Perception-based analytical technique of evacuation behavior under radiological emergency: An illustration of the Kori area

  • A simulation-based approach is proposed to study the protective actions taken by residents during nuclear emergencies using cognitive findings. Human perception-based behaviors are not heavily incorporated in the evacuation study for nuclear emergencies despite their known importance. This study proposes a generic framework of perception-based behavior simulation, in accordance with the ecological concept of affordance theory and a formal representation of affordance-based finite state automata. Based on the generic framework, a simulation model is developed to allow an evacuee to perceive available actions and execute one of them according to Newton's laws of motion. The case of a shadow evacuation under nuclear emergency is utilized to demonstrate the applicability of the proposed framework. The illustrated planning algorithm enables residents to compute not only prior knowledge of the environmental map, but also the perception of dynamic surroundings, using widely observed heuristics. The simulation results show that the temporal and spatial dynamics of the evacuation behaviors can be analyzed based on individual perception of circumstances, while utilizing the findings in cognitive science under unavoidable data restriction of nuclear emergencies. The perception-based analysis of the proposed framework is expected to enhance nuclear safety technology by complementing macroscopic analyses for advanced protective measures.

Evaluation of the classification method using ancestry SNP markers for ethnic group

  • Various probabilistic methods have been proposed for using interpopulation allele frequency differences to infer the ethnic group of a DNA specimen. The selection of the statistical method is critical because the accuracy of the statistical classification results vary. For the ancestry classification, we proposed a new ancestry evaluation method that estimate the combined ethnicity index as well as compared its performance with various classical classification methods using two real data sets. We selected 13 SNPs that are useful for the inference of ethnic origin. These single nucleotide polymorphisms (SNPs) were analyzed by restriction fragment mass polymorphism assay and followed by classification among ethnic groups. We genotyped 400 individuals from four ethnic groups (100 African-American, 100 Caucasian, 100 Korean, and 100 Mexican-American) for 13 SNPs and allele frequencies that differed among the four ethnic groups. Additionally, we applied our new method to HapMap SNP genotypes for 1,011 samples from 4 populations (African, European, East Asian, and Central-South Asian). Our proposed method yielded the highest accuracy among statistical classification methods. Our ethnic group classification system based on the analysis of ancestry informative SNP markers can provide a useful statistical tool to identify ethnic groups.

Mapping of Quantitative Trait Loci for Yield and Grade Related Traits in Peanut (Arachis hypogaea L.) Using High-Resolution SNP Markers

  • Yield and grade are the key factors that affect production value of peanut. The objective of this study was to identify QTLs for pod yield, hundred-seed weight, and total sound mature kernel (TSMK). A total of 90 recombinant inbred lines, derived from Tamrun OL07 and a breeding line Tx964117, were used as a mapping population and planted in Brownfield and Stephenville, Texas. A genetic map was developed using 1,211 SNP markers based on double digest restriction-site associated DNA sequencing (ddRAD-seq). A total of 10 QTLs were identified above the permutation threshold, three for yield, three for hundred-seed weight and four for TSMK, with LOD score values of 3.7 - 6.9 and phenotypic variance explained of 12.2% - 35.9%. Among those, there were several QTLs that were detected in more than one field experiment. The commonly detected QTLs in this study may be used as potential targets for future breeding program to incorporate yield and grade related traits through molecular breeding.

