Study on White Ginseng Extract Preparation for Cognition Improvement (인지능 개선 효과 증진을 위한 백삼 추출물 조제 연구)

  • Background: The study was conducted to elucidate the extraction conditions under which white ginseng has cognition-improving efficacy. Methods and Results: Extracts from white ginseng under different solvent and temperature conditions were analyzed for ginsenoside content and inhibitory effect on N-methyl-D-aspartate (NMDA) receptor and acetylcholinesterase. The total ginsenoside contents and amounts of ginsenoside Rb1 plus ginsenoside Rg1 from the 1st extracts (prepared with EtOH/$H_2O$ as solvent) were higher than those from the 2nd extracts (extracted with $H_2O$ after the 1st EtOH/$H_2O$ extraction). The contents in the 1st and 2nd extracts produced at $80^{\circ}C$ were also higher than those obtained at $50^{\circ}C$. Samples from the 1st extraction at $80^{\circ}C$ indicated higher inhibitory activities on NMDA receptors-whose excessive activation is thought to mediate the calcium-dependent neurotoxicity associated with several neurodegenerative diseases-than those from the 2nd extraction. Among the samples prepared at varying temperatures, the extract prepared at $50^{\circ}C$ showed the highest suppression activity on NMDA receptors. Note, however, that the extracts from the 2nd extraction at $50^{\circ}C$ inhibited acetylcholinesterase-whose inhibition could be a therapeutic strategy for neurodegenerative diseases with cognitive deficits and memory malfunction-more effectively than those from the 1st extraction. Conclusions: To enhance the cognition-improving activity of white ginseng extract, it is suggested that the extracts be utilized after being combined the 1st extracts (made with EtOH/$H_2O$ solvent) and the 2nd extracts (prepared with $H_2O$) at low temperature.

Composition of Saponin and Free Sugar of Some White Ginsengs with Processing Conditions (가공방법에 따른 몇가지 백삼 제품의 사포닌 및 유리당 조성 변화)

  • To analyse components of fresh ginseng and some white ginsengs with different processing conditions, approximate composition, extraction yield, total saponin content and composition and free sugar composition of fresh ginseng, white ginseng, Taeguksam A and Taeguksam B were examined. Yield of hot water extraction was two times higher than that of 80% methanol extraction. Hot water extraction yields of fresh ginseng, white ginseng, Taeguksam A and Taeguksam B were 56.4, 39.9, 42.9 and 46.6%, respectively, while the 80% methanol extraction yields ranged from 15.8% to 21.9%. Total saponin contents of the above were 2.40, 1.73, 1.45 and 1.79%, respectively, with hot water extraction and were 2.15, 2.99, 2.81 and 2.35%, respectively, with 80% methanol extraction. Ginsenoside compositions of the above varied with processing conditions and extraction solvents. Hot water and 80% methanol extracts of fresh and white ginseng composed of fructose, glucose, sucrose and maltose. Rhamnose was detected only in the extract of Taeguksam A and B.

Effect of Cultivation Using Plastic-Film House on Yield and Quality of Ginseng in Paddy Field (논토양에서 비닐하우스를 이용한 재배방법이 인삼의 수량 및 품질에 미치는 영향)

  • The average and maximum temperature were $29.5^{\circ}C$ and $33.2^{\circ}C$ at 2:00 p.m. respectively, in the plastic-film house covered with shade net, and both of temperature were lower $0.6^{\circ}C$ and $1.3^{\circ}C$ than those of conventional shade. Light transmittance was 14% in the plastic-film house, while 9.9% in conventional shade during growing season from May to October. Withering time of aboveground part was on October 3rd in conventional shade with 60% of withering leaf, while it was on November 10th with 3.7% of withering leaf in the plastic-film house, about 40 days longer survival. The main disease incidence were 15% of anthracnose, 17% of leaf spot, 5% of phytophthora blight and 3% of gray mold in the conventional shade, while 0 ~ 0.1% disease incidence and 95% of emergence rate in the plastic-film house. The growth in the aboveground and underground part of ginseng was totally better, particularly characteristics affecting yield such as root length, main root length and diameter in the plastic-film house. The fresh weight was increased by 128% compared to the conventional shade and harvested roots per $3.3m^2$ were 36 roots in the conventional shade and 58 roots in the plastic-film house and futhermore yield per $3.3m^2$ was increased by 216% compared to the conventional shade. As covering materials, the rice straw in the plastic-film house was excellent. The ginsenoside contents affecting the quality of ginseng were higher in the plastic-film house indicating 0.333% of Rg1, 0.672% of Rb1, 0.730% of Rc and rate of red rusty root was less than 4.0 ~ 6.1%. Above the results, the quality of ginseng grown in the plastic-film house covered with shade net was improved than that of the conventional shade.

