Occurrence of tomato late blight (Phytophthora infestans) has caused significant losses in tomato yield in all over the world. Evaluation of the level of resistance in tomato gene resources for main breeding and initiation of the resistance breeding program are important for control of this disease. Resistant assay of 78 tomato cultivars/lines to late blight in pots and field experiment was carried out under controlled and natural conditions in 2009. All commercial cultivars including 'Legend' were susceptible. However, 10 lines including KNU-2, KNU-6-1, KNU-11, KNU-13, KNU-14-1 lines distributed from University of California, Riverside and L3708, $AV107-4{\times}L3708$, $07-15{\times}L3708$, $BS67{\times}L3708$ lines which have resistant gene Ph-3 and $06-9-62A{\times}06-9-62A$ were highly resistant to late blight. These highly resistant lines can be used as resources of resistance to late blight in a tomato breeding program in future.
Using the abundant available information about the tomato genome, we developed DNA markers that are linked to disease resistant loci and performed marker-assisted selection (MAS) to construct multi-disease resistant lines and varieties. Resistance markers of Ty-1, T2, and I2, which are linked to disease resistance to Tomato yellow leaf curl virus (TYLCV), Tomato mosaic virus (ToMV), and Fusarium wilt, respectively, were developed in a co-dominant fashion. DNA sequences near the resistance loci of TYLCV, ToMV, and Fusarium wilt were used for primer design. Reported candidate markers for powdery mildew-resistance were screened and the 32.5Cla marker was selected. All four markers (Ty-1, T2, I2, and 32.5Cla) were converted to cleavage amplification polymorphisms (CAPS) markers. Then, the CAPS markers were applied to 96 tomato lines to determine the phenetic relationships among the lines. This information yielded clusters of breeding lines illustrating the distribution of resistant and susceptible characters among lines. These data were utilized further in a MAS program for several generations, and a total of ten varieties and ten inbred lines were constructed. Among four traits, three were introduced to develop varieties and breeding lines through the MAS program; several cultivars possessed up to seven disease resistant traits. These resistant trait-related markers that were developed for the tomato MAS program could be used to select early stage seedlings, saving time and cost, and to construct multi-disease resistant lines and varieties.

In order to develop chili pepper bacterial spot resistant cultivars and near-isogenic lines (NILs) to prompt the molecular mapping of the resistance gene, we have run backcross breeding program since 1994. Two resistance genes against Xanthomonas axonopodis pv. vesicatoria Bs2 from Fla. XVR 3-25 and Bs3 from our breeding line 25-11-3-2, were introduced into a land race, Chilseongcho (abbreviated to Chilseong hereafter) with good fruit guality. We report here the testing of $BC_4F_3\;to\;BC_4F_5$. We found that $BC_4F_5$ lines of the crosses were homozygous with respect to the respective genes of introduction. The lines, in which Bs2 gene was introduced, were hypersensitively resistant to both race 1 and race 3 of X. axonopodis pv. vesicatoria, whereas, those in which Bs3 was introduced were resistant to race 1.
This review describes the stratagies of development of virus-resistant Alstroemeria plants using the genetic modification system. Despite of increasing of its importance in cut flower market, improvements of some horticultuirally important traits such as fragrance, long vase-life, virus resistance and tolerance against abiotic stresses are lack of the breeding program in Alstroemeria. Of these traits, virus-resistance is quite difficult to develop in Alstroemeria plants due to the limitations of genetic variation in the existed germplasm. To extend the genetic variation, plant biotechnological techniques such as genetic transformation and tissue culture should be combined to develop virus-resistant line in Alstroemeria. In this review, several strategies for the generation of virus-resistance by using natural resistance genes, pathogen-derived genes and other sources including pathogen-derived proteins, virus-specific antibodies and ribosome-inactivating proteins are presented. Also, brief histories of breeding, tissue culture, and transformation system in Alstroemeria plants are described to inderstand of the application of transgenic approach for the development of virus-resistance in Alstroemeria species.
