Hydrolysis of Agricultural Residues and Kraft Pulps by Xylanolytic Enzymes from Alkaliphilic Bacillus sp. Strain BK

  • An alkaliphilic bacterium, Bacillus sp. strain BK, was found to produce extracellular cellulase-free xylanolytic enzymes with xylan-binding activity. Since the pellet-bound xylanase is eluted with 2% TEA from the pellet of the culture, they contain a xylan-binding region that is stronger than the xylan-binding xylanase of the extracellular enzyme. The xylanases had a different molecular weight and xylan-binding ability. The enzyme activity of xylanase in the extracellular fraction was 6 times higher than in the pellet-bound enzyme. Among the enzymes, xylanase had the highest enzyme activity. When Bacillus sp. strain BK was grown in pH 10.5 alkaline medium containing xylan as the sole carbon source, the bacterium produced xylanase, arabinofuranosidase, acetyl esterase, and $\beta$-xylosidase with specific activities of 1.23, 0.11, 0.06, and 0.04 unit per mg of protein, respectively. However, there was no cellulase activity detected in the crude enzyme preparation. The hydrolysis of agricultural residues and kraft pulps by the xylanolytic enzymes was examined at 50$^{\circ}C$ and pH 7.0. The rate of xylan hydrolysis in com hull was higher than those of sugarcane bagasse, rice straw, com cop, rice husk, and rice bran. In contrast, the rate of xylan hydrolysis in sugarcane pulp was 2.01 and 3.52 times higher than those of eucalyptus and pine pulp, respectively. In conclusion, this enzyme can be used to hydrolyze xylan in agricultural residues and kraft pulps to breach and regenerate paper from recycled environmental resources.

벼 상자육묘에서 규산코팅볍씨의 건묘육성과 벼키다리병 경감효과 (Effect of Silicate-Coated Rice Seed on Healthy Seedling Development and Bakanae Disease Reduction when Raising Rice in Seed Boxes)

  • 벼 직파재배에서 새 피해, 발아 및 입모불량 그리고 도복피해 등 기술보급저해요인을 해소하기 위하여 개발된 규산 코팅볍씨의 이앙용 상자육묘에서 건묘육성과 벼키다리병 발병 경감을 구명하기 위하여 호기조건인 상자육묘조건과 혐기조건인 Pot이앙조건으로 수행한 결과는 다음과 같다. 1. 코팅볍씨에서 묘의 출현은 무코팅볍씨에서 보다 2-3일 빨랐고 입고병과 벼키다리병 발생이 현저히 경감되었다. 2. 파종후 45일 생체중은 건전묘에서 11%, 이병묘에서 2.01배로 규산코팅볍씨의 건묘 육성효과가 뚜렷하였다. 3. 육묘 중 파종후 80일까지 벼키다리병 발생은 무코팅볍씨 91.6%에 비하여 7.8%로 현저하게 경감되었다. 4. 최대발병률을 보인 파종 후 45일에 이앙된 코팅볍씨에서 무코팅볍씨에 비하여 건전묘의 추가발병이 거의 없었고 이병묘의 정상생육 회복도 가능하였다. 5. 코팅볍씨에서 육묘된 토양과 식물체의 뿌리와 엽초기부조직에서 활동성 소형포자와 대형포자의 분포가 무코팅에서보다 현저하게 줄었다. 특히 코팅볍씨의 육묘토양과 이병묘/건전묘에서는 무코팅볍씨에서 나타난 전형적인 대형포자(3-7개의 격막과 양끝이 낫처럼 굽은)와는 다른 격막이 없고 두터운 세포벽을 갖는 장방형 미성숙 대형포자 출현이 발병 경감원인으로 주목되었다. 6. 이상의 결과를 종합하여 보면 코팅볍씨의 육묘 중 벼키다리병 발병경감과 이앙 후 이병묘의 정상생육 회복 그리고 건전묘의 이병화경감은 강알칼리성 수용성 규산과 다공성 지오라이트 그리고 종피 잠복 병원균 간의 물리화학적 특성과 건전묘의 균에 대한 저항성에 기인되었을 것으로 판단되었다.

Chitinolytic and Chitosanolytic Activities from Crude Cellulase Extract Produced by A. niger Grown on Apple Pomace Through Koji Fermentation

