Artificial insemination (AI) has been performed widely in swine industry using fresh liquid sperm instead of frozen type of sperm. However fresh sperm are not able to preserve more than three days with optimal motility and other sperm parameters for the successful fertilization, since in vitro stored sperm has an oxidative stress that resulted increase of abnormality and acrosome reation. To overcome these major problems, novel preservative formulation is needed to neutralize the oxidative stress and to provide suitable physiological environment for sperm in in vitro. In this study, naturally derived substances such as Poncirus trifoliate (Trifoliate orange), Garcinia mangostana (Mangosteen), pig placenta and testis extracts were tested as sperm preservative agents. Placenta extracts (PE), trifoliate orange extracts (TOE), testes extracts (TE) and mangosteen extracts (ME) were applied to analyze specific parameters for sperm motion characteristics individually and combinatorial. Each individual extract treatment can accelerate the sperm motility but noticeably TOE, TE and ME treatments exhibited the considerable and significant preservation of sperm motility. PE, TE and ME showed a significant (p<0.05) increase in ALH after one week. Further we evaluated the five different combinations of these extracts on sperm motility and its motion characteristics. Surprisingly even after one week ME, TOE and TE combination significantly preserved the sperm motility about 75%. It is noteworthy that unlike individual extract treatment, combination of ME, TOE and TE simultaneously protect the sperm motility and its motion characteristics. Taken together these data conclude that addition of ME, TOE and TE can be effective for preservation of pig sperm.
인간은 에너지 섭취와 소모에 있어 변동이 심한 편이지만 비교적 일정한 체중을 유지하고 적절하게 섭식행동을 조절한다. 그러나 이러한 조절기전에 문제가 발생하면 비만이나 그 이외의 다른 섭식장애를 일으키게 되며 다양한 후유증을 야기한다. 본 논문에서는 최근에 소개되기 시작한 호르몬인 leptin이 섭식의 조절과 에너지의 항상성을 조절하는데 있어 어떻게 중요한 작용을 하게되는지 조사하여 주로 정신과 영역에서 관심을 기울일 만한 부분에 대해 고찰하고자 한다. Leptin이 처음 발견되었을 때 그 역할은 주로 음식의 섭취를 제한하고 에너지의 소모를 증가시키는 호르몬으로 소개되었지만, 점차 그 이외의 다른 신체 기능 즉, 생식주기나 신경대분기계에도 영향을 미치는 것으로 보고되면서 정선과 영역에서 차지하는 비중도 확대되고 있다. 그러나 leptin의 작용기전은 매우 복잡하며 많은 부분이 아직도 불분명한 상태에 있다. 이에 저자들은 중추신경계에서 현재까지 알려진 leptin의 작용기전에 대해 살펴보고 섭식과 관련된 그 밖의 다른 신경전달물질들과의 상호작용 그리고 섭식장애와의 관련성에 대해서 최근 문헌들을 조사하여 함께 고찰하고자 한다.
The present study analysed the various productive and reproductive characteristics of 1/2 Pabna Milking Cows (PMC)-1/2 Sahiwal (S) and 1/4 PMC-3/4 S collected from the Pabna milkshed area at Baghabarighat, Sirajgonj, Bangladesh. The studied traits were birth weight (BWT), age at puberty (AP), number of services per conception (NSC), post partum heat period (PPHP), gestation period (GP), daily milk yield (DMY), lactational production (LP), lactation length (LL), fat percentage and solids-not-fat percentage (SNFP). Five individual Sahiwal sires were used for the upgrading of Pabna Milking Cows. The used data covered from 1987 to 1994. Least-squares analysis of variance showed that genetic group had a significant effect on BWT (P < 0.05), AP (p < 0.01), DMY (p < 0.01), LP (p < 0.001), LL (p < 0.05), FP (p < 0.05) and SNFP (p < 0.01). Genetic group had non-significant effect on NSC, GP and PPHP. The effect of sire was significant on BWT (p < 0.01), NSC (p < 0.01), LP (p < 0.05) and LL (p < 0.05). The AP, NSC, DMY, LP, LL and SNFP were higher in 1/2 PMC-1/2S cows; BWT and PPHP were higher in 1/4 PMC-3/4S but GP and FP were almost same in both genetic groups. From this study it may be concluded that production and use of 1/2 PMC-1/2S would seem more profitable for commercial milk production in the Bangladesh Milk Producers' Cooperative Union Limited (BMPCUL) area and at the same time emphasis should be given on rigorous sire selection.