Insects are among the most important abiotic and biotic constraints to rice production. National rice research programs are in various stages in the development and implementation of integrated pest management (IPM) stratagies for rice insect control. Among the various control tactics, insect resistant cultivars are sought as the major tactic in rice IPM. Through the activities of interdisciplinary teams of scientists significant progress has been made in the development and release of insect resistant cultivars to farmers. Because of its compatibility with other control tactics insect resistance has proven to fit well into the IPM approach to rice insect control agents and minimize the need for insecticide applications. The development of biotypes which overcome the resistance in rice plants has been a significant constraint in the breeding of rice for resistance to insects. Most notable examples in Asia are the green leafhopper, Nephotettix virescens, brown planthopper, Nilaparvata lygens and the Asian rice gall midge, Orseolia oryzae. The current breeding stratege is to develop rice cultivars with durable resistance on which virulent biotypes cannot adapt. In spite of the significant progress made in the breeding of insect resistant cultivars there are still numerous important rice insect species for which host plant resistance as a control tactic has not been fully utilized. Advances in biotechnology provide promise of solving some of the problems that have limited the use of host plant resistance as a major tactic in the integrated management of rice insect pests.

Bud rot (BR) is the most devastating disease affecting oil palm (Elaeis guineensis) crops in Colombia. Its causal agent, Phytophthora palmivora, initiates the infection in immature oil palm leaflets producing necrotic lesions, followed by colonization of opportunistic necrotrophs, which increases disease damage. To improve the characterization of the disease, we transformed P. palmivora using Agrobacterium tumefaciens-mediated transformation (ATMT) to include the fluorescent proteins CFP-SKL (peroxisomal localization), eGFP and mRFP1 (cytoplasmic localization). The stability of some transformants was confirmed by Southern blot analysis and single zoospore cultures; additionally, virulence and in vitro growth were compared to the wild-type isolate to select transformants with the greatest resemblance to the WT isolate. GFP-tagged P. palmivora was useful to identify all of the infective structures that are commonly formed by hemibiotrophic oomycetes, including apoplastic colonization and haustorium formation. Finally, we detected cell death responses associated with immature oil palm tissues that showed reduced susceptibility to P. palmivora infection, indicating that these tissues could exhibit age-related resistance. The aim of this research is to improve the characterization of the initial disease stages and generate cell biology tools that may be useful for developing methodologies for early identification of oil palm materials resistant or susceptible to BR.
Although zoysia grass (Zoysia spp.) has a lot of excellent chracteristics as warm-season grass, it have been limited in use due to slow establishment, low seed production, poor shade tolerance and other factors. Breeding trials have been continued from 1900's, much attentions have been paid especially in U.S.A., Korea and Japan recently. In U.S.A., more than 24 varieties had been evaluated at National Turfgrass Evaluation Program(NTEP) from 1991 to 1995 and some were regsistered as commercial. After the 6th International Turfgrass Research Conference at Japan in 1989, Japan Turfgrass In-corporation (JTI) sponsored by private companies and government carried out breeding programs for pest, salt, and shade tolerant and herbicide resistant varieties. JTI also has been trying to im-prove vigor and breed evergreen zoysia Korean breeders collected germplasms since 1960's. After USDA breeders came to Korean penesula in 1982, Korean breeders joined with USDA zoysia breeding project for several years. Many interspecific hybrids and natural selected varieties were breeded that period both in U.S.A and Korea. Breeding objectives were to extend green color period, improve leaf quality and density, and better leaf color at dormant stage. Since 1990's, zoysia grass breeding trials are getting more diverse in many points such as random amplified polymorphic DNA (RAPD) assay for ecotype identification. The objectives of this study are to evaluate germplasms in Korea, and also review the present status and future prospect in zoysia grass breeding in the world.
