Response of grain yield and milled-rice protein content to nitrogen topdress (N) timing at panicle initiation stage (PIS) is critical for quantifying real-time N requirement for target grain yield and milled-rice protein content. Two split-split-plot experiments with three replications, one in 2004 and the other in 2005, were conducted in Experimental Farm, Seoul National University, Suwon, Korea. The experiments included three N rates at tillering stage (TS), three N timing treatments at panicle initiation stage (PIS) and two rice cultivars. The N rates at TS, N timing at PIS, and rice cultivars were randomly assigned to main plot, sub plot, and sub-sub plot, respectively. Results showed that the delayed N application at PIS reduced grain yield in 2004 and increased milled-rice protein content in both years significantly at 0.05 probability level. The calculated optimum N timing at PIS from pooled data by N rates and rice cultivars in two years was at 28 days before heading (DBH). However, real-time of N timing at PIS was dependent on plant growth and N status around PIS that in turn was dependent on applied N rates at TS. The optimum N timing at PIS was at 30 DBH for no N treatments at TS while at 27 DBH for 3.6 and 7.2 kg N/10a treatments and at 27 and 29 DBH for Hwaseongbyeo and Daeanbyeo, respectively. In general, earlier applied N at PIS resulted in lower milled-rice protein content but the highest grain yield was expected to be obtained when N topdress at PIS was applied at the time when shoot N concentration started to drop below about 23 mg/g due to dilution effect after transplanting. In conclusion, the results of our experiments imply that the currently recommended N topdress time (24DBH) at PIS in Korea should be reconsidered for the higher grain yield and the better quality of rice.

The study was carried out to examine changes of ${\alpha}$-amylase activity and free proline contents during rice germination under low temperature($13^{\circ}C$). The plant height, root length, and germination rate were investigated during seed germination under the low temperature. Those growth chracteristics were the highest in Dongjin cultiver compared with other rice cultivars. The ${\alpha}$-amylase activity of the 9 days after germination was higher in Dongjin than those of Sangju and Koshihikari cultivars. The content of free proline was increased in all rice cultivars, when the germination of rice seeds was prolonged. As a result, it was concluded that ${\alpha}$-amylase activity and free proline content of Donjin cultivar were relatively increased higher under low temperature stress than those of other rice cultivars.
Bacterial leaf blight (BLB) caused by Xanthomonas oryzae pv. oryzae (Xoo) is one of the most serious threats to rice production. In this study, screening of rice for resistance to BLB was carried out at two different times and locations; that is, in a greenhouse during winter and in an open field during summer. The pathogenicity of Xoo race K1 was tested on 32 Korean rice cultivars. Inoculation was conducted at the maximum tillering stage, and the lesion length was measured after 14 days of inoculation. Five cultivars, Hanareum, Namcheon, Samgdeok, Samgang, and Yangjo, were found to be resistant in both the greenhouse and open-field screenings. Expression of the plant defense-related genes JAmyb, OsNPR1, OsPR1a, OsWRKY45, and OsPR10b was observed in resistant and susceptible cultivars by qRT-PCR. Among the five genes tested, only OsPR10b showed coherent expression with the phenotypes. Screening of resistance to Xoo in rice was more accurate when conducted in open fields in the summer cultivation period than in greenhouses in winter. The expression of plant defense-related genes after bacterial inoculation could give another perspective in elucidating defense mechanisms by using both resistant and susceptible individuals.
The experiment was carried out to determine Al tolerance ranges of two rice cultivars(Ilpumbyeo and Hyangmibyeo) during germination and early growth. The amount of 0, 300, 600 and 900ppm Al were treated in the germination media. Plant height and root length of rice cultivars were decreased significantly with Al concentration. The reduction of growth was more remarkable in root than in shoot. Germination rates in all Al concentrations were decreased comparing with untreated control. Two rice cultivars were not different in Al concentrations. As Al concentration was increased, chlorophyll content were decreased. Ca and Mg contents in 300ppm of Al treatment were sharply increased. Free proline contents of both rice cultivars grown at the 300, 600, and 900ppm of Al treatments were increased only in the 3 days after Al treatments.
An insertion sequence identified as a solo long terminal repeat (LTR) of a new rice copia-like retrotransposon was detected in the ORE of the Pi-b gene from the rice cv. Nipponbare, and was designated as Osr1. Osr1 consists of a 6386 bp nucleotide sequence including 965 bp LTRs on both ends with an 82% nucleotide sequence identity to the wheat Tarl retrotransposon on reverse transcriptase. Nucleotide divergence was noted among the individual LTRs, as well as the coding region of Osr1. Various restriction fragment length polymorphism (RFLP) of LTR were detected in indica cultivars, whereas, only a few could be detected in the japonica cultivars. The population of Osr1 is lower in the wild-type rice compared with that in the domesticated cultivars. The insertion of LTR sequence in the h-b gene in the susceptible cultivar suggested that retro-tyansposon-mediated insertional mutation might play an important role in the resistance breakdown, as well as in the evolution of resistance genes in rice.
Rice seeds of 4 cultivars including Whachung-giant embryonic rice and Nampung-giant embryonic rice, as a group of the non-waxy rice cultivars, and Shinsunchal-giant embryonic rice and Whachungchal-giant embryonic rice, as that of the waxy rice cultivars, were germinated at $27^{\circ}C$ for 3 days to compare the changes in some physicochemical properties of the starch granules and the starch-hydrolysing enzyme activities during germination, respectively. ${\alpha}-Amylase$ activity of rices germinated for 3 days found to be higher than that of malt. Especially, Whachung-giant embryonic rice and Shinsunchal-giant embryonic rice were greater in activity than other rice cultivars and possessed the activities double that of malt. In contrast, ${\beta}-amylase$ of germinated rice found to be considerably less active than malt, although the giant embryonic rice group showed prevalent activity as compared o the normal rice group. With the starch granules, the amount of long glucose chains from amylose molecules were reduced in the non-waxy type giant embryonic rices, while the chain length increase was found in the waxy type giant embryonic rices. For the distribution profile of the glucose chain length from amylopectin molecules, we could observed that the chain length with DP (degree of polymerization) ranged 33 to 66 and 14 to 32 increased with the decreasing rate of that above 67 and below 13 regardless of starch waxiness. With non-waxy type of giant embryonic rices, susceptibility for glucoamylase were found to reduce along with germination, however, increase in susceptibility was observed with waxy rice types. In addition, we found the reduction in both initiation and termination temperature, and enthalpy for gelatinization.
With the development of diverse agricultures worldwide, biofortified rice noted for its preferable marketability and palatability plays an important role in the world's agricultural economics and rice breeding programs. In this report, several $M_5$ of T-DNA inserted lines derived from the donor cultivars, 'Hwayong' and 'Dongjin', were selected for high or low protein, high lipid and low amylose content, respectively. The coefficients and ranges of variation for the chemical constituents between $M_4$ and $M_5$ T-DNA inserted lines were evaluated in comparison with those of the donor varieties. Results indicated that T-DNA insertion might be an effective way to generate useful variations for chemical composition of rice grains which could be used for the development of biofortified rice cultivars.

