이담자균 효모 Rhodosporidium toruloides에서 Rhodotorucine A에 의한 막단백질 인산화의 저해와 Trigger Peptidase의 관련성

  • [$\gamma$-$^{32}$P]ATP was used to test phosphorylation of membrane proteins of mating type a cells of heterobasidiomycetous yeast Rhodosporidium toruloides separated by non-denaturing electrophoresis. The phosphoprotein was observed in the membrane proteins. The phosphorylation was inhibited by the pheromone rhodotorucine A (Rh. A) secreted by mating type A of the yeast. Rh. A didn't inhibit the phosphorylation in the presence of a trigger peptidase (TPase) inhibitor, antipain. Partially digested Rh. A by trypsin maintained the phosphorylation inhibitory activity. These results show that TPase activity plays an important role in the transduction of pheromone signal in the yeast.

  • A yeast strain utilizing styrene epoxide as a sole carbon and energy source was isolated from soil samples for the production of enantiopure of styrene epoxide by kinetic resolution. The strain, identified as a Rhodosporidium toruloides SJ-4, expressed an epoxide hydrolase which preferentially converted (R)-styrene epoxide into the corresponding diol. A maximum activity of 135 U/L was observed when biomass (dry cell mass) reached 6.7 g/L at 21 h of batch culture. Under the partially optimized reaction conditions ($35^{\circ}C$ and pH 8.0), the optically pure (S)-styrene epoxide was obtained with the yield of 21% when the initial substrate concentration was 100 mM. The reaction was completed at 9 h.

  • When mating type A and a cells of heterobasidiomycetous yeast Rhodosporidium toruloides were mix-cultured, both of the mating type cells have shown strong agglutination. But this agglutination was not detactable when the A and a cell were cultured separately. From reagglutination made just after the result of disassembling the agglutination by sonication, we knew that the agglutination was sexual-agglutination, not simple physical cell agglutination. The sexual agglutination was progressed actively on logarithmic phase and, in addition, progressed faster on mating type a cell treated with rhodotorucine A. These sexual agglutination have been inhibited by several protease such as trypsin, pronase, chymotrpysin and thermolysin and inhibited by 5 mM DTT as well.

  • Microorganisms isolated from soil (150 strains), fungi (39 strains), yeasts (9 strains) and Streptomyces species (39 strains) were assayed for L-phenylalanine ammonia-lyase(PAL) activity. 17 strains of fungi and 46 strains of soil isolates were proved to produce PAL, Aspergillus panamensis, Penicillium varioti and 11 soil isolates showed comparatively large PAL activity. When PAL activity was assayed with cell-free extracts of these 13 strains and 7 strains of Rhodotorula and Rhodosporidium geni, Rhodosporidium toruloides (IFO 0559) showed the highest PAL activity with 0.333 units per g of the wet cell weight.

Phylogenetic Analysis of Genus Sporobolomyces Based on Partial Sequences of 26S rDNA

  • The sequences of the D1/D2 region of 26S rDNA from seven Sporobolomyces species, Bensingtonia subrosea, and Rhodosporicium toruloides were determined and compared with those from representatives of the genera Leucosporidium, Rhodosporidium, Rhodotorula, and Sporidiobolus. The five species of Sporobolomyces analyzed were distantly related to a monophyletic clade consisting of species of Sporidiobolaceae and Sporobolomycetaceae. Sporobolomyces falcatus was found to be closely related to Tremella exigua. The members of Sporidiobolaceae and Sporobolomycetaceae were divided into four groups. Group 1 was composed of Leucosporidium scottii and two Rhodotorula species, and group 2 contained three Rhodotorula species. Group 3 was designeate as the Sporobolomyces/Sporidiobolus core group, as it contained Sporidiobolus johnsonii, the type species of Sporidiobolus and the teleomorphic state of Sporobolomyces salmonicolor (the type species of Sporobolomyces). Group 4, named the Rhodotorula/Rhodosporidium core group, included Rhodosporidium toruloides and Rhodotorula glutinis, the type species of the genera Fhodosporidium and Rhodotorula, respectively. The four groups were differentiated on the basis of their physiological characteristics including the assimilation of D-glucosamine, glucuronate, 2-keto-gluconate, L-arabinitol, raffinose, methyl-$\alpha$-glucoside, and satrch. The taxonomy of the genera Leucosporidium, Rhodosporidium, Rhodotorula, Sporidiobolus, and Sporobolomyces will require a major revision when more data becomes available.