Characterization and optimization for beverage manufacture using Korean red ginseng extract (기호성 개선 인삼농축액 제조를 위한 추출 조건 설정)

  • This study was designed to develop a manufacturing process for ginseng concentrate with reduced unpleasant aroma and bitter taste. Two types of ginseng, white and red, were extracted under six different conditions (the 1st to the 6th step) of temperature ($65{\sim}95^{\circ}C$) and ethanol concentration (0~70%). Six extracts of each ginseng were evaluated by a sensory test, and assayed for crude saponin, ginsenosides, and acidic polysaccharides. The content of crude saponin in the extracts decreased with extraction time. There was no significant difference in the crude saponin content between white and red ginseng extracts. The yield of red ginseng extract was higher (45%) than that of white ginseng. No significant difference was observed in the acidic polysaccharide content between red and white ginseng extracts. $Rg_3$, a specific ginsenoside in red ginseng, was detected in the 1st to 6th extracts of red ginseng. Bitterness, astringency, and sourness of ginseng extracts decreased as the extraction steps proceeded. The composite of the 1st, 2nd, and 6th step extracts decreased bitterness and astringency, and the highest overall acceptance. Compared with commercial beverages, the composition of the three extracts is the desirable method to decrease the bitter and astringent tastes, and the overall unpleasant flavor of ginseng.

4 Weeks Repeated Oral Dose Toxicity Studies with LMK02-Jangwonhwan in SD Rats (LMK02의 Sprague-Dawley 랫드를 이용한 4 주간 반복 경구투여 DRF 독성시험)

  • The oriental medicine Jangwonhwan, which is a boiled extract of 12 medicinal herbs/mushroom, has been prescribed for patients with cognitive dysfunction and it is originally from the Korean medical text, DonguiBogam(amnesia chapter). Recently, a modified recipe of Jangwonhwan (LMK02-Jangwonhwan) consisting of seven medicinal plants/mushroom, was shown to reduce ${\beta}$-amyloid deposition in the brain of Tg-APPswe/PS1dE9 mouse model of Alzheimer disease. The toxicity of LMK02 was investigated in SD rats by oral repeated adminstration for 4 weeks and we tried to determine test does for 13 weeks repeated study. Quality control of tablet form of LMK02 was established by estimating indicative components, Ginsenoside Rg3 of Red Ginseng and Decursin of Angelicagigas Nakai. The toxicity of LMK02 was investigated in 6 weeks old specific pathogen free (SPF) Sprageu-Dawley rats by oral administration. Each test group were consist of 5 male and 5 female and they received doses of 500, 1,000 and 2,000 mg/kg/day of test substance for 4 weeks. The clinical signs, death rate, body weight, food consumption, ophthalmic examination, urinalysis, hematological and serum biochemistry, organ weight and pathological changes were examined and compared with those of control group. Urinalysis : We observed increase of PRO(p<0.01), SG(p<0.01) in female rats of 1,000 mg/kg/day and 2,000 mg/kg/day(p<0.01). Also, we observed increase of pH and KET in female rats of 1,000 mg/kg/day(p<0.05) and of 2,000 mg/kg/day(p<0.01). WBC in female rats in 1,000 mg/kg/day and 2,000 mg/kg/day were on increase. Hematological test : We observed increase of MCV in male rats of 250 mg/kg/day. (p<0.05) Serum biochemistry test : We found increase of CHO in female rats of 2,000 mg/kg/day(p<0.05). During the experimental period, there were no animals dead or moribund. There were no treatment related changes of general symptom, food and water consumption, organ weight and autopsy According to the results of 4-week repeated dose range finding study, the highest dose was established as 1000 mg/kg for 13-week repeated dose toxicity study and we determined to put 2 more groups by common ratio two.

