Molecular Identification and First Morphological Description of Larvae and Juveniles of Neosalanx anderssoni (Salangidae) Collected from the Southwestern Sea of Korea (한국 서해 남부해역에서 채집된 도화뱅어, Neosalanx anderssoni (뱅어과) 자치어의 분자 동정 및 첫 형태기재)

  • During ichthyoplankton survey in the southwestern sea of Korea, we collected six individuals of noodlefish larvae and juveniles between April and May 2023. They were identified as Neosalanx anderssoni by mitochondrial DNA cytochrome c oxidase subunit I or 16S ribosomal RNA sequences, and their external morphological traits were described for the first time. All six individuals have a slender and elongated body. When preflexion and flexion larval stages (10.24 mm notochord length, NL and 15.47 mm total length, TL, respectively), oval-shaped black melanophores were distributed in a row along the ventral side of the gut. However, when postflexion larval and juvenile stages (23.58~25.90 mm TL, and 29.20~31.26 mm TL, respectively), melanophores on the ventral side of the gut were disappeared, and dark spot-shaped melanophores appeared along the dorsal side of the gut in a single row. Also, from the postflexion larval stage (23.58 mm TL), two large black spots began to appear symmetrically on the caudal fin. Our results suggest that N. anderssoni may use coastal area as spawning and/or nursery ground unlike previous study (Kim and Park, 2002).

Identification of Antagonistic Bacteria, Pseudomonas aurantiaca YC4963 to Colletotri­chum orbiculare Causing Anthracnose of Cucumber and Production of the Antibiotic Phenazine-l-carboxylic acid (Colletotrichum orbiculare에 대한 길항세균 Pseudomonas aurantiaca YC4963의 분리 동정 및 항균물질 Phenazine-1-carboxylic acid의 생산)

  • A bacterial strain YC4963 with antifungal activity against Colletotrichum orbiculare, a causal organism of cucumber anthracnose was isolated from the rhizosphere soil of Siegesbeckia pubescens Makino in Korea. Based on physiological and biochemical characteristics and 16S ribosomal DNA sequence analysis, the bac­terial strain was identified as Pseudomonas aurantiaca. The bacteria also inhibited mycelial growth of several plant fungal pathogens such as Botrytis cinerea, Fusarium oxysporum and Rhizoctonia solani on PDA and 0.1 TSA media. The antifungal activity was found from the culture filtrate of this isolate and the active compound was quantitatively bound to XAD adsorption resin. The antibiotic compound was purified and identified as phenazine-l-carboxylic acid on the basis of combined spectral and chemical analyses data. This is the first report on the production of phenazine-l-carboxylic acid by Pseudomonas aurantiaca.

Development of Species-Specific PCR to Determine the Animal Raw Material (종 특이 프라이머를 이용한 동물성 식품원료의 진위 판별법 개발)

  • In this study, the detection method was developed using molecular biological technique to distinguish authenticity of animal raw materials. The genes for distinction of species about animals targeted at Cytochrome c oxidase subunit I (COI), Cytochrome b (Cytb), and 16S ribosomal RNA (16S rRNA) genes in mitochondrial DNA. The species-specific primers were designed by that Polymerase Chain Reaction (PCR) product size was around 200 bp for applying to processed products. The target 24 raw materials were 2 species of domestic animals, 6 species of poultry, 2 species of freshwater fishes, 13 species of marine fishes and 1 species of crustaceans. The results of PCR for Rabbit, Fox, Pheasant, Domestic Pigeon, Rufous Turtle Dove, Quail, Tree Sparrow, Barn Swallow, Catfish, Mandarin Fish, Flying Fish, Mallotus villosus, Pacific Herring, Sand Lance, Japanese Anchovy, Small Yellow Croaker, Halibut, Jacopever, Skate Ray, Ray, File Fish, Sea Bass, Sea Urchin, and Lobster raw materials were confirmed 113 bp ~ 218 bp, respectively. Also, non-specific PCR products were not detected in compare species by species-specific primers. The method using primers developed in this study may be applied to distinguish an authenticity of food materials included animal raw materials for various processed products.