Purification and Characterization of Internal Invertase in Rhodosporidum toruloides Mating Type a Cells

  • The internal invertase of Rhodosporidium toruloids mating type a cells was purified to a single band on SDS-PAGE from cell-free extract by acid precipitation, ion exchange chromatogaphy andgel filtration. The determined molecular weight of he purified enzyme was about 95,000 by gel filtration and 100,000 daltons on SDS-polyacryamide gel electrophoresis. This enzyme didn't show any activity change by several metal ions except 15.4% decrease by {TEX}$Mn^{2+}${/TEX} and was strongly inhibited by 2-mercaptoethanol and SDS. The invertase maintained its activity at high level until 70℃, but inactivated at 80℃ almost completely. The optimal temperature and pH of the enzyme were about 60℃ and pH 5.0, respectively. The stable pH range of invertase was narrow from pH 3.0 to 6.0. The Km value and isoelectric point of enzyme were 3.4×{TEX}$10^{3}${/TEX} M, pH 4.4, respectively.

Rhodosporidium toruloides를 활용한 비트 열수추출물의 중성지방 억제효과 (Inhibitory Effects of Hot Water Extract of Beta vulgaris L. on Triglyceride Biosynthesis Using Rhodosporidium toruloides)

  • 본 연구에서는 Rhodosporidium toruloides를 활용하여 비트 열수추출물의 중성지방 억제효과를 확인하고자 하였다. 대조구, 실험대조구, 비트 열수추출물, 30% 및 60% 에탄올 추출물을 첨가한 YPD 배지에서 배양한 유지효모의 중성지방의 함량은 각각 8.76, 3.43, 2.87, 3.16 및 3.64 mg/dL로 나타났으며, 유지 효모의 세포수는 각각 10.48, 8.46, 12.40, 12.80 및 $8.24{\times}10^3cell/mL$로 나타났다. 대조구, 실험대조구, 0.02, 0.1 및 0.5% 비트 열수추출물을 첨가한 유지효모의 총 지질 함량은 각각 112.29, 38.90, 147.87, 211.36 및 291.89 mg/g로 나타났으며, 유지효모의 중성지방의 함량은 각각 6.02, 2.83, 2.38, 1.37 및 0.73 mg/dL로 나타났다. 대조구, 실험대조구, 0.02, 0.1 및 0.5% 비트 열수추출물의 첨가 농도가 높아짐에 따라서 유지효모내 소량의 형광이 발현됨을 확인하였다. 실험대조구, 0.02, 0.1 및 0.5% 비트 열수추출물을 첨가한 유지효모의 세포수 증가율은 대조구에 비해 각각 -4, 21, 82 및 103%로 증가하였다. 유리지방산과 총 카로티노이드 함량은 비트 열수추출물의 첨가 농도에 따라 농도의존적으로 증가하였다.

Rhodosporidium toruloides에서의 적색 파프리카 에탄올 추출물의 중성지방 억제 효과 (Inhibitory Effects of Ethanol Extract of Red Sweet Pepper (Capsicum annuum L.) on Triglyceride Biosynthesis in Rhodosporidium toruloides)

