  • The present investigation has been undertaken to understand the mechanism of implantation process, by demonstrating the role of ovarian steroids in the differentiation of uterine endometrium for implantation. In particular, an attempt was made to examine the activity of alkaline phosphatase (ALP) in the either luminal, stroma or endometrium tissue sites under the pseudopregnant state induced by ovarian steroid hormones. Attempt was also made to demonstrate the correlate function of ovarian steroids with the cAMP concentration and prolactin level. The higher activity of ALP in the uterine endometrium was observed on day 3. However, the higher activity of ALP in the stroma and epithelium was observed on Day 6. This study, therefore, clearly demonstrates that progesterone is consecutive effect in stroma differ entiation. The cAMP concentrations on Day 3 treated with E or P was lower than those of control. On the other hand concentration on Day 6 treated with hormones was increased than those of control. It is, therefore, concluded that the concentration of cAMP in the uterine tissue undergoing differentiation is decreased. The prolactin level of the treated groups was the lower levels than those of the control groups. It is indicated that there is no effect of ovarian steroid hormone on the prolactin synthesis in this pseudopregnant state.

Altered expression of norepinephrine transporter and norepinephrine in human placenta cause pre-eclampsia through regulated trophoblast invasion

  • Objective: We investigated the norepinephrine transporter (NET) expression in normal and pre-eclamptic placentas and analyzed the invasion activity of trophoblastic cells based on norepinephrine (NE)-NET regulation. Methods: NET and NE expression levels were examined by western blot and enzyme-linked immunosorbent assay, respectively. Trophoblast invasion activity, depending on NE-NET regulation, was determined by NET-small interfering RNA (siRNA) and NET transfection into the human extravillous trophoblast cells with or without NE treatment and invasion rates were analyzed by zymography and an invasion assay. Results: NET mRNA was expressed at a low level in pre-eclamptic placentas compared with normal placentas and NE concentration in maternal plasma increased significantly in pre-eclamptic women compared to normal pregnant women (p<0.05). NET gene upregulation and NE treatment stimulated trophoblast cell invasion up to 2.5-fold (p<0.05) by stimulating matrix metalloproteinase-9 activity via the phosphoinositol-3-kinase/AKT signaling pathway, whereas NET-siRNA with NE treatment reduced invasion rates. Conclusion: NET expression is reduced by inadequate regulation of NE levels during placental development. This suggests that a complementary balance between NET and NE regulates trophoblast cell invasion activities during placental development.

The Expressional Pattern of Epididymal Protease Inhibitor (EPPIN) in the Male Syrian Hamsters

  • The spermatogenesis is the process by which spermatozoa are generated in the testes. The spermatozoa travel male reproductive tract during which they meet many substances secreted from reproductive organs. One of the substances is epididymal protease inhibitor (EPPIN) that is involved in the post-testicular maturation including capability of fertilizing the eggs. The expression of EPPIN gene was investigated in various tissues of sexually mature and regressed male Syrian hamsters by reverse transcription polymerase chain reaction (RT-PCR). The EPPIN gene was identified in the testis and epididymis of the male Syrian hamsters and compared to the genes reported previously. There was no expression of EPPIN gene in reproductively and completely regressed testes of Syrian hamster. These results suggest that the expressions of the EPPIN gene are associated with the reproductive capability in the Syrian hamsters.

Expressions of Semenogelin Gene in Male Syrian Hamsters according to Photoperiod

  • The morphogenetically matured spermatozoa (sperm) are generated in the testes by the spermatogenesis. They travel male reproductive tract with many substances secreted from the accessory reproductive organs. One of the substances is the semenogelin (SEMG) released from the seminal vesicles that is involved in the post-testicular maturation. The expression of SEMG gene was investigated in seminal vesicle tissues of sexually matured and regressed male Syrian hamsters by reverse transcription polymerase chain reaction (RT-PCR). The SEMG gene was uniquely identified in the seminal vesicles of the matured Syrian hamsters and compared to the genes reported previously. But the expression of SEMG gene was not observed in reproductively and completely regressed testes of Syrian hamsters. These results indicate that the expressions of the SEMG gene are related to the reproductive capability in the male Syrian hamsters.

