Isolation of Rhizopus sp. R2 Producing Protopectinase and Optimum Condition ofr Pre-paring Single Cells from Potato Tissues (Protopectinase 생산균주, Rhizopus sp. R2의 분리 및 감자조직의 단세포화를 위한 최적조건)

  • Several microorganisms capable of producing protopectinase, which catalyzes dissociation of plant tissue to single cells, were isolated from soils. Crude enzymes prepared from culture supernatants of the strains released potato cells as a single cell from potato tissues. One of the isolated strains showing higher activity of protopectinase was selected and identified as Rhizopus sp. from the morphological characteristics. For preparing single cells from potato tissues, optimum enzyme activity of protopectinase, which was prepared from culture filtrate of Rhizopus sp. R2, was obescrved at $40^{\circ}C$ and pH 4.

Studies on the Hydrolysis of Milk Fat by Microbial Lipases (미생물에서 추출된 Lipase의 유지방 분해)

  • To utilize microbial lipases for hydrolysis of milk fat, optimum reaction conditions and characteristics of enzymatic reactions of lipases originated from Rhizopus delemar, Mucor sp., and Candida cylindracea were investigated. Optimum pH and temperature were pH 5.6 and $45^{\circ}C$ for Rhizopus delemar lipase, pH7.5 and $35^{\circ}C$ for Mucor sp. lipase, and pH7.5 and $35^{\circ}C$ for Candida cylindracea lipase. Optimum lipase concentration and optimum substrate concentration were $600{\sim}800\;units/ml$ and 20% milk fat, regardless of their origin. Km values were 6.06% milk fat for Rhizopus delemar lipase, 7.69% for Mucor sp. lipase and 7.99% for Candida cylindracea lipase. Rate of lipid hydrolysis was Rhizopus delemar lipase>Mucor sp. lipase>Candida cylindracea lipase. As the reaction time was extended, liberation of short chain fatty acids was increased. After 8 hours reaction, capric acid content significantly increased with Candida cylindracea lipase, palmitic acid with Mucor sp. lipase and butyric acid with Rhizopus delemar lipase.

Enzymological Characteristics and Identification of Useful Fungi Isolated from Traditional Korean Nuruk (한국전통누룩에서 분리한 유용곰팡이의 효소학적 특성 및 동정)

  • For the standardization and quality improvement of traditional Korean Nuruk, 10 strains of fungi, which were isolated from Nuruks and showed good productivity of the saccharogenic and dextrinogenic enzymes, acid and flavor, were selected and their enzymological characteristics and identification were carried out. Aspergillus spp. and Rhizopus sp. showed a high liquefying activity without regard to cultivation time, whereas the majority of strains except for Rhizopus sp. had decreasing saccharifying activity in proportion to the increase in cultivation time. Aspergillus spp. No.17-2, No.17-6 and Rhizopus sp. No.18-1 showed high liquefying and saccharifying activity after 15 and 30 day cultivation. The optimum temperature of most of these saccharogenic and dextrinogenic enzymes was from 40$^{\circ}C$ to 60$^{\circ}C$, and their optimum pH was extensive between pH 3 and pH 11. But Penicillium spp.(2 strains) and Rhizopus sp. showed low activity under the alkalic and acidic conditions. Among these isolated strains, 5 strains which had shown the high productivity of materials were identified as Aspergillus oryzae NR3-6 and Aspergillus oryzae NR17-6, Aspergillus penicilloides NR12-1, Penicillium expansum NR7-7 and Rhizopus oryzee NRl8-1, respectively. Five kinds of mixed culture were carried out and all of them showed a better productivity of saccharogenic and dextrinogenic enzymes than single culture. These results indicate that it is possible to make traditional Korean liquors of good quality by using these fungi.

Identification of Filamentous Molds Isolated from Korean Traditional Nuruk and their Amylolytic Activties (한국전통누룩에 존재하는 사상균의 분리 동정 및 Amylolytic 효소 활성)

  • Through the study of identification of filamentous molds isolated from 12 traditional Nuruk, collected from several regions in Korea, 78 strains of Absidia spp., including Absidia corymbifera, Absidia ramosa, and Absidia sp. of which specific names were not identified, 19 strains of Rhizopus spp. including Rhizopus cohnii, Rhizopus arrhizus, and Rhizopus oryzae, 8 strains of Circinella spp., 1 strain of Actinomucor sp., 49 strains of Aspergillus spp., including Aspergillus oryzae, Aspergillus flavus, Aspergillus flavus var. columnaris, Aspergillus wentii, Aspergillus candidus, Aspergillus fumigatus, Aspergillus niger, and Aspergillus foetidus, 2 strains of Cladospoyium spp. and 2 strains Botryotrichum spp., etc. total 159 kinds of filamentous molds were isolated and identified. There were many differences in numbers and distributions of filamentous molds from each Nuruk according to their collected region. Absidia spp. were most frequently isolated from every Nuruk sample. Aspergillus oryzae, Aspergillus candidus, and Rhizopus cohnii showed even distribution. Penicillium sp. and Mucor sp. were not detected. Actinomucor sp., Aspergillus foetidus, Botryotrichum sp., and Cladosporium spp., which have not been reported by far, were found. Amylase activities, pH stability of amylase, and acid productivity of isolated strains were compared.