This study was conducted to obtain the basic information on rice cold damage and relationship to be connected with cold tolerance. The 20 rice varieties were screened to cold tolerance under low water temperature condition. The water gradient were controlled to $17^{\circ}C$ at the inlet and $21^{\circ}C$ at outlet of screening field. In the relationship between cold water irrigation and growth of rice varieties, effect on medium maturing cultivars were higher than early maturing cultivars. In relationship of shorting of culm length and panicle exsertion, culm length and panicle exsertion showed positive correlation. Yield of cultivars and culm length and percent of fertile grain showed positive correlation. However, heading date presented negative correlation. In the early maturing cultivars, except 'Kumobyeo', all cultivars range of discoloration value (1-3) was resistant. In the medium maturing cultivars, 'Donghaebyeo' was resistant in both the seedling stage whereas 'Hwajinbyeo' was resistant only in tiller stage. Cold water irrigation reduced spikelet number per panicle and percent of filled grain. Unhulled rice yield was increased according to water temperature gradient from inlet to outlet. There was varietal differences in head rice recovery by cold water treatment.
The effects of NaCl salinity on the leaf photosynthesis and water relation of two cultivars of rice(Oryza sativa L.) , the salt-tolerant cultivar Seohae and the salt-senstive cultivar Iri-380 were exam-ined. Two cultivars of rice were grown for 14 days in nutrient solution at SOmM NaCl. Comparing theieaf Na content of two cultlvars, Seohae showed high accumulation of Na content in the leaf blade, while Iri-380 showed low. The Na content in leaf blade reduced the rate of leaf photosynthesis. Salt-tolerant cultivar Seohae was less decreased the rate of leaf photosynthesis than salt- sensitive cultivarIri-380. And Seohae showed larger decreased the osmotic potential in the leaves than Iri-380. This in-dicates that in the salt-tolerant cultivar, osmotic adjustment is developed under saliniEation.

The indica ${\times}$ japonica rice (Oryza sativa L.) cultivars released in Japan since 1980 are high-yielding. However, occasionally in these cultivars their high yield potentials cannot be realized, particularly depending on climate conditions. The objectives of this study were to identify the reason for yield instability and the critical yield component factor causing lower grain yield in these high-yielding cultivars. Standard-yield japonica, high-yielding japonica-dominant, and indica-dominant cultivars were grown in western Japan. Rough grain yield (RY) in these high-yielding cultivars ranged from 450 to $980g\;m^{-2}$, and was positively correlated with potential grain yield (PRY). By fluctuations of solar radiation, RY changed with spikelet number (SNO), and SNO was correlated with cumulative radiation during the panicle formation period of 30 days before heading. Even if higher SNO was achieved under higher radiant conditions, RY was lower than PRY. The lower grain-filling in plants bearing higher SNO resulted from a lower filling percentage of spikelets (%F, RY/PRY), and %F was strongly correlated with spikelet fertility (%S) across all cultivars. %S was highly influenced by cumulative radiation per PRY during pollen development and establishment around heading. Inhibition of assimilation by leaf removal lowered %S. Conversely, stem thinning and removal of upper panicles around heading increased %S in spikelets of the lower part of the panicle in which sterility was higher. These results suggest that limitation of assimilate-supply due to low irradiance at the spikelet-formation and flowering stages restrains the potential of the high-yielding cultivars, depending on reduction of SNO and %S, respectively.