Comparative Analysis of Mitochondrial Genomes of the Genus Sebastes (Scorpaeniformes, Sebastidae) Inhabiting the Middle East Sea, Korea (한국 동해 중부해역에 서식하는 볼락속(Sebastes) 어류의 미토콘드리아 유전체 비교분석)

  • Sebastes minor, Sebastes trivittatus, Sebastes owstoni, and Sebastes steindachneri are indigenous fish species inhabiting the central part of the East Sea, Korea. In order to understand the molecular evolution of these four rockfishes, we sequenced the complete mitochondrial genomes (mitogenomes) of S. minor and S. trivittatus. To further analyze the phylogeny of Sebastes species, the mitogenomes of 16 rockfishes were comparatively investigated. The complete mitochondrial DNA (mtDNA) nucleotide sequences of S. minor and S. trivittatus were 16,408 bp and 16,409 bp in length, respectively. A total of 37 genes were found in mtDNA of S. minor and S. trivittatus, including 13 protein-coding genes, 2 ribosomal RNA genes, and 22 transfer RNA genes, which exhibited similar characters with other Sebastes species in the East Sea, Korea. In addition, we detected a conserved motif "ATGTA" in the control region of the four Sebastes species, but no tandem repeat units. Comparative analyses of the congeneric mitochondrial genomes were performed, which showed that control regions were more variable than the concatenated protein-coding genes. As a result of analysing phylogenetic relationships of four Sebastes species by using concatenated nucleotide sequences of 13 protein-coding genes, S. minor, S. trivittatus, S. owstoni and S. steindachneri were clustered into three clades. The phylogenetic tree exhibited that S. minor and S. steindachneri shared a closer relationship, whereas S. trivittatus and S. vulpes formed another distinct clade. Our results contribute to a better understanding of evolutionary patterns of Sebastes species inhabiting the middle East Sea, Korea.

ITS Sequence Variations in Populations of Ilex cornuta (Aquifoliaceae) (호랑가시나무(Ilex cornuta) 개체군의 ITS 염기서열 변이 분석)

  • Genetic variation of Ilex cornuta Lindley et Paxton was examined by sequence analyses of ITS for 65 individuals from Korea and China. The length of ITS 1 ranged from 253 to 259 bp. The 5.8S was 159 bp and ITS2 was observed to be 231 bp. A total of 8 different ITS types (Single Nucleotide Polymorphism haplotypes), which showed the difference of 1 - 6 bp, were detected from 65 individuals. The sequence polymorphisms of ITS appeared at 9 different sites. All of four individuals collected at Daejeong-eup in Jeju-do exhibited different types, but all individuals from Naju-si and Muan-gun in Jeollanam-do were identical. The variation of ITS was higher in Jeju-do population than in inland population. Since I. cornuta contains various types of ITS sequences, ITS analyses will provide important information on genetic diversity and conservation of this species.

Occurrence of Brown Patch on Kentucky Bluegrass Caused by Rhizoctonia solani AG-1 IB (Rhizoctonia solani AG-1 IB에 의한 Kentucky Bluegrass 갈색잎마름병 발생)

  • Brown patch caused by Rhizoctonia solani AG-1 IB occurred on Kentucky bluegrass during late May through early October 2010 at golf course in Gyeongbuk Province, Korea. Disease symptoms on the turfgrass for spring season were leaf blights dying from the leaf tip, which appeared patches of brown color in the field. However, it appeared patches of dark brown color or gray brown color in fall. The fungus (B-7 isolate) of brown patch was isolated from the diseased leaf tissue and cultured on potato-dextrose agar (PDA) for identification. The young hyphae had acute angular branching and few septa and mature hyphal branches showed about 90-degree angles and development of monilioid cells, which were morphologically identical to Rhizoctonia solani AG-1 IB reported previously. DNA sequences of ribosomal RNA gene (internal transcribed spacer) of the fungus were homologous with similarity of 99% to those of Rhizoctonia solani AG-1 IB isolates in GenBank database, confirming the identity of the causal agent of the disease. Pathogenicity of the fungus was also confirmed on the creeping bentgrass and Kentucky bluegrass by Koch's postulates. This is the first report of brown patch on Kentucky bluegrass caused by Rhizoctonia solani AG-1 IB in Korea.