Effects of Culture Conditions of Rhizopus sp. ZB9 on the Production of Saccharifying Amylase during the Preparation of Rice Koji (쌀 입국 제조시 Rhizopus sp. ZB9의 배양조건이 당화 아밀라아제 생성에 미치는 영향)

  • This study was conducted to determine the influence of cultural conditions such as temperature, time, water content, koji-thickness and agitation on the production of saccharifying amylase by Rhizopus sp. ZB9 isolated from Korean Nuruk during the preparation of rice koji, which is used in brewing Korean rice wines, Takju and Yakju. Rice kojies were made under different cultural conditions, and the saccharifying activities of each koji were tested. The temperature range suitable for the production of saccharifying amylase was $28{\sim}36^{\circ}C$. Based on the saccharifying activity and color, 60 hours of cultivation at $28^{\circ}C$ was believed to produce the optimum results. The water contents of steamed rice suitable for the production of saccharifying amylase were 35~40%. An increase in koji-thickness induced no adverse effects on the production of saccharifying amylase, but agitation-work during cultivation had a harmful effect.

The temperature range suitable for the production of organic acid was $28{\sim}32^{\circ}C$, and 36~48 hours of cultivation at that temperature range seemed to produce the optimum results. The production of organic acid increased in proportion to the increase in water content of steamed rice from 25% to 60%. An increase in koji-thickness induced no adverse effects on the production of organic acid, and agitation-work during cultivation showed very beneficial effects.

Rice koji was made under different culture conditions, and the proteolytic activity of each koji was tested. The temperature range suitable for the production of protease was $28~32^{\circ}C$. Based on the protease and color, 60 hours of cultivation at $28^{\circ}C$ was shown to produce optimum results. The production of protease increased in proportion to the increase in water content of steamed rice from 25% to 35%. An increase in koji-thickness induced no adverse effects on the production of protease, and agitation during cultivation showed beneficial effects.

Screening of a Potent, Raw Naked Barley Saccharifying Enzyme Producer and Its Application on the Uncooked Alcohol Fermentation (쌀보리 전분 당화효소 생산균의 분리 동정 및 무증자 알코올 발효에의 이용)

  • Microorganisms capable of degrading the raw naked barley were isolated from soil, and the amylase productivity of each strain was examined on plate contained 2% raw naked barley. Of the fungi and actinomycetes tested, 71 strains were subjected to subsequent testing for amylase production, and 4 strains were selected as potent amylase producers. Among them, Strain No. 281 produced the most potent raw naked barley saccharifying enzyme, and was identified as genus Rhizopus from morphological and physiological studies. The ratio of raw starch saccharifying activity (RDA) of the crude enzyme derived from the Rhizopus sp. No. 281 was showed 2-3 fold higher than that of commercial enzyme when the raw naked barley was used as the substrate. In the case of uncooked alcohol fermentation using Rhizopus sp. No. 281 glucoamylase preparation, the alcohol yield of the broth was 2% higher than that of the commercial enzyme.

  • A new method for manufacturing the active seed starter (Korean Kok-Ja) has been studied, designed new method consists of the boiling process of raw wheat material and pure culture process of Rhizopus fungi. The feasibility study for industrialization of these methods have been performed, the liquefying and saccharifying activities were put to analysis and compared with conventional product of Kok-Ja. The isolated strains of mold from raw wht and Kok-Ja were idetified as Rhizopus sp., Aspergillus sp., Aspergillus niger sp., Endomyces sp., and Fungi imperfecii. It is proved that the saccharifying and liquefying activities are much stronger than that of conventional product of Kok-Ja, and the growth of Rhizopus strain in boild what material are vigorous than that of old type Kok-Ja. Then it is considered the newly designed method is effective to manufacturing a fine better Kok-Ja.

Screening and Characteristics of Useful Fungi for Brewing from Commercial Nuruk in Chungcheong Provinces (충청지역 누룩에서 양조용 우수 곰팡이의 탐색 및 특성)

  • Studies on standardization and quality upgrade of nuruk which is a basic component in brewing are required to increase the quality level of Korean traditional rice wines and to develop the technology for practical use of it. It is important to isolate best strains, to improve the properties and effectively preserve them for brewing industry. In this study, 16 commercial nuruk samples were obtained from the commercial markets located in Chungcheong areas in Korea. 174 fungal strains were isolated from the samples on DG18 medium using a dilution plating method and then screened for enzyme activity and acid production. The active strains were identified based on the morphological characteristics and ITS sequence analysis. Out of 174 strains, 12 strains showed high amylase activity. Especially, Rhizopus sp. CN084, CN174, Aspergillus sp. CN161 and Mycocladus sp. CN042 showed high saccharogenic power and dextrinogenic enzyme activity on cooked wheat bran medium. On the other hand, Aspergillus sp. CN010, CN161, Rhizopus sp. CN105, CN168 and Rhizomucor sp. CN088 produced high acid production on the same medium. Our results showed that the active strains may be used as microbial sources for nuruk starter with good quality in brewing.