  • Enzyme extracts of cellulase [filter paper cellulase (FPase) and carboxymethyl cellulase (CMCase)], chitinase, and chitosanase produced by Aspergillus niger NRRL-567 were evaluated. The interactive effects of initial moisture and different inducers for FP cellulase and CMCase production were optimized using response surface methodology. Higher enzyme activities [FPase $79.24{\pm}4.22$ IU/gram fermented substrate (gfs) and CMCase $124.04{\pm}7.78$ IU/gfs] were achieved after 48 h fermentation in solid-state medium containing apple pomace supplemented with rice husk [1% (w/w)] under optimized conditions [pH 4.5, moisture 55% (v/w), and inducers veratryl alcohol (2 mM/kg), copper sulfate (1.5 mM/kg), and lactose 2% (w/w)] (p<0.05). Koji fermentation in trays was carried out and higher enzyme activities (FPase $96.67{\pm}4.18$ IU/gfs and CMCase $146.50{\pm}11.92$ IU/gfs) were achieved. The nonspecific chitinase and chitosanase activities of cellulase enzyme extract were analyzed using chitin and chitosan substrates with different physicochemical characteristics, such as degree of deacetylation, molecular weight, and viscosity. Higher chitinase and chitosanase activities of $70.28{\pm}3.34$ IU/gfs and $60.18{\pm}3.82$ to $64.20{\pm}4.12$ IU/gfs, respectively, were achieved. Moreover, the enzyme was stable and retained 92-94% activity even after one month. Cellulase enzyme extract obtained from A. niger with chitinolytic and chitosanolytic activities could be potentially used for making low-molecular-weight chitin and chitosan oligomers, having promising applications in biomedicine, pharmaceuticals, food, and agricultural industries, and in biocontrol formulations.

Optimization of Laccase Production from Bacillus sp. PK4 through Statistical Design of Experiments

  • Statistical design of experiments was employed to optimize the media composition for the production of laccase from Bacillus sp. PK4. In order to find the key ingredients for the best yield of enzyme production from the selected eleven variables viz yeast extract, glucose, zinc sulphate, copper sulphate, potassium chloride, magnesium sulphate, calcium chloride, ferrous sulphate, sodium chloride, potassium dihydrogen phosphate ($KH_2PO_4$) and dipotassium hydrogen phosphate ($K_2HPO_4$), Plackett-Burman design was applied. The $MgSO_4$, $FeSO_4$, and $CuSO_4$ showed positive estimate, and their concentration optimized further. The steepest ascent method and Box-Behnken method revealed that 1.5 mM $MgSO_4$, 0.33 g/l $FeSO_4$ and 1.41 mM $CuSO_4$ were optimal for the laccase production by Bacillus sp. PK4. This optimization strategy leads to enhancement of laccase production from 2.13 U/ml to 40.79 U/ml. Agro-wastes residues replace the carbon source glucose in the optimized media namely sugarcane bagasse, wheat bran, rice husk, and groundnut shell, among these groundnut shells (117 U/ml) was found to enhance the laccase production significantly. The laccase produced by Bacillus sp. PK4 was found to have the potential to degrade persistent organic pollutant benzo[a]pyrene.

Development of fiber reinforced self-compacting concrete (FRSCC): Towards an efficient utilization of quaternary composite binders and fibers

  • This study has been carried out in two-phases to develop Fiber Reinforced Self-Compacting Concrete (FRSCC) performance. In the first phase, the composition of the quaternary composite binder compromised CEM I 42.5N (58-70%), Rice Husk Ash (25-37%), quartz sand (2.5-7.5%) and limestone crushing waste (2.5-7.5%) were optimized. And in the second phase, the effect of two fiber types (steel brass-plated and basalt) was investigated on the SCC optimized with the optimum CB as disperse reinforcement at 6 different ratios of 1, 1.2, 1.4, 1.6, 1.8, and 2.0% by weight of mix for each type. In this study, the theoretical principles of the synthesis of self-compacting dispersion-reinforced concrete have been developed which consists of optimizing structure-formation processes through the use of a mineral modifier, together with ground crushed cement in a vario-planetary mill to a specific surface area of 550 m2 / kg. The amorphous silica in the modifier composition intensifies the binding of calcium hydroxide formed during the hydration of C3S, helps reduce the basicity of the cement-composite, while reducing the growth of portlandite crystals. Limestone particles contribute to the formation of calcium hydrocarbonate and, together with fine ground quartz sand; act as microfiller, clogging the pores of the cement. Furthermore, the results revealed that the effect of fiber addition improves the mechanical properties of FRSCC. It was found that the steel fiber performed better than basalt fiber on tensile strength and modulus of elasticity; however, both fibers have the same performance on the first crack strength and sample destruction of FRSCC. It also illustrates that there will be an optimum percentage of fiber addition.

실규모 심부화재 소화특성에 관한 연구 (A Study of Fire Extinguishment Characteristic for the Real Scale Deap-Seated Fire)

  • 본 연구는 침윤소화약제의 소화성능평가를 위해 실규모 소화실험을 수행하였다. 실험은 '수동식소화기의 형식승인 및 검정기술기준'에 명시되어 있는 A급 소화실험 방법에 따라 진행하였으며, 목재 외에도 목분과 왕겨를 대상으로 실험을 진행하였다. 소화용수는 물과 국내에서 사용되고 있는 3종의 침윤소화약제를 사용하여 성능평가 실험을 진행함으로써 물과 침윤소화약제의 명확한 변별력을 확인하고자 하였다. 실험 결과 물의 경우에서 내부 온도의 유지시간이 길게 유지되고 침윤소화약제의 경우 빠른 온도 하강을 나타냄을 확인하였으며, 시간에 따른 온도분포를 통해 소화능력의 변별력을 확인하였다. 본 실험 결과를 바탕으로 추후 최적화 침윤소화약제 성능평가 방법을 제시하는데 기초자료로 활용할 수 있을 것으로 판단된다.

