Rhei Rhizoma Extracts Have Antiproliferative Properties and Differential Effects on NO Production in Macrophages

  • Recently, Rhei Rhizoma extracts (RRE) have begun to receive more attention as potential biological response modifiers. In the present study, we studied the antiproliferative effect of RRE on tumor cells and the effect of RRE on macrophage function. A variety of tumor cells and macrophages were treated with RRE at various concentrations. The effect of RRE on cell proliferation was measured by MTT assay and the effect of RRE on the production of nitric oxide (NO) was determined in the macrophage-like cell lines Raw264.7, C6 and peritoneal macrophages (pMQ). RRE inhibited the growth of tumor cells (e.g., B16, HOS). However, the effects of RRE on the production of NO varied with macrophage types. RRE had no effect on C6 cell growth and slightly increased the growth of Raw264.7 cells. In addition, treatment of normal pMQ with RRE enhanced NO production in a concentration-dependent manner, whereas RRE suppressed NO production at $50\;{\mu}g/mL$ in both Raw264.7 and C6 cells. However, RRE suppressed NO production in LPS/IFN-$\gamma$-stimulated C6 cells. Overall, these results suggest that RRE elicits an antiproliferative property and differentially modulates NO production in various macrophages, and have a potential for therapeutic application.

Analysis of Prescriptions from Somunsunmyungronbang and Nansilbijang (소문선명논방(素問宣明論方)과 난실비장(蘭室秘藏)의 본초구성 비교 분석)

  • Objectives : To quantitatively analyze the academic characteristics of Yuhagan and Idongwon and compare it to existing qualitative analysis. Methods : The prescriptions listed in Somunsunmyungronbang and Nansilbijang were entered into a database. Commonly used combination of medical herbs were researched throughout the literature. Results : In Nansilbijang, among the combinations of 8 medical herbs, Glycyrrhizae Radix, Cimicifugae Rhizoma, Bupleuri Radix, Ginseng Radix, Citri Pericarpium, Atractyodis Rhizoma, Astragali Radix, Phellodendri Cortex combination was used 11 times out of the 277 prescriptions. In Somunsunmyungronbang, combinations of 7 medical herbs: (Glycyrrhizae Radix, Platycodi Radix, Rhei Radix et Rhizoma, Menthae Herba, Atractylodis Rhizoma Alba, Gypsum Fibrosum, Forsythiae Fructus), (Glycyrrhizae Radix, Angelicae gigantis Radix, Paeniae Radix Alba, Atractylodis Rhizoma Alba, Gypsum Fibrosum, Cnidii Rhizoma, Scutellariae Radix), (Glycyrrhizae Radix, Platycodi Radix, Angelicae gigantis Radix, Rhei Radix et Rhizoma, Atractylodis Rhizoma Alba, Gypsum Fibrosum, Schizonepete Herba) were each used 7 times out of the 350 prescriptions. Conclusions : Nansilbijang shows more prescriptions applying Cimicifugae Rhizoma, Bupleuri Radix, Ginseng Radix, Citri Pericarpium, Atractyodis Rhizoma, Astragali Radix, Phellodendri Cortex. Prescriptions aimed to tonify ql with Ginseng Radix and Astragali Radix, to upraise the middle qi with Bupleuri Radix and Cimicifugae Rhizoma, and to regulate qi with Citri Pericarpium and Atractyodis Rhizoma were common. Somunsunmyungronbang preferred the use of Angelicae gigantis Radix, Platycodi Radix, Rhel Radix et Rhizoma, Gypsum Fibrosum, Schizonepete Herba, Forsythiae Fructus and Menthae Herba so its focus was on the aggressive management of febrile diseases.

  • To clarify the botanical origins of oriental medicines which have been argued or confused for plant origins, species of original plants were investigated through the textural research for oriental medicines and the comparison of Chinese, Korean, Japanese and North Korean Pharmacopoeia. Twenty oriental medicines were studied; Angelicae Gigantis Radix, Rhei Rhizoma, Atractylodis Rhizoma Alba, Aconiti Lateralis Radix Preparata, Acanthopanacis Cortex, Osterici Radix, Cnidii Rhizoma, Saposhnikovae Radix, Magnoliae Cortex, Paeoniae Radix, Liriopis Tuber, Zanthoxyli Fructus, Achyranthis Radix, Sinomeni Caulis et Rhizoma, Polygonati Rhizoma, Cinnamomi Cortex, Visci Herba et Loranthi Ramulus, Fritillariae Thunbergii Bulbus, Pogostemonis Herba, and Curcumae Longae Radix.

The Neuroprotective Activities of the Sam-Hwang-Sa-Shim-Tang in the Transient Ischemic Model in Rats.