Male Mating History in Antheraea mylitta and its Effect on Ejaculation Size and Female Reproductive Fitness

  • Reproductive activity in the economically important insect, Antheraea mylitta is very important because the female reproduces only once in her life time and the aging is very rapid and costly. It is a capital breeder and strictly a monoandrous species. To know how strategically the insect behaves and the male allocates its ejaculate resource more prudently during its successive mating with virgin females to maximize its own fitness and the fitness of the female with whom it mated, both being most important to sericulture industry. So, the present study was undertaken and the results revealed fresh and virgin females always prefer to mate with fresh virgin males (84%) and receives high dose of ejaculates leading to higher hatchability than to virgin males of one day old (13.7%) and one day old males with mating experience (2.3%). The ejaculation size (as referred to eupyrene sperm count in the ejaculation) declined significantly over successive mating (r = -0.9931, P < 0.001), so also the male body weight (r = -0.9560, P < 0.001). The quantity of ejaculate passed to female also dramatically declined during aging (r = -0.9982, P < 0.001). It was found that male weight contribute substantially to the quantum of ejaculate (r = -0.9519, P < 0.001), so also higher fecund females receive relatively more ejaculate than the lower group to reach higher reproductive fitness. The life time fecundity was found to be $334{\pm}31$.

제주도 주요 하천에 서식하는 은어(Plecoglossus altivelis)의 번식 특성 비교 (Comparison of the Reproductive Characteristics of Sweetfish Plecoglossus altivelis in the Main Streams of Jeju Island)

  • We investigated the reproductive characteristics of the sweetfish Plecoglossus altivelis, including changes in the gonadosomatic index (GSI), frequency of gonad developmental stages, and abundance of drifting larvae, in three streams (Gangjeong, Yeonoi, and Ongpo) on Jeju Island from May 2004 to December 2005. The GSI values of female P. altivelis in all Jeju streams began to increase in September and reached a maximum in October and November. Peak GSI values in males occurred in Gangjeong from October to November, in Yeonoi from November to January, and in Ongpo from September to October. The gonadal development of P. altivelis was classified into four stages: growth (March to October), maturity (September to December in females; July to December in males), spawning (September to January), and degeneration (October to March in females; after November in males). Drifting larvae were collected from October to January. These results suggest that the main spawning activity of P. altivelis in Jeju streams occurs from October to November. The information about the reproductive characteristics of P. altivelis obtained in this study is critical to fishery management for this species.

Nesfatin-1 as a New Potent Regulator in Reproductive System

  • Nesfatin-1 is a recently discovered anorexigenic peptide which is distributed in several brain areas implicated in the feeding and metabolic regulation. Recently, it has been reported that nesfatin-1 is expressed not only in brain, but also in peripheral organs such as digestive organs, adipose tissues, heart, and reproductive organs. Nesfatin-1 is markedly expressed in the pancreas, stomach and duodenum. Eventually, the nesfatin-1 expression in the digestive organs may be regulated by nutritional status, which suggests a regulatory role of peripheral nesfatin-1 in energy homeostasis. Nesfatin-1 is also detected in the adipose tissues of humans and rodents, indicating that nesfatin-1 expression in the fat may regulate food intake independently, rather than relying on leptin. In addition, nesfatin-1 is expressed in the heart as a cardiac peptide. It suggests that nesfatin-1 may regulate cardiac function and encourage clinical potential in the presence of nutrition-dependent physio-pathologic cardiovascular diseases. Currently, only a few studies demonstrate that nesfatin-1 is expressed in the reproductive system. However, it is not clear yet what function of nesfatin-1 is in the reproductive organs. Here, we summarize the expression of nesfatin-1 and its roles in brain and peripheral organs and discuss the possible roles of nesfatin-1 expressed in reproductive organs, including testis, epididymis, ovary, and uterus. We come to the conclusion that nesfatin-1 as a local regulator in male and female reproductive organs may regulate the steroidogenesis in the testis and ovary and the physiological activity in epididymis and uterus.

Effect of Mating Delay and Mating Duration on Reproductive Performance of Antheraea mylitta

  • Tropical tasar silkmoth Antheraea mylilta D. is an economically important insect. The wild nature and above all unsynchronized emergence of moths lead to impairment in reproductive activity. The mating in moths is low (64%-70%), leaving about 30-36% of potential females unused for seed (= egg) production. Delay in mating adversely affects the reproductive performance such as mating behaviour, fecundity and fertility. Females are more severely affected by mating delay than males. Therefore, a study was under taken to develop a method to overcome the problem so that the production of layings can be increased to meet the ever increasing demand. It was revealed from the study that out of 16 possible age combinations (from the age groups of day 0 to day 3), female and male of day 0 (4-8 hours old) excel in their reproductive performance than others (with 94.4% mating success, 284 fecundity and 91.5% hatching). Reproductive performance declined significantly even if one of the parents was older. When either of the mating pair was 1 day old, the mating declined by 28%; fecundity by 32.8%; and hatching by 40.8%. The insemination of eupyrene sperm was declined by 35.2% when both the parents are 1 day old. Mating duration of 8 hours resulted in insemination of $8.3{\times}10^5$ eupyrene sperms, which was declined by 31.6%, 48% and 55% upon lowering of mating duration to 6 hr, 4 hr and 2 hr were maintained respectively.

