정근에 함유된 rubiadin의 추출 효율에 미치는 영향 (Effects of Extraction Condition on Extraction Efficiency of Rubiadin in Adventitious Roots of Noni (Morinda citrifolia))

  • 노니($Morinda$ $citrifolia$) 부정근의 주요 성분인 rubiadin은 간 보호 효과가 있어 제약산업에서 높은 가치가 있다. 노니 부정근의 주요 유효성분인 rubiadin의 효율적인 추출조건을 규명하고자 용매 종류, 물과 메탄올의 비율(물, 20%, 40%, 60%, 80%, 100%), 추출시간 및 추출방법을 달리하여 추출하였다. 노니 부정근에서의 rubiadin의 함량분석은 HPLC 이용 분석조건을 확립하였으며, C-18 컬럼을 사용하여 280nm에서 메탄올과 물로 농도구배를 주어 분석하였다. 용매별 추출효율은 메탄올(0.08%) > 에탄올(0.05%) > 아세토니트릴(0.03%) > 아세톤(0.02%) 및 메틸렌클로라이드(0.02%) 순으로 증가하였다. 메탄올에 물의 혼합 비율을 달리하여 초음파 추출기로 1시간 동안 추출한 결과 60% 메탄올(0.21%) > 80% 메탄올(0.13%) > 100% 메탄올(0.07%) 및 40% 메탄올(0.07%) 순으로 효과적이었으며, 환류냉각 추출기로 2시간 동안 추출한 결과 60% 메탄올(0.21%) > 40% 메탄올(0.17%) > 80% 메탄올(0.14%) 용매처리구 순으로 효율이 좋았다. 추출방법 및 추출시간에 따른 rubiadin의 추출 효율을 비교하기 위해 환류냉각추출 및 초음파추출, 진탕추출법으로 추출하였다. 추출방법 및 추출시간에 따른 rubiadin의 추출효율은 초음파추출 및 진탕추출에서 각각 8 및 24시간 동안 추출했을 때 효율적이었다.

여우구슬(Phyllanthus urinaria)의 부정근 유도 및 기내증식조건 (Induction and in vitro Proliferation of Adventitious Roots in Phyllanthus urinaria)

  • 본 실험은 여우구슬의 기내 부정근 유도 및 증식조건의 확립을 목적으로 수행되었다. 우선 여우구슬의 기내 발아체로부터 부위를 달리하여 부정근을 유도한 결과 줄기부위는 뿌리보다 양호한 부정근의 유도를 보였다. 또한 유도된 부정근을 이용하여 옥신의 종류(IAA, IBA, NAA와 2.4-D)에 따른 부정근 유도율을 조사한 결과 IBA와 NAA는 IAA와 2.4-D보다 높은 유도율을 보였다. IBA의 농도에 따른 유도율과 증식효율은 IBA가 0.5 mg/L첨가되었을 때 가장 높은 유도 및 증식효율을 보였다. 최적의 액체배지조건을 확인하고자 IBA의 농도는 0.5 mg/L로 첨가하고 sucrose의 농도를 달리하여 실험한 결과 sucrose는 30 g/L 첨가 되었을 때 가장 높은 생중량과 건중량을 나타냈다. 액체배양된 여우구슬의 부정근을 각각 MS, 1/2MS, 1/3MS배지에 30 g/L sucrose, 0.5 mg/L IBA가 첨가된 5 L 용량의 생물반응기에 4주간 배양한 결과 1/2MS 배지에서 양호한 생장을 보였다. 본 실험에서는 여우구슬의 종자발아체를 이용하여 부정근의 유도 및 증식조건에 필요한 기내배양조건과 2차적으로 유도된 부정근을 이용하여 플라스크와 생물반응기 배양을 통한 효율적인 증식조건을 확립하였다.

A Study on Transcriptome Analysis Using de novo RNA-sequencing to Compare Ginseng Roots Cultivated in Different Environments

  • Ginseng (Panax ginseng C.A. Meyer), one of the most widely used medicinal plants in traditional oriental medicine, is used for the treatment of various diseases. It has been classified according to its cultivation environment, such as field cultivated ginseng (FCG) and mountain cultivated ginseng (MCG). However, little is known about differences in gene expression in ginseng roots between field cultivated and mountain cultivated ginseng. In order to investigate the whole transcriptome landscape of ginseng, we employed High-Throughput sequencing technologies using the Illumina HiSeqTM2500 system, and generated a large amount of sequenced transcriptome from ginseng roots. Approximately 77 million and 87 million high-quality reads were produced in the FCG and MCG roots transcriptome analyses, respectively, and we obtained 256,032 assembled unigenes with an average length of 1,171 bp by de novo assembly methods. Functional annotations of the unigenes were performed using sequence similarity comparisons against the following databases: the non-redundant nucleotide database, the InterPro domains database, the Gene Ontology Consortium database, and the Kyoto Encyclopedia of Genes and Genomes pathway database. A total of 4,207 unigenes were assigned to specific metabolic pathways, and all of the known enzymes involved in starch and sucrose metabolism pathways were also identified in the KEGG library. This study indicated that alpha-glucan phosphorylase 1, putative pectinesterase/pectinesterase inhibitor 17, beta-amylase, and alpha-glucan phosphorylase isozyme H might be important factors involved in starch and sucrose metabolism between FCG and MCG in different environments.

