  • The effect of starvation on the growth and hepatocyte nuclear size of larval rockfish Sebastes schlegeli and spotted sea bass Lateolabrax sp. were studied. The growth of total length and wet weight in both rockfish and spotted sea bass starved were lower than their control counter-parts. The nuclei sizes of parenchymal cells in the liver of rockfish and spotted sea bass were correlated with the nutritional status of their first-feeding larvae. The result suggested that hepatocyte nuclear size in rockfish and spotted sea bass could be used as an alternative indicator for the identification of starving condition and such karyometry might be criteria for evaluating the successful transition from endogenous to exogenous feeding regime.

Effect of Dietary Proteins without Vitamin Premix Supplementation on the Growth and Body Composition of Juvenile Black Rockfish Sebastes schlegeli (비타민프리믹스 무첨가시 사료 중 단백질원이 조피볼락(Sebastes schlegeli) 치어의 성장과 체성분에 미치는 영향)

  • A feeding experiment was conducted to determine the nutritional value of various dietary protein sources without a vitamin premix supplementation in juvenile black rockfish, Sebastes schlegeli. Diets containing similar quantities of lipid and carbohydrate with 50% crude protein from fish meal (FM)/soybean meal (SBM)/corn gluten meal (CGM), FM only, casein/gelatin, high FM/low casein and low FM/high casein were fed to rockfish along with a commercial rockfish diet. The diet containing only 65.6% FM and the commercial rockfish diet produced significantly greater weight gain (WG) and feed efficiency (FE) than all other semipurified diets (SPDs); the economical diet with 52.8% FM, 10% SBM and 5% CGM produced lower WG than the diet containing FM and commercial rockfish diet, but FE and survival were similar both diets. Intermediate responses were observed for rockfish fed the SPD containing 32.8% FM and 27.4% casein, while SPDs containing 10% FM and 41.8% casein and 38.5% casein and 10% gelatin yielded the lowest WG and FE. Some differences in tissue indices and body composition of rockfish were induced by the diets containing various protein sources and levels without the vitamin premix supplementation.

  • A feeding trial was conducted to investigate the growth and body composition of juvenile sea cucumber Apostichopus japonicus in integrated culture with abalone Haliotis discus hannai or rockfish Sebastes schlegeli. Triplicate groups of sea cucumber averaging $1.2{\pm}0.05g$ were cultured alone or with abalone or rockfish for 12 weeks. Survival of sea cucumber was not affected by co-culturing (P>0.05). Weight gain of sea cucumber cultured with rockfish was significantly higher than that of sea cucumber cultured alone (P<0.05), and did not differ from that of those cultured with abalone (P>0.05). These findings indicate that co-culturing sea cucumber with rockfish effectively improves the growth of sea cucumber.

Effects of Sodium Cyanide (NaCN) on the Endogenous Rhythm of the Oxygen Consumption Rate in the Black Rockfish Sebastes schlegeli

  • Laboratory bioassays were conducted to test the acute toxicity effects of sudden exposure to sodium cyanide (NaCN) on the endogenous rhythm of the oxygen consumption rate (OCR) in the black rockfish Sebastes schlegeli. The OCR of the black rockfish(n = 14, total length = $20.4{\pm}1.16\;cm$, wet weight = $158{\pm}25\;g$) was measured with an automatic intermittent-flow-respirometer. OCR decreased significantly when experimental fish were exposed to NaCN. When exposed to 10 ppb NaCN, fish were able to recover their OCR rhythmic activities. When fish were exposed to 20 ppb, however, the metabolic activity rhythms were not recovered. These results suggest that exposure to NaCN concentrations over 20 ppb cause severe physiological damage to the endogenous rhythms of black rockfish.

Comparison of Xenobiotic Metabolism and Antioxidative Status in Cultivated and Stocked Rockfish Sebastes schlegeli (양식산과 방류산 조피볼락(Sebastes schlegeli)의 간장 중 해독 및 항산화 효소계의 비교)

  • This study compared the composition and texture of muscle and levels of detoxification and antioxidant enzymes in the livers of stocked and cultivated rockfish Sebastes schlegeli released after the primary culture stage in Tongyoung, South Korea. The crude lipid content of muscle was significantly higher ($P$<0.05) in cultivated rockfish than stocked rockfish, while the texture did not differ significantly ($P$>0.05). The condition factor (CF) and hepatosomatic index (HSI) did not differ significantly and the growth of stocked and cultivated rockfish was similar. The levels of the detoxification enzymes cytochrome P450 (CYP) and Ethoxyresorufin-O-deethylase (EROD) were significantly lower in the livers of stocked rockfish, perhaps because of their reduced exposure to xenobiotic compounds. In addition, stocked rockfish had a significantly ($P$<0.05) lower CAT and higher GR than cultivated rockfish, but similar levels of tGPx, SOD, GSH, and GSSG. The present study shows that the growth rates of stocked and cultivated rockfish are similar and that stocked rockfish are exposed to fewer xenobiotic compounds and less oxidative stress.