Tremendous progress has been made over the past quarter-century studying the genetics of gametogenesis and the resulting gametes and embryos. Studies merging molecular techniques and conventional cytogenetics are now beginning to bridge the gap between what we have learned about the meiotic process in males and females and what we know of the mitotic chromosomes of zygotes. Numerical abnormalities in sperm, oocytes and embryo can now diagnosed by fluorescence in situ hybridization (FISH). "At risk" couples can, therefore, have only unaffected embryos replaced in the sterus and avoid the possibility of terminating a pregnancy that might only be diagnosed as affected later gestation. Single-cell genetic analysis has also provided powerful tools for studying genetic defects arising during early human development. Recent studies of sperms, oocytes and cleavage-stage human embryos have revealed an unexpectedly high incidence. These genetic abnormalities are likely to contribute to early pregnancy loss and have important implications for improving pregnancy rates in infertile couples by assisted reproduction. The widespread use of preimplantation genetic diagnosis (PGD) awaits further documentatio of safety and accuracy. Other issues also must be addressed. First, the ethical issues regarding germ cell and embryo screening must be addressed including what diseases are serious enough to warrant the procedure. Another concern is the use of this technology for non-genetic disorders such as gender selection. Finally, the experimental nature of these procedure must continually be discussed with patients, and long-term follow-up studies must be undertaken. Development of more accurate and less expensive assays coupled with improved assisted reproductive technology success rates may make PGD a more widely use clinical tool. The future awaits these development.velopment.
Purpose: Genetic disorders can be prevented by basic public health measures and activities that focus primarily on education and approaches in Primary Health Care. Premarital screening is one such approach that can identify asymptomatic carriers of hemoglobinopathies and provide genetic counseling to couples for a healthy reproductive life. This study aimed to estimate the prevalence of beta thalassemia and sickle cell disorders in the adult population screened as a part of the United Arab Emirates Premarital Screening Program and to measure the effectiveness of the program in decreasing high-risk marriages in Ras Al Khaimah (RAK). Materials and Methods: A retrospective, population-based study was conducted at the RAK Primary Health Care Center, where the National Premarital Screening Program is implemented. The study included data collected from the premarital screening records of all couples who had applied for a marriage license during 2008-2015. Results: Of the 17,826 individuals screened during the studied period, 4.02% (717) were diagnosed as positive for hemoglobinopathies. The prevalence of beta thalassemia and sickle cell disorders among the total study population was 2.98% and 1.05%, respectively. The hemoglobinopathy trait was more prevalent than the disease. Among these 8,913 couples who were issued certificates for compatibility based on screening tests, 28 (0.31%) couples were declared high-risk (unmatched). Seventy percent of these unmatched couples reported consanguineous marriages. Conclusion: The program was successful in achieving its objective of identifying high-risk marriages. In spite of the counseling, however, all of the high-risk couples still married each other.

Regular monitoring on experimental animal management found the fluctuation of ART outcome, which showed a necessity to explore whether superovulation treatment is responsible for such unexpected outcome. This study was subsequently conducted to examine whether superovulation treatment can preserve ultrastructural integrity and developmental competence of oocytes following oocyte activation and embryo culture. A randomized study using mouse model was designed and in vitro development (experiment 1), ultrastructural morphology (experiment 2) and functional integrity of the oocytes (experiment 3) retrieved after PMSG/hCG injection (superovulation group) or not (natural ovulation; control group) were evaluated. In experiment 1, more oocytes were retrieved following superovulation than following natural ovulation, but natural ovulation yielded higher (p < 0.0563) maturation rate than superovulation. The capacity of mature oocytes to form pronucleus and to develop into blastocysts in vitro was similar. In experiment 2, a notable (p < 0.0186) increase in mitochondrial deformity, characterized by the formation of vacuolated mitochondria, was detected in the superovulation group. Multivesicular body formation was also increased, whereas early endosome formation was significantly decreased. No obvious changes in other microorganelles, however, were detected, which included the formation and distribution of mitochondria, cortical granules, microvilli, and smooth and rough endoplasmic reticulum. In experiment 3, significant decreases in mitochondrial activity, ATP production and dextran uptake were detected in the superovulation group. In conclusion, superovulation treatment may change both maturational status and functional and ultrastuctural integrity of oocytes. Superovulation effect on preimplantation development can be discussed.

The swine is one of the most widespread mammalian throughout the whole world. Presently, many studies concerning microsatellites in swine, especially domestic pigs, have been carried out in order to investigate general diversity patterns among either populations or breeds. Until now, a lot of time and effort spend into a single PCR method. But simple and more rapid multiplex PCR methods have been developed. The purpose of this study is to develop a robust set of microsatellites markers (MS marker) for traceability and individual identification. Using multiplex-PCR method with 23 MS marker divided 2 set, various alleles occurring to 5 swine breed (Berkshire, Landrace, Yorkshire, Duroc and Korea native pig) used markers to determine allele frequency and heterozygosity. MS marker found 4 alleles at SW403, S0227, SWR414, SW1041 and SW1377. The most were found 10 alleles at SW1920. Heterozygosity represented the lowest value of 0.102 at SWR414 and highest value of 0.861 at SW1920. So, it was recognized appropriate allele frequency for individual identification in swine. Using multiplex-PCR method, MS markers used to determine individual identification biomarker and breed-specific marker for faster, more accurate and lower analysis cost. Based on this result, a scientific basis was established to the existing pedigree data by applying genetics additionally. Swine traceability is expected to be very useful system and be conducted nationwide in future.