Effects of High Temperature Heating on the Some Physicochemical Properties of Korean Red Ginseng (Panax ginseng C.A. Meyer) Water Extract (고온 열처리가 홍삼물추출물의 이화학적 특성에 미치는 영향)

  • This study was carried out to investigate the some physicochemical properties of Korean red ginseng (Panax ginseng C.A. Meyer) water extract (RGWE) after heated with high temperatures above $100^{\circ}C$ for 2 hours. RGWEs were heated at 100, 110 and $120^{\circ}C$ for 2 hours by using autoclave. After RGWEs were heated at high temperature for 2 hours without not adjustment of pH, the changes of saponin, free sugars, mineral and color in the RGWEs were investigated. Total ginsenoside content in control was 1.99%, while those of RGWE were 1.65, 1.49 and 1.29% when treated at 100, 110 and $120^{\circ}C$, respectively. The contents of total ginsenoside showed decreased tendency as heating temperatures were increased. The ginsenoside-$Rh_{2}$ and $-Rg_{3}$, which have been reported as very stable red ginseng ginsenosides, showed relatively strong spots on TLC when RGWEs were heated at 110 and $120^{\circ}C$. In case of free sugars in RGWEs, fructose, glucose and maltose showed high contents when compared with control, while Fe, Ca and Mg ions showed very low contents. Value of L in RGWE treated with high temperature was almost the same with control, while values of a and b were increased. Values of a were increased from -0.86 of control to +0.04, +0.05 and +1.14 when treated with 100, 110 and $120^{\circ}C$, respectively. Values of b also were increased from 27.68 of control to 33.61, 33.61 and 37.42 when treated with 100, 110 and $120^{\circ}C$, respectively. Values of total color in RGWEs treated with high temperatures, E, were finally increased by values of a and b.

Effects of Cultured Wild Ginseng Roots on the Alcoholic Fermentation (산삼배양근 첨가가 알콜 발효에 미치는 영향)

  • In order to manufacture the alcoholic drinks using cultured wild ginseng roots(CWGR) of 5 and $10\%$ (w/v), sugar content of fermentation media was adjusted to 24-25 $^{\circ}$Brix with white sugar and glucose. And 3 kinds of yeast (S. cerevisiae(KCCM 50757), S. cerevisiae (KCCM 50583) and S. bayanus(ATCC 10601) were used and then the quality of alcoholic drinks was analyzed by physical, chemical and sensory evaluation. Alcohol content was highest value of $15.8\%$ in $10\%$ of CWGR, white sugar, and S. bayanus(ATCC 10601). Major alcohols were ethanol and 1-propanol. Number of yeast cells increased to 5 days fermentation and slightly decreased afterwards. The pH was decreased abruptly from 5.0 in initial fermentation to 3.1-4.1 in 5 days fermentation. Total sugar contents were decreased continuously with fermentation periods and showed 7.0-10.5 $^{\circ}$Brix in 20 days fermentation. Saponin patterns and contents were various and higher in wine treated with S. bayanus(ATCC 10601). From the sensory evaluation, the highest score of overall quality was observed in the alcoholic beverage of $10\%$(w/v) of CWGR, glucose, and S. cerevisiae(KCCM 50583).