Effect of formalin killed vaccine of atypical Aeromonas salmonicida for black rockfish (Sebastes schlegeli) (조피볼락(Sebastes schlegeli)에 대한 비정형 Aeromonas salmonicida 포르말린 사균 백신의 효과)

  • Atypical furnculosis caused by atypical Aeromonas salmonicida, is an emerging problem of farming of rockfish (Sebastes schlegeli) in Korea. In this study, protection against atypical furunculosis was compared in rockfish vaccinated with A. salmonicida formalin killed cell (FKC) and adjuvant containing FKC. The formalin inactivated A. salmonicida vaccine provided a low protection of 20% and 10% relative percent survival (RPS) at 44 and 58 days post vaccination. However, addition of adjuvant (squalene and aluminum hydroxide) into inactivated A. salmonicida vaccine clearly enhanced the level of protection showing 70% and 50% RPS at 44 and 58 days post vaccination.

  • The purpose of this research is to evaluate the effects of a commercial mixed extruded pellet(EP) and raw fish moist pellet(MP) diet on diseases prevalence and serological constituents and HSI of korean rockfish, Sebastes schlegeli grown in two commercial-scale aquaculture farms from June to December in 2009. The growth of MP-fed rockfish was slightly faster than EP-fed rockfish during experimental period, but did not show significant differences. The levels of TCHO, TG, Glucose and TP in the MP-fed rockfish group were higher than those of EP-fed group, whereas there was no significant difference in other biochemical factors of blood between two groups. In addition, bacteria and virus were not detected in both MP- and EP-fed rockfish groups, and the infection rates of gill fluke between two groups did not show clear difference. In conclusion, there was no comparable difference in growth and the health extents of one year rockfish by administration of the commercial mixed extruded pellet(EP) and raw fish moist pellet(MP) diets.

Critical Thermal Maximum (CTM) of Cultured Black Rockfish, Sebastes schlegeli

  • The critical thermal maximum (CTM) of black rockfish, Sebastes schlegeli, was evaluated. Black rockfish were acclimated at $24^{\circ}C$, and then exposed to temperatures from 24 to $33^{\circ}C$. Black rockfish were kept in constant darkness and subjected to a gradual temperature increase $(1 ^{\circ}C\;12^{-1})$. The oxygen consumption rate (OCR) was measured using an automatic intermittent­flow-respirometer (AIFR) during the exposure period (from 119.3 to 143.5 h). The OCR increased from 94.5 to 214.2mL $O_2 kg^{-1}\;ww\;h^{-1}$ as the temperature rose from 24 to $29.4-30.9^{\circ}C$. Subsequently, the OCR increased abruptly, reaching 245.8-412.7mL $O_2 kg^{-1}\;ww\;h^{-1}$ at $32^{\circ}C$. This study suggests that the CTM for black rockfish is $29.4-30.9^{\circ}C$ when temperature is increased at $1^{\circ}C\;12h^{-1}$ following acclimation at $24^{\circ}C$.

Activities of non-specific defense factors in cultured oblong rockfish(Sebastes oblongus) and rockfish(S. schlegeli) (양식 볼락류에서 비특이적 방어인자의 활성)

  • To understand the activity of non-specific defence factors in cultured Sebastes, the antibacterial effect of the serum, skin mucus and homogenate of various organs from cultured oblong rockfish (Sebastes oblongus) and rockfish(Sebastes schlegeli) against pathogenic bacteria, Aeromonas hydrophila, Edwardsiella tarda, Vibrio anguillarum, and Streptococcus sp. was compared with that of flounder(Paralichthys olivaceus) and seabass(Leteolabrax japonicus). And the activities of proteolytic enzyme, chitinolytic enzyme and haemolycin as non-specific defence factor were investigated on the oblong rockfish and rockfish. Samples from oblong rockfish showed the highest antibacterial activity by lysoplate assay on agar plate mixed with pathogens, followed in descending order by rockfish, seabass, and flounder. Turbidimetric assay was carried to evaluate the lysozyme activity of fish samples against lyophilized cells of Micrococcus lysodeiktikus. The serum, kidney, liver, stomach, intestine and eyeball of oblong rockfish and the mucus and gill of rockfish appeared to have the highest lysozyme activity among the fish strains investigated. All samples except skin mucus, liver, and eyeball of oblong rockfish and rockfish showed proteolytic enzyme activity. Chitinolytic enzyme activity was showed in random sampling and haemolytic activity was remarkable in oblong rockfish. Therefore, Sebastes strain was proved to have effective defense mechanisms based on the antibacterial activities, and lysozyme, proteolytic enzyme, chitinolytic enzyme, and haemolycin were considered to act as the non-specific defence factor of Sebastes.
