Comparisons of Protein-bound Polysaccharide Contents Obtained from Mycelial Cultured Broth and Fruit body of Coriolus versieolor (구름버섯의 균사배양액 및 자실체로부터 얻어진 단백다당체의 수율비교)

  • This study was performed to obtain higher yield rate of protein-bound polysac-charide extracted from cultured broth and fruit body of Coriolus versieolor, basidiomycetes. 16002 strain produced 0.58%, the highest yield rate among four strains, and there was no significantly different yield in other three strains. The optimal liquid media were CVT-III to obtain higher yield of protein-bound polysaccharide comparing with the various culture media. Compared with several materials for substitution of medium source, the residue extract media of Coriolus versieolor fruit-body were yield rate similar to control, and addition of the extracts of ginseng residues increased 11.4-48%. And also compared with fruit bodies that were artificially cultured on the oak longs, 16002 strain produced the highest yield similar to the result of cultured broth. Fruit body of the prematures stage was more effective to obtain high yield rate.

  • Pseudomonas sp. S-47 is capable of degrading 4-chlorobenzoate to produce 5-chloro-2-hydroxymuconic semialdehyde (5C-2HMS) by the enzymes encoding by xylXYZLTE cluster. In this study, the resulting 5C-2HMS was confirmed to be transformed to 5-chloro-2-hydroxymuconic acid (5C-2HMA) by 5C-2HMS dehydrogenase. The xylG gene encoding 5C-2HMS dehydrogenase was cloned from the chromosomal DNA of strain S-47. The nucleotide sequence of xylG showed to be composed of 1,600 base pairs with ATG initiation and TGA termination codons. A deduced amino acid sequence of the 5C-2HMS dehydrogenase (XylG) exhibited 98%, 93%, and 89% identity with those of the dehydrogenases from P. putida mt-2, P. putida G7, and Pseudomonas sp. CF600, respectively.

Fate of 14C - Carobofuran in Rice Plant and Paddy Soil (수도체(水稻體) 및 담수토양중(湛水土壤中) $^{14}C$-Carbofuran의 행적(行跡)에 관한 연구(硏究))

  • A study was undertaken to investigate the absorption, translocation and metabolism of carbofuran in rice paddies. Ring-3-$^{14}C-carbofuran$ applied onto the paddy soil surface was rapidly absorbed and translocated into rice plants. Within 2 days after treatment, it was observed that carbofuran reached shoot tips and accumulated. More than 15% of total radioactivity was recovered in rice plant from 3 to 20 days after treatment. In organic soluble fraction of rice plant extract, 3-hydroxycarbofuran was the major metabolite recording 43% and 4% of total organic soluble radioactivity in shoot and root at 20 days respectively. 3-Ketocarbofuran and phenolic metabolites including carbofuran phenol, 3-hydroxycarbofuran phenol and 3-ketocarbofuran phenol were also detected in the organic soluble fractions. Some glycosidic conjugates of carbofuran metabolites were found in water soluble fraction of rice plant extract and 3-hydroxycarbofuran was the most abundant aglycone. Radioactivity in paddy soil was rapidly decreased until 3 days after treatment and then maintained almost constant level. A significant portion (42∼56 %) of the total radioactivity remained in soil as nonextractable residue from 5 to 20 days after treatment. The nonextractable radioactivity was mainly located in soil organic matter distributing in humin, fulvic acid and humic acid fractions with the decreasing order. Evolution of $^{14}CO_2$ from ring cleavage of $3-^{14}C-carbofuran$ was negligible recording only 1.8% of total radioactivity during 20 days after treatment.

Determination of Oxolinic Acid in Paddy Soil by HPLC Coupled with UV Detector (HPLC-UV검출기를 이용한 논토양 중 oxolinic acid 분석)

  • This study was performed to examine analytical method of a quinolone compound, oxolinic acid in paddy soil by HPLC coupled with UV detector. Two types of soil texture in different regions were used for this experiment. Oxolinic acid was extracted by a 4 M-KOH : MeOH(1 : 3, v/v) mixtures and acidified followed by liquid-liquid partitioning in dichloromethane. Dichlormethane layer was dehydrated, evaporated and analyzed by HPLC (262 nm). Retention time was 10.2 min. The standard calibration curve of oxolinic acid showed linearity ($r^2>0.999^{**}$, y=378.99x+135.08) in the range of $1{\sim}40$ ng. The mean recoveries, evaluated from fortified soil samples at two concentration levels of 0.2 mg/kg and 1.0 mg/kg, were $90.9{\pm}4.52%$(C.V. 4.97%) and $95.0{\pm}0.23%$(C.V. 0.24%) for soil 1 and $92.2{\pm}1.15%$(C.V. 1.25%) and $93.1{\pm}0.31%$ (C.V. 0.33%) for soil 2, respectively The detection limits of two types of soils were same as 0.05 ppm. Overall, the present analytical method of oxolinic acid by HPLC coupled with UV detector seems to be used reasonably.

