Reanalysis of discarded blastocysts for autosomal aneuploidy after sex selection in cleavage-stage embryos

  • Objective: The goal of the present study was to investigate the rate of chromosomal aneuploidies in surplus embryos after sex determination at the cleavage stage. Then, the same chromosomal aneuploidies were evaluated in blastocysts after extended culture. Methods: Sixty-eight surplus embryos were biopsied at the cleavage stage and incubated for an additional 3 days to allow them to reach the blastocyst stage. The embryos were reanalyzed via fluorescence in situ hybridization (FISH) to examine eight chromosomes (13, 15, 16, 18, 21, 22, X, and Y) in both cleavage-stage embryos and blastocysts. Results: Although the total abnormality rate was lower in blastocysts (32.35%) than in cleavage-stage embryos (45.58%), the difference was not significant (p=0.113). However, when we restricted the analysis to autosomal abnormalities, we observed a significant difference in the abnormality rate between the cleavage-stage embryos (44.11%) and the blastocysts (17.64%, p=0.008). A higher rate of sex chromosomal abnormalities was also observed in cleavage-stage embryos (29.4%) than in blastocysts (14.70%, p=0.038). Conclusion: The data indicated that embryo biopsy should be conducted at the blastocyst stage rather than the cleavage stage. The results also emphasized that examination of common chromosomal aneuploidies apart from sex selection cycles can be conducted in the blastocyst stage with the FISH method.

Reproductive Biology of the Pen Shell, Atrina(Servatrina) pectinata on the Boryeong Coastal Waters of Korea

  • The gonad index, condition index, reproductive cycle and spawning of the pen shell Atrina (Servatrina) pectinata were investigated using samples from the subtidal zone of Nokdo on the Boryeong coastal waters of Korea. Samples were collected monthly by SCUBA divers for one year from January to December, 2001. A. (Servatrina) pectinata is dioecious and oviparous. The spawning season of this species occurred once a year from June to August, with the main spawning occurring between June and July when the seawater temperature was around $20^{\circ}C$. Ripe oocytes were about 60-65 ${\mu}m$ in diameter. The reproductive cycle of this species could be classified into five successive stages; early active stage (November to March), late active stage (February to May), ripe stage (April to July), partially spawned stage (June to August), and spent/inactive stage (August to October). Monthly changes in the gonad index reached a maximum (4.6) in May (ripe stage), thereafter, the GI values gradually decreased from June to August when spawning occurred continuously. Therefore, monthly changes in the GI values showed a similar pattern to the gonadal phase. The condition index (CI) of the meat part without the posterior adductor muscle reached the maximum in June (ripe and partially spawned stage) and the minimum in September (spent/inactive stage), Accordingly, monthly changes in the condition indice of the meat part without the posterior adductor muscle coincided with the gonadal phases.

Reproductive Cycle of BlueStriped Angelfish, Chaetodontoplus septentrionalis in Jeju Coastal Waters

  • Reproductive cycle of the blue-striped angelfish, Chaetodontoplus septentrionalis were histologically investigated. Fish were monthly collected in the coastal waters of Munseom, Seogwipo, Jeju-Island from February to December 2018. The gonadosomatic index (GSI) increased from May and maintained high values in August. The reproductive cycle of female fish can be classified by the characteristics observed during gonadal development as followed: growing stage (November to June), early mature stage (May to June), mature and spawning stage (June to September), and degenerative and recovery stage (September to December). In the male, testicular development period was similar to that of ovarian development period, but mature and spawning period was one month longer from June to October. Fecundity of mature female ranged from 4,601 to 22,840 and was correlated positively with total length and body weight. The histological analysis of gonadal development indicated that the C. septentrionalis was summer-spawning type and is considered a multiple spawner during spawning season.

  • We investigated the gonadosomatic index (GSI), germ cell development, reproductive cycle of the Afriean lungfish Protorierus annecteus (Owen) by histological observations and morphometric data. Samples were collected from the river Orie and its flood of Nigeria, from January to December 2000. The fish is dioecious and oviparous. Monthly changes in the gonadosomatic index (GSI) showed a similar pattern to change in the mean oocyte diameter and the reproductive cycle. The reproductive period occurred from March to July-August; the spawning period was once a year between truly and August, and the main spawning occurred in August when active and voracious feeding occurred during the rainy season. In the resting (dormant) stage after spawning, fish stopped feeding and aestivated during the dry season from December to February. The reproductive cycle of the species can be divided into five successive stages, quiescent stage (March to April), developing/maturing stage (April to lune), ripe/spawning stage (July to August), post-spawning stage (September to November), and resting (dormant) stage (December to February).

Reproductive Cycle of the Ark Shell, Scapharca subcrenata, on the West Coast of Korea

  • Monthly changes in the gonad index (GI), egg-diameter composition, gonadal development, reproductive cycle of the ark shell, Scapharca subcrenata, were investigated by histological method and morphometric data. This species is dioecious and oviparous. The gonad is located among the subregion of mid-intestinal gland, digestive diverticula and the outer fibromuscular layers compacted by the fibrous connective tissues and muscle fibers. The gonad index sharply increased in May, reached the maximum value in June, and then gradually decreased from July to December. The reproductive cycle of this species can be divided into six successive stages: early active stage (January to May), late active stage (June to July), ripe stage (June to August), partially spawned stage (July to September), degenerative stage (August to December), and resting stage (January to April). S. subcrenata spawns once a year between July and early September, and the main spawning occurred between July and August when the water temperatures were above $20^{\circ}C$. This evidence suggest that timings of maturation and spawning are closely related to water temperatures. Even though the spawning period was once a year, it is assumed that the number of spawning frequencies (broods) may occur more than twice during the spawning season.