For initiation of resistance breeding program of the red pepper, 21 PR ($Phytophthora$ resistance) cultivars, 36 cultivars collected from USA and 'Supermanitta' which is a susceptible cultivar against phytophthora blight were assayed against phytophthora blight, powdery mildew, and anthracnose. For seedling assay of phytophthora blight, three different mating type strains of $Phytophthora$$capsici$ were used (A1, A2, Sterile). The result showed that most of the pepper of PR cultivars were resistance or moderately resistance at each mating type. 'Yeokganghongjanggun' was resistant to all three $P.$$capsici$ strains and 'PR-Datta' and 'PR-Manitta' were resistant or moderately resistant at each type. In case of the collected cultivars, 'NuMex J.E.Parker', 'Omni Color', and 'SCM334' were resistant to all the three types and some cultivars including 'Sweet Banana' and 'Tabasco' were moderately resistant to each type fungi. 'Orange Habanero' and 'Black Cuban' were resistant to powdery mildew and 'Supermanitta' and 'PR Keumdong' were moderately resistant, while 'Santa Fe Grande', 'NuMex Pinata' and 'Puya' were very susceptible. In the case of anthracnose, 'Aji Limon' and 'Capsicum baccatum var. pendulum 3-4' were resistant and 'Pobalno', 'Omni Color', 'Negro', 'Mesilla', 'Mulato', 'Bhut Jolokia', 'Big Dipper', 'Black Cuban', 'NuMex Pinata', and 'NuMex Big Jim' were moderately resistant. The most PR cultivars except 'Taesan' were susceptible or very susceptible. These resistant individuals identified through this experiment can be used as sources of resistance to pepper pathogens in the future breeding programs.
Phytophthora blight caused by Phytophthora capsici Leonian is a dangerous disease threatening pepper growers worldwide. The efficacy of chemical control is generally low as the pathogen is soil-borne and rapidly spread by zoospores during the rainy season. Thus, based on the demand for resistant varieties, various good resistant sources, such as CM334, AC2258, and PI201234, have been reported and their inheritance of resistance studied by many different authorities. However, the mode of inheritance remains unclear, as 1 or 2 independent dominant genes, 3 genes, or multiple genes have all been reported as responsible for resistance. Recently, QTL mappings of the gene factors for resistance have been reported, and molecular markers for resistance used in breeding programs. With the release of many resistant commercial hybrid cultivars, differentiation of pathotypes of the pathogen is attracting interest among breeders and plant pathologists. Various authorities have already classified the pathogen strains into different races according to the inter-action between resistant host plants, including the source of resistance, such as CM334 and PI201234, and resistant commercial varieties and P. capsici isolates. However, no standard differential host sets have yet been established, so the results are good only for the pathogen strains used in the experiments. Thus, for breeding varieties with durable resist-ance, it is important to introduce resistance from different sources and use diverse local pathogen strains collected in the target area for distribution in a breeding program.
Rice blast is one of the most serious disease threatening stable production of rice. Breeding of resistant cultivars has been used as the most effective and useful method to controll rice blast caused by Magnaporthe oryzae. To collect rice blast isolates in fields and test their pathogenicity on new cultivars are important for establishment of new resistant cultivars breeding program of rice. Pathotypes of Korean rice blast isolates have been categorized to Korean differential race system developed in 1985. However, it is little known about genetic background of Korean differential cultivars, so that it is hard to understand for relationship between each pathogen and each host plant at genetic level. In this study, we suggested necessity of a new differential system by analyzing pathogenic responses between 24 monogenic rice lines and 200 Korean rice blast isolates. In addition, we determined the nine representative resistant genes based on the resistance responses of the monogenic lines to rice blast isolates, indexed resistant responses of the monogenic lines to ten representative rice blast isolates and selected 30 Korean representative rice blast isolates proper to Korean system. We think the newly developed differential race system can be broadly used to select resistant cultivars to rice blast in Korea.
