This study was performed in order to investigate the antioxidant properties of hot water extract from the root and aerial part of the Syneilesis palmata in respect to its potential use as food, cosmetics material, or medicinal resource. The results showed that the S. palmata root hot water extract (RHW) possessed a higher content of total flavonoid compounds (4.58 mg/g) and total polyphenol compounds (59.11 mg/g). The SOD-like activities of the RHW and APHW were 23.74% and 21.61%, respectively, at a concentration $2,000{\mu}g/mL$. In the nitrite scavenging ability of a $2,000{\mu}g/mL$ concentration, the RHW showed 63.06% (pH 1.2) and 47.16% (pH 3.0). The $IC_{50}$ values of the nitrite scavenging abilities were $99.93{\mu}g/mL$ (ascorbic acid), $1,150.85{\mu}g/mL$ (RHW), and $1,610.25{\mu}g/mL$ (APHW). The $IC_{50}$ values of DPPH free radical scavenging abilities were $99.87{\mu}g/mL$ (RHW) and $118.29{\mu}g/mL$ (APHW). The inhibition values ($IC_{50}$) of xanthine oxidase were $139.62{\mu}g/mL$ (RHW) and $111.11{\mu}g/mL$ (APHW). In all of the experiments, the S. palmata root hot water extracts have higher activities than the aerial hot water extract, except for the xanthine oxidase inhibitory activity. These results suggest that the S. palmata is a potentially useful antioxidant source for the development of functional nutraceuticals, cosmetics and medicine.
The indigenous Saccharomyces cerevisiae strains S13 and D8 were isolated at the microbial succession stage during spontaneous fermentation of Campbell Early wine as a resistant to potassium metabisulfite and a high sugar concentration. In this study, the fermentation characteristics of Campbell Early wine were investigated and compared with those of S. cerevisiae W-3, an industrial wine yeast. Alcohol production by the two strains was delayed at the initial fermentation stage, but increased fast when the fermentation continued. After the fermentation, the S13 and D8 wines contained 12.6% and 13.2% (v/v) alcohol, respectively, which were significantly higher than the alcohol content of the W-3 wine (12%, v/v). No marked differences were observed in the residual soluble solid content and the pH. However, the S13 and D8 wines showed high levels of total acid content, including malic and lactic acids. Especially, the lactic acid content was 8.9-fold in the S13 wine and six-fold in the D8 wine, compared with that of the W-3 wine. The two strains produced a higher level of acetaldehyde and a lower amount of methanol in the wine than the W-3 strain. The iso-Butanol content was lower in the two indigenous yeast wines with similar levels of n-propanol and iso-amyl alcohol contents than that in the W-3 wine. In the sensory evaluation, the S13 and D8 wines had higher scores for their color, flavor, taste and overall preferences than the W-3 wine. Especially, the S13 and D8 wines had much higher scores than the W-3 wine for flavor and color, respectively.
This study was aimed to test harvest time effect with polyethylene (PE) film liner, 1-methylcyclopropene (1-MCP) and aminoethoxyvinylglycine (AVG) treatments on fruit quality attributes in 'Sangjudungsi' persimmon fruit during cold storage. The fruits were harvested 10 days earlier in 2016 than the mature harvest time in 2015. The ethylene production was significantly lower in early harvested fruits than in mature harvested ones. Flesh firmness was maintained higher in 1-MCP treated fruit than in other treatments during cold storage. The rate of fruit weight loss was significantly inhibited by PE film liner treatment during storage, regardless of harvest time. 1-MCP treatment showed less change in fruit peel color variables ($L^*$ and $b^*$) from the calyx-end and equatorial regions during cold storage, compared with those from the control and PE film treatments. The incidence rate of fruit decay and softening was higher in PE film treated fruits than in the other treatments. However, there was no decay detected in AVG treated fruit. The early harvested fruits were maintained higher flesh firmness, compared with mature harvested fruits. Nevertheless, the mature harvested fruits showed much higher soluble solids content, the redness (Hunter a value) of the fruit peel and respiration rate, compared with early harvested fruits. Furthermore, the rate of weight loss in the fruit was remarkably inhibited in the PE film treatment.