Mobility of pesticides in different soil textures and gravel contents under soil column (토양 column을 이용한 토성 및 자갈함량별 농약 이동특성)

  • This study was investigated to elucidate the pesticide mobility in three different soil textures(Heongseong sandy loam; Chuncheon, loam; Taeback, silty clay loam) and four different gravel contents(0, 20, 40, 60%) of Taebaek soil using soil column. Carbofuran, which ranks the highest water solubility among 7 pesticides(carbendazim, carbofuran, chlorpyrifos, cypermethrin, dimethomorph, diniconazole and endosulfan) was defected over 87% in leachate samples within all soil types from early sampling time. Amount of 5 residual pesticides excluding carbendazim and carbofuran were ordered silty clay loam > loam > sandy loam, indicating pesticide residues are related to percentage of clay contents in soils. Comparing the amount of residual pesticides in soil column(upper, middle and lower layer), 6 pesticides apart from carbofuran were found in the range of $50{\sim}92%$ on the upper layer of silty clay loam and loam. Mobility of pesticides either in soil or leachate samples is dependant on water solubility of pesticide and clay content of soil. The results obtained from four different gravel contents of Tacback soil were similar to the results of three different soil textures. Also it was found that more the gravel contents, faster the flow velocity of leachate water. These results possibly provide an idea to select proper pesticides and to reduce soil and water contamination at alpine and sloped-land.

Reduction of pesticide residues in/on mini-tomato by washing at the different harvest days after pesticide spray (약제 살포 후 경과일수별 세척에 의한 방울토마토의 잔류농약경감)

  • The objective of this study was to compare the reduction of residues in/on mini-tomato by washing methods and to evaluate the relation of reduction rates in different pesticides. Mini-tomato was sprayed with three commercial pesticides, oxadixyl, thiophanate-methyl and chlorothalonil, and harvested for washing test as 0, 5 and 7 days elapsed. The reduction rates of oxadixyl in/on mini-tomato were 61, 11 and 4%, those of thiophanate-methyl were 89, 89 and 79%, and in case of chlorothlaonil, reduction rates were 84, 84 and 88%, respectively When mini-tomato at 0 day and 5 days after spray was washed by 0.02% of $Sur-ten^{(R)}$ solution, its pesticide residues were reduced by 81 and 55% on oxadixyl, 95 and 91% on thiophanate-methyl, and 97 and 98% on chlorothlaonil. When three wettable powdered pesticides with the same recipe were sprayed, their reduction rates in/on mini-tomato by washing with water at 0, 5 and 7 days were 76, 66 and 43% on oxadixyl, 91, 83 and 82% on thiophanate-methyl, and 65, 68 and 82% on chlorothlaonil, suggesting that the washing effect may be dependent on the amounts of active ingredients and inert types of the pesticides used.

Extraction of pesticide residues in medical herbs by microwave (Microwave를 이용한 한약재 중의 잔류농약 추출)

  • This study was conducted to elucidate extraction efficiency by microwave in comparison with Soxhlet for extraction of pesticide residues in dried medical herbs; red-ginseng, white-ginseng, Bupleuri Radix, Angelica gigas Nakai, Rehmannia glutinosa. The acetone extraction by microwave of tolclofos-methyl and quintozene in medical herbs was efficient. The extraction efficiency by microwave with power 45 to 150 watts, extraction time 1 to 5 minutes and solvent volume 30 ml was compared with that of Soxhlet with extraction time 7 hours and solvent volume 150 ml. The extraction efficiency by microwave with extraction time 3 to 5 minutes was similar with extraction time of 7 hours by Soxhlet. When medical herbs spiked with tolclofos-methyl and quintozene was analyzed to how the extraction efficiency of microwave by kind of medical herbs, the extraction efficiency by microwave with extraction time of 3 to 5 minutes was the same as Soxhlet extraction. The optimal condition for extraction of tolclofos-methyl and quintozene in medical herbs by microwave was 45 to 90 watts of power supply, 3 to 5 minutes of extraction time and acetone 30 ml of solvent volume.