Cultivation of Lactic Acid Bacteria for the Development of Probiotic Products using Red Ginseng Starch (프로바이오틱스 개발을 위한 홍삼 전분을 활용한 유산균 배양)

  • To reduce the production cost of Lactobacillus, discarded red ginseng starch was collected from a factory of red ginseng extract in order to develop the Lactobacillus culture medium. According to the analysis of the gensenoside composition of red ginseng starch, the total gensenoside content of starch was 2.73 mg/g, and the gensenoside $Rb_1$, $Rb_2$ and $Rg_3$ contents were 0.1, 0.29 and 0.52 mg/g, respectively. For the preparation of the liquid media, red ginseng starch was added at rates of 0, 5, 10 and 20%. Further, Lactobacillus plantarum 15357 and Leuconostoc mesenteroides sub sp. strains were then inoculated to these prepared broths. With the red ginseng starch medium, the growth rates ($1.42{\times}10^7$ and $2.96{\times}10^{10}$ CFU/mL) and the final cell concentrations were higher than the MM medium ($1.0{\times}10^7$ CFU/mL). The optimal concentration of red ginseng starch and yeast extract as a medium were 20% and 10 g/L, respectively. Under these conditions, the cell mass of L. plantarum 15357 and L. mesenteroides sub sp. reached $5.11{\times}10^{10}$ and $8.17{\times}10^{10}$ CFU/mL. These results show a great possibility for the utilization of red ginseng starch as economic medium sources in the production of cell mass of lactic acid bacteria. This is the first trial of development of economic LAB growth medium using discarded red ginseng starch.

Difference of the Ginsenosides Contents According to the Planting Location in Panax ginseng C. A. Meyer (인삼의 재식 위치에 따른 진세노사이드 함량 차이)

  • The difference of ginsenosides content according to placement of ginseng planting (line) under shading net in 5-year-old ginseng roots were examined. The total saponin ($Rb_1$, $Rb_2$, Rc, Rd, Re, and $Rg_1$) contents were 15.01 mg/g and 21.79 mg/g in the main roots, 35.93 mg/g and 43.32 mg/g in the lateral roots, 87.85 mg/g and 105.51 mg/g in the fine roots for the front $1st{\sim}2nd$ lines in Yunpoong and Landrace variety (purple-stem variant), respectively. In the middle $3rd{\sim}5th$ lines the total saponin contents were 18.73 mg/g and 23.19 mg/g in the main roots, 44.92 mg/g and 43.50 mg/g in the lateral roots, 92.97 mg/g and 110.70 mg/g in the fine roots in Yunpoong and Landrace variety, respectively. In the rear $6th{\sim}7th$ lines the total saponin contents were 21.88 mg/g and 26.68 mg/g in the main roots, 38.41 mg/g and 44.89 mg/g in the lateral roots, 101.03 mg/g and 107.06 mg/g in the fine roots in Yunpoong and Landrace variety, respectively. The differences in total and individual ginsenosides content in the main, lateral and fine roots among the lines were not significant but total ginsenosides contents in the main roots were different in case of Yunpoong variety. The ratios of protopanaxadiol (PD) type saponin to protopanaxatriol (PT) type saponin in roots were lower in the front lines compared to the middle and rear lines and the ratios were significantly different among the parts of roots.

  • The Immersion Grating Infrared Spectrometer (IGRINS) is an unprecedentedly minimized infrared cross-dispersed echelle spectrograph with a high-resolution and high-sensitivity optical performance. A silicon immersion grating features the instrument for the first time in this field. IGRINS will cover the entire portion of the wavelength range between 1.45 and $2.45{\mu}m$ accessible from the ground in a single exposure with spectral resolution of 40,000. Individual volume phase holographic (VPH) gratings serve as cross-dispersing elements for separate spectrograph arms covering the H and K bands. On the 2.7m Harlan J. Smith telescope at the McDonald Observatory, the slit size is $1^{\prime\prime}{\times}15^{\prime\prime}$. IGRINS has a $0.27^{\prime\prime}$ pixel-1 plate scale on a $2048{\times}2048$ pixel Teledyne Scientific & Imaging HAWAII-2RG detector with SIDECAR ASIC cryogenic controller. The instrument includes four subsystems; a calibration unit, an input relay optics module, a slit-viewing camera, and nearly identical H and K spectrograph modules. The use of a silicon immersion grating and a compact white pupil design allows the spectrograph collimated beam size to be 25mm, which permits the entire cryogenic system to be contained in a moderately sized rectangular vacuum chamber. The fabrication and assembly of the optical and mechanical hardware components were completed in 2013. In this presentation, we describe the major design characteristics of the instrument and the early performance estimated from the first light commissioning at the McDonald Observatory.