Evaluation of polyglycolic acid as an animal-free biomaterial for three-dimensional culture of human endometrial cells

  • Objective: Animal-free scaffolds have emerged as a potential foundation for consistent, chemically defined, and low-cost materials. Because of its good potential for high biocompatibility with reproductive tissues and well-characterized scaffold design, we investigated whether polyglycolic acid (PGA) could be used as an animal-free scaffold instead of natural fibrin-agarose, which has been used successfully for three-dimensional human endometrial cell culture. Methods: Isolated primary endometrial cells was cultured on fibrin-agarose and PGA polymers and evaluated various design parameters, such as scaffold porosity and mean fiber diameter. Cytotoxicity, scanning electron microscopy (SEM), and immunostaining experiments were conducted to examine cell activity on fabricated scaffolds. Results: The MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assay and SEM results showed that endometrial cells grew and proliferated on both scaffolds. Immunostaining showed cytokeratin and vimentin expression in seeded cells after 7 days of culture. On both scaffolds, an epithelial arrangement of cultured cells was found on the top layer and stromal arrangement matrix on the bottom layer of the scaffolds. Therefore, fibrin-agarose and PGA scaffolds successfully mimicked the human endometrium in a way suitable for in vitro analysis. Conclusion: Both fibrin-agarose and PGA scaffolds could be used to simulate endometrial structures. However, because of environmental and ethical concerns and the low cost of synthetic polymers, we recommend using PGA as a synthetic polymer for scaffolding in research instead of natural biomaterials.

  • The study was conducted to assess the repeated dose and reproduction and developmental toxicities of acetanilide, an intermediate for drug production, as a part of OECD Screening Information Data Set (SIDS) program. The test agent was administered by gavage at dose levels of 0, 22, 67, 200 and 600 mg/kg to Sprague-Dawley rats (12/group/sex) during pre-mating and mating period for males(up to 30 days) and females and pregnancy and early lactation period for females (up to 39-50 days). At 22 mg/kg, decreases in HGB, HCT (males) and MCHC (females), hyperplasia of spleen red pulp, hyperplasia of femur bone marrow (both sexes) were observed. At 67 mg/kg, salivation (males), reduced food consumption (both sexes), decreases in RBC, HGB, HCT and MCHC (males), increases in MCV (males) and spleen weight (males), hyperplasia of spleen red pulp and femur bone marrow (both sexes) were observed. At 200 mg/kg, decreases in locomotor activity and salivation (both sexes), reduced food consumption (both sexes), decreases in RBC, HGB, HCT and increases in MCV, MCH, BUN, T-BIL (males), enlargement of spleen (both sexes), increased weight of spleen (males), hyperplasia of spleen red pulp and femur bone marrow and extramedullary hematopoiesis of liver (both sexes), atrophy of thymus and corpus luteum hyperplasia of ovary (females) were observed. At 600 mg/kg, decreases in locomotor activity, cyanosis (both sexes), reddish tear, and salivation (males), mortality (4 out of 12 females), decreased body weight (females), reduced food consumption (both sexes), decreases in RBC, HGB, HCT and MCHC and increases in WBC, MCV, MCH, reticulocyte, neutrophil, lymphocyte, monocyte, AST, ALT, BUN, T-BIL, ALB, Ca and A/G ratio (males), enlargement of spleen, increased weights of spleen (both sexes), liver (males), kidney and ovary, decreased weights of thymus (females), hyperplasia of spleen red pulp, hyperplasia of femur bone marrow and extramedullary hematopoiesis of liver (both sexes), and atrophy of thymus and corpus luteum hyperplasia of ovary (females) were observed. Regarding the reproduction and development toxicities, there were no treatment-related changes in precoital time, mating index, fertility index and pregnancy index at all doses tested. At 22 and 67 mg/kg, there were no adverse effects on all the parameters observed. At 200 mg/ kg, decreased body weight of pups (day 4 p.p.) were observed. At 600 mg/kg, decreased body weight of pups (day 0 and 4 p.p.) and viability index (day 4 p.p.), increased incidence of newborns dead or with abnormal clinical signs were observed. The results suggest that the NOAELs for general toxicity are < 22 mg/kg, LOAELs are 22 mg/kg and the NOAELs for reproductive toxicity are 67 mg/kg.