CURVES WITH MAXIMAL RANK, BUT NOT ACM, WITH VERY HIGH GENERA IN PROJECTIVE SPACES

  • A curve $X{\subset}\mathbb{P}^r$ has maximal rank if for each $t{\in}\mathbb{N}$ the restriction map $H^0(\mathcal{O}_{\mathbb{P}r}(t)){\rightarrow}H^0(\mathcal{O}_X(t))$ is either injective or surjective. We show that for all integers $d{\geq}r+1$ there are maximal rank, but not arithmetically Cohen-Macaulay, smooth curves $X{\subset}\mathbb{P}^r$ with degree d and genus roughly $d^2/2r$, contrary to the case r = 3, where it was proved that their genus growths at most like $d^{3/2}$ (A. Dolcetti). Nevertheless there is a sector of large genera g, roughly between $d^2/(2r+2)$ and $d^2/2r$, where we prove the existence of smooth curves (even aCM ones) with degree d and genus g, but the only integral and non-degenerate maximal rank curves with degree d and arithmetic genus g are the aCM ones. For some (d, g, r) with high g we prove the existence of reducible non-degenerate maximal rank and non aCM curves $X{\subset}\mathbb{P}^r$ with degree d and arithmetic genus g, while (d, g, r) is not realized by non-degenerate maximal rank and non aCM integral curves.

  • The noise problem became more serious by the rapid urban population growth and traffic. This study estimates the noise level considering actual traffic volume and building shape and location. Based on the state of the arts noise level, changing amount of noise that was controlled by the speed control and truck and passenger car restriction is calculated. Some conclusion is found out. The 2-D type more than 3-D type is more than over standard of noise pollution and night time's over rate is larger than day time. And in the case of various solutions are used instead of applying one solution, noise pollution is effectively reduced.

Helicobacter pylori Strain 51 (Korean Isolate): Ordered Overlapping BAC Library, Combined Physical and Genetic Map, and Comparative Analysis with H. pylori Strain 26695 and Strain J99

  • We constructed a defined physical and genetic map of H. pylori strain 51, previously isolated from a Korean patient with a duodenal ulcer, by combining a restriction analysis by pulse-field gel electrophoresis with the construction of a BAC library. A Notl-digest of H. pylori strain 51 genome yielded seven fragments, from which the genomic size was estimated to be 1,698$\pm$24 kb. The BAC library was constructed from 50 to 200 kb fragments of HindIII-digested genomic DNA. From 700 BAC clones, an ordered overlapping maxi-set of 82 BAC clones was assembled that covered the entire genome. The positions of 15 genes were localized in the strain 51 genome with 4-22 kb of resolution and were compared with their orthologues in strain 26695 and strain J99. The arrangement of the 15 genes was identical in strain 51 and strain J99, except for flaA and hpaA. The plasticity zone of strain 51, like that of strain J99, was located in the single region, and was shorter than those of strain 26695 and strain J99. The strain 51 plasticity zone consisted of ORFs common only to strain 51 and J99 or to strain 51 and 26695, as well as strain 51-specific ORFs. Three genetic translocations and/or inversions were found between orthologue ORFs in strain 51 and strain J99. These results show that the chromosomal organization of strain 51 differs from Western strains such as strain 26695 and strain J99.

Construction of Genetic Linkage Map and Identification of Quantitative Trait Loci in Populus davidiana using Genotyping-by-sequencing (Genotyping-by-sequencing 기법을 이용한 사시나무(Populus davidiana) 유전연관지도 작성 및 양적형질 유전자좌 탐색)

  • Tree species within the Populus genus grow rapidly and have an excellent capacity to absorb carbon, conferring substantial ability to effective purify the environment. Poplar breeding can be achieved rapidly and efficiently if a genetic linkage map is constructed and quantitative trait loci (QTLs) are identified. Here, a high-density genetic linkage map was constructed for the control pollinated progeny using the genotyping-by-sequencing (GBS) technique, which is a next-generation sequencing method. A search was also performed for the genes associated with quantitative traits located in the genetic linkage map by examining the variables of height and diameter at root collar, and resilience to insect damage. The height and diameter at root collar were measured directly, while the ability to recover from insect damage was scored in a 4-year-old breeding population of aspen hybrids (Odae19 × Bonghyeon4 F1) established in the research forest of Seoul National University. After DNA extraction, paternity was confirmed using five microsatellite markers, and only the individuals for which paternity was confirmed were used for the analysis. The DNA was cut using restriction enzymes and the obtained DNA fragments were prepared using a GBS library and sequenced. The analyzed results were sorted using Populus trichocarpa as a reference genome. Overall, 58,040 aligned single-nucleotide polymorphism (SNP) markers were identified, 17,755 of which were used for mapping genetic linkages. The genetic linkage map was divided into 19 linkage groups, with a total length of 2,129.54 cM. The analysis failed to identify any growth-related QTLs, but a gene assumed to be related to recovery from insect damage was identified on linkage group (chromosome) 4 through genome-wide association study.