  • Sam-Hwang-Sa-Shim-Tang(SHSST), a traditional Chinese medicine, composed of Rhei rhizoma, Scutellaria radix, and Coptidis rhizoma were used in the several disease including hypertension, constipation, and hemorrhage. In the present study, we investigated the neuroprotective effects of SHSST and its ingredients on the ischemia/ reperfusion-induced brain injury was evaluated in the rat brain. Ischemia was induced by intraluminal occlusion of the right middle cerebral artery for 120 min and reperfusion was continued for 22 h. SHSST (450 mg/kg), Rhei rhii oma (100 mg/kg), Coptidis rhizoma (100 mg/kg), and Scutellaria radik (100 mg/kg) were orally administered twice, promptly prior to reperfusion and 2 h after the repefusion. Total infarction volume in the ipsilateral hemisphere of ischemia/ reperfusion rats was significantly lowed by the treatments of SHSST (39.2%) and Scutellaria radix (66.5%). However, Coptidis rhizoma did not show any significant effects on the total infarct volume. The inhibiting effect of Scutellaria radix on the total infarct volume was more potent than that of SHSST. In addition, Scutellaria radix significantly inhibited myeloperoxidase (MPO) activity, an index of neutrophil infiltration in ischemic brain tissue. However, there was marked mismatch between total infarct volume and MPO activity in the Scutellaria radix-treated rats. Our findings suggest that Scutellaria radix as an ingredient of SHSST plays a protective role in ischemia-induced brain injury by inhibiting neutrophil infiltration. The effects of Rhei rhizoma on transient brain ischemia-induced neuronal injury are under study.

A Study on Cytotoxicity of Dangkwi-Yonghoe-Hwan (당귀용회환의 세포독성(細胞毒性)에 관(關)한 연구(硏究))

  • The purpose of this research was to investigate cytotoxicity of DangkwiYonghoe-Hwan(DYH) and the constitutive crude drugs on several cancer cell-lines, thymocytes, splenocytes and 3T3 cells. The DYH consists of Coptidis Rhizoma, Scutellariae Radix, Phellodendri Cortex, Gardeniae Fructus, Gentianae scabrae Radix, Indigo pulverata Levis, Aloe, Rhei Rhizoma, Moschus, Saussureae Radix and Angelicae Gigantis Radix. The cytotoxicity was determined by MTT method. The DYH inhibited the proliferation of MOLT-4, K562, HL-60, Jurkat, L1210, P815, S180 and Yac-1, thymocyte, splenocyte and 3T3 cells. The cytotoxicity of Coptidis Rhizoma on the cancer cell-lines was the most potent in the constitutive crude drugs. The proliferation of cancer cell-lines was partly inhibition and partly increase by the treatment of Scutellariae Radix, Gardeniae Fructus, Gentianae scabrae Radix, Indigo pulverata Levis, Aloe, Rhei Rhizoma, Moschus and Angelicae Gigantis Radix. Phellodendri Cortex and Saussureae Radix had a poor cytotoxicity on cancer cell-lines. Coptidis Rhizoma and Phellodendri Cortex inhibited the proliferation of thymocyte, splenocyte and 3T3 cells.

Purgative Activities of Seunggitangs

  • The purgative activity of Seunggitang prescriptions (Deseunggitang, Soseunggitang and Joweseunggitang) was measured to compare the laxative potency of these herbal prescriptions. Daeseunggitang and Jowiseunggitang more potently stimulated the transportation of small and large intestine than Rhei Rhizoma alone. However, the small and large intestine transportation activities of Soseunggitang were similar to those of Rhei Rhizoma alone. Soseunggitang inhibited nitrite production in LPS-induced RAW 264.7 cells and trypsin than the other Seunggitangs. The inhibitory activity of Jowiseunggitang was more potent on nitrite production in LPS-induced RAW 264.7 cells than those of Daeseunggitang. These results suggest that Daeseunggitang can be used as a emergent purgative for patients with severe fever and constipation, Soseunggitang can be as a mild purgative for chronic constipation with inflammation and Jowiseunggitang can be as a potent purgative for patients for severe constipation and weak colitis.

Effects of Rhei Rhizoma and Moutan Cortex on Inflammation and Insulin Resistance in Endothelial Cells Stimulated with Palmitic Acid (팔미트 지방산으로 자극된 혈관내피세포에서 대황 및 목단피가 염증 및 인슐린 저항성에 미치는 효과)

  • Rhei Rhizoma (RR) and Moutan cortex (MC) have been reported to have anti-inflammatory effects. However, little is known about the effects of RR and MC on endothelial inflammation and insulin resistance (IR). This study aims to investigate whether the water extracts of RR and MC could exert protection against palmitic acid (PA)-induced inflammation and IR in human umbilical vein endothelial cells (HUVECs). HUVECs were pretreated for 6 h with RR or MC, and then exposed to PA for 24 h. The levels of interleukin-6 (IL-6) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) were determined by enzyme-linked immunosorbant assay kits. Western blot analysis was performed for activation of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) and insulin receptor substrate-1 (IRS-1). In HUVECs stimulated with PA, both RR and MC significantly inhibited the production of TNF-${\alpha}$ and IL-6 and the activation of NF-${\kappa}B$. At the same concentrations, the inhibitory effects of RR were more potent than those of MC. PA reduced insulin-induced phosphorylation of IRS-1, which was reversed by RR and MC. The results suggest that RR and MC are effective in inhibiting PA-associated endothelial inflammation and ameliorating IR by beneficial regulation of NF-${\kappa}B$ and IRS-1 activation.