Rhizopus Soft Rot on Lily Caused by Rhizopus oryzae in Korea (Rhizopus oryzae에 의한 백합 무름병)

  • Rhizopus soft rot of lily (Lilium longiflorum) caused by Rhizopus oryzae was observed in the experimental field in Taean Lily Experiment Station in Korea, 2012. The typical symptoms were water-soaked lesions on bottom stem and leaf rot. The lesion rapidly expanded and the plant was softened totally. The fungus grew vigorously at an optimum temperature ($25^{\circ}C$) and brownish colony and black sporangia were formed on potato dextrose agar medium. Sporangiophores formed on end of sporangia were sub-globose, brownish and $6-10{\mu}m$ in size. Sporangia were globose, blackish and $87-116{\mu}m$ in size. Sporangiospores were irregularly oval and sub-globose, brownish $4-8{\mu}m$ in size. On the basis of mycological characteristics, analyzing sequences of internal transcribed spacer region of ribosomal DNA, and pathogenicity test on host plants, the causal fungus was identified as R. oryzae. This is the first report of Rhizopus soft rot on lily caused by R. oryzae in Korea.

Phylogenetic Diversity and Antibacterial Activity in Bacterium from Balloon Fish (Diodon holocanthus) of Jeju Island (제주 연안의 가시복(Diodon holoanthus)에서 분리된 세균의 다양성 및 항균활성 효과)

  • Over the past 20 years, global warming has transformed the marine ecosystem of the Jeju Island into a subtropical zone making it conducive to the production of tropical fishes. Recently, the balloon fish (Diodon holoanthus) has been found off the coast of the Jeju Island. In this study, we analyzed the diversity of its intestinal microorganisms as a representative for the surrounding environment. In addition, the isolates were evaluated for their antibacterial activity. A total of 161 strains of various species were identified and isolated using 16S ribosomal RNA gene sequence analysis. They were separated into three groups, of which Phylum Proteobacteria was found to be the most dominant with 91% sequence similarity. This includes the class γ-proteobacteria that is made up of twelve genera and twenty-four hundred species. The second group comprised strains of the genus Vibrio, made up of 35% Photobacteria, 32% Shewanella, and 6% Psychrobacter. It was also determined that 4% of the isolates were Acinetobacter, 3% were Enterovibrio, while Moraxella_g2 accounted for 1% of the total isolates. Class α-proteobactera includes five genera and five species; Brevundimonas, Allorhizobium, Pseudoceanicola and Erythrobcter, each accounting for 1% of the total isolates. The Firmicute strains belonged to six genera and ten species. 5% of the strains were Terribacillus, while Paenibacillus, Salinicoccus, Staphylococcus and Streptococcus accounted for 1% each of the total isolates. Actinobacteria accounted for the final phylum with strains belonging to three genera and ten species with Janibacter, Micrococcus and Isoptericola each accounting for 1% of the total isolates.

Phylogenetic Relationships between the Genus Inonotus and its Related Genera Based on the Nucleotide Sequences of Internal Transcribed Spacers (ITS 염기서열에 기초한 차가버섯과 근연속간 유연관계분석)

  • In this study the ITS1, ITS2 and 5.8S ribosomal DNA sequences from 29 strains of the Genus Inonotus and its related genera were compared with 31 strains obtained from GenBank database. Using the neighbor-joining (NJ) method and most parsimonious analysis the phylogenetic tree was constructed. The hymenochaetales formed no monophyletic group and several non-hymenochaetales appeared as intermingled with the Hymenochaetales. Strains 6, 46, 49, 50, 53, 55 showed no certain affinities within the Hymenochaetales, whereas Inonotus sp. (51) was closely related to Phellinus baumii, and Inonotus sp. (52), and Inonotus glomeratus (10) was related to Phellinus linteus, and Fomes fomentarius (30) was related to Ganoderma lucidum. Inonotus sp. and Phellinus sp. formed no monophyletic groups and a subdivision in the following genera is accepted: Inonotus sp. Phellinus baumii, Phellinus linteus, Phellinus igniarius, Phellinus pini, Hericium erinaceum, Ganoderma lucidum and Sparassis sp. were confirmed and separated genera. The taxonomic status of Inonotus remained uncertain. Eight new combinations are proposed.