혼화재 종류 및 활성황토 대체율별 콘크리트의 공학적 특성에 관한 실험적 연구 (Experimental Study on the Properties of Concrete by the Kinds of Admixture and the Replacement Ratios of Activated Hwangtoh)

  • 본 연구에서는 천연재료인 황토의 활용성을 넓히기 위해, 황토를 혼화재로 사용한 경우 혼화재 종류 및 황토 대체율에 따른 콘크리트의 기초물성을 비교.검토하였다. 연구 결과, 황토를 사용할 경우 유동성은 다소 저하하는 것으로 나타났으며, 혼화재를 대체하지 않은 콘크리트에 비해 공기량도 다소 저하하는 것으로 나타나, 고성능 감수제의 선택성 및 AE제의 사용에 의한 공기량 보정이 필요할 것으로 판단된다. 황토와 시멘트간의 포졸란반응에 의한 강도발현성상을 검토한 결과, 황토를 콘크리트용 혼화재로서 사용하기 위해서는 가열에 의한 활성화공정이 필요한 것으로 나타났으며, 활성황토를 콘크리트용 혼화재료로 사용할 경우 활발한 포졸란반응에 의해 압축 강도 발현이 플레인콘크리트보다 높게 나타나, 건설 신재료로서의 황토의 개발이 가능할 것으로 사료된다. 활성황토의 대체율에 따른 콘크리트의 특성은 대체율이 증가할수록 강도발현수준이 높게 나타났으나, 대체율 30%의 경우 무혼입에 비해 강도가 저하하는 것으로 나타나, 활성황토의 적정 대체율에 대한 보다 심도 있는 연구가 필요할 것으로 사료되며, 본 연구의 범위에서 활성황토의 적정 대체율의 범위는 10~20%내외에 존재할 것으로 사료된다.

  • 바이오매스계 연료의 고위발열량(HHV) 예측을 성분원소분석 데이터와 제안된 대표적인 상관관계식을 이용하여 수행하였고, 각 상관식에 대한 그 타당성을 실측 HHV와 비교하여 검토하였다. 실험에 사용된 샘플들은 산림계 바이오매스인 노송나무의 톱밥과, 농업계 바이오매스인 왕겨, 그리고 여기에 유기성 폐기물인 polystyrene, waste paper, polypropylene을 각각 10, 30, 50wt%로 혼합하여 제작하였다. 성분원소의 측정과 HHV의 측정은 각각 KS 표준방법에 따랐으며, 예측된 HHV값의 평균 오차는 약 880 kJ/kg으로 이는 측정된 HHV의 약 3.8%에 해당하였다. 실험을 통한 값과 예측된 HHV 값 사이의 대응 상관계수의 곱($R^2$)은 $0.957{\sim}0.996$의 값을 나타내었다.

  • 본 연구에서는 자연분해재 펠릿과 polyethylene들을 이용해 LDPE와 신장율과 인장강도가 유사한 생붕괴성 플라스틱 필름들을 제조하였다. 식품 포장재로서의 안전성, 광분해성, 열분해성 및 곰팡이 생분해성을 평가한 결과 제작된 생붕괴성 플라스틱 필름들은 식품 포장재로 사용하기에 안전하고 LDPE 필름에 비해 상대적으로 우수한 분해 특성을 나타내어 환경친화적인 식품 포장 소재로 사용될 가능성을 보여주었다.

Assessment of flowing ability of self-compacting mortars containing recycled glass powder

  • This paper investigates the effect of recycled glass powder (RGP) on flowing properties of self-compacting mortars (SCMs) containing different ratios of fillers and superplasticizer dosages. Fly ash (FA), nano-silica (NS), micro-silica (MS), metakaolin (MK) and rice husk ash (RHA) are used as fillers and their synergistic effect with RFP is studied. The effects of fillers and high-range water reducer (HRWR) on flowing ability of mortars are primarily determined by slump flow and V-funnel flow time tests. The results showed that for composites with a higher RGP content, the mortar flowing ability increased but tended to decrease when the composites containing 10% MK or 5% RHA. However, the flowing ability of samples incorporating 5% RGP and 10% SF or 25% FA showed an opposite result that their slump flow spread decreased and then increased with increasing RGP content. For specimens with 3% NS, the influence of RGP content on flowing properties was not significant. Except RHA and MS, the fillers studied in this paper could reduce the dosage of HRWR required for achieving the same followability. Also, the mixture parameters were determined and indicated that the flowability of mixtures was also affected by the content of sand and specific surface area of cement materials. It is believed that excess fine particles provided ball-bearing effect, which could facilitate the movement of coarse particles and alleviate the interlocking action among particles. Also, it can be concluded that using fillers in conjunction with RGP as cementitious materials can reduce the material costs of SCM significantly.