Litchi (Litchi chinensis Sonn.) is an important subtropical fruit crop with high commercial value due to its high nutritional values and favorable tastes. However, irregular bearing attributed to unstable flowering is a major ongoing problem for litchi producers. Previous studies indicate that low-temperature is a key factor in litchi floral induction. In order to reveal the genetic and molecular mechanisms underlying the reproductive process in litchi, we had analyzed the transcriptome of buds before and after low-temperature induction using RNA-seq technology. A key flower bud differentiation associated gene, a homologue of FLORICAULA/LEAFY, was identified and named LcLFY (GenBank Accession No. KF008435). The cDNA sequence of LcLFY encodes a putative protein of 388 amino acids. To gain insight into the role of LcLFY, the temporal expression level of this gene was measured by real-time RT-PCR. LcLFY was highly expressed in flower buds and its expression correlated with the floral developmental stage. Heterologous expression of LcLFY in transgenic tobacco plants induced precocious flowering. Meantime, we investigated the sub-cellular localization of LcLFY. The LcLFY-Green fluorescent protein (GFP) fusion protein was found in the nucleus. The results suggest that LcLFY plays a pivotal role as a transcription factor in controlling the transition to flowering and in the development of floral organs in litchi.
Streptomyces coelicolor is a gram-positive soil bacterium which is well known for the production of several antibiotics used in various biotechnological applications. But numerous proteins from its genome are considered hypothetical proteins. Therefore, the present study aimed to reveal the functions of a hypothetical protein from the genome of S. coelicolor. Several bioinformatics tools were employed to predict the structure and function of this protein. Sequence similarity was searched through the available bioinformatics databases to find out the homologous protein. The secondary and tertiary structure were predicted and further validated with quality assessment tools. Furthermore, the active site and the interacting proteins were also explored with the utilization of CASTp and STRING server. The hypothetical protein showed the important biological activity having with two functional domain including POD-like_MBL-fold and rhodanese homology domain. The functional annotation exposed that the selected hypothetical protein could show the hydrolase activity. Furthermore, protein-protein interactions of selected hypothetical protein revealed several functional partners those have the significant role for the bacterial survival. At last, the current study depicts that the annotated hypothetical protein is linked with hydrolase activity which might be of great interest to the further research in bacterial genetics.

목적: 거대세포바이러스(Cytomegalovirus [CMV])는 정상 면역을 가진 사람에서는 대부분 불현성 감염이지만, 면역저하자 또는 태아의 선천 감염의 경우 위중한 합병증을 유발한다. 본 연구는 21년 동안의 시기 및 연령에 따른 CMV 항체 양성률을 조사하였다. 방법: 1995년 1월부터 2015년 12월까지 성균관대학교 의과대학, 삼성서울병원에서 CMV immunoglobulin G (IgG) 검사를 시행한 환자를 대상으로 하였다. 검사 결과는 성별, 연령별, 전국 지역별로 분석하였고, 두 시기에 따른 (1995-2005년; 1기, 2006-2015년; 2기) 항체 양성률의 변화를 분석하였다. 결과: 총 11,584명의 환자 중 CMV IgG 양성결과를 보인 총 환자는 10,900명 (94.1%)이었다. 전체 환자에서 두 시기에 따른 CMV 양성률의 차이는 없었다 (94.2% vs. 94.1%) (P=0.86). 10년 간격으로 비교를 했을 때 다른 연령군은 유의하지 않았으나 11-20세의 연령군의 CMV 양성률은 1기에 비해 2기에 더 낮았다 (P<0.001). 이 연령에 따른 항체 양성률 분석에서 31-40세 연령군의 항체 양성률 (97.4%)은 이보다 어린 연령군에 비하여 유의하게 높았으며 (P<0.001, odds ratio [OR]<1), 더 높은 연령군에 비하여 유의하게 낮았다 (P<0.001, OR>1). 가임기 여성은 총 2,441명이었으며 (15-49세) 이 중 CMV IgG 양성률은 2,367 (97%)명이었다. 해당 연령군의 시기별 항체 양성률의 유의한 차이는 없었다. 가임기 여성 중 20-24세의 연령군은 해당 연령의 남자에 비하여 유의한 높은 항체 양성률을 보였다 (97.6% vs. 85.6%, OR, 6.78; 95% CI, 1.585-28.968; P=0.0028). 지역별 항체 양성률의 큰 차이는 보이지 않았다. 결론: 본 논문은 한국인의 전 연령의 CMV 항체 양성률 변화에 대한 조사를 시행한 최초 논문이며 전체적으로 약 94%의 높은 CMV 항체 양성률과 가임기 여성은 97%의 항체 양성률을 보이는 것으로 관찰되었다. 향후, 가임기 여성을 포함한 한국인의 CMV 항체 양성률의 변화 추세에 대한 모니터링과 국내 임산부에서의 CMV 감염에 대한 연구가 필요할 것으로 사료된다.
