Association of the RORA Gene Polymorphism and Diurnal Preference in a Young Korean Population (RORA 유전자 다형성과 한국인 젊은 성인에서 일중선호도와의 연관성)

  • Objectives: The retinoid-related orphan receptor A (RORA) gene has been reported to have an impact on circadian rhythm regulation. In this study, we analyzed the relationship between the RORA gene polymorphism and diurnal preference in Korean young adults. Methods: A population of 504 young adults was included in the study. All subjects were given and completed a 13-item composite scale for morningness (CSM). The RORA gene rs11071547 single-nucleotide polymorphism (SNP) was genotyped by PCR-based methods. Results: CSM score was not associated with genotype or allele carrier status of the RORA rs11071547 SNP. Conclusion: This result indicates that the RORA rs11071547 SNP does not play a role in diurnal preference.

T-Cell Death-Associated Gene 51 Is a Novel Negative Regulator of PPARγ That Inhibits PPARγ-RXRα Heterodimer Formation in Adipogenesis

  • The nuclear receptor peroxisome proliferator-activated receptor γ (PPARγ) is the master transcriptional regulator in adipogenesis. PPARγ forms a heterodimer with another nuclear receptor, retinoid X receptor (RXR), to form an active transcriptional complex, and their transcriptional activity is tightly regulated by the association with either coactivators or corepressors. In this study, we identified T-cell death-associated gene 51 (TDAG51) as a novel corepressor of PPARγ-mediated transcriptional regulation. We showed that TDAG51 expression is abundantly maintained in the early stage of adipogenic differentiation. Forced expression of TDAG51 inhibited adipocyte differentiation in 3T3-L1 cells. We found that TDAG51 physically interacts with PPARγ in a ligand-independent manner. In deletion mutant analyses, large portions of the TDAG51 domains, including the pleckstrin homology-like, glutamine repeat and proline-glutamine repeat domains but not the proline-histidine repeat domain, are involved in the interaction with the region between residues 140 and 506, including the DNA binding domain, hinge, ligand binding domain and activation function-2 domain, in PPARγ. The heterodimer formation of PPARγ-RXRα was competitively inhibited in a ligand-independent manner by TDAG51 binding to PPARγ. Thus, our data suggest that TDAG51, which could determine adipogenic cell fate, acts as a novel negative regulator of PPARγ by blocking RXRα recruitment to the PPARγ-RXRα heterodimer complex in adipogenesis.

  • Hepatic stellate cells (HSCs) play essential roles in normal and pathophysiological function in liver. In steady state, HSCs contribute to retinoid storage, immune tolerance, and extracellular matrix (ECM) homeostasis. Upon liver injury, they become activated and lead to morphological and functional changes. Studies have demonstrated that activation of HSCs by various stimuli such as toxins, microbial infection, or metabolic overload can promote the fibrotic changes in liver by production of ECM. Herein, we provide current knowledge about the basic characteristics of HSCs and the mechanism by which they are activated.

  • Objective : The patient presented with a week history of orange-eryhtematous follicular hyperkeratotic papules in facial and skull part.Methods : Korean medicinePalMul-tang八物湯 was used and Aroma ointment and Retinoid lotion were used together.Results : After 6 months therapy, she experienced remarkable improvement of symptoms.Conclusions : Pityriasis Rubra Pilaris (PRP) is belong to 'Pityriasis Rubra Pilaris (PRP) is belong to 'Honyoja狐尿刺'in Korean medicine, and we can expect a relatively good treatment effect.