Health of human intestine has deteriorated due to excessive stress and western diet. In this study, Vigna angularis was fermented by Bacillus subtilis KCCM11965P in order to develop prebiotic resource for improving bowel movement. The contents of ash and crude protein were $3.35{\pm}0.04%$ and $21.1{\pm}0.19%$ respectively. Vigna angularis extract (1, 3, 5%) were incubated with 3% (v/v) Bacillus subtilis KCCM 11965P for 0, 24, 48, and 72 h. Total bacterial numbers showed that the combination of 3% powdered seeds and 72 h incubation time was optimum condition for this experiment. Total polyphenol content increased from $0.18{\pm}0.010mg/mL$ in pre-incubation to $0.23{\pm}0.007mg/mL$ in post-incubation with the condition mentioned above. DPPH radical scavenging activity also increased from $36.1{\pm}6.0%$ to $63.6{\pm}5.2%$. Analysis of protease activity showed $2.69{\pm}0.003unit/mL$ in combination of 5% powdered seeds and 72 h incubation time. Amylase activity increased from $1.0{\pm}0.1unit/mL$ in pre-incubation to $26.0{\pm}0.2unit/mL$ in post-incubation. The analysis of free amino acids after incubation with Bacillus subtilis KCCM 11965P showed that leucine increased from 5.22 mg/L to 67.59 mg/L and tyrosine, one of non-essential amino acid also increased 10.08 mg/L to 259.35 mg/L by incubation with 5% powdered seeds. Most of organic acid were reduced by incubation for 72 h. These results suggest that Vigna angularis could be utilized most as a prebiotic resources.

This study was carried out to investigate the effects of ultrasonic thawing on physicochemical characteristics of frozen pork sirloin. To determine the velocity of an ultrasonic thawing machine, $-80^{\circ}C$ frozen pork sirloins were thawed using 132, 580, and 1,000 kHz ultrasonic thawing machines, and the physicochemical properties of pork sirloin thawed with $15^{\circ}C$ tap water and those using the ultrasonic thawing machines were compared. As a result, thawing speed by ultrasonic thawing was three times faster than that of tap water, but drip weight loss rate is increased by about 5% during ultrasonic thawing compared to that during thawing in tap water. However, biochemical properties, such as pH, volatile basic nitrogen, thiobarbituric acid, and total aerobic bacteria, were not improved by ultrasonic thawing. Hunter values, such as total color change and redness, improved slightly after 580 kHz ultrasonic thawing. The hardness and chewiness of the pork sirloin decreased significantly after 580 kHz ultrasonic thawing; however, springiness was not changed. These results indicate that ultrasonic thawing is faster than thawing in tap water, but the physicochemical characteristics of frozen pork sirloin was not significantly improve by ultrasonic sound.
Vaccinium bracteatum Thunb. is an evergreen plant that belongs to the Genus Vaccinium distributed in Korea, and its fruit has been used for food by the common people and thus has a high possibility of commercialization. Therefore, the habitat environment and individual characteristics of V. bracteatum in Jeju Island were investigated to obtain the basic data for commercialization and preservation. The results showed that V. bracteatum is mostly distributed on the southern slope of Mt. Halla at an altitude of 100 to 500 m. V. bracteatum that occurred in the quadrat were total 30 individuals, which had an average height of 6.0 m, an average DBH of 7.8 cm, and an average number of trunk of 4.3. As for the community structure of the habitat, the average percentage of vegetation cover for the tree layer was 36%, while that for the subtree layer was 78%, which emphasized the importance of the subtree layer. The total number of species that occurred in the quadrat was 76. For the tree layer, total 10 species occurred, which include 2 species of evergreen coniferous trees, 3 species of evergreen broad-leaved trees, and 5 species of deciduous broad-leaved trees. For the subtree layer, total 24 species occurred, which include 1 species of evergreen coniferous tree, 11 species of evergreen broad-leaved trees, and 10 species of deciduous broad-leaved trees. Among these, Eurya japonica Thunb. was the species with a high relative contribution, which threatened V. bracteatum For the shrub layer, 27 species occurred including Cinnamomum japonicum Siebold, and for the herbaceous layer, 64 species occurred including Ardisia crenata Sims. The similarity between the habitats of V. bracteatum was very low, and it did not prefer a characteristic community. The V. bracteatum community around Shinrei stream was distributed on the surrounding area adjacent to the slope of the stream rather than the slope. There were a lot of basins, but the branches, which did not receive sunlight, had withered. It is thought that V. bracteatum would decline if the forest formation continues.