생강 저장굴에서 발생한 건강 피해 (Health hazards occurred in the underground storing places of ginger roots)

  • Ginger has been cultivated for thousands of years. It is very popular in the food industry as an additive to ginger ale, candies, pastries and cakes. The root has a strong, sweet scent. It contains protein, fat, carbohydrates, ash, calcium, potassium, vitamin A. thiamine, riboflavin, niacin and ascorbic acid. In korea, farmers keep ginger roots into underground holes from the harvest time to the seedtime. They dig 6 meters under the ground and preserve the cultivated ginger roots. This study was conducted to evaluate the health hazards occurred in the underground storing places of ginger roots. Authors reviewed the accidents occurred in those places, therefore transported by the public emergency system in Seo-cheon city from Jan, 1 1996 to Dec, 31 1997. We could confirm 8 persons on transport records. Among them, 5 persons were due to poisoning, 3 persons were due to injuries. We would like to suggest that poisoning could be induced by hypoxia and injuries occurred from a fall into the underground hole. These should be prevented by the health education for farmers. And further investigations are needed to understand their magnitude and the pathogenic mechanisms.

오옥신 처리가 에키네시아의 부정근 및 생리활성물질 생산에 미치는 영향 (Auxin Affects on Production of Adventitious Roots and Secondary Metabolites in Echinacea angustifolia)

  • The production of adventitious roots derived from root explant of Echinacea angustifolia and its secondary metabolite content were assessed in different types and levels of auxin. The induction of adventitious roots from root explant cultured in Murashige and Skoog solid medium supplemented with 1.0 mg/L indole -3-butyric acid (IBA) attained highest as 20.87 mg fresh weight and 3.07 mg dry weight per culture but root suspension culture at the same concentration of IBA enhanced biomass production as 3.07 g fresh weight and 0.38 g per culture after 4 weeks in culture. 3.0 mg/L ${\alpha}$-naphthalene acetic acid (NAA) treatment had similar effect on root biomass production as 3.07 g fresh weight and 0.38 g per culture with liquid suspension culture, whereas adventitious roots exposed to over 3.0-5.0 mg/L IBA or 5.0 mg/L NAA were less responsive by reducing the number of adventitious roots and/or changing root morphology such as short and thick. The content of secondary metabolites such as phenolic, flavonoids and total caffeic acid in adventitious roots cultured on MS medium supplemented with 1.0 mg/L IBA were attained highest as 27.20, 9.60. 10.67 mg/g dry weight, respectively. Overall, the best production of root biomass and secondary metabolites were given by 1.0 mg/L IBA.

엽록체기반 SSR marker를 이용한 당귀의 기원 판별 (Determination of the Origin of Angelica Roots using Angelica gigas Chloroplast Based SSR Markers)

  • Background: In the herbal medicinal industry, Angelica gigas Nakai, Angelica sinensis (Oliv.) Diels. and Angelica acutiloba (Siebold & Zucc.) Kitag. are often confused, because the roots of the three species can not be distinguished by their appearance. This confusion can cause serious side effects. In this study, we determined the origins of Angelica roots distributed in the Korean market using the simple sequence repeat (SSR) markers developed based on the A. gigas chloroplast DNA sequence. Methods and Results: We collected twenty seven A. gigas and three A. acutiloba samples from the Seoul, Daegu, and Cheongju herbal medicinal markets. Fifty sections of one collection were mixed and ground to make a powder, which was used for DNA extraction using the cetyl trimethylammonium bromide (CTAB) method. Chloroplast based SSR markers were applied to the DNA for the determination of the species. In addition, polymorphism was found in eight samples. The phylogenetic analysis showed that the A. gigas roots collected from herbal medicinal markets were clearly discriminated from A. sinensis and A. acutiloba even though they were grouped into four clusters. Conclusions: This study showed that chloroplast based SSR markers would help the discrimination of Angelica roots in the Korean herbal medicinal industry and the markers are useful to prevent confusion between Angelica roots.

Biosafety and Toxicological Evaluation of Tissue-Cultured Echinacea purpurea Adventitious Roots

  • Echinacea purpurea (L.) Moench (purple cone flower) is an important medicinal plant; it can enhance immunity, relieve pain, and reduce inflammation, and also has hormonal, antiviral, and antioxidant effects. Adventitious root biomass of Echinacea purpurea was produced in commercial-scale bioreactors for use as a dietary supplement in the food industry and in traditional medicine. Biosafety and toxicological evaluations of tissue-cultured Echinacea purpurea adventitious roots (TCEPARs) were performed. Reverse mutation and chromosomal aberration tests showed no significant mutagenicity. Furthermore, repeated four-week oral dose tests performed in Sprague-Dawley rats did not show any notable changes in the general behavior of the rats, in the gross appearance of their internal organs, or in their mortality rate. There were no differences between the control group and the treatment group in parameters such as absolute body weight, hematology, blood chemistry, and absolute and relative organ weights. These findings indicate that TCEPARs are safe and nontoxic when consumed at an average dietary level and can be used as raw material for traditional medicine and the food industry.