Monitoring and Safety Assessment of Pesticide Residues on Agricultural Products Sold via Online Websites (온라인 판매 농산물 잔류농약 실태 및 안전성 평가)

  • This study was carried out to monitor the current status of pesticide residues in selling agricultural products via online and assessed their safety in 2014. A total of 124 samples were purchased six times from March to August 2014 twenty online shopping malls randomly. These samples were analysed 208 pesticides by multiresidue method using a GC-ECD/NPD and a LC-MS/MS and confirmed by a GC-MSD. As a result of analysis, residual pesticides samples were 11 (8.9%) such as leek, young radish, welsh onion etc, of which 2 samples (1.6%) such as sesame bud (Chlorothalonil), artemisia (Chlorpyrifos) were violated Korea Maximum Residue limits (MRLs). 11 kinds of pesticides (19 times) were detected in 11 samples. Risk assessment evaluated human health exposure with the ratio of EDI (Estimated daily intake) to ADI (Acceptable daily intake) of pesticides detected. %ADI (the ratios of EDI to ADI) were 0.04~95.70% and some samples represented a fairly dangerous levels. In particular, Chlorothalonil in the sesame bud was shown as a significant risk close to 100% of %ADI. Accordingly, it is recommended to strengthen a safety check on agricultural products in online sales.

Biotinoyl Domain of Human Acetyl-CoA Carboxylase;Structural Insights into the Carboxyl Transfer Mechanism

  • Acetyl-CoA carboxylase (ACC) catalyzes the first step in fatty acid biosynthesis: the synthesis of malonyl-CoA from acetyl-CoA. As essential regulators of fatty acid biosynthesis and metabolism, ACCs are regarded as therapeutic targets for the treatment of metabolic diseases such as obesity, In ACC, the biotinoyl domain performs a critical function by transferring an activated carboxyl group from the biotin carboxylase domain to the carboxyl transferase domain, followed by carboxyl transfer to malonyl-CoA. Despite the intensive research on this enzyme, only the bacterial and yeast ACC structures are currently available, To explore the mechanism of ACC holoenzyme function, we determined the structure of the biotinoyl domain of human ACC2 and analyze its characteristics using NMR spectroscopy. The 3D structure of the hACC2 biotinoyl domain has a similar folding topology to the previously determined domains from E. coli and P. Shermanii, however, the 'thumb' structure is absent in the hACC2 biotinoyl domain. Observations of the NMR signals upon the biotinylation indicate that the biotin group of hACC2 does not affect the structure of the biotinoyl domain, while the biotin group for E. coli ACC interacts directly with the thumb residues that are not present in the hACC2 structure. These results imply that, in the E. coli ACC reaction, the biotin moiety carrying the carboxyl group from BC to CT can pause at the thumb of the BCCP domain. The human biotinoyl domain, however, lacks the thumb structure and does not have additional non-covalent interactions with the biotin moiety; thus, the flexible motion of the biotinylated lysine residue must underlie the "swinging arm" motion. This study provides insight into the mechanism of ACC holoenzyme function and supports the "swinging arm" model in human ACCs.

Synergistic Effect of Citric Acid on Antioxidant Property of Red Pepper (고추의 산화성(酸化性)에 대한 Citric Acid의 상승효과)

  • The antioxidant activity of ground red pepper, ethyl alcohol extracts of ground red pepper peel and alcohol extracts plus organic acid was studied by using edible soybean oil as a substrate. All the substrates were stored in a dark place at $45.0{\pm}0.5^{\circ}C$ respectively. The results of the study were as follow: 1. Ground red pepper, black pepper and cassia exhibited slight antioxidant activity and there was no significant difference in antioxidant activity among them. 2. Although both ground red pepper peel and red pepper seed showed antioxidant activity, ground red pepper peel exhibited greater antioxidant activity than ground red pepper peel did. 3. Ethyl alcohol extracts of red pepper peel showed strong antioxidant activity, but the residue did not show the antioxidant activity. 4. The antioxidant activity of the ethyl alcohol extracts of red pepper peel increased in direct proportion to the concentration of the extracts. 5. In connection with the synergistic effect of organic acid to the ethyl alcohol extracts of red pepper peel, citric acid showed a very strong synergistic effect on the antioxidant activity of red pepper, while ascorbic acid showed a weak effect. On the other hand, phosphoric acid showed an accelerating effect on the oxidation of oils. 6. The maximum antioxidant activity of the ethyl alcohol of ground red pepper peel was observed at the citric acid concentration of 0.04%, when tested at various concentration of citric acid.