  • 본 연구는 유지효모 R. toruloides를 활용하여 적색 파프리카 추출물의 중성지방 억제 효과를 확인하기 위하여 수행되었다. 대조구, 양성대조구, 0.02, 0.1 및 0.5% 적색 파프리카 70% 에탄올 추출물을 첨가한 실험구의 총 지질 함량은 각각 80.45±2.40, 86.61±11.58, 79.82±2.80, 85.30±0.91 및 92.40±12.40 mg/g으로 나타났으며, 유지효모의 중성지방 함량은 각각9.89±0.36, 7.58±1.11, 7.28±0.17, 6.68±0.35 및 5.23±0.67 mg/dL으로 나타났다. 대조구, 양성대조구, 0.02, 0.1 및 0.5% 적색 파프리카 70% 에탄올 추출물을 첨가한 실험구의 지방소적 형성을 관찰한 결과, 적색 파프리카 70% 에탄올 추출물의 첨가농도가 증가함에 따라 대조구에 비해 소량의 형광이 관찰되는 것으로 보아 유지효모 내 생성되는 지방소적이 감소됨을 확인하였다. 대조구, 양성대조구, 0.02, 0.1 및 0.5% 적색 파프리카 70% 에탄올 추출물을 첨가한 실험구의 세포수는 각각 1.90±0.16, 2.17±0.25, 1.85±0.11, 2.28±0.21 및 2.49±0.03×108 cell/mL으로 나타났으며, 유리지방산의 함량은 각각2.44±0.05, 2.71±0.07, 2.46±0.16, 2.52±0.10 및 2.98±0.14 mg/g으로 나타났다. 본 연구결과에서 적색 파프리카 70% 에탄올 추출물의 첨가농도에 따른 유지효모 내 중성지방 억제 효과, 지방소적의 감소, 세포수 및 유리지방산의 함량이 증가한 결과를 통해 적색 파프리카 70% 에탄올 추출물이 중성지방 억제 효과에 유의적인 영향을 미치는 것으로 확인되었으며, 추후 활성 물질의 규명에 관한 추가적인 연구가 필요할 것으로 사료된다.

  • We have studied the relation between Ca$^{2+}$-ATPase and trigger peptidase(TPase) which are membeane protein well known as their significant role for signal transduction of mating pheromone in heterobasidiomycetous yeast. Rhodosporidium toruloides. We found out that there were Ca $^{2+}$-ATPase and TPase together in isolated calmodulim binding protein(CBP), usion calmodulin affinity column chromatography after solubilization of mation type a cell membrane protein, and that the dependence of enzyme activity of both the enzymes on Ca$^{2+}$, phospholipid and nonionic detergent are similar. However, Ca$^{2+}$-ATPase hed quite absolute dependence on calmodulin and, on the other hand, TPase didn't have any dependence. Judging from the fact that there are both enzymes in CBP which the dependence of calmodulin are quite different, we found out that both enzymes were made to their compound and existed in mating type a cell membrane.

이담자 효모균의 성접합형에 따른 세포내 Invertase의 성질 비교 (The Comparison of the Characteristics of Partially Purified Internal Invertase by Mating Type in the Heterobasidiomycetous Yeast)

  • 서로 다른 성접합형 A와 a를 가지는 Rhodosporidium toruloides가 생산하는 invertase를 조사한 결과, 접합형 A는 세포내, 세포벽 결합, 세포의 분비성 invertase의 3가지를 생산하였으나, 접합형 a는 세포외 invertase를 생산하지 않는 접합형 특이성이 있는 것을 알았다. 양 접합형 세포로부터 세포내 invertase를 부분정제하여 이들의 성질을 비교한 결과, 접합형 A세포 유래의 효소는 $Zn^{2+}$에 의하여 11의 활성 상승효과와Mn^{2+}$에 의한 42.2%의 감소를 보이는 반면, a세포 유래의 효소는 $Mn^{2+}$에 의한 15의 활성 감소만을 보였다. 효소 저해제의 효과에서 a세포의 invertase는 2-mercaptoethanol, sodium dodecyl sulfate, pheonl에만 약간 저해를 받을 뿐이었다. 이들 양 효소의 열안정성은 모두 $80^{\circ}C$ 이상에서 급격히 실활되는 양상을 보였으며 최적 온도가 $60^{\circ}C$로 둘다 비슷하였다. a세포 유래의 invertase는 pH 3에서 pH 10까지 넓은 pH의 범위에서 안정하며 최적 pH가 4.0인반면, A 세포의 invertase는 pH 3에서 pH 6까지 산성의 조건에서만 안정하였으며 최적 pH는 5.0으로 나타났다.그리고, A 세포와 atpvh 유래 invertase의 $K_m$값은 각각 $2.5{\times}10^3$M 과 $3.4{\times}10^3$M과 이였다.