Effect of a Mixture of Rhei Rhizoma and Scutellariae Radix Extract on Acute Reflux Esophagitis Rats (대황(大黃)과 황금(黃芩) 추출물 혼합물이 급성 역류성 식도염 흰쥐에 미치는 효과)

  • Objective : Reflux esophagitis is a disease caused by reflux of stomach contents, stomach acid, and pepsin into the esophagus, and is currently increasing worldwide. This study was conducted to evaluate the effect of a mixture of Rhei Rhizoma and Scutellariae Radix (RS) extract on acute reflux esophagitis in rats. Methods : Rats were divided into five groups for examination: Normal group (Nor, n=8), water-treated acute reflux esophagitis rats (Con, n=8), tocopherol 30 mg/kg body weight/day-treated acute reflux esophagitis rats (Toco, n=8), RS 100 mg/kg body weight/day-treated acute reflux esophagitis rats (RS100, n=8), RS 200 mg/kg body weight/day-treated acute reflux esophagitis rats (RS200, n=8). All rats fasted for 18 h and then were derived by linking the metastatic junction between pylorus and forestomach and corpus. And rats were sacrificed 5 h after surgery. We analyzed the expression of NADPH, MAPK, inflammatory, anti-inflammatory, and tight junction related proteins by western blot in esophageal tissue and observed the level of reactive oxygen species (ROS), alanine aminotransferanse (ALT), and aspartate aminotransferase (AST) in serum. Results : RS administration significantly protected the esophageal mucosal damage of reflux esophagitis, and ROS, AST, and ALT levels were significantly reduced in RS administration compared to Con group. In addition, RS administration effectively suppressed MAPK and NF-κB pathways and upregulated protein expressions of tight junction protein. Conclusions : These results suggest that RS protected the esophageal mucosa by inhibiting the MAPK and NF-κB pathways and upregulating tight junctions.

Effects of Radix et Rhizoma Rhei on the Renal Function and Vessel in the Diabetic Rats (대황이 당뇨병 백서의 신장 및 혈관에 미치는 효과)

  • Objectives : This study aimed at investigating whether aqueous extract of Radix et Rhizoma Rhei (AR) ameliorates renal and vascular complications in diabetic rats. Methods : The experiment operated for 6 weeks. The rats were divided into 4 groups: normal group, diabetic group (control group), diabetic group treated with AR (100 mg/kg/day) for the last 3 weeks, and diabetic group treated with AR (200 mg/kg/day) for the last 3 weeks. Results : There were no significant changes in the renal functional parameters by treatment of AR in the diabetic rats. Aorta segment in the diabetic rats revealed a thickening of intima and media, which was ameliorated by treatment with AR. The aortic expression level of endothelin-1 was also significantly attenuated by treatment with AR. Conclusions : Treatment with AR could not ameliorate renal functional defects, but improved vascular complication in diabetic rats.

Effects of Several Herbal Medicines on the Replication of Hepatitis B Virus (수종(數種)의 한약재(韓藥材)가 B형 간염(肝炎)바이러스 증식억제(增殖抑制)에 미치는 효과(效果))

  • This study was performed to investigate an anti-HBV activities of the aqueous extracts from 10 Korean herbal medicines in the HepG2 2.2.15 cell culture system and the results were as follows: 1. The extracts of 6 plants (Herba Artemisiae Capillaris, Radix et Rhizoma Rhei, Cortex Cinnamomi, Fructus Chebulae, Fructus Rubi and Radix Rubi) decreased, significantly and dose-dependently, the levels of extracellular HBV virion in the concentrations (10, 100, 500 and $1,000\;{\mu}g/m{\ell}$) tested. 2. However, others (Radix lsatidis, Lignum Sappan, Herba Lysimachiae and Fructus Lycii) did not show any effect either on the replication of HBV or on the levels of virion DNA in the culture media of HepG2 2.2.15 cell. 3. Among the 6 plants which showed the inhibitory potency on the production of extracellular HBV virion, Radix et Rhizoma Rhei, Cortex Cinnamomi, Fructus Chebulae, Fructus Rubi and Radix Rubi except Herba Artemisiae Capillaris also showed the inhibition of the replication of intracellular HEV DNA in the range of $100{\sim}500\;{\mu}g/m{\ell}$. Considering the above results, it is thought that 6 plants(Herba Artemisiae Capillaris, Radix et Rhizoma Rhei, Cortex Cinnamomi, Fructus Chebulae, Fructus Rubi and Radix Rubi) possess the anti-HBV activities in the HepG2 2.2.15 cell culture system. We thus suggest that these plants possess a potential as a therapeutic agent for the chronic viral hepatitis. These results might be useful as a basic data for the development of the new preventive drugs for HBV diseases.
