Effect of Saussurea Lappa Root Extract on Proliferation and Hair Growth-related Signal Pathway in Human Hair Follicle Dermal Papilla Cells (당목향 뿌리 추출물의 인체 모유두세포 증식 및 모발 성장 관련 신호전달에 미치는 영향)

  • In this study, Saussurea Lappa roots were extracted using ethanol and n-hexane, and also the effects on proliferation of human hair dermal papilla cells and fibroblast and related signaling pathway were evaluated. 3-[4, 5-dimethylthiazol-2-yl]-2, 5 diphenyl tetrazolium bromide (MTT) assay was conducted for cell proliferation effect of Saussurea Lappa root extract, and extracellular signal-related kinase (ERK), serine/threonine protein kinase (Akt), wingless-related integration site (Wnt)/𝛽-catenin signaling pathway, and 5𝛼-reductase expression through western blot analysis were measured. Saussurea Lappa root extract significantly increased human hair dermal papilla cells and propagation of fibroblast, promoted phosphorylation of ERK and Akt that get involved in cell proliferation. Additionally, Saussurea Lappa root extract significantly decreased promotion of Akt phosphorylation and cell proliferation by MEK/ERK inhibitor PD98059 and PI3K/Akt inhibitor LY294002. Also, Saussurea Lappa root extract induced intranuclear 𝛽-catenin accumulation by promoting phosphorylation of 𝛽-catenin (Ser552, 675) through phosphorylation of GSK-3𝛽 (Ser9), and suppressed activation of 5𝛼-reductase type I and II. Overall, Saussurea Lappa root induces cell proliferation through vitalization of ERK and Akt route of human hair dermal papilla cells and fibroblast and apoptosis defense mechanism, and can be helpful in hair loss prevention and hair growth by vitalizing the 𝛽-catenin signaling pathway and inhibiting activation of 5𝛼-reductase, which can be used as a potential hair care products.

Experimental Studies on the Hair Growth Activity of Drynariae Rhizoma Extracts in C57BL/6N Mice (C57BL/6N 생쥐에서 골쇄보(骨碎補) 추출물을 이용한 발모(發毛)효과에 대한 실험적 연구)

  • Drynariae Rhizoma has been used for promotes mending of the sinews and bone, tonifies the kidney for such symptoms as weak low back and knees, and stimulates the growth of hair as a tinctute for alopecia in oriental medicine. This experiment examined the effect of an acetone extracts of Drynariae Rhizomas(GSB-1), its EtoAc fraction(GB-2) and n-buOH fraction(GSB-3), on hair growth activity of the C57BL/6L mice after topical application to skin. First, We examined on hair growth activity of extracts of Drynariae Rhizomas compare to control and 1 % minoxidil groups. Second, We investigated on the number of hair follicle and mast cells after topical application of extracts of the Drynariae Rhizomas to skin for 16 day. Third, We investigated immunoreactive density of vascular endothelial growth factor(VEGF), protein kinase C-${\alpha}$(PKC-${\alpha}$) and stem(mast) cell factor(SCF) in skin of C57BL/6N mice by immunohistochemical methods. The results were as follows : Hair growth effect of acetone extracts of Drynariae Rhizomas, its EtoAc fraction and n-BuOH fraction was observed in 98 %, 96 % and 60 % in hair removed skin area in 16 day respectively, Immunoreactive density of VEGF in skin of GSB-1 group was weakly stained compare to control group in 10 day, But GSB-2 and GSB-3 groups were mildy stained in bulge and root sheath of skin. Immunolocalization of SCF antigens was observed weakly stained density in epidermis, bulge, stem cells and dermal papilla of control gruop. but in experimental group, immunoreactivity of SCF antigens was observed mildly stained density in bulge, epidermis and root sheath of GSB-1 gruop, heavily stained density in epidermis, bulge and root sheath of GSB-2 and GSB-3 groups to the hair removal skin of C57BL/6N mice on day 10. These experiment suggest that acetone extracts of Drynariae Rhizomas and its EtoAc fraction may be used for topical treatment of alopecia areata.