  • This study was carried out to assess the reproductive toxicity of 2-bromopropane (S-BP) using spermatogenesis stage classification and Sertoli cell indices (SCI).Vehicle control olive oil and 2-BP doses of 125, 250 and 500 mg/kg of body weight were injected in the interaperitoneum of 12 weeks male Sprague-Dawley rats for 28 days respectively of SCI on germ cells including the spermatogonia of stages II-III, Ⅵ,Ⅹ, XII, ⅩIII, and spermatocytes of stages VIII (preleptotene), Ⅹ (leptotene), XII (leptotene), V and Ⅵ (pachytene), and the round spermatids of stage Ⅵ. Considering the process of maturation depletion in spermatonesis, spermatogonia may be the primary target cells of 2-BP toxicity.

  • Spermatogenesis and the reproductive cycle in male Spisula sachalinensis were investigated by cytological and histological observations. The morphology of the spermatozoon has a primitive type and is similar to those of other bivalves in that it contains a short midpiece with four mitochondria surrounding the centrioles. But spermatozoon of this species has not axial rod and satellite body in the midpiece. The morphologies of the sperm nucleus type and the acrosome shape of this species have a globe-shape type and modified cap-like shape, respectively. The spermatozoon is approximately $40-45{\mu}m$ in length including the sperm nucleus length (about $1.35{\mu}m$), acrosome length (about $1.50{\mu}m$) and tail flagellum. The axoneme of the sperm tail flagellum consists of nine pairs of microtubules at the periphery and a pair at the center. The axoneme of the sperm tail shows a 9+2 structure. The spawning period of these species lasts from June to July, and the main spawning occurs in July when seawater temperatures are greater than $20^{\circ}C$. The male reproductive cycle of this species can be categorized into five successive stages: early active stage (October to January), late active stage (February to April), ripe stage (April to June), partially spawned stage (June and July), and spent/inactive stage (August to September).

Reproductive Cycle and First Sexual Maturity of Sinonovacula constricta(Lamarck, 1818)(Bivalvia: Pharidae) in Western Korea

  • The gonad index, reproductive cycle and first sexual maturity of Sinonovacula constricta collected from Simpo, Kimje-gun, Korea were investigated by histological analysis. The gonad index(GI) in both sexes of S. constricta increased from April and reached a maximum in July when the water temperature rapidly increased. And then, the GI values gradually decreased by spawning from August through October. Monthly variations in the GI showed a close relationship with ovarian development. The reproductive cycle in females and males can be classified into five successive stages: early active stage(March to June), late active stage(May to July), ripe stage(July to September), partially spawned stage(August to October), spent/inactive stage(October to March). The percentage of first sexual maturations in female and male clams of 50.1-60.0 mm in shell length was over 50%, and for clams over 70.1 mm in shell length, it was 100%. Because harvesting clams < 50.1 mm in shell length could potentially cause a drastic reduction in recruitment, a measure including a prohibitory fishing size should be taken for adequate improved fisheries resource management.

  • Maturation and spawning of the Blackthroat seaperch, Doderleinia berycoides were investigated based on the samples captured in Korean waters from January 2008 to December 2009. Gonadosomatic index began to increase in June, and reached maximum between July to September. After spawning it began to decrease from October. Reproductive season was estimated to July-September, with peak in August. Fecundity was proportional to the size of the female, with the clutch size varying from 115,500 eggs in the smallest female (TL〓28.2cm) to 652,000 eggs in the largest (TL〓33.5cm). Size at 50% sexual maturity ($TL_{50}$), determined from mature females, was 29.6cm. Annual reproductive cycles of this species could be divided into six successive stages; immature stage (October-May), nucleolus stage (June-July), yolk vesicle stage (July-August), vitellogenic stage (June-September), ripe and spent stage (August-October).

Ultrastructure of Germ Cells during Spermatogenesis and the Reproductive Cycle in Male Meretrix petechialis on the West Coast of Korea

  • Gonad index, spermatogenesis and the reproductive cycle of Meretrix petechialis were investigated by cytological, histological observations. Monthly changes in the gonad index coincides the gonadal development. The morphology of the spermatozoon had a primitive type and is similar to that of other bivalves having a short mid-piece with five to six mitochondria surrounding the centrioles. The morphology of the sperm nucleus type and the acrosome shape of this species were cylindrical type and cap-like shape, respectively. The spermatozoon was approximately 40-45 ${\mu}m$ in length including the sperm nucleus length (about 1.50 ${\mu}m$), acrosome length (0.60 ${\mu}m$) and tail flagellum. The axoneme of the tail flagellum consisted of nine pairs of microtubules at the periphery and a pair at the center. The axoneme of the sperm tail showed 9 + 2 microtubular arrangement. The spawning period was from June to September and the main spawning occurred from July to August when seawater temperatures were higher than $20^{\circ}C$. The reproductive cycle of this species could be categorized into five successive stages: early active stage (February to March), late active stage (February to May), ripe stage (April to July), partially spawned stage (June to September), and spent/inactive stage (September to February).