The United States Department of Agriculture (USDA) funded collaborative project on The Economic Feasibility Analysis of Crop Cultivation Practice Project in Pirganj and Kurigram Districts in Bangladesh will started during 2008-2012, for 4 years with total project cost of US$ 571,270. The project will be implemented in 6 villages; has 1,097 hectares areas which is divided into 948 hectares of agricultural land, 52 hectares of forest land and 345 hectares of other land, covered 1,059 households equal to 5,305 persons in Pirganj and Kurigram districts The project has proposed to be implemented in joint collaboration by Bangladesh Agricultural Research Council (BARC), Bangladesh Agricultural Research Institute (BARI) and Rangpur Dinajpur Rural Service (RDRS) Bangladesh with full participation of the farmers' groups of respective project site. The specific objectives of the project are: (1) to estimate the productivity of paddy, wheat, maize, tobacco and sugarcane (2) to determine the cost of production and returns to the above mentioned crops (3) to study the interrelationship between inputs and output of the above mentioned crops and (4) to examine the resource utilization patterns at farm level. In this project analysis, the net incremental profit is US$33,028. The expected incremental project benefit and incremented production cost are estimated as US$ 219,959 and US$ 186,931 respectively. The financial decision making criteria would be followed in this crop cultivation practice project. After the project implementation, the expected project benefits are assumed to be continued for 15 years. The benefit cost ratio (B/C) of the project is estimated at 1.077 (table 11) when using discount rate of 10% as an opportunity cost of capital in Bangladesh. FIRR of project is estimated at 26.15% which is bigger than the opportunity cost by more than double. So this project is financially feasible and acceptable. Therefore, this project should be extended to other areas to increase the farm income and economic growth of marginal poor farmers in Bangladesh.
This study analyzed the physicochemical characteristics of Astragalus membranaceus (AM) fermented with seven different mushroom mycelia. Physicochemical characteristics, such as contents of moisture, pH, total reducing sugars, free sugar, and isoflavonoid, were investigated. The moisture content was increased in most of the samples. The pH values of AM fermented with Phellinus linteus and Flammulina velutipes were increased, while the pH of other samples were similar to that of non-fermented AM. The reducing sugar content was in the range of 211.69~391.74 mg/100 g. The extraction yield using water was higher than that when extracted with 80% ethanol. The free sugar content was increased through fermentation with mushroom mycelia. However, the glucose contents of the 80% ethanol and water extracts were decreased. Finally, the calycosin and formononetin contents in 80% ethanol and water extracts of AM fermented with Phellinus linteus were 2,549.24 mg/g, and 827.66 mg/g for calycosin, and 1,366.69 mg/g and 221.28 mg/g for formononetin, respectively. These results suggest that fermentation with mushroom mycelia could be used to increase the bioactivity of AM. The mycelium-fermented AM might be a valuable source of functional material and edible resource for industry.
Null mutants generated by targeted gene replacement are frequently used to reveal function of the genes in fungi. However, targeted gene deletions may be difficult to obtain or it may not be applicable, such as in the case of redundant or lethal genes. Constitutive expression system could be an alternative to avoid these difficulties and to provide new platform in fungal functional genomics research. Here we developed a novel platform for functional analysis genes in Magnaporthe oryzae by constitutive expression under a strong promoter. Employing a binary vector (pGOF1), carrying $EF1{\beta}$ promoter, we generated a total of 4,432 transformants by Agrobacterium tumefaciens-mediated transformation. We have analyzed a subset of 54 transformants that have the vector inserted in the promoter region of individual genes, at distances ranging from 44 to 1,479 bp. These transformants showed increased transcript levels of the genes that are found immediately adjacent to the vector, compared to those of wild type. Ten transformants showed higher levels of expression relative to the wild type not only in mycelial stage but also during infection-related development. Two transformants that T-DNA was inserted in the promotor regions of putative lethal genes, MoRPT4 and MoDBP5, showed decreased conidiation and pathogenicity, respectively. We also characterized two transformants that T-DNA was inserted in functionally redundant genes encoding alpha-glucosidase and alpha-mannosidase. These transformants also showed decreased mycelial growth and pathogenicity, implying successful application of this platform in functional analysis of the genes. Our data also demonstrated that comparative phenotypic analysis under over-expression and suppression of gene expression could prove a highly efficient system for functional analysis of the genes. Our over-expressed transformants library would be a valuable resource for functional characterization of the redundant or lethal genes in M. oryzae and this system may be applicable in other fungi.
Dollar spot, caused by Sclerotinia homoeocarpa, is the major disease in cool-season turfgrasses. Understanding the distribution of this pathogen in soil and thatch is important to developing disease control strategies. In this study, toothpicks were used to detect S. homoeocarpa in the turfgrass canopy, thatch, and soil at different distances from dollar spot infection centers. The effect of penetrant and contact fungicide applications with different water volumes on distribution of S. homoeocarpa was also investigated. S. homoeocarpa was detected in 100% of samples taken from the leaf canopy, 83.3% in thatch area, and 0% in the soil from within the infection center. S. homoeocarpa was isolated in 100% of samples taken from the edge of the infection center, but was only detected in 13% of the samples taken at 1.5 cm away from the infection center edge. S. homoeocarpa was isolated at a higher frequency in the propiconazole treated plots than those treated with chlorothalonil and was not detected in leaf canopy samples when either fungicides was applied with 6.78 L of water. In conclusion, the toothpick-aided detection technique has improved our understanding of S. homoeocarpa epidemiology and could be used as a diagnostic tool to detect for fungicide resistance on golf courses.