  • This study was carried out to find out the optimum hardening condition for ginseng plantlets redifferentiated by tissue culture method. While a lot of root hair were observed on the root of seedling grown on the soil, few root hair were observed on the root of plantlet redifferentiated in vitro. On the medium solidified with $0.1{\%}$ phytagel, root hair was not observed and root weight, root width and laternal root development were also very poor. While raising the phytagel concentration on the media, root hair began to increase and root weight, root width and latemal root development were improved. Vascular tissue of plantlet grown on the medium with $0.1{\%}$ phytagel was very poor, but that of plantlet grown on the medium with $0.8{\%}$ phytagel was very good.

Studies on the effect of Sophora flavescens extract on the hair growth stimulation and acne inhibition (苦蔘抽出物이 毛髮成長 促進 및 面疱 抑制에 미치는 영향)

  • In the course of screening natural extracts for hair growth, we found that the extract of dried root of Sophora flavescens has the prominent hair growth promoting effect. After topical application of Sophora flavescens extract to the back of C57BL/6 mice, the earlier conversion of telogen-to-anagen phase was induced. In addition, the Sophora flavescens extract revealed to possess potent inhibitory effect on $5{\alpha}$-reductase Ⅰ and Ⅱ activity. The growth of dermal papilla cells and mouse vibrissae hair follicle cultured in vitro, however, was not affected by Sophora flavescens extract treatment. RT-PCR analysis showed that Sophora flavescens extract induced mRNA levels of growth factors such as insulin-like growth factor-Ⅰ and keratinocyte growth factor in dermal papilla cells, suggesting hair growth promoting effect of Sophora flavescens extract is mediated through inhibition of $5{\alpha}$-reductase type Ⅱ activity and the regulation of growth factors in dermal papilla cells. Furthermore, Sophora flavescens extract also showed anti-bacterial effect on Propionibacterium acnes. These results suggest that Sophora flavescens can be used as a potent treatment agent for helping hair growth stimulation and acne inhibition.

Immunohistochemical Study on the Hair Growth Promoting Effect of Yonnyuniksoogobon-dan

  • Objectives : This study evaluates the hair growth promoting effect of Yonnyuniksoogobon-dan on shaved C57BL6 mice. Methods : Yonnyuniksoogobon-dan was administered orally (Group I) and both orally and by skin application (Group II) once a day for 12 days. The experimental groups were compared to Control, which was orally administered physiological saline solution. Hair regrowth was photographically and histologically determined during the experimental period. The levels of hair growth cycle related factors (EGF, TGF-${\beta}$1) and vascular factors (VEGF, iNOS) were also determined by immunohistochemistry. On gross observation of hair growth, both Group I and Group II shaved C57BL6 mice showed accelerated hair regrowth. Results : The hair regrowth index of Group I increased significantly from 7 days (P<0.05) to 12 days (P<0.01) after shaving and that of Group II was significantly higher at 12 days (P<0.05). On histological observation, both Group I and Group II demonstrated histological improvement and increases in the number and diameter of the hair follicles. EGF expression on the root sheath of hair follicles was up-regulated in both Group I and II. TGF-${\beta}$1 expression on the root sheath of hair follicles was unchanged in both Group I and II. VEGF expression in the tissues surrounding hair follicles was up-regulated in both groups. iNOS expression in the tissues surrounding hair follicles was down-regulated in both groups. Conclusions : These results suggest that Yonnyuniksoogobon-dan promotes hair growth and this effect is related to up-regulation of EGF and VEFG expression and down-regulation of TGF-${\beta}$1 and iNOS expression on hair roots.