광노화의 탄력소의 변화

  • 최근 오존층의 파괴로 인한 지구 자외선량의 증가로 피부암 및 광노화의 발생가능성이 증가되고있다. 광노화의 주요한 현상중의 하나는 탄력섬유상 물질이 축적되는 일광탄력 섬유증이다. 자외선은 탄력소 유전자의 전사과정증가를 유도하며 결국 비정상적인 탄력소의 증가를 유발한다는 보고가 있다. 본 연구에서는 피부 섬유아세포 및 각질형성세포를 배양하여 자외선이 탄력소 유전자발현에 미치는 영향 및 전사조절인자에 미치는 영향을 알아보고자 하였다. 피부섬유아세포에 UVB를 30mJ-200J/m 조사하여 nothern blotting한 결과 UV양이 증가함에 따라 탄력소 전사물이 증가되었고, 1001/$m^2$ 에서 최대의 증가를 보였다. 탄력소 promoter와 CAT을 접합시킨 pEP62 vector을 섬유아세포에 형질전환 시키고 UV에 의한 Promoter 활성을 본 결과 UV조사량이 증가할수록 활성이 증가되었고 200J/$m^2$ 에서 대조군에 비해 5배의 활성 증가를 보였다. 이 system에 광노화 억제물질로 알려진 retinoid를 5 $\times$ $10^{-6}$M처리하였을 때 UV에 의한 탄력소 promoter활성을 약 1/3로 감소시켰다. 각질 형성세포에서 UVB에 의한 transcription factor의 활성을 mobility shift assay에 의해 조사한 결과 AP-1 및 NFkB가 활성화됨을 볼 수 있었다.

  • Mycosis fungoides is the most common type of cutaneous T-cell lymphoma. Patients with early stage disease usually respond well to conventional therapies, with a relatively favorable prognosis. However, a few patients are refractory to treatment and need alternative strategies, even at the patch and plaque stages. We report the case of a middle-aged woman with long-standing and refractory mycosis fungoides that responded to combination therapy with the 308-nm excimer laser and oral alitretinoin.

  • Subcorneal pustular dermatosis (SPD) is a rare pustular skin disorder of dogs. Dapsone is treatment of choice, but ineffective in some cases. Therapeutic alternatives are limited and less effective. In humans, successful results with oral retinoid have been reported. To describe successful treatment of a dog that developed SPD with acitretin as an alternative drug of dapsone. A 7-year-old male neutered miniature schnauzer was presented with generalized pustules and crusts. SPD was diagnosed based on physical, cytological, and histopathological examination with direct immunofluorescence test. In this report, we describe a case of canine SPD treated initially by dapsone with poor response that improved with acitretin. Acitretin could be considered as an alternative drug in canine SPD.

Effect of retinoic acid on the bystander effect in gene therapy using the Herpes Simplex Virus thymidine kinase (Herpes Simplex Virus thymidine kinase gene을 이용한 유전자 치료에서 retinoic acid가 bystander effect에 미치는 영향)

  • Background : Metabolic cooperation via gap junctional intercellular communication (GJIC) is an important mechanism of the bystander effect in gene therapy using the Herpes Simplex Virus thymidine kinase/ganciclovir (HSVtk) "prodrug" system. Since retinoids have been reported to increase GJIC by induction of connexin 43 expression, we hyporthesized that treatment of tumor cells with retinoic acid could augment the bystander effect of the HSVtk/GCV system and result in improved tumor cell killing by enhancing GJIC. Methods : We transferred HSVtk gene to SKHep-J cell line that does not express connexin43, and also transferred the gene to human and murine mesothelioma cell lines that express connexin43. We verified that retinoic acid enhanced GJIC utilizing a functional double-dye transfer study and evaluated the effects of retinoic acid on the growth rate of tumor cells. We then tested the effects of retinoic acid on bystander-mediated cell killing. Results : Addition of all-trans retinoic acid (RA) increased GJIC in cell lines expressing connexin 43 and was asspciated with more efficient in vitro bystander killing in cells transduced with HSVtk via adenoviral and retroviral vectors. In contrast, there was no increase in the efficiency of the bystander effect after exposure to RA in a cell line which had no delectable connexin 43. Conclusion : These results provide evidence that retinoids can augment the efficiency of cell killing with the HSVtk/GCV system by enhancing bystander effect and may thus be a promising new approach to improve responses in gene therapy utilizing the HSVtk system to treat tumors.