  • Most of former studies for car license plate detection restrict the image acquisition environment. The aim of this research is to diminish the restrictions by proposing a new method of using SIFT and neural network. SIFT can be used in diverse situations with less restriction because it provides size- and rotation-invariance and large discriminating power. SIFT extracted from the license plate image is divided into the internal(inside class) and the external(outside class) ones and the classifier is trained using them. In the proposed method, by just putting the various types of license plates, the trained neural network classifier can process all of the types. Although the classification performance is not high, the inside class appears densely over the plate region and sparsely over the non-plate regions. These characteristics create a local feature map, from which we can identify the location with the global maximum value as a candidate of license plate region. We collected image database with much less restriction than the conventional researches. The experiment and evaluation were done using this database. In terms of classification accuracy of SIFT keypoints, the correct recognition rate was 97.1%. The precision rate was 62.0% and recall rate was 50.2%. In terms of license plate detection rate, the correct recognition rate was 98.6%.

Rock Joint Trace Detection Using Image Processing Technique (영상 처리를 이용한 암석 절리 궤적의 추적)

  • The investigation on the rock discontinuity geometry has been usually undergone by direct measurement on the rock exposures. But this sort of field work has disadvantages, which we, for example, restriction of surveying areas and consuming excessive times and labors. To cover these kinds of disadvantages, image processing could be regarded as an altemative way, with additional advantages such as automatic and objective tools when used under adequate computerized algorithm. This study was focused on the recognition of the rock discontinuities captured in the image of rock exposure by digital camera and the production of the discontinuity map automatically. The whole process was written using macro commands builtin image analyzer, ImagePro Plus. ver 4.1(Media Cybernetic). The procedure of image processing developed in this research could be divided with three steps, which are enhancement, recognition and extraction of discontinuity traces from the digital image. Enhancement contains combining and applying several filters to remove and relieve various types of noises from the image of rock surface. For the next step, recognition of discontinuity traces was executed. It used local topographic features characterized by the differences of gray scales between discontinuity and rock. Such segments of discontinuity traces extracted from the image were reformulated using an algorithm of computer decision-making criteria and linked to form complete discontinuity traces. To verify the image processing algorithms and their sequences developed in this research, discontinuity traces digitally photographed on the rock slope were analyzed. The result showed about 75~80% of discontinuities could be detected. It is thought to be necessary that the algorithms and computer codes developed in this research need to be advanced further especially in combining digital filters to produce images to be more acceptable for extraction of discontinuity traces and setting seed pixels automatically when linking trace segments to make a complete discontinuity trace.

Expression of Glucose Isomerase Gene from Bacillus licheniformis in Escherichia coli. (Bacillus licheniformis 포도당 이성화 효소 유전자의 Excherichia coli에 발현)

  • A Bacillus licheniformis ATCC31667 gene coding for a glucose isomerase has been cloned and expressed in glucose isomerase negative mutant of Escherichia coli. A recombinant plasmid, constructed by ligation of a EcoRI fragment of B.licheniformis chromosomal DNA to vector plasmid pBR322, was expressed glucose isomerase positive in E.coli LE392-6 with growth on minimal medium containing xylose as a sole carbon source. This recombinant plasmid, designated pBGI6, had the insery of 4.1Kb of Bacillus gene in EcoRI site, and restriction map of the plasmid was established. The plasmid pBG16 was very stable after 10days of serial transfer to a fresh medium. The activity of glucose isomerase from the transformed cell containing pBGI6 was increased about 20 fold than its wild type of host.