The Immunohistochemical Changes of Skin during Hair Follicle Cycle after Depilation in Mice

  • We have examined the histological changes of skin during hair follicle growth after depilation in C57BL/6N mice. We first studied on histological changes of number of mast cells and thickness of skin during hair follicle growth periods (telogen, 1 day, 3 day, 5 day, 10 day, 14 day, 17 day and 21 day after depilation) by toluidine blue, Giemsa and H&E staining methods. We second studied immunoreactive density of cytokines and Brdu labeled cells in skin during hair follicle growth periods after depilation in C57BL/6N mice by immunohistochemical methods. The histological changes on skin thickness was increased from telogen to 14 day. The number of mast cells was decreased in 3,5 and 10 day and increased in 14, 17 and 20 day after depilation. Immunoreactive density of cytokines [protein kinase C-${\alpha}$ (PKC-${\alpha}$), c-kit, and vascular endothelial growth factor (VEGF)] in 1, 3, 5, 10, and 14 day after depilation was mildly stained in bulge and cutaneous trunci m., but immunoreactive density of cytokines in 17 and 21 day was heavily stained in epidermis, bulge, outer root sheath (ORS), inner root sheath (IRS) and cutaneous trunci m.. Immunoreactive density of Brdu labeled cells in skin in 1 and 3 day was heavily stained in bulge, epidermis and connective tissue under the cutaneous trunci m.. In all periods, immunoreactive density of Brdu labeled cells in skin was heavily stained in bulge, subcutaneous tissue, cutaneous trunci m, ORS and IRS. These experiments suggest that histological changes related to hair follicle growth elevated mast cell counts, skin thickness and epidermis thickness and heavily stained immunoreactive density of cytokines and Brdu labeled cutaneous trunci m. and connective tissue under the cutaneous trunci m. after depilation in C57BL/6N mice.

The Experimental Study on the Effect of Herbal Exrtacts on Hair Growth and Acnes (복분자(覆盆子), 석창포(石菖蒲), 상침자 및 숙지황(熟地黃)이 모발성장(毛髮成長)과 면포에 미치는 실험적(實驗的) 연구(硏究))

  • The effects of four extracts of medicinal herbs, Rubus coreanum, Acorus calamus, Morus alba and Rehmannia glutinosa on hair growth activity and acnes control were investigated. In the course of screening natural extracts for hair growth, we found that the extract of dried root of Rubus coreanum has the hair growth promoting effect. After topical application of these extracts to the back of C57BL/6 mice, the earlier conversion of telogen-to-anagen phase was induced. The growth of dermal papilla cells and mouse vibrissae hair follicle cultured in vitro, however, was not affected by treatment of these extracts. Furthermore these extracts do not possesspotent inhibitory effect on $5{\alpha}-reductase$ I and II activity and anti-bacterial effect on Escherichia coli , Propionibacterium acnes, Pityrosporum ovale, Staphylococcus aureus, Staphylococcus epidemidis, and Candida albicans. RT-PCR analysis showed that these extracts did notinduce mRNA levels of growth factors such as insulin-like growth factor-I, keratinocyte growth factor, hepatocyte growth factor and vascular endothelial growth factor in dermal papilla cells. These results suggest that Rubus coreanum has hair growth promoting effect. However, the effects of these materials on the hair growth promotion are not mediated through inhibition of $5{\alpha}-reductase$ I and II activity, stimulation of hair follicle cells and expression of growth factors in the dermal papilla cells.

Development of in vitro 3D hair growth model using tissue engineering technology

  • The recent development of methods for culturing hair follicles in vitro has proved an important tool to investigate many aspects of drug screening. Hair follicles develop as a result of epithelial-mesenchymal interactions between epidermal keratinocytes and dermal cells. We isolated some follicle cells using explantation and enzymatic digestion method from human scalp hair follicles. So we could culture some follicular cells, such as outer root sheath (ORS) cells, dermal papilla (DP) cells, dermal sheath (DS) cells, matrix cells and melanocytes. To induce hair morphogenesis in vitro the cells were 3-D cultured as skin structures. Moreover, to develop hair follicel organ culture model, we applied dermal equivalent (DE) to culturing hair follicles to expand hair growth period.

  • Objectives: Yonnyuniksoogobon-dan (Yan Nian Yi Shou Gu Ben Dan 延年益壽固本丹) is composed of 11 herbs (Polygoni Mutiflori Radix, Lycii Radicis Cortex, Polia, Rehmanniae Radix, Rehmanniae Radix Preparat, Asparagi Radix, Liriopis Tuber, Lycii Fructus, Acori Graminei Rhizoma, Angelicae Acutiloba Radix, and Pini Folium) based on Yonryunggobon-dan (Yan Ling Gu Ben Dan 延齡固本丹) and Yonnyuniksoobulrho-dan (Yan Nian Yi Shou Bu Lao Dan 延年益壽不老丹). This study evaluated hair growth promoting effect of Yonnyuniksoogobon-dan on the shaved C57BL/6 mice. Methods: Yonnyuniksoogobon-dan was treated by oral administration (Sample I) and oral administration plus skin application (Sample II) once a day for 12 days. Hair regrowth was photographically and histologically determined during the experimental period. Hair growth cycle related factors (EGF, TGF-${\beta}1$) and vascular factors (VEGF, iNOS) were also determined with immunohistochemistry. Results: 1. On gross observation of hair regrowth, Sample I and Sample II groups demonstrated acceleration of hair regrowth. 2. The hair regrowth index of the Sample I group increased significantly from 7 days (P<0.05) to 12 days (P<0.01) after the shave while that of the Sample II group significantly increased at 12 days (P<0.05). 3. On histological observation, both Sample I and Sample II groups demonstrated histological improvement and increases of number and diameter of the hair follicles. 4. EGF expressions on the root sheath of hair follicles were up-regulated in both Sample I and Sample II groups. 5. TGF-${\beta}1$ expressions on the root sheath of hair follicles were not regulated in Sample I or Sample II groups. 6. VEGF expressions in the surrounding tissues of hair follicles were up-regulated in both Sample I and Sample II groups. 7. iNOS expressions in the surrounding tissues of hair follicles were down-regulated in both Sample I and Sample II groups. Conclusions: These results suggest that Yonnyuniksoogobon-dan has hair growth-promoting activity and these effects relate to up-regulations of EGF and VEFG expressions and down-regulations of TGF-${\beta}1$ and iNOS expressions on hair roots.

Effects of Microneedle Therapy System(MTS) and Hwangryeonhaedoktang Pharmacopuncture Solution on Hair Growth in an Alopecia Model of C57BL/6N Mouse (C57BL/6N 마우스 탈모모델에서 미세다륜침(MTS)과 황련해독탕 약침액 도포가 모발성장에 미치는 영향)

  • Objective : This study was carried out to investigate the effect of Hwangryeonhaedoktang pharmacopuncture solution (HRHDT) and microneedle therapy system (MTS) on hair growth in an alopecia model of C57BL/6N mice.Methods : Six-week old mice were depilated and separated in 4 groups ; CON (saline), MXD (3% Minoxidil), MTS and HRHDT+MTS. The treatments were applied twice a week for 17 days. The hair growth was determined photographically. The hair density, thickness and length were identified by Folliscope and the weights of body and organs were measured. In dorsal skin tissue, the expression of hair growth-related gene and protein was analyzed by RT-PCR. In addition, the hair follicles in the dermis were observed by H&E staining.Results : The promotion of hair growth was observed in HRHDT+MTS and MTS compared to CON. The hair density, thickness and length were also improved in HRHDT+MTS and MTS compared to CON. The mRNA expression of IGF-1, PRL and PL and the protein expression of VEGF and IGF-1 were increased in HRHDT+MTS and MTS compared to CON. The hair follicles and hair root growth were improved in HRHDT+MTS and MTS compared to CON. In the above results, HRHDT+MTS were more effective than MTS.Conclusions : These results suggest that HRHDT and MTS have a hair growth activity and can be useful for the treatment of alopecia.
